Abstract:This manuscript covers the latest advancements and persisting challenges in the domain of tissue engineering, with a focus on the development and engineering of hydrogel scaffolds. It highlights the critical role of these scaffolds in emulating the native tissue environment, thereby providing a supportive matrix for cell growth, tissue integration, and reducing adverse reactions. Despite significant progress, this manuscript emphasizes the ongoing struggle to achieve an optimal balance between biocompatibility, biodegradability, and mechanical stability, crucial for clinical success. It also explores the integration of cutting-edge technologies like 3D bioprinting and biofabrication in constructing complex tissue structures, alongside innovative materials and techniques aimed at enhancing tissue growth and functionality. Through a detailed examination of these efforts, the manuscript sheds light on the potential of hydrogels in advancing regenerative medicine and the necessity for multidisciplinary collaboration to navigate the challenges ahead.

Abstract:Hydrogels have emerged as versatile biomaterials with remarkable applications in biomedicine and tissue engineering. Here, we present an overview of recent and ongoing research in Italy, focusing on extracellular matrix-derived, natural, and synthetic hydrogels specifically applied to biomedicine and tissue engineering. The analyzed studies highlight the versatile nature and wide range of applicability of hydrogel-based studies. Attention is also given to the integration of hydrogels within bioreactor systems, specialized devices used in biological studies to culture cells under controlled conditions, enhancing their potential for regenerative medicine, drug discovery, and drug delivery. Despite the abundance of literature on this subject, a comprehensive overview of Italian contributions to the field of hydrogels-based biomedical research is still missing and is thus our focus for this review. Consolidating a diverse range of studies, the Italian scientific community presents a complete landscape for hydrogel use, shaping the future directions of biomaterials research. This review aspires to serve as a guide and map for Italian researchers interested in the development and use of hydrogels in biomedicine.

Abstract:It gives us great pleasure to prepare this invited editorial for the Research Topic in Frontiers in Chemistry titled "Biocompatible Hydrogels: Properties, Synthesis and Applications in Biomedicine". Due to the favourable mechanical properties, biocompatibility and biodegradability features, the hydrogels with three-dimensional (3D) networks have displayed great potentials in biomedical applications, including drug delivery and tissue engineering. In recent years, several strategies and techniques on biocompatible polymer constructions and hydrogel preparation have been developed, guaranteeing the biocompatible hydrogels in several biomedical applications, including wound healing, cancer therapy, etc. In this Research Topic, a number of interesting studies were written by 41 authors, related to biocompatible hydrogels and polymeric nano-materials with functional characteristics in tissue engineering and biomedicine applications. Niclosamide has been widely used in treating cancer, viral infections, and metabolic disorders due to its ability to regulate signalling pathways and biological processes. To improve the efficacy of niclosamide, Tai et al. have fabricated niclosamide conjugated microspheres based on poly(lactic-co-glycolic acid) (PLGA) and hyaluronic acid (HA) via microfluidic approach. The obtained microspheres with a drug-loading efficiency of 8.70% displayed a sustained release behavior at pH 7.4 and 5.0, showing a quasi-first-order release kinetic. Furthermore, the niclosamide conjugated microspheres could increase the cellular uptake using Caco-2 cells and the inhibitory effect on Caco-2 cells, showing potential in targeted drug delivery in the intestine. In recent years, cancer immunotherapy using the hydrogels strategies has made significant progress. Liu et al. have synthesized a novel polymer for drug delivery. We believe that the encouraging approaches could advance the biocompatible hydrogels in tissue engineering and drug delivery, as well as for future clinical applications. Once again, we sincerely hope the researchers in related areas will enjoy reading all the papers in this special edition.

Tailoring the Swelling-Shrinkable Behavior of Hydrogels for Biomedical Applications.

