Abstract:AbstractFunctional soft materials, especially hydrogels have been widely developed to achieve various soft structures and machines. However, synthetic hydrogels commonly show formula‐dependent mechanical properties to fulfill the requirements of mechanical elasticity, stiffness, toughness, and tearing‐resistance for adapting to complex application scenario. Inspired by heterostructures and materials found in nature such as leaves and insect wings, a sequential photopolymerization process combined with site‐selective patterning exposure is reported to prepare programmable hydrogels with locally heterogeneous reinforcement skeletons, i.e., interpenetrating double networks. The heterogeneous interface between soft matrices and stiff skeletons is seamlessly interlocked through strong multiple hydrogen bonds induced by phase transition. By harnessing the size, shape, and distribution of the patterned stiff skeletons, a wide range of mechanical properties of hydrogels including modulus (0.32–5.92 MPa), toughness (0.15–18 kJ m−2), dissipated energy (1–100 kJ m−3), impact resistance, and mechanical anisotropy can be readily sculpted within one material system without needing design and optimization of the complex and elusive material formulation on demand. It is believed that this simple yet powerful method relying on heterogenous patterning would guide the development of functional hydrogel materials with programmable mechanical properties toward potential engineering applications, such as damping and flexible circuits.

Abstract:Elastomeric, robust, and biocompatible hydrogels are rare, while the need for these types of biomaterials in biomedical-related uses remains high. Here, a new family of genetically engineered silk-elastin copolymers (SELPs) with encoded enzymatic crosslinking sites is developed for a new generation of stimuli-responsive yet robust hydrogels. Input into the designs is guided by simulation, and realized via genetic engineering strategies. The avoidance of gamma irradiation or chemical crosslinking during gel fabrication, in lieu of an enzymatic process, expands the versatility of these new gels for the incorporation of labile proteins and cells. In the present study, the new SELP hydrogels offers sequence dependent, reversible stimuli-responsive features. Their stiffness covers almost the full range of the elasticity of soft tissues. Further, physical modification of the silk domains provided a secondary control point to fine-tune mechanical stiffness while preserving stimuli-responsive features, with implications for a variety of biomedical materials and device needs.
Strong and Ultra-Tough Supramolecular Hydrogel Enabled by Strain-Induced Microphase Separation

Abstract:Architected hydrogels are widely used in biomedicine, soft robots, and flexible electronics while still possess big challenges in strong toughness, and shape modeling. Here, inspired with the universal hydrogen bonding interactions in biological systems, a strain-induced microphase separation path toward achieving the printable, tough supramolecular polymer hydrogels by hydrogen bond engineering is developed. Specifically, it subtly designs and fabricates the poly (N-acryloylsemicarbazide-co-acrylic acid) hydrogels with high hydrogen bond energy by phase conversion induced hydrogen bond reconstruction. The resultant hydrogels exhibited the unique strain-induced microphase separation behavior, resulting in the excellent strong toughness with, for example, an ultimate stress of 9.1 +/- 0.3 MPa, strain levels of 1020 +/- 126%, toughness of 33.7 +/- 6.6 MJ m(-3), and fracture energy of 171.1 +/- 34.3 kJ m(-2). More importantly, the hydrogen bond engineered supramolecular hydrogels possess dynamic shape memory character, i.e shape fixing at low temperature while recovery after heating. As the proof of concept, the tailored hydrogel stents are readily manufactured by 3D printing, which showed good biocompatibility, load-bearing and drug elution, being beneficial for the biomedical applications. It is believed that the present 3D printing of the architected dynamic hydrogels with ultrahigh toughness can broaden their applications.

Abstract:The molecular engineering of dissipative energy to construct the hyperelastic hydrogels with outstanding swelling resistance, good biocompatibility, excellent hydrophilicity and skin‐like elastic moduli is introduced. Noticeable progress has been made in mechanically strong and tough of polymer gels, whereas this consistency between mechanical robustness and perfect elasticity remains a challenge. Here, a simple method is conducted for fabricating hyperelastic hydrogels based on the proposed molecular engineering of dissipative energy taking advantage of two distinct interactions with great difference of binding energy. This molecular engineering of dissipative energy refers to the complementarity at the molecular level between strong and weak interactions: the strong interaction provides the mechanical strength, the weak interaction eliminates the yielding and dense chain entanglements are further implanted, resulting in the perfect hyperelasticity. Such a complementarity in reformation time and binding energy of two interactions triggers a wide range of controllable window for regulating mechanical performances, filling the current gap in the design of hyperelasticity using only high‐order structures. The resultant hyperelastic hydrogels exhibit remarkable tensile strength and linear elastic deformation, but also outstanding swelling resistance, biocompatibility, hydrophilicity, and skin‐like elastic moduli. The work thus provides a promising concept for designing elastic and robust polymer gels and brings a new perspective in the combination of mutually incompatible mechanical properties, which will likely promote innovative applications in bioengineering and bioelectronics.

