Abstract:Exogenous photoacoustic contrast agents such as gold nanoparticles, are widely utilized in photoacoustic imaging. Enhancing the photoacoustic performance of gold nanoparticles is pivotal for improving the quality and expanding the application scope of photoacoustic imaging. In this study, the influence of pulse duration (10 ps to 10 ns) on photothermal and photoacoustic responses of gold nanospheres is investigated quantitatively. The influence of the electron-lattice coupled effect of gold nanospheres on the photoacoustic signal was specifically considered. The results reveal that the electron-lattice coupled process significantly affects the photoacoustic signal when the pulse duration is smaller than 5 ns. Furthermore, as the pulse duration decreases, increasing the interfacial thermal conductivity can substantially enhance heat transfer between the gold nanospheres and the surrounding water. However, this approach does not effectively enhance photoacoustic performance. Interestingly, when increasing the thermal conductivity, it was found that there is an optimal pulse duration within the range of 10 ps-10 ns. Employing an incident pulse laser with this optimal pulse duration can maximize the enhancement of the photoacoustic signal.

Abstract:By combining the advantages of high resolution in optical imaging and deep penetration depth in ultrasound imaging, photoacoustic(PA) imaging enables high resolution deep imaging in vivo. A nanoprobe with high conversion efficiency is usually used in order to increase the amplitude of the photoacoustic signal. Here, highly efficient PA conversion is demonstrated in metal-insulator-metal(MIM) nanostructures. A magnetic resonance can be formed to achieve nearly 100% absorption of incident light near the resonant wavelength. In this paper, the absorbance of the MIM structure in the visible and near-infrared wavelength is demonstrated at first. Then, multiphysics coupling approach is used to solve the electromagnetic, thermodynamic and transient acoustic pressure physics. The results show that the photoacoustic signal amplitude of the MIM structures is much higher than that of the same structures without the top metal strips. Due to its high PA conversion efficiency, the MIM nanostructures can generate strong PA signals with low laser incident power, resulting in better biocompatibility. In this way, it can be applied not only as a PA probe to biomedical PA imaging, photoacoustic tomography, but also to medical related fields such as photothermotherapy and precision drug delivery.
Heat Transfer at Solid–Gas Interfaces by Photoacoustics at Brillouin Frequencies

Abstract:We use time-resolved ellipsometry to investigate the rate of heat transfer at solid-gas interfaces through measurements of the amplitude and phase of acoustic waves at Brillouin frequencies, 100-400 MHz, at pressures 2 orders of magnitude higher than earlier comparable studies. An ultrafast optical pulse heats a thin metal film deposited on a sapphire substrate. Heat flow from the substrate into the gas causes expansion of the gas and generates an acoustic wave that is probed by off-null ellipsometry with subpicosecond time resolution. We compare the amplitudes and phases of photoacoustic signals generated in inert gases Ne, Ar, Kr, and Xe to a continuum theoretical model that includes the thermal accommodation coefficient a at the gas-solid interface. For the surfaces we have studied, bare Au and Au coated by a self-assembled monolayer of 1-octadecanethiol (ODT), this comparison between experiment and theory for the amplitude of the photoacoustic waves suggests that alpha values for bare and ODT-terminated Au are similar and alpha > 0.3. This conclusion is tentative, however, because the phases of the photoacoustic waves show systematic differences that are not predicted by the model. For tetrafluoroethane vapor (R-134a refrigerant), the photoacoustic signal generated by a Au surface coated with a hydrophilic (COOH-terminated) self-assembled monolayer is a factor of 2 larger than the photoacoustic signal generated by a hydrophobic (CH3-terminated) monolayer. We also report measurements of the ultrafast ellipsometry signals generated by the sudden desorption of physisorbed methanol and water on hydrophobic and hydrophilic self-assembled monolayers.
Low Temperature-Mediated Enhancement of Photoacoustic Imaging Depth