Abstract:Hydrogels with tailor-made swelling-shrinkable properties have aroused considerable interest in numerous biomedical domains. For example, as swelling is a key issue for blood and wound extrudates absorption, the transference of nutrients and metabolites, as well as drug diffusion and release, hydrogels with high swelling capacity have been widely applicated in full-thickness skin wound healing and tissue regeneration, and drug delivery. Nevertheless, in the fields of tissue adhesives and internal soft-tissue wound healing, and bioelectronics, non-swelling hydrogels play very important functions owing to their stable macroscopic dimension and physical performance in physiological environment. Moreover, the negative swelling behavior (i.e., shrinkage) of hydrogels can be exploited to drive noninvasive wound closure, and achieve resolution enhancement of hydrogel scaffolds. In addition, it can help push out the entrapped drugs, thus promote drug release. However, there still has not been a general review of the constructions and biomedical applications of hydrogels from the viewpoint of swelling-shrinkable properties. Therefore, this review summarizes the tactics employed so far in tailoring the swelling-shrinkable properties of hydrogels and their biomedical applications. And a relatively comprehensive understanding of the current progress and future challenge of the hydrogels with different swelling-shrinkable features is provided for potential clinical translations.

Abstract:A single because of their distinctive traits, hydrogels have become the most preferred materials for biomedical applications today. Hydrogels are three-dimensional hydrophilic polymeric networks with a high water-holding capacity as well as the ability to store biological fluids. The specific structure and swelling behavior of hydrogel largely influence the water holding and biological properties. The advantageous properties of hydrogels like flexibility, versatility, stimuli-responsive and soft structure make them versatile materials for medical applications. However, extreme water content, porous nature, and smooth in consistency are closely simulated living tissue than other alternative biomaterials. Such a close resemblance of hydrogels triggers several chances in the biomedical field. Hydrogels have a wide footprint in day today products but still their significant use yet not completely explored. Hydrogels have a well-demonstrated appreciable theatrical role in wound healing, contact lenses, tissue engineering, and many hygiene products. But commercially still hydrogels have not been well-explored in tissue engineering and drug delivery. In the meanwhile, a variety of drug-delivery-related devices and scaffolds have been developed, reviewed, and a few have been patented, but few have reached the market. Abstract:Nowadays, there are still numerous challenges for well-known biomedical applications, such as tissue engineering (TE), wound healing and controlled drug delivery, which must be faced and solved. Hydrogels have been proposed as excellent candidates for these applications, as they have promising properties for the mentioned applications, including biocompatibility, biodegradability, great absorption capacity and tunable mechanical properties. However, depending on the material or the manufacturing method, the resulting hydrogel may not be up to the specific task for which it is designed, thus there are different approaches proposed to enhance hydrogel performance for the requirements of the application in question. The main purpose of this review article was to summarize the most recent trends of hydrogel technology, going through the most used polymeric materials and the most popular hydrogel synthesis methods in recent years, including different strategies of enhancing hydrogels' properties, such as cross-linking and the manufacture of composite hydrogels. In addition, the secondary objective of this review was to briefly discuss other novel applications of hydrogels that have been proposed in the past few years which have drawn a lot of attention.

Abstract:Hydrogel is a new class of functional polymer materials with a promising potential in the biomedical field. The purpose of this article is to review recent advancements in several types of biomedical hydrogels, including conductive hydrogels, injectable hydrogels, double network hydrogels, responsive hydrogels, nanocomposite hydrogels, and sliding hydrogels. In comparison with traditional hydrogels, these advanced hydrogels exhibit significant advantages in structure, mechanical properties, and applications. The article focuses on different methods used to prepare advanced biomedical hydrogels and their diversified applications as drug delivery systems, wound dressings, biosensors, contact lenses, and tissue replacement. These advances are rapidly overcoming current limitations of hydrogels, and we anticipate that further research will lead to the development of advanced hydrogels with ubiquitous roles in biomedicine and tissue replacement and regeneration.

