Abstract:Herein we report a novel strategy for the detection and identification of proteins using unmodified noble metal nanoparticles. Five gold nanoparticles (AuNPs) and two silver nanoparticles (AgNPs) with different sizes were utilized as sensing elements to create a colorimetric sensor array. In the presence of proteins, the UV-vis absorbance of the noble metal nanoparticles changed due to the interactions between the protein and nanoparticles, producing distinct absorbance response patterns which can be visually detected by the naked eye. The color pattern of the array is a unique "fingerprints" for each protein sample, which can be differentiated by linear discriminant analysis (LDA). Ten different proteins at concentrations of 0.5, 5 and 50 μM have be successfully discriminated. Moreover, the array was also able to discriminate different bacteria at a concentration of 0.05 OD in 200 μL, as well as cancer cells at the level of 5000 cells in 200 μL. This work demonstrates that an unmodified noble metal nanoparticle-based protein detection array has potential for applications in medical diagnostics.

Abstract:We report a simple and novel colorimetric sensor array for rapid identification of microorganisms. In this study, four gold nanoparticles (AuNPs) with diverse surface charges were used as sensing elements. The interactions between AuNPs and microorganisms led to obvious color shifts, which could be observed by the naked eye. A total of 15 microorganisms had their own response patterns and were differentiated by linear discriminant analysis (LDA) successfully. Moreover, microorganism mixtures could also be well discerned. The method is simple, fast (within 5 s), effective, and visual, showing the potential applications in pathogen diagnosis and environmental monitoring.

Abstract:Antibiotic contamination has become a serious global problem due to abuse and misuse. Therefore, it is important to develop an efficient detection method to monitor the rational use of antibiotics. In this study, fluorescent gold nanoclusters with 11-mercaptoundecanoic acid as ligands (MUA-AuNCs) were synthesized by a one-step method firstly. Rare earth ions (Re3+) can enhance the fluorescence of MUA-AuNCs through inducing the aggregation of MUA-AuNCs, but antibiotics decrease the fluorescence of the Re3+-MUA-AuNCs to different degrees through coordination with Re3+ and competitive absorption with AuNCs. Therefore, a sensor array was obtained on the basis of the above mechanism, which can detect and discriminate six different antibiotics with a detection range from 40 to 300 μM. A 100% correct classification was achieved. The fluorescent sensor array showed high selectivity for tetracycline antibiotics and good anti-interference performance was demonstrated. Combined with pattern recognition methods, the proposed sensor array can be used for the discrimination of different antibiotics and binary antibiotic mixtures. Furthermore, the excellent performance of this sensor array in quantitation and blind sample recognition further validates its potential for practical applications.

Abstract:A series of dual-ligand cofunctionalized fluorescent gold nanodots with similar fluorescence and diverse surface properties has been designed and synthesized to build a protein sensing array. Using this chemical nose/tongue strategy, fluorescence response patterns can be obtained on the array and identified via linear discriminant analysis (LDA). Eight proteins have been well distinguished at low concentration (A(280) = 0.005) based on this sensor array. The practicability of this sensor array was further validated by high accuracy (100%) examination of 48 unknown protein samples.

Abstract:Convenient, precise, and high-throughput discrimination of multiple bioanalytes is of great significance for an early diagnosis of diseases. Array-based pattern recognition has proven to be a powerful tool to detect diverse analytes, but developing sensing elements featuring favorable surface diversity still remains a challenge. In this work, we presented a simple and facile method to prepare programmable metal-nanoparticle (NP)-supported nanozymes (MNNs) as artificial receptors for the accurate identification of multiple proteins and oral bacteria. The in situ reduction of metal NPs on hierarchical MoS2 on polypyrrole (PPy), which generated differential nonspecific interactions with bioanalytes, was envisaged as the encoder to break through the limited supply of the receptor's quantity. As a proof of concept, three metal NPs, i.e., Au, Ag, and Pd NPs, were taken as examples to deposit on PPy@MoS2 as colorimetric probes to construct a cross-reactive sensor array. Based on the principal component analysis (PCA), the proposed MNN sensor array could well discriminate 11 proteins with unique fingerprint-like patterns at a concentration of 250 nM and was sufficiently sensitive to determine individual proteins with a detection limit down to the nanomolar level. Remarkably, two highly similar hemoglobins from different species (hemoglobin and bovine hemoglobin) have been precisely identified. Additionally, five oral bacteria were also well separated from each other without cross-classification at the level of 107 CFU mL-1. Furthermore, the sensor array allowed effective discrimination of complex protein mixtures either at different molar ratios or with minor varying components. Most importantly, the blind samples, proteins in human serums, proteins in simulated body fluid environment, the heat-denatured proteins, and even clinical cancer samples all could be well distinguished by the sensor array, demonstrating the real-world applications in clinical diagnosis.

