Abstract:Acoustic signals are constrained by background noise. Visual signals are an alternative or complementary communication mode in noisy habitats and play a fundamental role in anuran communication. The Bornean rock-skipper frog, Staurois latopalmatus, is a diurnal species living along fast-flowing streams and waterfalls. Males perform foot-flagging displays with either one or two legs to advertise their readiness to defend their territories. In quantitative video analyses of visual displays during 14 male-male agonistic interactions, totaling 106 minutes, foot flagging performed in the direction of the interacting male was the most common display and was performed at a higher rate than advertisement calls. According to a dyadic transition matrix, foot flagging was preceded by foot-flagging displays of interacting males. Advertisement calls were temporally coupled with foot flaggings and act as introductory components to direct the receiver's attention to the subsequent visual display. We conclude that foot flagging acts as a spacing mechanism and may have resulted from the ritualization of agonistic male behavior to minimize physical attacks.

Abstract:How diverse animal communication signals have arisen is a question that has fascinated many. Xenopus frogs have been a model system used for three decades to reveal insights into the neuroendocrine mechanisms and evolution of vocal diversity. Due to the ease of studying central nervous system control of the laryngeal muscles in vitro, Xenopus has helped us understand how variation in vocal communication signals between sexes and between species is produced at the molecular, cellular, and systems levels. Yet, it is becoming easier to make similar advances in non-model organisms. In this paper, we summarize our research on a group of frog species that have evolved a novel hind limb signal known as 'foot flagging.' We have previously shown that foot flagging is androgen dependent and that the evolution of foot flagging in multiple unrelated species is accompanied by the evolution of higher androgen hormone sensitivity in the leg muscles. Here, we present new preliminary data that compare patterns of androgen receptor expression and neuronal cell density in the lumbar spinal cord – the neuromotor system that controls the hind limb – between foot-flagging and non-foot-flagging frog species. We then relate our work to prior findings in Xenopus , highlighting which patterns of hormone sensitivity and neuroanatomical structure are shared between the neuromotor systems underlying Xenopus vocalizations and foot-flagging frogs' limb movement and which appear to be species-specific. Overall, we aim to illustrate the power of drawing inspiration from experiments in model organisms, in which the mechanistic details have been worked out, and then applying these ideas to a non-model species to reveal new details, further complexities, and fresh hypotheses.

Glomerular structural quantities in baseline biopsies from cadaveric donor kidney pairs.

Abstract:Glomerular structural parameters were estimated in kidney biopsies from 11 cadaveric donors at the time of transplantation. The main purpose was to compare the biopsies from the right and left kidney from each donor. Mean glomerular volume was determined by light microscopy. The ultrastructural parameters were basement membrane thickness and volume fractions of mesangial regions with either the tuft or total glomerulus, (i.e. the tuft plus the urinary space in between the capillaries) as reference spaces. Further, the mesangial composition was determined as the volume fraction of basement membrane-like material to total mesangial regions (Vv(BMLM/mes]. The structural quantities conformed with those obtained in living kidney donors. The coefficient of variation (CV) for the group of 11 cases was highest for the mean glomerular volume (0.26) and lowest for the Vv(BMLM/mes) (0.10). The coefficient of error (CE) of the two independent determinations of structural parameters in each individual was well below 5%. The findings therefore demonstrate that, with the methods used, a renal biopsy gives valid and reliable information for that particular individual - at least in cases with 'normal' structures.

Abstract:Being aggressive and by extension, dominant, is an important mechanism for determining access to resources such as mates or territories. While predictors of contest outcome and dominance are increasingly studied, we have a poor understanding of how they vary across populations. Here, I use the widely distributed Australian agamid lizard, the Jacky dragon (Amphibolurus muricatus), to quantify variation in features predicting contest outcome among males of different populations. I measured physical attributes, maximal physiological performance capacity (sprint speed, endurance, bite force) and visual displays during staged encounters. I found that morphology, performance capacity and the type and frequency of visual displays used during agonistic interactions varied significantly across populations. Contest winners from the Cann River State Forest population favored tail‐flicks and push‐up/body‐rocks, while those from Royal National Park were more likely to chase and individuals from Yarratt State Forest performed more bite‐lunges than other populations. The losers of contests also differed in their displays. Individuals from the Cann River population were dominant over the others based on behavioral attributes (i.e., aggressive visual displays, chases and bite‐lunges). I suggest that population differences in signal form and function could have implications for range dynamics as populations come into contact in an era of rapid environmental change.

