Abstract:Owing to flexible structural design and rapid prototyping manufacturing, 3D printable hydrogels with solid-like and fluid-like characters have emerged as a promising candidate for biomedical engineering, soft robotics, and flexible electronics. However, existing 3D printing of hydrogel place substantial constraints on hydrogel high-fidelity of the printed hydrogel complex structures due to the shape deformation and collapse caused by water evaporation. Herein, we propose a mixed solvent printing strategy followed with water dialysis to achieve vat photopolymerization 3D printing of high-fidelity hydrogel structure by restraining water evaporation. Concretely, dimethyl sulfoxide (DMSO) is introduced into the hydrogel ink as co-solvent to improve the anti-dehydration performance during 3D printing. The as-prepared gels are then transformed into hydrogels with good fidelity and excellent performance through solvent exchange and water dialysis. Enabled by these characteristics, various functional devices with complex structures are demonstrated with the present high-fidelity 3D printing of diverse hydrogels. The solvent assisted high-fidelity hydrogel 3D printing protocol combining with the compatibility to various high-performance hydrogels is believed to be promising for wearable devices, wound dressings, electronic skin, and intelligent robots.

Abstract:As the biocompatible materials, hydrogels have been widely used in three- dimensional (3D) bioprinting/organ printing to load cell for tissue engineering. It is important to precisely control hydrogels deposition during printing the mimic organ structures. However, the printability of hydrogels about printing parameters is seldom addressed. In this paper, we systemically investigated the printability of hydrogels from printing lines (one dimensional, 1D structures) to printing lattices/films (two dimensional, 2D structures) and printing 3D structures with a special attention to the accurate printing. After a series of experiments, we discovered the relationships between the important factors such as air pressure, feedrate, or even printing distance and the printing quality of the expected structures. Dumbbell shape was observed in the lattice structures printing due to the hydrogel diffuses at the intersection. Collapses and fusion of adjacent layer would result in the error accumulation at Z direction which was an important fact that could cause printing failure. Finally, we successfully demonstrated a 3D printing hydrogel scaffold through harmonize with all the parameters. The cell viability after printing was compared with the casting and the results showed that our bioprinting method almost had no extra damage to the cells.

Abstract:<p>3D printing alias additive manufacturing can transform 3D virtual models created by computer-aided design (CAD) into physical 3D objects in a layer-by-layer manner dispensing with conventional molding or machining. Since the incipiency, significant advancements have been achieved in understanding the process of 3D printing and the relationship of component, structure, property and application of the created objects. Because hydrogels are one of the most feasible classes of ink materials for 3D printing and this field has been rapidly advancing, this Review focuses on hydrogel designs and development of advanced hydrogel-based biomaterial inks and bioinks for 3D printing. It covers 3D printing techniques including laser printing (stereolithography, two-photon polymerization), extrusion printing (3D plotting, direct ink writing), inkjet printing, 3D bioprinting, 4D printing and 4D bioprinting. It provides a comprehensive overview and discussion of the tailorability of material, mechanical, physical, chemical and biological properties of hydrogels to enable advanced hydrogel designs for 3D printing. The range of hydrogel-forming polymers covered encompasses biopolymers, synthetic polymers, polymer blends, nanocomposites, functional polymers, and cell-laden systems. The representative biomedical applications selected demonstrate how hydrogel-based 3D printing is being exploited in tissue engineering, regenerative medicine, cancer research, in vitro disease modeling, high-throughput drug screening, surgical preparation, soft robotics and flexible wearable electronics. Incomparable by thermoplastics, thermosets, ceramics and metals, hydrogel-based 3D printing is playing a pivotal role in the design and creation of advanced functional (bio)systems in a customizable way. An outlook on future directions of hydrogel-based 3D printing is presented.</p>

Abstract:This study explores the dynamic field of 3D-printed hydrogels, emphasizing advancements and challenges in customization, fabrication, and functionalization for applications in biomedical engineering, soft robotics, and tissue engineering. It delves into the significance of tailored biomedical scaffolds for tissue regeneration, the enhancement in bioinks for realistic tissue replication, and the development of bioinspired actuators. Additionally, this paper addresses fabrication issues in soft robotics, aiming to mimic biological structures through high-resolution, multimaterial printing. In tissue engineering, it highlights efforts to create environments conducive to cell migration and functional tissue development. This research also extends to drug delivery systems, focusing on controlled release and biocompatibility, and examines the integration of hydrogels with electronic components for bioelectronic applications. The interdisciplinary nature of these efforts highlights a commitment to overcoming material limitations and optimizing fabrication techniques to realize the full potential of 3D-printed hydrogels in improving health and well-being.

