Abstract:In this paper, we constructed a label-free ratiometric fluorescence nanoprobe for ascorbic acid (AA) on the basis of the change in dual-emission signals by the specific reaction between cobalt oxyhydroxide (CoOOH) and AA. CoOOH nanoflakes were used as the recognition unit while polymer dots (PFO dots) and gold nanoclusters (AuNCs) served as luminescent units of the nanoprobe. After electrostatic assembly of PFO dots and AuNCs on the surface of CoOOH nanoflakes, the energy transfer between the PFO dots and CoOOH could occur and thereby quench the fluorescence of the PFO dots. When AA was present, CoOOH nanoflakes were reduced to Co2+, resulting in a recovery of the fluorescence of the PFO dots. In the meantime, the generated Co2+ could combine with the surface ligands of AuNCs, accompanied by a reduction in the luminescence intensity of the AuNCs. Therefore, the fluorescence intensity ratio of PFO dots to AuNCs would be related to the concentration of AA. The nanoprobe enabled highly sensitive analysis of AA with a detection limit of 1.9 μM and showed great performance in human serum samples and living cells. This report may not only endow a CoOOH-based nanoprobe with improved ability but also offer a novel strategy for the construction of ratiometric nanoprobes for AA and have potential applications in research into and clinical diagnosis of AA-related diseases.

Abstract:Herein, we demonstrate a facile, straightforward and green method for synthesis of highly red-emitting fluorescent bimetallic Au/Ag nanoclusters (NCs) employing lipoic acid as both a reducing agent and a protecting agent. The as-prepared Au/Ag NCs exhibited bright red emission with a strong peak centered at 630 nm (quantum yield measured as 6.4%), and showed excellent stability and remarkable temperature-dependent fluorescent property. The fluorescence responses were reversible and linear to the environment temperature from 20 to 65 C with a resolution as high as 1.34 degrees C, which allows its potential application as a fluorescent nanothermometer. Moreover, Au/Ag NCs were also explored for selective and sensitive determination of Fe3+ based on the specific quenching of the fluorescence by Fe3+, and explored for fluorescence imaging Fe3+ in living HeLa cells thanks to their very low cytotoxicity. One-pot Green Synthesis of Highly Fluorescent Glutathione-Stabilized Copper Nanoclusters for Fe3+ Sensing

Abstract:Herein, we reported a simple one-pot green method for preparation of fluorescent copper nanoclusters (Cu NCs) with a high quantum yield up to 8.6%, using glutathione as the stabilizing agent. The as-prepared Cu NCs had good water solubility, excellent stability and ultrafine size through the characterization of UV-vis absorption spectroscopy, fluorescence spectroscopy, transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared (FTIR) spectroscopy. Experimental results showed that the fluorescence of Cu NCs was linearly quenched by the Fe3+ concentrations in the range of 1-100 mu M, with the detection limit of 0.3 mu M. Abstract:In this paper, growth hormone releasing peptide-6 (GHRP-6) is a bioactive polypeptide and acts as the reducing agent and capping ligand for synthesis of bright green fluorescent gold nanoclusters (GHRP-6-Au NCs) by a simple and environmental-friendly aqueous method, with the assistance of NaBr as the fluorescent sensitizer. The obtained GHRP-6-Au NCs had high fluorescent quantum yield (10.7%), and the fluorescence was strongly quenched by the existence of trace Fe3+. Thus, a new and highly sensitive sensor for the assay of Fe3+ was constructed based on the analyte-induced fluorescent quenching mechanism. The sensor had a low detection limit of 1.4µM (S/N=3) and a wide linear range of 2-1000µM. Besides, GHRP-6-Au NCs exhibited low cytotoxicity and high biocompatibility for cell imaging.
Highly sensitive AuNCs@GSH/Ch-PtNPs metal nanoprobes for fluorescent and colorimetric dual-mode detection of ascorbic acid in drink

Abstract:Fluorescence detection is a commonly used analytical method with the advantages of fast response, good selectivity and low destructiveness. However, fluorescence detection, a single-mode detection method, has some limitations, such as background interference that affects the accuracy of the fluorescence signal, lack of visualization of the detection results, and low sensitivity for detecting low-concentration samples. In order to overcome the shortcomings of fluorescence single-mode detection, we used the dual-mode method of fluorescence and colorimetry to detect ascorbic acid. The dual-mode detection of AA by fluorescence and colorimetry in the probe system enhances the specificity and accuracy of the detection. This bimodal detection method solved the problem of low detection sensitivity in the low concentration range of the analytes to be tested, and was linear in the lower (0-50 {\mu}M) and higher (50-350 {\mu}M) concentration ranges, respectively, and had a lower detection limit (0.034 {\mu}M). This glutathione-based gold cluster assay is characterized by simplicity, rapidity and accuracy, and provides a new way for the quantitative analysis of ascorbic acid. In addition, the method was validated during the determination of AA in beverages, which has the advantages of high sensitivity and fast response time.