Abstract:Sexual selection can result in the evolution of extreme armaments and ornaments, and the development and maintenance of these traits can come at a considerable cost. These costs have been implicated in enforcing an upper limit on trait divergence and promoting condition-dependent traits, such that only individuals in sufficiently high condition can effectively wield these armaments and advertise these ornaments. Numerous studies demonstrate the condition-dependence of sexually selected traits, especially those used by males to advertise to females. In this study, we investigated condition-dependent mating calls in the túngara frog Physalaemus ( = Engystomops ) pustulosus . We manipulated male condition in the laboratory over a nine-day period by restricting food availability. We then documented: the relationship between male condition (the relative change in body mass from night 1 to night 9) and acoustic parameters of his mating call; how male condition influenced the male's responses to call playbacks; and finally, how male condition influenced the attractiveness of the male's calls to females. Males who were not fed during this period showed significant changes in call frequency, duration, and amplitude. In response to playbacks, unfed males called less, and made fewer complex calls. Finally, in phonotaxis experiments, females were more attracted to the calls of unfed males on night 1 to the calls of the same males on night 9. Fed males, on the other hand, showed no significant differences between nights 1 and 9 in call parameters, calling effort, and call attractiveness. This study shows the pervasive effects of condition on three aspects of sexual communication: signal parameters, behavioral response to vocal competition, and mating call attractiveness.

Abstract:Significance Communication often is a major component of social interactions. Signaling individuals are faced with the challenge of capturing the attention of intended receivers while limiting eavesdropping by potential predators. We conducted an experiment in nature to evaluate the hypothesis that prey species can modulate the physical properties of movement-based displays in response to the presence of predators. We found that male brown anoles dramatically decreased the amplitude of their head-bob displays in the presence of a predatory lizard, resulting in less conspicuous signals. Although less conspicuous signals may be safer for the signaling individuals, they also reduce the distance from which potential mates or competitors can detect them, which might affect the territory size and reproductive success of signaling males.

Abstract:Animal communication can be complex, often involving multiple static and dynamic traits. The extent to which these traits are correlated can elucidate their function as either redundant or multiple messages. Using the agamid lizard, Psammophilus dorsalis, as a model system we examined patterns of trait expression and the role of steroid hormones in mediating these traits during social interactions. We staged male-male interactions in the lab and measured the repertoire of display behaviour and colours, which change dynamically in the visible and ultraviolet ranges in different body regions. Additionally, we measured testosterone and corticosterone levels before and after the social trials. Our results show that within behaviour and colour trait categories, components were strongly correlated within individuals, suggesting either a shared physiological pathway or redundant information content. However, across trait categories, correlation patterns varied. The chromatic contrast of the (yellow) dorsal region of lizards was correlated with both body size and level of aggression, whereas the size of UV patches was correlated with body size only. We also found a negative association between baseline corticosterone levels, body size and dorsal yellow chromatic contrast, suggesting a mechanistic link between these traits. However, social interaction induced testosterone and corticosterone levels were uncorrelated with the expression of the dynamic behavioural and colour displays during the social interactions itself. Notably, the intensity of colour and behavioural displays of males were matched by their opponents. Overall, our results suggest that multiple signalling traits can ensure both redundancy as well as provide multiple messages to receivers, thus improving the robustness of information transfer, particularly during competitions which have high fitness consequences.