Abstract:Fluorescent gold nanoclusters (AuNCs) have recently emerged as a novel kind of promising fluorescent probes for high-performance sensors and bioimaging because of their ultrasmall size (<3 nm), strong luminescence, good photostability, and excellent biocompatibility. Over the past decade, we have witnessed growing popularity of AuNCs in analytical applications and enormous efforts have been devoted to their development. In this review, we provide an update on recent advances in the development of AuNCs in terms of physicochemical properties, synthesis strategies, and bioapplications. The optical, electrochemical, catalytical, and solvatochromic properties of AuNCs are first summarized, which are followed by different ligands or template-assisted controllable synthetic methods. Afterwards functionalization of AuNCs is described in terms of ligand exchange, bioconjugation, and noncovalent interaction. We then focus on the applications of AuNCs as fluorescent probes for detection of metal ions, inorganic anions, small biomolecules, proteins, nucleic acids, drug molecules, pH, and temperature. We also summarize the usage of metal NCs in cellular and in vivo targeting and imaging. Finally, we conclude with a brief look at the future challenges and prospects of the development of AuNCs.

Abstract:Herein, we demonstrate a facile, straightforward and green method for synthesis of highly red-emitting fluorescent bimetallic Au/Ag nanoclusters (NCs) employing lipoic acid as both a reducing agent and a protecting agent. The as-prepared Au/Ag NCs exhibited bright red emission with a strong peak centered at 630 nm (quantum yield measured as 6.4%), and showed excellent stability and remarkable temperature-dependent fluorescent property. The fluorescence responses were reversible and linear to the environment temperature from 20 to 65 C with a resolution as high as 1.34 degrees C, which allows its potential application as a fluorescent nanothermometer. Moreover, Au/Ag NCs were also explored for selective and sensitive determination of Fe3+ based on the specific quenching of the fluorescence by Fe3+, and explored for fluorescence imaging Fe3+ in living HeLa cells thanks to their very low cytotoxicity. Abstract:Gold nanoclusters (Au NCs) exhibit broad fluorescent spectra from visible to near-infrared regions and good enzyme-mimicking catalytic activities. Combined with excellent stability and exceptional biocompatibility, the Au NCs have been widely exploited in biomedicine such as biocatalysis and bioimaging. Especially, the long fluorescence lifetime and large Stokes shift attribute Au NCs to good probes for fluorescence sensing and biological detection. In this review, we systematically summarized the molecular structure and fluorescence properties of Au NCs and highlighted the advances in fluorescence sensing and biological detection. The Au NCs display high sensitivity and specificity in detecting iodine ions, metal ions, and reactive oxygen species, as well as certain diseases based on the fluorescence activities of Au NCs. We also proposed several points to improve the practicability and accelerate the clinical translation of the Au NCs.

Abstract:Rationale: Rapid and facile detection of pathogenic bacteria is challenging due to the requirement of large-scale instruments and equipment in conventional methods. We utilize D-amino acid as molecules to selectively target bacteria because bacteria can incorporate DADA in its cell wall while mammalian cells or fungi cannot. Methods: We show a broad-spectrum bacterial detection system based on D-amino acid-capped gold nanoparticles (AuNPs). AuNPs serve as the signal output that we can monitor without relying on any complex instruments. Results: In the presence of bacteria, the AuNPs aggregate and the color of AuNPs changes from red to blue. This convenient color change can distinguish between Staphylococcus aureus (S. aureus) and methicillin-resistant Staphylococcus aureus (MRSA). This system can be applied for detection of ascites samples from patients. Conclusion: These D-amino acid-modified AuNPs serve as a promising platform for rapid visual identification of pathogens in the clinic.

Abstract:The growing co-contamination of multiple metal ions seriously influences human health due to their synergistic and additive toxicological effects, whereas the rapid discrimination of multiple heavy metal ions in complex aquatic systems remains a major challenge. Herein, a high- throughput fluorescence sensor array was fabricated based on three gold nanoclusters (GSH-Au NCs, OVA-Au NCs, and BSA-Au NCs) for the direct identification and quantification of seven heavy metal ions (Pb 2+ , Fe 3+ , Cu 2+ , Co 2+ , Ag + , Hg 2+ and As 3+ ) from environmental waters without sample pretreatment other than filtration. At the detection system, three gold nanoclusters with various ligands possessed distinct binding capacities against metal ions and induced aggregation-induced fluorescence enhancement and quenching, resulting in a unique pattern of fluorescence variations. Meanwhile, integrated the collected fluorescence fingerprints with linear discriminant analysis (LDA) and hierarchical cluster analysis (HCA), a discrete database was obtained for the accurate recognition and sensitive detection of metal ions. Under the optimized conditions, the limit of detection (LOD) of the proposed fluorescence sensor array for metal ions detection at nM concentration level along with a satisfactory accuracy. Importantly, our study indicated that the fluorescence sensor array could be widely used as a general platform in environmental monitoring against multiple targets at low concentrations.

