Abstract:Abstract Background Acoustic communication is important for the survival and reproduction of anurans and masking background noise is a critical factor for their effective acoustic communication. Males of the concave-eared frog ( Odorrana tormota ) have evolved an ultrasonic communication capacity to avoid masking by the widespread background noise of local fast-flowing streams, whereas females exhibit no ultrasonic sensitivity. However, the molecular mechanisms underlying the high-frequency hearing differences between the sexes of O. tormota are still poorly understood. Results In this study, we sequenced the brain transcriptomes of male and female O. tormota , and compared their differential gene expression. A total of 4,605 differentially expressed genes (DEGs) between the sexes of O. tormota were identified and eleven of them were related to auditory based on the annotation and enrichment analysis. Most of these DEGs in males showed a higher expression trend than females in both quantity and expression quantity. The highly expressed genes in males were relatively concentrated in neurogenesis, signal transduction, ion transport and energy metabolism, whereas the up-expressed genes in females were mainly related to the growth and development regulation of specific auditory cells. Conclusions The transcriptome of male and female O. tormota has been sequenced and de novo assembled, which will provide gene reference for further genomic studies. In addition, this is the first research to reveal the molecular mechanisms of sex differences in ultrasonic hearing between the sexes of O. tormota and will provide new insights into the genetic basis of the auditory adaptation in amphibians during their transition from water to land.

Male Serrate-Legged Treefrogs Adjust Competition Strategies According to Visual or Chemical Cues from Females.

Abstract:There is increasing evidence that many anurans use multimodal cues to detect, discriminate and/or locate conspecifics and thus modify their behaviors. To date, however, most studies have focused on the roles of multimodal cues in female choice or male-male interactions. In the present study, we conducted an experiment to investigate whether male serrate-legged small treefrogs (Kurixalus odontotarsus) used visual or chemical cues to detect females and thus altered their competition strategies in different calling contexts. Three acoustic stimuli (advertisement calls, aggressive calls and compound calls) were broadcast in a randomized order after a spontaneous period to focal males in one of four treatment groups: combined visual and chemical cues of a female, only chemical cues, only visual cues and a control (with no females). We recorded the vocal responses of the focal males during each 3-min period. Our results demonstrate that males reduce total number of calls in response to the presence of females, regardless of how they perceived the females. In response to advertisement calls and compound calls, males that perceived females through chemical cues produced relatively fewer advertisement calls but more aggressive calls. In addition, they produced relatively more aggressive calls during the playback of aggressive calls. Taken together, our study suggests that male K. odontotarsus adjust their competition strategies according to the visual or chemical cues of potential mates and highlights the important role of multisensory cues in male frogs' perception of females.
Multisensory modalities increase working memory for mating signals in a treefrog

Abstract:Animal choruses, such as those found in insects and frogs, are often intermittent. Thus, females sampling males in the chorus might have to remember the location of the potential mates' calls during periods of silence. Although a number of studies have shown that frogs use and prefer multimodal mating signals, usually acoustic plus visual, it is not clear why they do so. Here we tested the hypothesis that preference for multimodal signals over unimodal signals might be due to multimodal signals instantiating longer memories than unimodal signals, particularly during the inter-chorus intervals. We tested this hypothesis in serrate-legged small treefrogs Kurixalus odontotarsus whose males produce advertisement calls accompanied by conspicuous vocal sac inflation. Females were tested with acoustic and acoustic + visual (video of inflating-deflating vocal sac) mating calls. We found that females prefer multimodal calls over unimodal, audio-only calls. Furthermore, multimodal calls are still preferred after a silent period of up to 30 s, a time that spans the average silent period of the chorus. This was not true of unimodal calls. Our results demonstrate that a multimodal signal can engage longer working memory than a unimodal signal, and thus female memory might favour the evolution of multimodal signals in males through sexual selection. Selection might also favour female preference for multimodal signals if longer memory facilitates mate searching and assessment. Our study does not allow us to elucidate the sequence of evolution of this trait and preference.