Abstract:We describe a method for the preparation of water-soluble gold nanoclusters (Au-NCs) from chloroauric acid using denatured-casein as both a reducing and stabilizing agent. The resulting Au-NCs were characterized by photoluminescence, UV–vis absorption, and X-ray photoelectron spectroscopies, and by transmission electron microscopy. The Au-NCs have an average diameter of 1.7 ± 0.2 nm and exhibit orange-red fluorescence emission peaking at 600 nm (with a Stokes’ shift as large as 237 nm), a quantum yield of 4.3 %, and good stability over the physiologically relevant range of pH values and ionic strength. Cytotoxicity studies showed the Au-NCs to display negligible effects in terms of altering cell proliferation or triggering apoptosis. Fluorescence imaging of HeLa cancer cells was accomplished by loading such cells with the Au-NCs. The fluorescence of the Au-NCs is found to be strongly quenched by Hg(II) ions, and thus the Au-NCs can be used for detecting and, possibly, imaging of Hg(II). An assay was worked out for the determination of Hg(II), and its limit of detection is 1.83 nM, which is 5.5 times lower than the maximum allowed concentration of Hg(II) in drinking water as defined by the US EPA.

Abstract:The development of dual-emission ratiometric fluorescent probes with aggregation-induced emission enhancement (AIEE) overcomes the limitations of gold nanocluster (Au NC)-based probes, particularly their weak intrinsic fluorescence, in real-world applications. These AIEE probes also exhibit superior detection limits and enhanced sensitivity. A novel combination for the reliable fluorometric detection of glutathione (GSH) was proposed, utilizing aggregation-induced emission enhancement (AIEE) facilitated by electrostatic interaction and spatial confinement. The probe consists of a ratiometric combination of negatively charged α-lipoic acid-modified Au NCs (LA@Au NCs) and positively charged silicon nanoparticles (SiNPs). The addition of SiNPs causes aggregation of LA@Au NCs, enhancing the fluorescence of LA@Au NCs through the AIE effect under electrostatic interaction and spatial confinement. The addition of Cu 2+ quenched the emission of LA@Au NCs as a result of charge transfer. The fluorescence emissions of LA@Au NCs were restored upon the addition of GSH due to the interaction between GSH and Cu 2+ . Simultaneously, the emission signal of SiNPs remains unchanged, serving as an internal reference signal during GSH measurement. It was found that the fluorescence ratio ( F 680 / F 465 ) is directly proportional to the concentration of GSH in the range of 0.05–100 μM, with a detection limit of 1.7 nM (S/N = 3). The proposed system was applied to detect GSH in real samples, including dietary supplements, human serum, and saliva samples. This work opens new avenues for constructing novel sensors based on AIEE for detecting biomolecules.

Abstract:We established a simple spectrophotometric and colorimetric method for detection of ascorbic acid based on the growth of silver nanoparticles in bovine serum albumin protected-silver nanoclusters (BSA–AgNCs) and Ag+ mixture. Due to the catalysis of BSA–AgNCs, ascorbic acid could reduce Ag+ to silver nanoparticles (NPs) at room temperature. The color of the mixture changed from colorless to yellow and a strong absorption band near 420nm could be found in their absorption spectra owing to localized surface plasmon resonance (LSPR) of produced silver NPs. The absorbance changes at 420nm had a good relationship with ascorbic acid concentration. Thus, we proposed a spectrophotometric and colorimetric method to determine ascorbic acid in concentration range from 2.0 to 50.0μM, with the corresponding limits of determination (3σ) of 0.16μM.

Abstract:In this report, a novel one-step chemical reduction method was reported for synthesis of water-soluble and stable fluorescent glutathione-templated silver nanocluster (GSH-Ag NCs) with ascorbic acid as an environmental-friendly reducing agent. On the basis of an oxidoreduction-induced fluorescence quenching mechanism, the prepared GSH-Ag NCs found to act as a cheap, non-toxic and highly sensitive “turn-off” fluorophore for ascorbic acid (AA). Furthermore, the fluorescence of the fluorophore/AA system could be recovered through addition of arginine (Arg), which made the system function as a highly selective “turn-on” sensor for arginine. Therefore, a “turn-off-on” switch sensor was proposed for detection of AA and Arg. Under optimized conditions, the probe gives a fluorescent response that is linear in the 2–300 μM concentration range of AA, with a detection limit of 0.1 μM. The probe for Arg, in turn, has a linear range in the 10–180 μM concentration range, and the limit of detection is 0.5 μM. In addition, the developed method showed great accuracy when employed to detect AA and Arg in human urine and serum, which shows its great potential in biological molecular recognition applications.