Abstract:Hydrogels have found widespread use in tissue engineering and functional devices because of their biomimetic properties and multifunctionalities. As a promising fabrication technique, three-dimensional (3D) printing is able to construct hydrogels into complex artificial tissues and soft robotics with precise control. In this chapter, we first provide a brief introduction of 3D printing methods and general principles that make a hydrogel useful for 3D printing. Subsequently, we discuss the relationships of structure and property of hydrogels (based on natural polymers and synthetic polymers) and hydrogel composites (double network hydrogels, particle-reinforced hydrogels, and fiber-reinforced hydrogels), the control of key factors (viscosity, shear thinning, thixotropy, and interfacial bonding) in 3D printing, and its effect on printability of hydrogels and fidelity of constructs. Additionally, we also present the applications of 3D printed hydrogels as well as recent progress of hydrogel and hydrogel composites in 4D printing applications. Finally, challenges and future perspectives on 3D/4D printing of hydrogels will be discussed.

Abstract:Hydrogels are a promising material for a variety of applications after appropriate functional and structural design, which alters the physicochemical properties and cell signaling pathways of the hydrogels. Over the past few decades, considerable scientific research has made breakthroughs in a variety of applications such as pharmaceuticals, biotechnology, agriculture, biosensors, bioseparation, defense, and cosmetics. In the present review, different classifications of hydrogels and their limitations have been discussed. In addition, techniques involved in improving the physical, mechanical, and biological properties of hydrogels by admixing various organic and inorganic materials are explored. Future 3D printing technology will substantially advance the ability to pattern molecules, cells, and organs. With significant potential for producing living tissue structures or organs, hydrogels can successfully print mammalian cells and retain their functionalities. Furthermore, recent advances in functional hydrogels such as photo- and pH-responsive hydrogels and drug-delivery hydrogels are discussed in detail for biomedical applications.

Abstract:Hydrogels with tailor-made swelling-shrinkable properties have aroused considerable interest in numerous biomedical domains. For example, as swelling is a key issue for blood and wound extrudates absorption, the transference of nutrients and metabolites, as well as drug diffusion and release, hydrogels with high swelling capacity have been widely applicated in full-thickness skin wound healing and tissue regeneration, and drug delivery. Nevertheless, in the fields of tissue adhesives and internal soft-tissue wound healing, and bioelectronics, non-swelling hydrogels play very important functions owing to their stable macroscopic dimension and physical performance in physiological environment. Moreover, the negative swelling behavior (i.e., shrinkage) of hydrogels can be exploited to drive noninvasive wound closure, and achieve resolution enhancement of hydrogel scaffolds. In addition, it can help push out the entrapped drugs, thus promote drug release. However, there still has not been a general review of the constructions and biomedical applications of hydrogels from the viewpoint of swelling-shrinkable properties. Therefore, this review summarizes the tactics employed so far in tailoring the swelling-shrinkable properties of hydrogels and their biomedical applications. And a relatively comprehensive understanding of the current progress and future challenge of the hydrogels with different swelling-shrinkable features is provided for potential clinical translations.

Abstract:3D printing hydrogels with a good resolution, diversified geometries, and a tunable mechanical performance is still a big challenge, which is mainly due to the low photo-reactivity of the high-water containing precursor and the low modulus of the photo-cured hydrogel. In the present work, an acryloylmorpholine - acrylic acid - PEG diacrylate drygel was firstly printed with a good fidelity and subsequently post- treated with an aqueous solution containing specific ions, during which hydration of the hydrophilic network and coordination between the carboxyl and ions occurred simultaneously. The obtained dual-crosslinked hydrogel combined a complex geometry and a good stretchability. This approach decoupled the printability and mechanical properties of the hydrogels. To furtherly show this advantage over conventional direct hydrogel 3D printing, ultrasoft hydrogel lattice structures which cannot be self-supported and purely ionic crosslinked hydrogels with a good self-weldability were demonstrated.

Abstract:3D printing technology has been widely explored for the rapid design and fabrication of hydrogels, as required by complicated soft structures and devices. Here, a new 3D printing method is presented based on the rheology modifier of Carbomer for direct ink writing of various functional hydrogels. Carbomer is shown to be highly efficient in providing ideal rheological behaviors for multifunctional hydrogel inks, including double network hydrogels, magnetic hydrogels, temperature-sensitive hydrogels, and biogels, with a low dosage (at least 0.5% w/v) recorded. Besides the excellent printing performance, mechanical behaviors, and biocompatibility, the 3D printed multifunctional hydrogels enable various soft devices, including loadable webs, soft robots, 4D printed leaves, and hydrogel Petri dishes. Moreover, with its unprecedented capability, the Carbomer-based 3D printing method opens new avenues for bioprinting manufacturing and integrated hydrogel devices.