Abstract:Hydrogels based on natural polymers have bright application prospects in biomedical fields due to their outstanding biocompatibility and biodegradability. However, the poor mechanical performances of pure natural polymer‐based hydrogels greatly limit their application prospects. Recently, a variety of strategies has been applied to prepare natural polymer‐based hydrogels with enhanced mechanical properties, which generally exhibit stiffening, strengthening, and stretchable behaviors. This article summarizes the recent progress of natural polymer‐based hydrogels with enhanced mechanical properties. From a structure point of view, four kinds of hydrogel are reviewed; double network hydrogels, nanocomposite hydrogels, click chemistry‐based hydrogels, and supramolecular hydrogels. For each typical hydrogel, its preparation, structure, and mechanical performance are introduced in detail. At the end of this article, the current challenges and future prospects of hydrogels based on natural polymers are discussed and it is pointed out that 3D printing may offer a new platform for the development of natural polymer‐based hydrogels.

Abstract:Hydrogels provide a versatile platform for biomedical material fabrication that can be structurally and mechanically fine‐tuned to various tissues and applications. Applications of hydrogels in biomedicine range from highly dynamic injectable hydrogels that can flow through syringe needles and maintain or recover their structure after extrusion to solid‐like wound‐healing patches that need to be stretchable while providing a selective physical barrier. In this study, a toolbox is designed using thermo‐responsive poly(N‐isopropylacrylamide) (PNIPAM) polymeric matrices and nanocelluloses as reinforcing agent to obtain biocompatible hydrogels with altering mechanical properties, from a liquid injectable to a solid‐like elastic hydrogel. The liquid hydrogels possess low viscosity and shear‐thinning properties at 25°C, which allows facile injection at room temperature, while they become viscoelastic gels at body temperature. In contrast, the covalently cross‐linked solid‐like hydrogels exhibit enhanced viscoelasticity. The liquid hydrogels are biocompatible and are able to delay the in vitro release and maintain the bioactivity of model drugs. The antimicrobial agent loaded solid‐like hydrogels are effective against typical wound‐associated pathogens. This work presents a simple method of tuning hydrogel mechanical strength to easily adapt to applications in different soft tissues and broaden the potential of renewable bio‐nanoparticles in hybrid biomaterials with controlled drug release capabilities. Abstract:Nature uses combined covalent (chemical bonds) and non‐covalent (physical bonds) synthesis in a highly, controlled multi‐step fashion to create functional materials with different mechanical and dynamic properties out of similar building blocks. Surprisingly, this control in fully synthetic systems remains elusive, even though the effects of formulation pathways on the assembly processes have been emphasized—highlighting the importance of and relationship between energy landscapes and function in synthetic systems. Here, we control multiple, coherent supramolecular assembly processes (fiber formation and crosslinking) to formulate hydrogels with tunable mechanical and dynamic properties. Hydrogels are prepared via two different formulation methods using similar building blocks (monofunctional and bifunctional supramolecular monomers), including (1) the mixing of the supramolecular monomers under basic conditions (Fbasic) and (2) the mixing of the supramolecular monomers under neutral conditions (Fneutral). In Fbasic, network formation is induced via simultaneous fiber formation and crosslinking, yielding homogeneously mixed networks which are dense, stiff (~10 kPa) and robust. In Fneutral, network formation is induced through sequential fiber formation and crosslinking, yielding heterogenous, soft (~2 kPa) and dynamic networks. With these results, we advance towards the controlled, multistep, non‐covalent synthesis to create larger hierarchical, functional structures, similar to nature.

Digital Light Processing 4D Printing of Transparent, Strong, Highly Conductive Hydrogels

Abstract:Hydrogels have excellent biocompatibility, transparency, stretchability, and ionic conductivity, but their fabrication through photopolymerization-based 3D printing is limited due to the low solubility of hydrophobic photoinitiators and lack of efficient hydrophilic photoinitiators. Herein, a type of microemulsion is synthesized and the common hydrophobic photoinitiator can be adopted and finally, a series of transparent hydrogels with high strength (up to 22.9 MPa), elasticity (up to 583%), and ionic conductivity (up to 9.64 S m<sup>–1</sup>) are fabricated through digital light processing 3D printing technology. Objects with complex structures and a high printing resolution are printed. Hydrogels with both high strength and high ionic conductivity are obtained through chemical crosslinking and ion coordination effect. Dual-material 3D printing is applied to package the hydrogel with elastomers. Due to the high sensitivity and reliability under both stretching and compressive deformation, the hydrogel sensors are applied to monitor various human motions. A Facile Method To Fabricate Tough Hydrogel With Ultra-Wide Adjustable Stiffness, Stress, And Fast Recoverability