Abstract:Glutathione-Au nanoclusters (GSH-AuNCs) are known as typical luminophores with aggregation-induced emission (AIE) features, which can be triggered by low temperature, low pH values, and metal ions. However, these strict "triggers" restrict the application of this AIE probe under mild and neutral conditions. In this work, fluorescent amino-modified silicon nanoparticles (SiNPs) and GSH-AuNCs can self-assemble into well-designed nanospheres through electrostatic interactions under physiological conditions. The hybrid probe of SiNPs@GSH-AuNCs exhibits dual emissions located at 450 nm from SiNPs and 570 nm originating from the aggregation enhanced emission (AEE) of GSH-AuNCs. Interestingly, the addition of Zn2+ or Cd2+ can connect the nanospheres to form large aggregates, so a unique second increase of AEE from GSH-AuNCs appears, while the emission of SiNPs remains constant to act as an internal reference. The fluorescence ratios (I570/I450) of SiNPs@GSH-AuNCs are positively correlated with Zn2+ or Cd2+ with the linear range from 1.5 μM to 500 μM. Most importantly, besides the advantages of dual emissions, high fluorescence brightness, and good stability, the SiNPs@GSH-AuNCs also exhibit excellent biocompatibility and low cytotoxicity, and have been successfully applied in zinc imaging in live cells. Furthermore, the proposed method is also used to measure Zn2+ and Cd2+ in real samples with satisfactory recoveries.

Abstract:Sensing Hg 2+ is significant to protecting human health and environmental ecosystems, for its toxicity and genotoxicity. Here, highly stable fluorescent folic acid (FA)-protected Au nanoclusters (FA-AuNCs) were synthesized by optimizing the reactive parameters with high quantum yield of 34.7%. Main components of Au 4 L were confirmed by MALDI-TOF, and the electron-rich residues of FA shell enabled FA-AuNCs excellent photostability. FA-AuNCs exhibited sensitive response behavior to Hg 2+ with a minimum detectability of 1.3 nM, and presented extreme effect to the detection of Hg 2+ in real water. Notably, the cellular imaging and in-situ detection of Hg 2+ in cells can be achieved visually. The high selectivity was attributed to the chemical bond formed between Au + (4f 14 5d 10 ) and Hg 2+ (4f 14 5d 10 ). And the internal filter effect and static quenching effect were proved triggering the quenching of FA-AuNCs. The ultra-stable FA-AuNCs provide a potential promising opportunity for the in-situ tracing Hg 2+ from environmental and biological samples.

A simple and feasible fluorescent approach for rapid detection of hexavalent chromium based on gold nanoclusters

Abstract:Hexavalent chromium (Cr6+) owns hypertoxicity, non-biodegradability, and carcinogenicity, thus the detection of Cr6+ is of paramount significance for environmental monitoring and human health maintenance. Herein, a simple, rapid, and feasible fluorescent method based on gold nanoclusters (AuNCs) was established for determination of Cr6+. The AuNCs was coated by a simple and fast one-pot method using d-histidine (d-His) and polylysine (P-Lys) as stabilizers and reductants, which could be quenched by the addition of Cr6+ owing to the aggregation of AuNCs and fluorescence resonance energy transfer (FRET) between AuNCs and Cr6+. Under the optimal conditions, the fluorescent sensor exhibited good linearity within 10-10000 μg/L with limit of detection of 7.2 μg/L. The developed sensor possessed favorable sensitivity and selectivity. Additionally, the proposed method also had favorable recovery with good precision and accuracy within the actual sample, including celery cabbage, rice, capsule shell, and river water.
Fluorescent Sensing of Ascorbic Acid Based on Iodine Induced Oxidative Etching and Aggregation of Lysozyme-Templated Silver Nanoclusters

