Abstract:Herein, we demonstrate a facile, straightforward and green method for synthesis of highly red-emitting fluorescent bimetallic Au/Ag nanoclusters (NCs) employing lipoic acid as both a reducing agent and a protecting agent. The as-prepared Au/Ag NCs exhibited bright red emission with a strong peak centered at 630 nm (quantum yield measured as 6.4%), and showed excellent stability and remarkable temperature-dependent fluorescent property. The fluorescence responses were reversible and linear to the environment temperature from 20 to 65 C with a resolution as high as 1.34 degrees C, which allows its potential application as a fluorescent nanothermometer. Moreover, Au/Ag NCs were also explored for selective and sensitive determination of Fe3+ based on the specific quenching of the fluorescence by Fe3+, and explored for fluorescence imaging Fe3+ in living HeLa cells thanks to their very low cytotoxicity. Dual-emission ratio fluorescence for selective and sensitive detection of ferric ions and ascorbic acid based on one-pot synthesis of glutathione protected gold nanoclusters

Abstract:A fluorometric method was proposed for the determination of Fe3+ and ascorbic acid (AA) based on blue and red dual fluorescence emissions of glutathione (GSH) stabilized-gold nanoclusters (AuNCs). AuNCs were synthesized from GSH and tetrachloroauric acid. The fluorescence peaks of AuNCs were at 425 nm and 585 nm, respectively. In the presence of Fe3+, the fluorescence peak at 425 nm can be enhanced and that at 585 nm can be quenched. There is a good linear relationship between the fluorescence intensity ratio for the 425 and 585 nm peaks (F 425/F 585) and the concentration of Fe3+ in the range of 0.75-125 μM. However, when AA was added to the AuNCs-Fe3+ system, the value of F 425/F 585 decreased consistently with the concentration of AA in the range of 0.25-35 μM. The limit of detection for Fe3+ and AA was 227 and 75.8 nM, respectively. The interaction between AuNCs and Fe3+ can induce the ligand-metal charge transfer (LMCT) effect leading to the fluorescence increment at 425 nm, while AA can reduce Fe3+ to Fe2+. The production of Fe2+ can not enhance or quench the fluorescence of AuNCs. By comparison with previous literature, the AuNCs prepared here show two fluorescence peaks without additional fluorescence labels. Furthermore, the method was successfully applied in the determination of Fe3+ and AA in some real samples, such as water, human serum and tablets.

Abstract:Glutathione-Au nanoclusters (GSH-AuNCs) are known as typical luminophores with aggregation-induced emission (AIE) features, which can be triggered by low temperature, low pH values, and metal ions. However, these strict "triggers" restrict the application of this AIE probe under mild and neutral conditions. In this work, fluorescent amino-modified silicon nanoparticles (SiNPs) and GSH-AuNCs can self-assemble into well-designed nanospheres through electrostatic interactions under physiological conditions. The hybrid probe of SiNPs@GSH-AuNCs exhibits dual emissions located at 450 nm from SiNPs and 570 nm originating from the aggregation enhanced emission (AEE) of GSH-AuNCs. Interestingly, the addition of Zn2+ or Cd2+ can connect the nanospheres to form large aggregates, so a unique second increase of AEE from GSH-AuNCs appears, while the emission of SiNPs remains constant to act as an internal reference. The fluorescence ratios (I570/I450) of SiNPs@GSH-AuNCs are positively correlated with Zn2+ or Cd2+ with the linear range from 1.5 μM to 500 μM. Most importantly, besides the advantages of dual emissions, high fluorescence brightness, and good stability, the SiNPs@GSH-AuNCs also exhibit excellent biocompatibility and low cytotoxicity, and have been successfully applied in zinc imaging in live cells. Furthermore, the proposed method is also used to measure Zn2+ and Cd2+ in real samples with satisfactory recoveries.