Abstract:Hydrogels, being hydrophilic polymer networks capable of absorbing and retaining aqueous fluids, hold significant promise in biomedical applications owing to their high water content, permeability, and structural similarity to the extracellular matrix. Recent chemical advancements have bolstered their versatility, facilitating the integration of the molecules guiding cellular activities and enabling their controlled activation under time constraints. However, conventional synthetic hydrogels suffer from inherent weaknesses such as heterogeneity and network imperfections, which adversely affect their mechanical properties, diffusion rates, and biological activity. Abstract:Hydrogels are biomolecules made of artificial and natural polymers. Their quasi-three-dimensional structure has created unique features. They are very hydrophilic, and in addition to the high inflation rate, they also have excellent water maintenance capacity, biodegradability, biocompatibility, and strong mechanical properties. These properties are used in many tissue engineering applications. All these features have made these scaffolds widely used as attractive structures in the world of tissue engineering and regeneration medicine. In addition to research, scaffolds entered the field of medicine and are expected to play a significant role in the repair of many tissues in the future. Abstract:Novel advanced biomaterials have recently gained great attention, especially in minimally invasive surgical techniques. By applying sophisticated design and engineering methods, various elastomer-hydrogel systems (EHS) with outstanding performance have been developed in the last decades. These systems composed of elastomers and hydrogels are very attractive due to their high biocompatibility, injectability, controlled porosity and often antimicrobial properties. Moreover, their elastomeric properties and bioadhesiveness are making them suitable for soft tissue engineering. Herein, we present the advances in the current state-of-the-art design principles and strategies for strong interface formation inspired by nature (bio-inspiration), the diverse properties and applications of elastomer-hydrogel systems in different medical fields, in particular, in tissue engineering. The functionalities of these systems, including adhesive properties, injectability, antimicrobial properties and degradability, applicable to tissue engineering will be discussed in a context of future efforts towards the development of advanced biomaterials.

Abstract:Biomedical research, known as medical research, is conducive to support and promote the development of knowledge in the field of medicine. Hydrogels have been extensively used in many biomedical fields due to their highly absorbent and flexible properties. The smart hydrogels, especially, can respond to a broad range of external stimuli such as temperature, pH value, light, electric and magnetic fields. With excellent biocompatibility, tunable rheology, mechanical properties, porosity, and hydrated molecular structure, hydrogels are considered as promising candidate for simulating local tissue microenvironment. In this review article, we mainly focused on the most recent development of engineering synthetic hydrogels; moreover, the classification, properties, especially the biomedical applications including tissue engineering and cell scaffolding, drug and gene delivery, immunotherapies and vaccines, are summarized and discussed.

Abstract:Shear‐thinning and self‐healing hydrogels are being investigated in various biomedical applications including drug delivery, tissue engineering, and 3D bioprinting. Such hydrogels are formed through dynamic and reversible interactions between polymers or polypeptides that allow these shear‐thinning and self‐healing properties, including physical associations (e.g., hydrogen bonds, guest–host interactions, biorecognition motifs, hydrophobicity, electrostatics, and metal–ligand coordination) and dynamic covalent chemistry (e.g., Schiff base, oxime chemistry, disulfide bonds, and reversible Diels–Alder). Their shear‐thinning properties allow for injectability, as the hydrogel exhibits viscous flow under shear, and their self‐healing nature allows for stabilization when shear is removed. Hydrogels can be formulated as uniform polymer and polypeptide assemblies, as hydrogel nanocomposites, or in granular hydrogel form. Abstract:A comprehensive review of the strategies of hydrophobized hydrogel constructions is presented, with an emphasis on their properties such as improved hydrophobic drugs solubilization, underwater adhesion, abnormal swelling behavior, self‐healing behavior, and enhanced mechanical properties. The current challenges and future research directions in this field are also discussed. Hydrogels, as 3D networks containing huge amount of water, display similarity to soft tissues, and thus they are of wide interest in tissue engineering. Hydrogels, due to biocompatibility and porous structure, are valuable therapeutic platforms for hydrophilic drugs. Over the last decade, there has been a strong emphasis on the development of hydrogel platforms with the ability to increase the solubility of hydrophobic drugs. However, the pronounced discrepancy between the hydrophilic character of hydrogels and the hydrophobic nature of numerous pharmacologically active compounds is problematic. In recent years, different strategies are applied using special polymer constructs or composite materials exploiting the advanced scientific knowledge in the area of polymer and lipid‐based nano‐ and microcarriers hydrophobization of the hydrogel turns out to be not only valuable in terms of achieving the ability to dissolve poorly soluble drugs in water, but also proves to be crucial in obtaining bioadhesion in wet conditions, but also, unexpected abnormal water swelling behavior, as well as in mechanical properties such as the dissipation mechanism and self‐healable hydrogel properties. This review is mainly focused on recent advances in the usage of hydrophobized hydrogels in biomedical applications.