Abstract:Acoustic signals are constrained by background noise. Visual signals are an alternative or complementary communication mode in noisy habitats and play a fundamental role in anuran communication. The Bornean rock-skipper frog, Staurois latopalmatus, is a diurnal species living along fast-flowing streams and waterfalls. Males perform foot-flagging displays with either one or two legs to advertise their readiness to defend their territories. In quantitative video analyses of visual displays during 14 male-male agonistic interactions, totaling 106 minutes, foot flagging performed in the direction of the interacting male was the most common display and was performed at a higher rate than advertisement calls. According to a dyadic transition matrix, foot flagging was preceded by foot-flagging displays of interacting males. Advertisement calls were temporally coupled with foot flaggings and act as introductory components to direct the receiver's attention to the subsequent visual display. We conclude that foot flagging acts as a spacing mechanism and may have resulted from the ritualization of agonistic male behavior to minimize physical attacks.

Abstract:Most mosquito and midge species use hearing during acoustic mating behaviors. For frog-biting species, however, hearing plays an important role beyond mating as females rely on anuran calls to obtain blood meals. Despite the extensive work examining hearing in mosquito species that use sound in mating contexts, our understanding of how mosquitoes hear frog calls is limited. Here, we directly investigated the mechanisms underlying detection of frog calls by a mosquito species specialized on eavesdropping on anuran mating signals: Uranotaenia lowii. Behavioral, biomechanical and neurophysiological analyses revealed that the antenna of this frog-biting species can detect frog calls by relying on neural and mechanical responses comparable to those of non-frog-biting species. Our findings show that in Ur. lowii, contrary to most species, males do not use sound for mating, but females use hearing to locate their anuran host. We also show that the response of the antennae of this frog-biting species resembles that of the antenna of species that use hearing for mating. Finally, we discuss our data considering how mosquitoes may have evolved the ability to tap into the communication system of frogs.
Social regulation of androgenic hormones and gestural display behavior in a tropical frog

Abstract:Many animals use forms of gesture and dance to communicate with conspecifics in the breeding season, though the mechanisms of this behavior are rarely studied. Here, we investigate the hormone basis of such visual signal behavior in Bornean rocks frogs (Staurois parvus). Our results show that males aggregating at breeding waterfalls have higher testosterone (T) levels, and we speculate that this hormone increase is caused by social cues associated with sexual competition. To this end, we find that T levels in frogs at the waterfall positively predict the number waving gestures-or "foot flags"-that males perform while competing with rivals. By contrast, T does not predict differences in male calling behavior. In these frogs, vocal displays are used largely as an alert signal to direct a rival's attention to the foot flag; thus, our results are consistent with the view that factors related to reproductive context drive up T levels to mediate displays most closely linked to male-male combat, which in this case is the frog's elaborate gestural routine.
Lung mediated auditory contrast enhancement improves the Signal-to-noise ratio for communication in frogs

Abstract:Environmental noise is a major source of selection on animal sensory and communication systems. The acoustic signals of other animals represent particularly potent sources of noise for chorusing insects, frogs, and birds, which contend with a multi-species analog of the human "cocktail party problem" (i.e., our difficulty following speech in crowds). However, current knowledge of the diverse adaptations that function to solve noise problems in nonhuman animals remains limited. Here, we show that a lung-to-ear sound transmission pathway in frogs serves a heretofore unknown noise-control function in vertebrate hearing and sound communication. Inflated lungs improve the signal-to-noise ratio for communication by enhancing the spectral contrast in received vocalizations in ways analogous to signal processing algorithms used in hearing aids and cochlear implants. Laser vibrometry revealed that the resonance of inflated lungs selectively reduces the tympanum's sensitivity to frequencies between the two spectral peaks present in conspecific mating calls. Social network analysis of continent-scale citizen science data on frog calling behavior revealed that the calls of other frog species in multi-species choruses can be a prominent source of environmental noise attenuated by the lungs. Physiological modeling of peripheral frequency tuning indicated that inflated lungs could reduce both auditory masking and suppression of neural responses to mating calls by environmental noise. Together, these data suggest an ancient adaptation for detecting sound via the lungs has been evolutionarily co-opted to create auditory contrast enhancement that contributes to solving a multi-species cocktail party problem.
Sexy fingers: Pheromones in the glands of male dendrobatid frogs