Abstract:The advent of 3D printing technology in the 1980s enabled the fabrication of patient-specific products with precise shape, size, and complexity. A variety of 3D printing techniques, such as direct ink writing by extrusion, particle fusion printing, light-induced printing, volumetric bioprinting, and inkjet printing, have been explored recently for the purpose of developing desired 3D printed objects. Among these techniques, direct ink writing has been used extensively due to its affordability, ease of use, potential for scaling up, and faster speed. This approach saves time and resources while allowing for the flexible use of various materials. The lack of a variety of varied printing inks, however, continues to be an impediment to its widespread commercial use. Owing to their quick gelling behaviour, extrudability, and shape fidelity, injectable hydrogels have become interesting alternatives for printing inks. Hydrogel inks can be developed by either natural or synthetic polymers, or a combination of the both, to achieve the desired rheological and mechanical properties utilizing various strategies of crosslinking and incorporation of nanoparticles like nanoclay, bioactive glass, graphene oxide and silica nanoparticles. Further, various stimuli responsive hydrogel inks responding to various external stimuli (such as temperature, pH, magnetic field) can be developed to prepare engineered biostructures that can be used in healthcare sector. This review provides a spotlight on recent advancement in developing hydrogel inks and their different physicochemical aspects for developing desired engineered biostructures. Finally, we discuss the opportunities of 4D printing, current difficulties and future prospects for hydrogel inks-based 3D printing to emphasize the significant scope for future innovations.

Abstract:Granular hydrogels are the conglomerations of micrometer-sized hydrogel particles that have recently become promising in tissue growth and three-dimensional (3D) bioprinting. Recent advances in the use of jamming transition of granular hydrogels represent a potential paradigm shift in the extrusion-based 3D bioprinting. These dynamic granular hydrogels are shear thinning and self-healing, enable higher printing performance, and the creation of better physiological conditions for heterocellular constructs. Here, we review the current efforts to explore materials to produce granular hydrogels with novel functional properties, focusing on the granular hydrogels that can be used for supporting baths and bioinks in the extrusion-based 3D bioprinting. The recent advances, benefits, and challenges in this emerging area are highlighted.

Abstract:Three-dimensional (3D) printing enables the production of personalized tissue-engineered products with high tunability and complexity. It is thus an attractive and promising technology in the pharmaceutical and medical fields. Printable and biocompatible hydrogels are attractive materials for 3D printing applications because they offer favorable biomimetic environments for live cells, such as high water content, porous structure, bioactive molecule incorporation, and tunable mechanical properties and degradation rates. However, most conventional hydrogel materials are brittle and mechanically weak and hence cannot meet the mechanical needs for handling and soft and elastic tissue use. Thus, the development of printable, high-strength, and elastic hydrogel materials for 3D printing in tissue repair and regeneration is critical and interesting. In this review, we summarized the recent reports on high-strength and elastic hydrogels for printing use and categorized them into three groups, namely double-network hydrogels, nanocomposite hydrogels, and single-network hydrogels. The reinforcing mechanisms of these high-strength hydrogels and the strategies to improve their printability and biocompatibility were further discussed. These high-strength and elastic hydrogels may offer opportunities to accelerate the development of 3D printing technology and provide new insights for 3D-printed product design in biomedicine. STATEMENT OF SIGNIFICANCE: Biocompatible and biodegradable hydrogels are highly attractive in 3D printing because of their desirable printability and friendly environment for loading bioactive molecules and living cells. The development of high-strength and elastic hydrogels changes the conventional impression of weak and brittle hydrogels and provides new opportunities and inspirations for 3D printing and biomedical applications. In this review, we analyzed the hydrogel reinforcement mechanisms, summarized recent progresses in developing high-strength and elastic hydrogels for 3D printing, and discussed the strategies to improve the printability and biocompatibility of the hydrogel inks.
Chemical modification and printability of shear-thinning hydrogel inks for direct-write 3D printing

Abstract:Shear-thinning hydrogels are often employed in direct-write 3D printing, however, the viscoelastic behaviors that define a printable hydrogel have not been fully established. Herein, we demonstrate a library of hydrogel inks based on the incorporation of water-soluble reactive meth(acrylate) monomers into F127-dimethacrylate hydrogels. This strategy afforded printed hydrogels with a broad range of chemical functionalities and mechanical properties. A systematic investigation was also performed to correlate the printability and mechanical properties to the viscoelastic properties of the hydrogel ink formulations. The materials with a high dynamic yield stress afforded extruded filaments that correlated well with the inner diameter of the printing nozzle. The static yield stress of the material was correlated to the extrusion pressure and print speed required for optimal printing. Thus, this study provides a guide for the future development of hydrogel inks for direct-write 3D printing along with a new set of functional hydrogel inks.