Abstract:The current review gives a broad spectrum of hybrid hydrogels with its importance and significance and highlights the strategies of generation of hybrid hydrogels reinforced with nanomaterials, biomaterials, and nanobiocomposites along with their advantages and disadvantages. Advancements in polymers have made significant contribution in diverse application‐oriented fields. The multidisciplinary application of polymers generates a range of strategies, which is applicable in a wide range of biomedicines. Polymeric compounds such as hydrogels have been explored globally as a potent biomaterial that can furnish essential attributes due to their three‐dimensional (3D) soft and highly hydrophilic polymeric network. These hydrogels have been used extensively in various applications due to their biocompatible, biodegradable, and biosorption nature. However, they have some limitations, owing to their soft nature, low mechanical properties, and unsatisfactory degradation profile. Therefore, there is a need to develop novel, stronger, and more durable hybrid hydrogels with enhanced biocompatible and multifunctional properties. The current review gives a broad spectrum of hybrid hydrogels with its importance and significance. Further, the review highlights the strategies of generation of hybrid hydrogels reinforced with nanomaterials, biomaterials, and nanobiocomposites along with their advantages and disadvantages. Abstract:Hyaluronic acid (HA) is widely used in the design of engineered hydrogels, due to its biofunctionality, as well as numerous sites for modification with reactive groups. There are now widespread examples of modified HA macromers that form either covalent or physical hydrogels through crosslinking reactions such as with click chemistry or supramolecular assemblies of guest-host pairs. HA hydrogels range from relatively static matrices to those that exhibit spatiotemporally dynamic properties through external triggers like light. Such hydrogels are being explored for the culture of cells in vitro, as carriers for cells in vivo, or to deliver therapeutics, including in an environmentally responsive manner. The future will bring new examples of HA hydrogels due to the synthetic diversity of HA.

Abstract:The popularity of hydrogels as biomaterials lies in their tunable physical properties, ability to encapsulate small molecules and macromolecular drugs, water holding capacity, flexibility, and controllable degradability. Functionalization strategies to overcome the deficiencies of conventional hydrogels and expand the role of advanced hydrogels such as DNA hydrogels are extensively discussed in this review. Different types of cross-linking techniques, materials utilized, procedures, advantages, and disadvantages covering hydrogels are tabulated. The application of hydrogels, particularly in buccal, oral, vaginal, and transdermal drug delivery systems, are described. The review also focuses on composite hydrogels with enhanced properties that are being developed to meet the diverse demand of wound dressing materials. The unique advantages of hydrogel nanoparticles in targeted and intracellular delivery of various therapeutic agents are explained. Furthermore, different types of hydrogel-based materials utilized for tissue engineering applications and fabrication of contact lens are discussed. The article also provides an overview of selected examples of commercial products launched particularly in the area of oral and ocular drug delivery systems and wound dressing materials. Hydrogels can be prepared with a wide variety of properties, achieving biostable, bioresorbable, and biodegradable polymer matrices, whose mechanical properties and degree of swelling are tailored with a specific application. These unique features give them a promising future in the fields of drug delivery systems and applied biomedicine.