Abstract:The development of label-free electrochemiluminescence (ECL)-based sensing technology for biomarkers detection has a congenital defect compared to noncompetitive sandwich-type biosensors due to the lack of detection antibody conjugated with a signal label. Nevertheless, it is still not difficult to realize the ultrasensitive analysis benefit from the exploration of efficient sensing substrates and signal transducers. In this work, an innovative sensing system is purposed utilizing Fe2O3 nanoarrays (Fe2O3 NAs) as a well-ordered coreaction accelerator and polypeptide-biomineralized gold nanoclusters (Au NCs) as a signal transducer. Bifunctional peptide ligands of H2N-MMYYHFRRHL-COOH (MYH-10) are self-designed; it cannot only play a role of reductant and coupling reagent for cluster formation using the MMYY sequence root in the N-terminal but also act as a connection for coupling carriers and immune molecules via the HFRRHL region of the C-terminal. In addition to intramolecular ECL emission between Au NCs and tris(3-aminoethyl)amine (TAEA), all strategies undoubtedly reduce the spatial hindrance of the sensing interface and increase the effectiveness of the electron transfer and immune recognition. With CYFRA21-1 as a target, the biosensor exhibits a linear ECL response in a wide range (10 fg mL(-1) to similar to 100 ng mL(-1)) and an ultralow detection limit of 1.33 fg mL(-1)(S/N = 3). With convincing experimental data, these innovative strategies will be more eye-catching in peptide-based nanocluster synthesis and expansion of a more novel thought for sensing platform fabrication.

Abstract:Gold nanoclusters (Au NCs) exhibit broad fluorescent spectra from visible to near-infrared regions and good enzyme-mimicking catalytic activities. Combined with excellent stability and exceptional biocompatibility, the Au NCs have been widely exploited in biomedicine such as biocatalysis and bioimaging. Especially, the long fluorescence lifetime and large Stokes shift attribute Au NCs to good probes for fluorescence sensing and biological detection. In this review, we systematically summarized the molecular structure and fluorescence properties of Au NCs and highlighted the advances in fluorescence sensing and biological detection. The Au NCs display high sensitivity and specificity in detecting iodine ions, metal ions, and reactive oxygen species, as well as certain diseases based on the fluorescence activities of Au NCs. We also proposed several points to improve the practicability and accelerate the clinical translation of the Au NCs.

Abstract:Gold nanoclusters (AuNCs) possess weak intrinsic fluorescence, limiting their sensitivity in biosensing applications. This study addresses these limitations by developing a spatially confined dual-emission nanoprobe composed of silicon nanoparticles (SiNPs) and AuNCs. This amplified and stabilized fluorescence mechanism overcomes the limitations associated with using AuNCs alone, achieving superior sensitivity in the sensing platform. The nanoprobe was successfully employed for ratiometric detection of bleomycin (BLM) in serum samples, operating at an excitation wavelength of 365 nm, with emission wavelengths at 480 nm and 580 nm. The analytical performance of the system is distinguished by a linear detection range of 0-3.5 μM, an impressive limit of detection (LOD) of 35.27 nM, and exceptional recoveries ranging from 96.80 to 105.9%. This innovative approach significantly enhances the applicability and reliability of AuNC-based biosensing in complex biological media, highlighting its superior analytical capabilities.

Abstract:Gold nanoclusters (AuNCs) are among the most promising organic-inorganic hybrid luminescent materials for various applications. The current development of AuNCs majorly focuses on controlling their luminescence properties. Herein, we report a new strategy to facilely construct two different nanocomposites featuring enhanced photoluminescence based on mercaptopropionic acid-protected AuNCs (MPA-AuNCs). Through co-assembly with Zn2+ and 2-methylimidazole (2M-IM), the weak luminescence of MPA-AuNCs evolved into either intense blue-green or orange emission at different concentration ratios of additives. HR-TEM and spectroscopic characterization studies revealed that the intense blue-green emission was ascribed to the formation of ZnS quantum dots (QDs) on the outer surface of AuNCs (AuNCs@ZnS), while the strong orange emission originated from the primitive MPA-AuNC core encapsulated by a cubic ZIF-8 shell (AuNCs@ZIF-8). The AuNCs@ZnS nanocomposite was further applied as an exceptional chemical sensor for selective detection of Pb2+ and Fe3+via different quenching mechanisms, and the AuNCs@ZIF-8 composite was applied for fabricating light-converting devices. The co-assembly of AuNCs with Zn2+ and imidazole derivatives provides a facile strategy for acquiring differentiated nanomaterials that have versatile potential applications in chemical detection and light-converting devices.

Abstract:A creatively designed and constructed a multifunctional ratiometric fluorescence probe is reported by assembling glutathione (GSH)-protected gold nanoclusters (AuNCs) with fluorescein-doped mesoporous silica nanoparticle (FS) for the detection of Cu 2+ and Ag + ions, which could eliminate most interferences by self-calibration. Under the excitation at 450 nm, the fluorescence connected with AuNCs can rapidly respond by quenching or enhancement, respectively, for Cu 2+ and Ag + ions, while the fluorescein isothiocyante (FITC) fluorescence served as reference with negligible change. The fluorescence intensity ratio showed good linear relationships with Cu 2+ and Ag + concentrations in the range 0.5–10 μM and 0.1–8 μM, respectively. The detection limits were as low as 140 nM and 60 nM for Cu 2+ and Ag + ions, respectively. The color change induced by fluorescent intensity ratio variation could also be employed for visual discrimination. The AuNC-embedded FS (FS-Au) nanoprobe was successfully used for Cu 2+ and Ag + ion determination in drinking water and intracellular Cu 2+ imaging, which exhibits promising prospects in cost-effective and rapid determination of both Cu 2+ and Ag + with good sensitivity and selectivity.