Abstract:We study the temperature dependence of the underlying mechanisms related to the signal strength and imaging depth in photoacoustic imaging. The presented theoretical and experimental results indicate that imaging depth can be improved by lowering the temperature of the intermediate medium that the laser passes through to reach the imaging target. We discuss the temperature dependency of optical and acoustic properties of the intermediate medium and their changes due to cooling. We demonstrate that the SNR improvement of the photoacoustic signal is mainly due to the reduction of Grüneisen parameter of the intermediate medium which leads to a lower level of background noise. These findings may open new possibilities toward the application of biomedical laser refrigeration.
Photoacoustic signal enhancements from gold nano-colloidal suspensions excited by a pair of time-delayed femtosecond pulses

Abstract:Photoacoustic signal enhancements were observed with a pair of time-delayed femtosecond pulses upon excitation of gold nanosphere colloidal suspension. A systematic experimental investigation of photoacoustic intensity within the delay time, t = 0 to 15 ns, was carried out. The results revealed a significant enhancement factor of ~2 when the pre-pulse energy is 20-30% of the total energy. Pre-pulse and main pulse energy ratios, E(1)p-pol. :E(2)s-pol. were varied to determine the optimal ratio that yields to maximum photoacoustic signal enhancement. This enhancement was ascribed to the initial stage of thermalization and bubble generation in the nanosecond time scale. Pre-pulse scattering intensity measurements and numerical finite-difference time-domain calculations were performed to reveal dynamics and light field enchancement, respectively.

Abstract:Exploiting the variances in light absorption among different components of biological tissues and utilizing the light absorption coefficient as an imaging contrast, photoacoustic imaging achieves remarkable imaging resolution. Within the laser wavelength range commonly employed in photoacoustic imaging, biological tissue components like hemoglobin and melanin exhibit higher light absorption coefficients compared to other components. Consequently, these components generate stronger photoacoustic signals during the imaging process. However, this heightened signal strength obscures components of biological tissues with low light absorption coefficients, making it challenging to clearly visualize them in the resulting images. To address this limitation and enhance the visibility of low absorption coefficient components against a background of intense photoacoustic signals produced by high absorption coefficient components, we propose a method that utilizes lowintensity unfocused ultrasound to induce the temperature difference (TD) effect in biological tissues during photoacoustic imaging. By analyzing the disparities between the photoacoustic signals collected before and after introducing the TD effect, we establish a novel imaging parameter that improves the imaging contrast. To validate the feasibility and effectiveness of inducing photoacoustic effects in biological tissues, we conducted simulations using the finite element method, which yielded positive results. Subsequent simulations of photoacoustic imaging incorporating the TD effect demonstrated enhanced visualization of components with low light absorption coefficients in biological tissue. The method proposed in this paper is expected to provide a research idea for improving the photoacoustic imaging contrast of biological tissues.

Abstract:Purpose: The purpose of this work is to demonstrate that higher amplitude of ultrashort laser induced photoacoustic signal can be achieved by multiple-pulse excitation when the temporal duration of the pulse train is less than the minimum of the medium's thermal relaxation time and stress relaxation time. Thus, improved signal-to-noise ratio can thus be attained through multiple-pulse excitation while minimizing the energy of each pulse.Methods: The authors used a Michelson interferometer together with a picoseconds laser system to introduce two 6 ps pulses separated by a controllable delay by introducing a path length difference between the two arms of the interferometer. The authors then employed a series of three interferometers to create a pulse train consisting of eight pulses. The average pulse energy was 11 nJ and the temporal span of the pulse train was less than 1 ns.Results: The detected peak-to-peak amplitude of the multiple-pulse induced photoacoustic waves were linearly dependent on the number of pulses in the pulse train and such a linearity held for different optical absorption coefficients. The signal-to-noise ratio improved when the number of pulses increased. Moreover, nonlinear effects were not detected and no photoacoustic saturation effect was observed.Conclusions: The authors have shown that multiple-pulse excitation improves the signal-to-noise ratio through an accumulated energy deposition effect. This method is invaluable for photoacoustic measurements that require ultrashort laser pulses with minimized pulse energy to avoid laser damage.
Tuning photoacoustics with nanotransducers via Thermal Boundary Resistance and Laser Pulse Duration