Phenotypic variation reveals sites of evolutionary constraint in the androgenic signaling pathway

Abstract:Steroid hormone systems play an important role in shaping the evolution of vertebrate sexual traits, but several aspects of this relationship remain unclear. For example, we currently know little about how steroid signaling complexes are adapted to accommodate the emergence of behavior in response to sexual selection. We use downy woodpeckers (Dryobates pubescens) to evaluate how the machinery underlying androgen action can evolve to accommodate this bird's main territorial signal, the drum. We focus specifically on modifications to androgenic mechanisms in the primary neck muscle that actuates the hammering movements underlying this signal. Of the signaling components we examine, we find that levels of circulating testosterone (T) and androgen receptor (AR) expression are consistently increased in a way that likely enhances androgenic regulation of drumming. By contrast, the expression of nuclear receptor co-factors-the 'molecular rheostats' of steroid action-show no such relationship in our analyses. If anything, co-factors are expressed in directions that would presumably hinder androgenic regulation of the drum. These findings therefore collectively point to T levels and AR as the more evolutionarily labile components of the androgenic system, in that they are likely more apt to change over time to support sexual selection for territorial signaling in woodpeckers. Yet the signaling elements that fine-tune AR's functional effects on the genome-namely the receptor's transcriptional co-factors-do not change in such a manner, and thus may be under tighter evolutionary constraint.

Abstract:Intraspecific chemical communication in frogs is understudied and the few published cases are limited to externally visible and male-specific breeding glands. Frogs of the family Odontobatrachidae, a West African endemic complex of five morphologically cryptic species, have large, fatty gland-like strands along their lower mandible. We investigated the general anatomy of this gland-like strand and analysed its chemical composition. We found the strand to be present in males and females of all species. The strand varies in markedness, with well-developed strands usually found in reproductively active individuals. The strands are situated under particularly thin skin sections, the vocal sac in male frogs and a respective area in females. Gas-chromatography/mass spectrometry and multivariate analysis revealed that the strands contain sex- and species-specific chemical profiles, which are consistent across geographically distant populations. The profiles varied between reproductive and non-reproductive individuals. These results indicate that the mandibular strands in the Odontobatrachidae comprise a so far overlooked structure (potentially a gland) that most likely plays a role in the mating and/or breeding behaviour of the five Odontobatrachus species. Our results highlight the relevance of multimodal signalling in anurans, and indicate that chemical communication in frogs may not be restricted to sexually dimorphic, apparent skin glands.

Abstract:RNA interference is a post-transcriptional silencing mechanism triggered by the bioavailability and/or exogenous introduction of double-stranded RNA (dsRNA) into cells. Here we describe a novel method for the synthesis of siRNA in a single vessel. The method employs in vitro transcription and a single-stranded DNA (ssDNA) template and design, which incorporates upon self-annealing, two promoters, two templates, and three loop regions. Using this method of synthesis we generated efficacious siRNAs designed to silence both exogenous and endogenous genes in mammalian cells. Due to its unique design the single-stranded template is easily amenable to adaptation for attachment to surface platforms for synthesis of siRNAs. A siRNA synthesis platform was generated using a 3' end-biotinylated ssDNA template tethered to a streptavidin coated surface that generates stable siRNAs under multiple cycles of production. Together these data demonstrate a unique and robust method for scalable siRNA synthesis with potential application in RNAi-based array systems.

Abstract:Testosterone is known as a "male" hormone; however, females also synthetize testosterone, which influences female sexual and aggressive behavior. In female vertebrates, as in males, testosterone levels can vary seasonally. However, female testosterone levels may also be related with female anogenital distance (AGD) length phenotype (a proxy of prenatal androgen exposure), and the social group environment. We used data from a long-term rodent study (2009–2019) in a natural population of degus ( Octodon degus) to examine the potential associations between female serum testosterone levels, season, female AGD phenotype, and social group composition. We quantified female serum testosterone levels during the mating and offspring rearing seasons, and we determined the number of females and males in social groups, as well the composition of groups, in terms of the AGD of the female and male group mates. Our results indicate that female testosterone levels vary with season, being highest during the offspring rearing season. Additionally, female testosterone levels were associated with the number of male group-members and the AGD of male group-members but were not associated with female social environment and focal female AGD phenotype. Together, our results suggest that female testosterone levels are sensitive to intersexual interactions. Our results also reveal that female and male testosterone levels do not differ between the sexes, a finding previously reported only in rock hyraxes. We discuss how the complex social system of degus could be driving this physiological similarity between the sexes.