Abstract:Traditional single sensor is designed based on the "lock-and-key" mode, which only relies on the most dominant interactions between the sensing element and the target. Although it exhibits high selectivity, there are challenges in detecting multiple analytes at the same time. Here, a sensor array with three sensing elements is developed to detect multiple heavy metal ions simultaneously and quickly. In our experiment, bovine serum albumin-encapsulated gold nanoclusters (BSA-AuNCs) were used as fluorescence probes and three different dopamine (DA) concentrations as nonspecific receptors. As we know, self-polymerized polydopamine (PDA) can quench part of the fluorescence of BSA-AuNCs. Upon the addition of the heavy metal ions, the diverse non-specific interactions between DA and heavy metal ions result in the difference in the number of the remaining PDA. Therefore it would lead to different degrees of fluorescence recovery behavior. This unique "turn-on" fluorescence response mode can be analyzed by linear discriminant analysis (LDA) and hierarchical cluster analysis (HCA). Two-dimensional, three-dimensional and even four-dimensional mixed ions detection and quantitative detection have also been achieved. Moreover, by using this fluorescence array mode, heavy metal ions in tap water or blood samples can be detected.
Confinement of gold nanoclusters on hydroxy double salt for versatile fluorescent sensing of bio-enzymes

Abstract:Fluorescent gold nanoclusters (AuNCs) have generated significant research interest because of their fascinating physicochemical properties. However, their biosensing applications are limited by the relatively low quantum yield (QY). Herein, a simple confinement effect based on AuNCs as guest molecules and Zn-containing hydroxy double salt (HDS) as host material (AuNCs@HDS) was used to significantly improve the fluorescence properties of AuNCs. The QY increased from 1.24 % to 22.80 %, and the time-tedious preparation or purification procedures were avoided. Further, the validated excellent fluorescence properties of AuNCs enabled the construction of a versatile fluorescent sensing strategy for bio-enzymes for the first time. The p-nitrophenol (pNP), a product of many bio-enzymes-catalyzed hydrolyzes of pNP-substrate, can effectively quench the fluorescence of AuNCs@HDS through an inner filter effect (IFE) process. Therefore, the AuNCs@HDS successfully detected several enzymatic disease biomarkers with robust analysis capabilities by varying the pNP-substrate, including alkaline phosphatase (ALP), β-glucosidase (β-GLU) and β-galactosidase (β-GAL). This study will inspire the design of highly fluorescent AuNCs as tools for biosensing.

Rare-Earth Eu3+/Gold Nanocluster Ensemble-Based Fluorescent Photoinduced Electron Transfer Sensor for Biomarker Dipicolinic Acid Detection

Abstract:The use of metal ions to bridge the fluorescent materials to target analytes has been demonstrated to be a promising way to sensor design. Herein, the effect of rare-earth ions on the fluorescence of L-methioninestabilized gold nanoclusters (Met-AuNCs) was investigated. It was found that europium (Eu3+) can significantly suppress the emission of Met-AuNCs, while other rare-earth ions showed a negligible impact. The mechanism on the observed fluorescence quenching of Met-AuNCs triggered by Eu' was systematically explored, with results revealing the dominant role of photoinduced electron transfer (PET). Eu3+ can bind to the surface of MetAuNCs by the coordination effect and accepts the electron from the excited Met-AuNCs, which results in Met-AuNC fluorescence suppression. After introducing dipicolinic acid (DPA), an excellent biomarker for spore-forming pathogens, Eu3+ was removed from the surface of Met-AuNCs owing to the higher binding affinity between Eu3+ and DPA. Consequently, an immediate fluorescence recovery occurred when DPA was present in the system. Based on the Met-AuNC/Eu3+ ensemble, we then established a simple and sensitive fluorescence strategy for turn-on determination of biomarker DPA, with a linear range of 0.2-4 mu M and a low limit of detection of 110 nM. The feasibility of the proposed method was further validated by the quantitative detection of DPA in the soil samples. We believe that this study would significantly facilitate the construction of metal-ion-mediated PET sensors for the measurement of various interested analytes by applying fluorescent AuNCs as detection probes.