Abstract:Fluorescent gold nanoclusters (AuNCs) have shown promise as metal ion sensors. Further research into surface ligands is crucial for developing sensors that are both selective and sensitive. Here, we designed simple tripeptides to form fluorescent AuNCs, capitalizing on tyrosine's reduction capability under alkaline conditions. We investigated tyrosine's role in both forming AuNCs and sensing metal ions. Two tripeptides, tyrosine–cysteine–tyrosine (YCY) and serine–cysteine–tyrosine (SCY), were used to form AuNCs. YCY peptides produced AuNCs with blue and red fluorescence, while SCY peptides produced blue-emitting AuNCs. The blue fluorescence of YCY- and SCY-AuNCs was selectively quenched by Fe3+ and Cu2+, whereas red-emitting YCY-AuNC fluorescence remained stable with 13 different metal ions. The number of tyrosine residues influenced the sensor response. DLS measurements revealed different aggregation propensities in the presence of various metal ions, indicating that chelation between the peptide and target ions led to aggregation and fluorescence quenching. Highlighting the innovation of our approach, our study demonstrates the feasibility of the rational design of peptides for the formation of fluorescent AuNCs that serve as highly selective and sensitive surface ligands for metal ion sensing. This method marks an advancement over existing methods due to its dual capability in both synthesizing gold nanoclusters and detecting analytes, specifically Fe3+ and Cu2+.

Abstract:With the rapid development of materials technology, fluorescent gold nanoclusters (AuNCs) are emerging as novel functional materials for diagnostic applications including the detection of biomarkers and bioimaging due to the advantages of their ultra-small size, tunable emissions, size-dependent fluorescence and excellent biocompatibility. In this review, we introduced the synthetic methods, and physical and chemical properties of AuNCs. Subsequently, we described the AuNCs-based design strategies for the detection of biomarkers including small molecules, DNA and proteins. The applications of AuNCs for tumor imaging in vitro and in vivo were also presented. Finally, we discussed the challenges and potential solutions of AuNCs-based nanosensors.

Abstract:There is a continuing high demand to design effective sensors for the determination of heavy metal ions (HMIs) since they are hazardous to both human health and the environment. In this study, we reported a facile fluorescent sensor array for rapid discrimination of HMIs based on a single gold nanocluster (AuNC) probe. This AuNC probe was prepared by using 2-mercapto-1-methylimidazole (MMI) as a ligand and polyvinypyrrolidone (PVP) as a dispersing agent. The fluorescence emission of PVP/MMI-AuNC was observed to be closely related to the pH value of the aqueous solution, which displays yellow (λmax = 512 nm) and red (λmax = 700 nm) fluorescence at pH 12.0 and 6.0, respectively. Further experiments indicated that different HMIs can produce differential effects on the photoluminescence of PVP/MMI-AuNC and thus generate distinct fluorescent responses at 512 and 700 nm. On the basis of this phenomenon, a fluorescent sensor array based on the PVP/MMI-AuNC was then built by simply changing pH value in the sensor element. A total of seven HMIs had their unique response patterns and were successfully distinguished by hierarchical cluster analysis and linear discriminant analysis both in buffer solution and spiked water samples, achieving 100% identification accuracy. This study provides a simple and powerful fingerprinting sensing platform for multiple HMIs, showing broad application prospects in the field of environmental monitoring.

Abstract:A colorimetric sensor array is proposed for ultrasensitive detection and identification of bacteria by using Cl− at various concentrations as sensing elements and triangular silver nanoparticles (T-AgNPs) as a single sensing nanoprobe. T-AgNPs are easily etched by Cl−. However, in the presence of bacteria, the etching process will be hindered. Different bacteria have differential protective effects on T-AgNPs due to their interactions, resulting in different etching degrees of T-AgNPs by Cl−, and visual color changes. By adjusting the antagonistic action between bacteria protection on T-AgNPs and the etching by using Cl− at various concentrations, different bacteria had their own color response patterns. Combined with linear discriminant analysis (LDA) and orthogonal partial least squares-discriminant analysis (OPLS-DA), the bacteria could be identified. The method was also used for bacteria mixtures identification and showed high sensitivity (OD600 = 1.0 × 10−6) for V. parahaemolyticus detection. Finally, the sensor array was successfully utilized in the identification of bacteria in pure and mineral bottled water. The method is low-cost, simple, sensitive, visual, and has potential application in point-of-care testing of bacteria.

Abstract:We report on a method for sensitive detection and discrimination of multiple proteins. The peroxidase-like activity of gold nanoparticles (AuNPs) is employed to catalyze the oxidation of 3′,3′,5′,5′-tetramethylbenzidine (TMB) in the presence of H2O2. Depending on the protein, this produces colorations with different absorbance. The catalytic activity of the reaction system is affected because different proteins are absorbed onto the surface of the AuNPs. The absorbance values at 370, 450, and 650 nm are different in the presence of different proteins. The array can discriminate 8 proteins at 50 nM concentration levels with 100% accuracy. It also can discriminate proteins being present in different concentrations and mixtures with different molar ratios of proteins. Human urine spikes with 8 proteins (Concanavalin A), Cytochrome C, Horseradish peroxidase, Human serum albumin, Myoglobin, Pepsin, Trypsin, Papain) can be differentiated by applying hierarchical cluster analysis.