Abstract:Animal courtship and aggressive displays are dramatic, often sexually dimorphic behaviors that require the coordinated modulation of diverse motivational and motor circuits. In Alstons singing mice (Scotinomys teguina), a novel and elaborate advertisement vocalization is sexually dimorphic and steroid sensitive. Males sing more often than females, and on average male songs have more notes. Song is influenced by circulating androgens, but how such hormonal differences influence the diverse brain regions involved in vocal display is not understood. To characterize androgen-sensitive sites in the vocal motor pathway, we used two isoforms of pseudorabies virus (PRV) to double-label circuits ending in laryngeal and jaw muscles involved in vocalization, and co-labeled these neurons for androgen receptor (AR). Next we manipulated circulating androgens and observed the effects on AR distribution and male song. We find androgens drive coordinated changes in AR abundance across motor and motivational circuits, and both individual and group differences in song are associated with AR abundance. The results reveal how circulating androgens and the auto-regulation of androgen receptors can influence the diverse circuits necessary for elaborate advertisement displays.

Male Serrate-Legged Treefrogs Adjust Competition Strategies According to Visual or Chemical Cues from Females.

Abstract:There is increasing evidence that many anurans use multimodal cues to detect, discriminate and/or locate conspecifics and thus modify their behaviors. To date, however, most studies have focused on the roles of multimodal cues in female choice or male-male interactions. In the present study, we conducted an experiment to investigate whether male serrate-legged small treefrogs (Kurixalus odontotarsus) used visual or chemical cues to detect females and thus altered their competition strategies in different calling contexts. Three acoustic stimuli (advertisement calls, aggressive calls and compound calls) were broadcast in a randomized order after a spontaneous period to focal males in one of four treatment groups: combined visual and chemical cues of a female, only chemical cues, only visual cues and a control (with no females). We recorded the vocal responses of the focal males during each 3-min period. Our results demonstrate that males reduce total number of calls in response to the presence of females, regardless of how they perceived the females. In response to advertisement calls and compound calls, males that perceived females through chemical cues produced relatively fewer advertisement calls but more aggressive calls. In addition, they produced relatively more aggressive calls during the playback of aggressive calls. Taken together, our study suggests that male K. odontotarsus adjust their competition strategies according to the visual or chemical cues of potential mates and highlights the important role of multisensory cues in male frogs' perception of females.

Abstract:Understanding why females prefer exaggerated male mating displays, when males offer little more than sperm in lek mating systems, has been an important challenge in behavioral ecology and evolutionary biology. The Hamilton-Zuk hypothesis proposes that the expression of mating displays is limited by parasitism and females choose parasite-resistant males that can produce costly, exaggerated displays. We used Pacific treefrogs ( Pseudacris regilla ) and natural infections by the fungal pathogen Batrachochytrium dendrobatidis ( Bd ), to test predictions of the Hamilton-Zuk hypothesis. Specifically, we predicted that (i) call properties, especially those that are more likely under selection by female choice, correlate with parasitic infection, and (ii) females discriminate against highly infected males based on the properties of their mating call. We found that energetically costly call properties that are likely under selection by female choice, like call rate and call effort, were not influenced by Bd infection. However, males with high levels of Bd infection produced calls with faster pulse rates than males with lower levels of infection. In two-choice tests, females did not show a preference between calls that resemble males with either high or low levels of Bd infection. Our results provide little support for the Hamilton-Zuk hypothesis and suggest that Bd infection may have little effect on female choice in natural settings. We discuss our results in terms of the effects of Bd and other parasites in anuran mating behavior.