Abstract:Many animals exchange chemicals during courtship and mating. In some amphibians, sexual chemical communication is mediated by pheromones produced in male breeding glands that are transferred to the female's nostrils during mating. This has been mostly studied in salamanders, despite frogs having similar glands and courtship behaviours suggestive of chemical communication. In Neotropical poison frogs (Dendrobatidae and Aromobatidae), males of many species develop breeding glands in their fingers, causing certain fingers to visibly swell. Many also engage in cephalic amplexus, whereby the male's swollen fingers are placed in close contact with the female's nares during courtship. Here, we investigate the possible roles of swollen fingers in pheromone production using whole-transcriptome sequencing (RNAseq). We examined differential gene expression in the swollen versus non-swollen fingers and toes of two dendrobatid species, Leucostethus brachistriatus and Epipedobates anthonyi, both of which have specialised mucous glands in finger IV, the latter of which has cephalic amplexus. The overwhelming pattern of gene expression in both species was strong upregulation of sodefrin precursor-like factors (SPFs) in swollen fingers, a well-known pheromone system in salamanders. The differentially expressed SPF transcripts in each species were very high (>40), suggesting a high abundance of putative protein pheromones in both species. Overall, the high expression of SPFs in the swollen fingers in both species, combined with cephalic amplexus, supports the hypothesis that these traits, widespread across members of the subfamilies Colostethinae and Hyloxalinae (ca. 141 species), are involved in chemical signalling during courtship.
Conspecific odor cues induce different vocal responses in serrate-legged small treefrogs, but only in the absence of acoustic signals

Abstract:Background Signal detection is crucial to survival and successful reproduction, and animals often modify behavioral decisions based on information they obtained from the social context. Undeniably, the decision-making in male-male competition and female choice of anurans (frogs and toads) depends heavily on acoustic signals. However, increasing empirical evidence suggests that additional or alternative types of cue (e.g., visual, chemical, and vibratory) can be used to detect, discriminate and locate conspecifics in many anuran species. Nevertheless, few studies have investigated whether conspecific odor cues affect male's calling behavior. In this study, we conducted an experiment to investigate whether and how different chemical cues (male odors, female odors, and stress odors) from conspecifics affect male's calling strategies in serrate-legged small treefrogs (Kurixalus odontotarsus), and whether the combined chemical and acoustic stimuli have additive effects on calling behavior or not. Results We found that compared with female odors, male K. odontotarsus reduced calling investment in response to male odors or stress odors, in the absence of rival's advertisement calls. When odor stimuli and advertisement calls were presented simultaneously, however, there were no differences in the vocal response of focal males among odor groups. Conclusions These results provide evidence that male treefrogs switch calling investment according to different odor cues from conspecifics, and further demonstrate that calling behavior can be affected by chemical cues in anuran species. Our study highlights the potential role of airborne chemical cues in sex identification and contributes to increase our understanding of anuran communication.

Abstract:Communication during parental care is important in the context of offspring defence and parent-offspring recognition. In aquatic environments, chemical communication is usually the most effective and plays different roles in parental behaviour in many groups of animals. Attending females of some Neotropical anurans ( Leptodactylus ) lead entire schools of tadpoles through the water. However, little is known about how attending females communicate with the tadpoles or whether this communication is mother-offspring specific. We therefore conducted behavioural experiments with Leptodactylus podicipinus to address these questions. Initially, we investigated how visual, tactile, and chemical stimuli from attending females affect the aggregation behaviour of tadpoles. Next, we compared the effects of the chemical stimulus from attending females with those from other conspecifics (non-attending females and males) on schooling behaviour. Finally, we tested whether tadpoles preferred to be aggregated next to familiar attending females instead of unfamiliar ones. Our results indicated that only the chemical stimulus from attending females, and not visual or tactile, increased the time that tadpoles remained aggregated. The chemical stimuli from non-attending females and males did not affect tadpoles' behaviour. However, contrary to our expectations, we did not find a school preference for familiar attending females. Our study is the first showing that chemical stimulus is important for mother-tadpole communication in a Leptodactylus species, playing a crucial role in tadpole aggregation and attraction. Future studies including other species should address the nature of the chemical compounds and their role, if any, in kin recognition in these frogs with aquatic maternal care.