Abstract:Herein, a dual-emission metal-organic framework based ratiometric fluorescence nanoprobe was reported for detecting copper(II) ions. In particular, carbon dots (CDs) and gold nanoclusters (AuNCs) were embedded into ZIF-8 (one of the classical metal-organic frameworks) to form CDs/AuNCs@ZIF-8 nanocomposites, which exhibited dual-emission peaks at UV excitation. In the presence of Cu2+, the fluorescence attributed to AuNCs can be rapidly quenched, while the fluorescence of CDs serves as reference with undetectable changes. Therefore, the CDs/AuNCs@ZIF-8 nanocomposites were utilized as a ratiometric fluorescence nanoprobe for sensitive and selective detection of Cu2+. A good linear relationship between the ratiometric fluorescence signal of CDs/AuNCs@ZIF-8 and Cu2+ concentration was obtained in the range of 10-3-103 μM, and the detection limit was as low as 0.3324 nM. The current ratiometric fluorescence nanoprobe showed promising prospects in cost-effective and rapid determination of Cu2+ ions with good sensitivity and selectivity. Furthermore, this nanoprobe has been successfully applied for the quantitative detection of Cu2+ in serum samples, indicating its value of practical application. Abstract:Ultrasmall gold nanoclusters (NCs) have been engineered as a new kind of functional material due to their excellent photoluminescence properties. However, the synthesis of highly luminescent water-soluble nanoclusters with near-infrared (NIR) emission remains limited. Herein, we developed a pH-regulated strategy to facilitate the construction of self-assemblies with enhanced luminescence based on aggregation-induced emission (AIE) strategy. Using 2-mercaptobenzoic acid (MBA) as reductant and stabilizer, the original weakly luminescent AuNCs exhibited intense emission by adjusting pH controllably. The formation of compact organized nanostructures could effectively restrict the rotation and vibration of capping ligands by non-covalent interactions, which reduced the nonradiative relaxation from excited states and finally improved the emission properties of AuNCs. Moreover, the assemblies possess many intriguing features including bright NIR luminescence and excellent biocompatibility, which could be used as luminous probes in biological molecules sensing (tyrosinase (TYR) and dopamine (DA)) and promising candidates for cell imaging. This study provides a simple and feasible strategy for developing metal NCs-based smart optical materials in the field of bioscience.

Abstract:The use of metal ions to bridge the fluorescent materials to target analytes has been demonstrated to be a promising way to sensor design. Herein, the effect of rare-earth ions on the fluorescence of l-methionine-stabilized gold nanoclusters (Met-AuNCs) was investigated. It was found that europium (Eu3+) can significantly suppress the emission of Met-AuNCs, while other rare-earth ions showed a negligible impact. The mechanism on the observed fluorescence quenching of Met-AuNCs triggered by Eu3+ was systematically explored, with results revealing the dominant role of photoinduced electron transfer (PET). Eu3+ can bind to the surface of Met-AuNCs by the coordination effect and accepts the electron from the excited Met-AuNCs, which results in Met-AuNC fluorescence suppression. After introducing dipicolinic acid (DPA), an excellent biomarker for spore-forming pathogens, Eu3+ was removed from the surface of Met-AuNCs owing to the higher binding affinity between Eu3+ and DPA. Consequently, an immediate fluorescence recovery occurred when DPA was present in the system. Based on the Met-AuNC/Eu3+ ensemble, we then established a simple and sensitive fluorescence strategy for turn-on determination of biomarker DPA, with a linear range of 0.2-4 μM and a low limit of detection of 110 nM. The feasibility of the proposed method was further validated by the quantitative detection of DPA in the soil samples. We believe that this study would significantly facilitate the construction of metal-ion-mediated PET sensors for the measurement of various interested analytes by applying fluorescent AuNCs as detection probes.

Abstract:With the rapid development of materials technology, fluorescent gold nanoclusters (AuNCs) are emerging as novel functional materials for diagnostic applications including the detection of biomarkers and bioimaging due to the advantages of their ultra-small size, tunable emissions, size-dependent fluorescence and excellent biocompatibility. In this review, we introduced the synthetic methods, and physical and chemical properties of AuNCs. Subsequently, we described the AuNCs-based design strategies for the detection of biomarkers including small molecules, DNA and proteins. The applications of AuNCs for tumor imaging in vitro and in vivo were also presented. Finally, we discussed the challenges and potential solutions of AuNCs-based nanosensors.