Abstract:In this work, we developed a sensitive and highly selective fluorescent approach for the detection of ascorbic acid (AA) by taking advantage of the oxidative etching effect of iodine (I-2) on the lysozymestabilized silver nanoclusters (dLys-AgNCs) with fluorescence quenching. I-2 could be produced from the redox reaction between iodate (IO3-) and AA, and thus the fluorescence intensity of dLys-AgNCs was turned off significantly in the coexistence of IO3- and AA. The fluorescence quenching of dLys-AgNCs had a good linear relationship with AA concentration, which allowed the detection of AA in the range from 0.05 to 45.0 mmol L-1 with a detection limit of 20 nmol L-1. The quenching mechanism was elucidated by Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM), zeta potential, and dynamic light scattering (DLS) measurements, confirming that the fluorescence quenching of the dLys-AgNCs was attributed to the oxidative etching of the in situ generated I-2, inducing aggregation of the dLys-AgNCs probe by forming Ag@ AgI nanocomposite. The dLys-AgNCs probe exhibited excellent selectivity for AA sensing over several common reducing agents tested. Moreover, this approach was extended to the detection of AA in orange juice and urine with recovery rates in the range of 96.0% (RSD: 4.11) to 100.9% (RSD: 3.28) and 94.5% (RSD: 6.40) to 99.2% (RSD: 5.36), respectively. A novel fluorescence sensor based on Al3+-mediated aggregation of gold nanoclusters for determination of citric acid in beverages

Abstract:This paper revealed a new strategy for citric acid (CA) detection using aggregation-induced emission (AIE)-based fluorescent gold nanoclusters (AuNCs). AuNCs was synthesized using glutathione (GSH) as the template and reducing agent and used as the fluorescent probe to detect CA under aluminum ion (Al 3+ ) mediation. The fluorescence intensity of AuNCs increased about 4 times with the addition of Al 3+ , but the enhanced fluorescence was quenched after the addition of CA. Based on this fluorescence phenomenon, an "on–off" fluorescence strategy was designed for the sensitive determination of CA and a linear detection range for CA was achieved within 0–80.0 μM. In addition, the developed probe exhibited high selectivity and accuracy for determination of CA. The mechanism of fluorescence enhancement and quenching of AuNCs was explored in detail. The established probe was used successfully for CA detection in beverages. The spiked recoveries from 97.50% to 103.67% were gratifying, which indicated the probe had potential prospects for detecting CA in food.

Synthesis of ficin-protected AuNCs in a droplet-based microreactor for sensing serum ferric ions

Abstract:A droplet-based microfluidic synthesis approach for preparation of ficin capped gold nano clusters (AuNCs) was developed. Well dispersed AuNCs could be procured within 8 min. Upon excitation wavelength at 340 nm, the resultant AuNCs exhibited a strong blue fluorescence with the maximum emission at 450 nm. Due to the aggregation-induced "turn-off" fluorescence mechanism, the synthesized AuNCs as a fluorescent probe displayed high sensitivity and good selectivity for sensing ferric ions. The relative fluorescence intensity versus ferric ions concentration yielded a good linear calibration in the range of 10.0-1000.0 μM (R2 = 0.998) and the limit of detection was 4.1 μM. Moreover, the possible mechanism for abated fluorescence intensity of AuNCs by adding ferric ions was discussed briefly. Further, the as-prepared fluorescent AuNCs was successfully applied for the detection of serum ferric ions. The results indicated that the droplet-based microfluidic synthesis system could provide a new way for the rapid preparation of AuNCs with good polydispersity and have potential as the sensing probes for the analysis of ferric ions in real biological samples.

Abstract:A water-soluble, stable, simple, and dual ligands (bovine serum albumin and lysozyme) and Ag doping co-enhanced Au nanoclusters (Lys/BSA-Ag/AuNCs) was synthesized by one-step wet chemical synthesis. The introduction of Lys-and Ag could improve significantly the quantum yields of AuNCs (QYs, 2.54% to 12.73%). A new ratio fluorescent probe (RF-probe) was successfully obtained for sensitively and selectively mensurating 4-hexylresorcinol (4-HR) and Cr 6+ . That is, the fluorescence (FL) intensity of nanoclusters at 610 nm was used as the built-in reference, while dopamine (DA) reacting with 4-HR could produce a new fluorescent substance, and its FL intensities at 490 nm were enhanced gradually with increasing the concentrations of 4-HR. On the other hand, Cr 6+ will react with DA and consume the amount of DA reacting with 4-HR, resulting in the weakening of FL intensity at 490 nm, so the ratiometric fluorescence detection of Cr 6+ was also realized. The contents of 4-HR and Cr 6+ could be monitored by the RF-probe with linear ranges of 1.0–140.0 and 3.0–110.0 nM, and corresponding values of LOD (limit of detection) were calculated to be 0.16 and 0.37 nM (3σ/slope). Moreover, the proposed method was extended to the detections 4-HR and Cr 6+ in practical samples, and the results were consistent with those of the standard HPLC and UV–vis spectrophotometric methods. In addition, Lys/BSA-Ag/AuNCs exhibited an excellent temperature sensing performance in the range of 10.0–64.0 °C.