Abstract:How diverse animal communication signals have arisen is a question that has fascinated many. Xenopus frogs have been a model system used for three decades to reveal insights into the neuroendocrine mechanisms and evolution of vocal diversity. Due to the ease of studying central nervous system control of the laryngeal muscles in vitro, Xenopus has helped us understand how variation in vocal communication signals between sexes and between species is produced at the molecular, cellular, and systems levels. Yet, it is becoming easier to make similar advances in non-model organisms. In this paper, we summarize our research on a group of frog species that have evolved a novel hind limb signal known as 'foot flagging.' We have previously shown that foot flagging is androgen dependent and that the evolution of foot flagging in multiple unrelated species is accompanied by the evolution of higher androgen hormone sensitivity in the leg muscles. Here, we present new preliminary data that compare patterns of androgen receptor expression and neuronal cell density in the lumbar spinal cord – the neuromotor system that controls the hind limb – between foot-flagging and non-foot-flagging frog species. We then relate our work to prior findings in Xenopus , highlighting which patterns of hormone sensitivity and neuroanatomical structure are shared between the neuromotor systems underlying Xenopus vocalizations and foot-flagging frogs' limb movement and which appear to be species-specific. Overall, we aim to illustrate the power of drawing inspiration from experiments in model organisms, in which the mechanistic details have been worked out, and then applying these ideas to a non-model species to reveal new details, further complexities, and fresh hypotheses.

Abstract:Abstract Anurans use different sensory modalities to communicate and interact socially, including acoustic, chemical, tactile, and visual signals. In a multimodal context, these sensory modes can transmit different information to the receiver or even reinforce the same message. In this study, we hypothesized that body colour traits and vocal sac movements of male eastern gray treefrogs [Dryophytes versicolor (= Hyla versicolor)] serve as secondary visual cues that increase their attractiveness while calling to attract mates. We tested this hypothesis using playback trials combining synthetic advertisement calls with a variety of visual cues, including differently coloured resin frog models, simulated vocal sacs, and live frogs. None of the visual stimuli tested impacted female choice, and there was no difference in response rate, response time, choice angle, or distance covered in the arena between multimodal and unimodal stimuli. However, females showed a strong preference for longer calls even if the less attractive calls were paired with a visual stimulus. The study provides comprehensive insights into the role of visual stimuli in the mate attraction of eastern gray treefrogs and suggests that call traits play a dominant role in the male-selection process of this species, contrary to what was observed for visual stimuli.

Abstract:Abstract Environmental noise has a significant negative impact on acoustic communication in most situations, as it influences the production, transmission, and reception of acoustic signals. However, how animals respond to conspecific sounds when there is interference from environmental noise, and whether males and females display convergent behavioral responses in the face of noise masking remain poorly understood. In this study, we investigated the effects of conspecific male advertisement calls with different signal-to-noise ratios on male–male competition and female choice in the Anhui tree frog Rhacophorus zhoukaiyae using playback and phonotaxis experiments, respectively. The results showed that (1) female Anhui tree frogs preferentially selected the conspecific calls with higher SNR compared to calls with lower SNR; (2) males preferentially responded vocally to the conspecific calls with higher SNR compared to calls with lower SNR; and (3) males’ competitive strategies were flexible in the face of noise interference. These results suggest that preferences of both sexes converge in outcome, and that male competitive strategies may depend on predictable female preferences. This study will provide an important basis for further research on decision-making in animals.
Tadpoles rely on mechanosensory stimuli for communication when visual capabilities are poor

Abstract:The ways in which animals sense the world changes throughout development. For example, young of many species have limited visual capabilities, but still make social decisions, likely based on information gathered through other sensory modalities. Poison frog tadpoles display complex social behaviors that have been suggested to rely on vision despite a century of research indicating tadpoles have poorly-developed visual systems relative to adults. Alternatively, other sensory modalities, such as the lateral line system, are functional at hatching in frogs and may guide social decisions while other sensory systems mature. Here, we examined development of the mechanosensory lateral line and visual systems in tadpoles of the mimic poison frog (Ranitomeya imitator) that use vibrational begging displays to stimulate egg feeding from their mothers. We found that tadpoles hatch with a fully developed lateral line system. While begging behavior increases with development, ablating the lateral line system inhibited begging in pre-metamorphic tadpoles, but not in metamorphic tadpoles. We also found that the increase in begging and decrease in reliance on the lateral line co-occurs with increased retinal neural activity and gene expression associated with eye development. Using the neural tracer neurobiotin, we found that axonal innervations from the eye to the brain proliferate during metamorphosis, with little retinotectal connections in recently-hatched tadpoles. We then tested visual function in a phototaxis assay and found tadpoles prefer darker environments. The strength of this preference increased with developmental stage, but eyes were not required for this behavior, possibly indicating a role for the pineal gland. Together, these data suggest that tadpoles rely on different sensory modalities for social interactions across development and that the development of sensory systems in socially complex poison frog tadpoles is similar to that of other frog species.