Abstract:In this article, we report a synergistic strategy to develop dual physically cross-linked tough hydrogels via one-pot bulk copolymerization of N-vinyl-2-pyrrolidone, acrylic acid, and stearyl methylacrylate (SMA) without any adscititious surfactant. Due to synergic effects of hydrogen bonding and hydrophobic association, the resulted dual physically cross-linked hydrogels (DP Gel) with ultra-wide range adjustable Young's modulus (0.08-45.6 MPa), tensile stress (0.7-6.9 MPa), and toughness (3.3-23.1 MJ m(-3)). Stretching to 300%, DP Gel exhibited fast recoverability that remained similar to 95% of initial dissipated energy after resting in 60 degrees C for 3 min. Finally, scanning electron microscopy revealed that the microstructure of hydrogel changed from phase separation structure to micro phase separation as SMA added, which accounted for excellent performance of DP Gel. Rationally Designed Synthetic Protein Hydrogels with Predictable and Controllable Mechanical Properties

Abstract:A key challenge in biomaterials research is to produce synthetic hydrogels that can replicate the diverse mechanical properties of the naturally occurring tissues for various biomedical applications, including tissue engineering, stem cell and cancer research, cell therapy, and immunomodulation. However, currently, the methods that can be used to control the mechanical properties of hydrogels are very limited and are mainly focused only on the elasticity of hydrogels. In this work, combining single molecule force spectroscopy, protein engineering and theoretical modeling, we show that synthetic protein hydrogels with predictable mechanical properties can be rationally designed using protein building blocks with known mechanical properties at the molecular level. We have successfully tuned the elasticity, extensibility, toughness and self-healing of hydrogels using this approach [1]. We have also demonstrated the engineering of physically crosslinked muscle-mimic hydrogels which are strong, tough, elastic and able to function under significant load and spontaneously self-heal when damaged [1]. Next, by using light-responsive proteins as building blocks, we have been able to engineer hydrogels whose mechanical properties are reversibly tunable by light illumination [2]. These hydrogels can capture the dynamic mechanical environments of naturally occurring extracellular matrices and are potentially suitable for cell culture and tissue engineering. Finally, we show that the hydrogel network structure is also important for the mechanical properties of hydrogels. The hydrogels made of the same protein building blocks but with different network structures can have distinct mechanical responses. These studies provide a biomechanical basis for the rational design of synthetic proteins hydrogels with predictable and controllable mechanical properties.
Hydrophobic Association Mediated Physical Hydrogels with High Strength and Healing Ability

Abstract:Supramolecular polymeric hydrogels, combining the responsiveness of noncovalent interactions and good mechanical properties of polymers, have received increasing attentions due to their high strength, multi-responses to stimuli, and good processability. In this article, we develop transparent physical hydrogels by simply casting and reswelling the films of copolymers synthesized by polymerization of a hydrophilic monomer of acrylic acid (AA) with a small fraction of hydrophobic monomer of stearyl acrylate (SA). The long alkyl chains of SA units segregate in water to form hydrophobic associations, serving as the physical crosslinking junctions of gels. The gels are robust with tensile strength up to similar to 2 MPa; the mechanical properties can be tuned by varying the composition of copolymers and depend on the deformation rate. The gels are responsive to temperature, pH, and ethanol solvent, which mediate the stability of the hydrophobic associations and even lead to gel-to-sol transition. Based on this reversible transition, the gels show stimuli-mediated healing ability. Gels with complex structures can be constructed by localized welding. In addition, the gels exhibit shape memory behaviour; the shape can be memorized by forming strong coordination between carboxylic acid groups and additional Fe3+ ions and relaxed by reduction of the Fe3+ ions. Abstract:Hydrogels, promising biological materials, need to have both strong mechanical properties and also inherent self-healing properties. In this work a double cross-linked network (DN) hydrogel was designed and prepared by combining a Diels-Alder click reaction and coordination effects. This DN hydrogel had good thermodynamic properties, anti-EDTA performance and self-healing properties. In addition, the mechanical properties, swelling properties and surface morphology of DN hydrogels can be controlled by adjusting the ratio of Fe3+-catechol. The adjustment of pH value can change the color, crosslinking mode and mechanical properties of the DN hydrogel. This smart hydrogel created from DA click chemistry and coordination effects has significance for guiding the design of new hydrogels with good mechanical properties, self-healing properties and controlled cross-link density.