Abstract:Label-free detection of ascorbic acid (AA) with high sensitivity and specificity based on the effects of AA on fluorescence quenching of carbon nitride quantum dots (CNQDs) is described. The CNQDs with a high fluorescence quantum yield of 21% and good dispersibility in water are prepared by reacting g-C3N4 sheets with ethylenediamine (EN). Fluorescence at 510 nm from the CNQDs is quenched gradually by addition of AA. As the AA concentration increases, the activity of the lone pair (LP) state of the CNQDs diminishes resulting in reduced fluorescence from the CNQDs and the mechanism of the static quenching effect is discussed. Since the CNQDs have a large specific surface area and abundant amino groups and AA exists in the anionic form at the physiological pH, the electrostatic interaction between CNQDs and AA inhibits excitation and emission of the LP states in the CNQDs. Owing to steric effects and hydrogen bonding, the CNQDs constitute a sensitive and selective detection platform for AA in a wide range from 0.5 to 200 μM with a detection limit of 150 nM (signal-to-noise ratio of 3). More importantly, the strategy can successfully be applied to the detection of AA concentrations in serum samples. This simple method which provides quantitative AA determination has large potential in clinical and health-related applications and the mechanism provides insights into intracellular AA monitoring.
Silver-enhanced fluorescence of bimetallic Au/Ag nanoclusters as ultrasensitive sensing probe for the detection of folic acid

Abstract:<p>Folic acid (FA) is the natural form of water-soluble vitamins widely found in most plants and animal products and its deficiency leads to several human body abnormalities. The advancements of metal nanoclusters are highly increasing due to their molecule-like optical properties and attractive applications. Because of increasingly demand of noble metal nanoclusters as sensing templates, different synthesis methods have been developed for facile synthesis of noble metal nanoclusters. Herein, red-emitting fluorescent bovine serum albumin (BSA)-capped Au-Ag bimetallic NCs are facilely synthesized through green one-pot synthetic approach. The effect of silver on the fluorescence properties of Au NCs was investigated and it was found that introduction of silver can enhance the fluorescence intensity. The fluorescence intensity of the as-prepared Au-Ag nanoclusters gets quenched in the presence of folic acid in an aqueous medium and it was used as ultrasensitive sensing probe for FA detection. The developed Au-Ag NCs-based sensing probe shows linear response in the wide range of 0-100 μM and the detection limit is as low as 0.47 nM. Its applicability has also been confirmed successfully in real human serum, urine and FA tablet samples. Due to the high stability, sensitivity and selectivity, the developed bimetallic cluster sensing system is highly promising to be applied in the pharmaceutical and clinical laboratories.</p>

Abstract:By using Fe 7 S 8 nanosheets (NSs) as catalyst, O -phenylenediamine (OPD) as chromogenic substrate, H 2 O 2 as oxidant, and ascorbic acid (AA) as regulator, a dual ratio colorimetric and fluorescence sensing system for determining AA was constructed. The inherent peroxidase like activity of Fe 7 S 8 NSs enables it to catalyze the oxidation of OPD in the existence of H 2 O 2 , resulting in the formation of yellow-colored 2, 3-diaminophenazine (DAP) with a characteristic absorption peak at 450 nm and a fluorescence emission peak at 560 nm. However, the presence of AA could suppress the oxidation of OPD and induce the generation of quinoxaline derivative (QXD) 3-(dihydroxyethyl) furo [3,4-b] quinoxaline-1-one with absorption and fluorescence emission peaks located at 345 and 435 nm, respectively. Thus, employing the absorption of QXD at 345 nm and emission at 435 nm as the reporting signals, and the absorption of DAP at 450 nm and emission at 560 nm as the reference signals, a ratio colorimetric ( A 345 / A 450 ) and ratio fluorescence ( F 435 / F 560 ) dual signal detection method for AA was developed. Under the optimized conditions, the value of A 345 / A 450 was found to be linear with AA concentration in the ranges of 1.0–40 μM and 40–100 μM, and the value of F 435 / F 560 was strongly correlated with AA concentration in the ranges of 1.0–30 μM and 30–100 μM. When applied for the assay of AA in human serum samples, an acceptable recovery ranging from 93.1 to 106.6 % was obtained, confirming the potential of the sensing system in practical applications.