Abstract:Noise is one of the main factors that can influence the processes of sound communication across a wide range of animal groups. Although the effects of ambient noise on animal communication, including anthropogenic noise, have received increasing attention, few studies have examined changes in the fine structure of acoustic signals produced by vocalizing species in constantly noisy environments. Here, we used natural recordings to determine the associations between stream noise and call parameters in the little torrent frog (<i>Amolops torrentis</i>). We also used playbacks of stream noise recorded in natural habitats and playbacks of white noise to examine how male vocal signals change with increasing noise levels. The results show that noise intensity has a significant effect on male call frequency, but not on call amplitude or other call characteristics. Based on this evidence, we suggest that in streamside species stream noise drives males to alter call frequency and call as loudly as possible in order to improve discriminability. These findings provide insights into the role played by ecological selection in the evolution of noise-dependent anuran vocal plasticity.

Abstract:Intraspecific chemical communication in frogs is understudied and the few published cases are limited to externally visible and male-specific breeding glands. Frogs of the family Odontobatrachidae, a West African endemic complex of five morphologically cryptic species, have large, fatty gland-like strands along their lower mandible. We investigated the general anatomy of this gland-like strand and analysed its chemical composition. We found the strand to be present in males and females of all species. The strand varies in markedness, with well-developed strands usually found in reproductively active individuals. The strands are situated under particularly thin skin sections, the vocal sac in male frogs and a respective area in females. Gas-chromatography/mass spectrometry and multivariate analysis revealed that the strands contain sex- and species-specific chemical profiles, which are consistent across geographically distant populations. The profiles varied between reproductive and non-reproductive individuals. These results indicate that the mandibular strands in the Odontobatrachidae comprise a so far overlooked structure (potentially a gland) that most likely plays a role in the mating and/or breeding behaviour of the five Odontobatrachus species. Our results highlight the relevance of multimodal signalling in anurans, and indicate that chemical communication in frogs may not be restricted to sexually dimorphic, apparent skin glands.

Abstract:In most anuran species, vocalizations often consist of different notes with various temporal and spectral acoustic attributes which play a crucial role in their survival and reproductive success. Although the first call note might be necessary for anuran communication, we know little about how different notes of the calls influence female choice. The present study used phonotaxis experiments to examine the effects of the notes on female choice in the Anhui tree frog ( Rhacophorus zhoukaiyae ). The stimulus pair consisted of the original male advertisement call (OC) and one of four revised versions, where the first (WN1), second (WN2), or fifth (WN5) notes were replaced by band-limited white noise (WN), and the second note was replaced by a period of silence (SN2), played back antiphonally. The results showed that (1) the females preferred OC compared with WN1, suggesting the first call note plays an important role in female choice, and (2) the females preferred WN2 and SN2 compared with OC. We discuss the possibility that the second note might be the result of the combined effects of physiological constraint and avoiding backward masking of the second note on the first one over evolutionary time. These results support the notion that the first two functionally antagonistic call notes may influence female choice in this species.

Abstract:Male frogs court females from within crowded choruses, selecting for mechanisms allowing them to call at favourable times relative to the calls of rivals and background chorus noise. To accomplish this, males must continuously evaluate the fluctuating acoustic scene generated by their competitors for opportune times to call. Túngara frogs produce highly frequency- and amplitude-modulated calls from within dense choruses. We used similarly frequency- and amplitude-modulated playback tones to investigate the sensory basis of their call-timing decisions. Results revealed that different frequencies present throughout this species' call differed in their degree of call inhibition, and that lower-amplitude tones were less inhibitory. Call-timing decisions were then driven by fluctuations in inhibition arising from underlying frequency- and amplitude-modulation patterns, with tone transitions that produced steeper decreases in inhibition having higher probabilities of triggering calls. Interactions between the varied behavioural sensitivities to different conspecific call frequencies revealed here, and the stereotyped amplitude- and frequency-modulation patterns present in this species' calls, can explain previously surprising patterns observed in túngara frog choruses. This highlights the importance of understanding the specific sensory drivers underpinning conspecific signalling interactions, and reveals how sensory systems can mediate the interplay between signal perception and production to facilitate adaptive communication strategies.