Abstract:Background Signal detection is crucial to survival and successful reproduction, and animals often modify behavioral decisions based on information they obtained from the social context. Undeniably, the decision-making in male-male competition and female choice of anurans (frogs and toads) depends heavily on acoustic signals. However, increasing empirical evidence suggests that additional or alternative types of cue (e.g., visual, chemical, and vibratory) can be used to detect, discriminate and locate conspecifics in many anuran species. Nevertheless, few studies have investigated whether conspecific odor cues affect male's calling behavior. In this study, we conducted an experiment to investigate whether and how different chemical cues (male odors, female odors, and stress odors) from conspecifics affect male's calling strategies in serrate-legged small treefrogs (Kurixalus odontotarsus), and whether the combined chemical and acoustic stimuli have additive effects on calling behavior or not. Results We found that compared with female odors, male K. odontotarsus reduced calling investment in response to male odors or stress odors, in the absence of rival's advertisement calls. When odor stimuli and advertisement calls were presented simultaneously, however, there were no differences in the vocal response of focal males among odor groups. Conclusions These results provide evidence that male treefrogs switch calling investment according to different odor cues from conspecifics, and further demonstrate that calling behavior can be affected by chemical cues in anuran species. Our study highlights the potential role of airborne chemical cues in sex identification and contributes to increase our understanding of anuran communication.
Ecological Correlates of Small Territories and Intra-Seasonal Variation in the Social Context of Displays in Male Water Dragons, Intellagama lesueurii

Abstract:Size and location of territories have important fitness consequences for breeding males in many species, including lizards. Australian Water Dragons (Intellagama lesueurii) are an interesting study system in which to examine socio-ecological aspects of territory size and male behavior because, despite their large body size, these lizards form dense aggregations within urban riparian habitats. In one such population, we found that males defended territories smaller than have been reported for most other large lizards. Surprisingly, smaller males defended larger territories, but larger territories did not increase access to females. Instead, territory size was negatively correlated with conspecific density, including females, which in turn was highest near a creek that provided abundant prey and refuges. That is, territories were smaller in sites offering more resources for reproduction, feeding, and predator evasion. The relative proportion of displays males gave during contests with rivals did not vary within the reproductive season. However, male travel and the context of stereotypical head displays showed a surprising pattern of seasonal variation. As the season progressed, males increased the proportion of displays to females, but decreased the proportion of undirected territorial advertisement displays. Because Water Dragons are long-lived and philopatric, increased display during interactions with postbreeding females may enhance mating opportunities in future reproductive seasons.

Abstract:Abstract Vocalization is the main form of communication in many animals, including frogs, which commonly emit advertisement calls to attract females and maintain spacing. In noisy environments such as streams, mechanisms to maximize signaling efficiency may include vocal plasticity and/or movement of individuals to quieter sections, but which strategy is used is still uncertain. We investigated the influence of stream geophony on the advertisement call of the torrent frog Hylodes perere in the Atlantic Rainforest, in southeastern Brazil. In a mark-recapture study, we tested if males remain in their territories and thus adjust their advertisement calls to maximize their communication. We ran linear mixed models to verify the relationship of call parameters and stream geophony, body size, and environmental temperature. We found that males remained in the same location across time, increased call intensity in noisier environments, but did not reduce call effort. Males also increased the dominant frequency in these situations, suggesting a modulation in this parameter. Our results indicate that territoriality is an important factor for males to increase call intensity to surpass stream noise instead of repositioning along the stream. However, because call effort was maintained, we suggest that sexual selection is crucial in this system, favoring males that better detect others and adjust their call efficiency. This is the first study to evaluate simultaneously frog movements and adaptations to geophony, which contributes to the investigation of the concomitant environmental and sexual selective pressures in species that communicate in noisy environments.

Abstract:Males in many vertebrate species have colorful ornaments that evolved by sexual selection. The role of androgens in the genesis and maintenance of these signals is unclear. We studied 21 adult high-ranking male rhesus macaques from nine social groups in the free-ranging population on Cayo Santiago, Puerto Rico, and analyzed facial and genital skin luminance and redness, fecal androgens, rates of mating behaviors, and offspring sired. Facial and genital coloration varied in relation to age, mating behavior, reproductive success, and testosterone concentration. Our results indicate that skin coloration in high-ranking male rhesus macaques is a sexually-selected trait mediated by androgens. These results add to the growing literature on the proximate and ultimate causes of male sexual signals and highlight the need to examine how these characteristics change with age in other species.