Abstract:<p>Supramolecular hydrogels display unique dynamic features such as self-healing and injectability which are endowed by their non-covalent crosslinking. These extraordinary dynamic properties enable them to be broadly applied in various fields, but also decrease their mechanical strength. This inherent contradiction between mechanical strength and dynamic features has posed great challenge to their practical applications. This article provides an overview of emerging techniques for mechanical strengthening of supramolecular hydrogels with preserved dynamic features. Inspired by the "strong means slow" principle of transient supramolecular networks, particular focus is placed on how the exchange kinetics of the supramolecular crosslinkers can be engineered to tune their mechanical strength. Some other effective strengthening strategies are also reviewed, which are usually combined to realize the coexistence of mechanical strength and dynamics. Meanwhile, we point out the future development of supramolecular hydrogels with unique mechanical properties and their potential challenges.</p>

Abstract:Hydrogels with good mechanical properties, excellent biocompatibility and designable shapes are of great importance for their biomedical applications. Herein, a series of high strength, biocompatible hydrogels have been synthesized by integrating sodium citrate into the thermally reversible chitosan/gelatin to form multiple physically crosslinking networks. Besides the ideal formability, a thermal etching or welding method has been developed to program the surface morphology and fabricate hydrogels with complicated shapes freely. More impressively, the special hollow "cup-shaped and tubular" structure has also been constructed by applying an interrupted gelation process in controlled ion crosslinking time and the subsequent dissolving process at 37°C in deionized water. The high strength, biocompatible hydrogels with special internal and external shape adjustable characters, potentially useful in vascular repair and substitutes of cartilage, may further broaden our understanding of the plasticity of the hydrogels.
Plastic-Like Supramolecular Hydrogels with Polyelectrolyte/Surfactant Complexes as Physical Cross-links

Abstract:Developing hydrogels with new structures and extraordinary performances is fundamental and mission-critical for the advancements of gel materials. Here, we report a class of tough supramolecular hydrogels facilely prepared by polymerizing methacrylic acid precursor solution in the presence of hexadecyltrimethylammonium chloride micelles. After swelling the as-prepared hydrogels in water, strong polyelectrolyte/surfactant complexes (PESCs) are formed between the weakly charged polymer chains and oppositely charged surfactants, serving as the physical cross-links of the gel matrix. The equilibrated hydrogels are transparent with a water content of 50–85 wt % and possess excellent mechanical properties, with a tensile breaking stress of 0.1–5 MPa, a breaking strain of 600–1200%, and Young's modulus of 1–70 MPa. Typical yielding is observed at a small tensile strain of ∼10%, followed by forced elastic deformation of the hydrogels, which are in a glassy state due to the reduced segmental mobility of the matrix highly cross-linked by PESCs. The plastic-like mechanical properties of hydrogels could be well tuned by pH, temperature, and ionic strength that influence the ionization of polymer chains and the strength of PESCs. These dynamic behaviors render the hydrogels with self-recovery and shape-memory properties. Furthermore, the nanosized hydrophobic pockets within the hydrogels afford the capacity of loading functional hydrophobic molecules with promising applications as fluorescent materials and drug delivery systems. Abstract:A novel hydrogel with a new, well‐ defined network structure is prepared through a two‐step method in which the radiation‐peroxidized macromolecular microspheres act as both initiators and crosslinkers. The macromolecular microsphere composite hydrogel (see figure) can effectively dissipate applied mechanical stress and has extremely high mechanical strength. Some of the hydrogels can nearly completely recover their original shapes, even after an extremely high strain (99.7%) in compression tests.

Abstract:3D printing hydrogels with a good resolution, diversified geometries, and a tunable mechanical performance is still a big challenge, which is mainly due to the low photo-reactivity of the high-water containing precursor and the low modulus of the photo-cured hydrogel. In the present work, an acryloylmorpholine - acrylic acid - PEG diacrylate drygel was firstly printed with a good fidelity and subsequently post- treated with an aqueous solution containing specific ions, during which hydration of the hydrophilic network and coordination between the carboxyl and ions occurred simultaneously. The obtained dual-crosslinked hydrogel combined a complex geometry and a good stretchability. This approach decoupled the printability and mechanical properties of the hydrogels. To furtherly show this advantage over conventional direct hydrogel 3D printing, ultrasoft hydrogel lattice structures which cannot be self-supported and purely ionic crosslinked hydrogels with a good self-weldability were demonstrated.