Abstract:The photoacoustic effect in liquids, generated by metal nanoparticles excited with short laser pulses, offers high contrast imaging and promising medical treatment techniques. Understanding the role of the thermal boundary resistance (TBR) and the laser pulse duration in the generation mechanism of acoustic waves is essential to implement efficient photoacoustic nanotransducers. This work theoretically investigates, for the paradigmatic case of water-immersed gold nanocylinders, the role of the TBR and of laser pulse duration in the competition between the launching mechanisms: the thermophone and the mechanophone. In the thermophone, the nanoparticle acts as a nanoheater and the wave is launched by water thermal expansion. In the mechanophone, the nanoparticle directly acts as a nanopiston. Specifically, for a gold-water interface, the thermophone prevails under ns light pulse irradiation, while the mechanophone dominates shortening the pulse to the 10 ps regime. For a graphene-functionalized gold-water interface, instead, the mechanophone dominates over the entire range of explored laser pulse durations. Results point to high-TBR, liquid-immersed nanoparticles as potentially efficient photoacoustic nanogenerators, with the advantage of keeping the liquid environment temperature unaltered.

Influence of the Temperature-Dependent Dielectric Constant on the Photoacoustic Effect of Gold Nanospheres

Abstract:Photoacoustic imaging techniques with gold nanoparticles as contrast agents have received a great deal of attention. The photoacoustic response of gold nanoparticles strongly depends on the far-field optical properties, which essentially depend on the dielectric constant of the material. The dielectric constant of gold not only varies with wavelength but is also affected by temperature. However, the effect of the temperature dependence of the dielectric constant on gold nanoparticles' photoacoustic response has not been fully investigated. In this work, the Drude-Lorentz model and Mie theory are used to calculate the dielectric constant and absorption efficiency of gold nanospheres in aqueous solution, respectively. Then, the finite element method is used to simulate the heat transfer process of gold nanospheres and surrounding water. Finally, the one-dimensional velocity-stress equation is solved by the finite-difference time-domain method to obtain the photoacoustic response of gold nanospheres. The results show that under the irradiation of a high-fluence nanosecond pulse laser, ignoring the temperature dependence of the dielectric constant will lead to large errors in the photothermal response and the nonlinear photoacoustic signals (it can even exceed 20% and 30%). The relative error of the photothermal and photoacoustic response caused by ignoring the temperature-dependent dielectric constant is determined from both the temperature dependence of absorption efficiency and the maximum temperature increase of gold nanospheres. This work provides a new perspective for the photothermal and photoacoustic effects of gold nanospheres, which is meaningful for the development of high-resolution photoacoustic detectors and nano/microscale temperature measurement techniques.

Abstract:Photoacoustic effect refers to the acoustic generation induced by laser irradiation, where nanosecond pulsed laser source is normally used to provide instantaneous heating and thermoelastic expansion of the sample. More generally, photoacoustic generation requires active intensity modulation of laser source to produce sharp temperature gradient, which is key to generate acoustic wave. In this paper, we propose a novel photoacoustic effect from moving droplet, which generates photoacoustic wave by a non-modulated continuous-wave laser. When the droplet moves through the laser spot area, it can be heated up and cooled down instantaneously, generating photoacoustic waves. We name it passive photoacoustic effect. Theoretical analysis and simulation study validated the existence of passive photoacoustic effect. This phenomenon may find potential application in high-throughput photoacoustic cytometry.

Abstract:The photothermal phenomenon involves material photon heating in the form of continuous waves, modulated waves, or pulses. The resulting temperature rise carries very rich information about the material's structures and thermophysical properties. This review is constructed to cover some of the extended ideas of the photothermal phenomenon for measuring a material's thermophysical properties and interface thermal conductance/resistance. For frequency-domain photothermal phenomena, the photoacoustic (PA) and photothermal radiation (PT) techniques provide great ways to measure coatings and suspended samples, which can also be measured using the laser flash and time-domain thermoreflectance (TDTR) techniques. Thermal probing based on electrical thermal sensing is successfully implemented in the transient photo-electro-thermal (TPET) and pulsed laser-assisted thermal relaxation (PLTR) techniques, which significantly extend the capability of laser flash technique to micro/nanoscale 1D structures. The energy transport state-resolved Raman (ET-Raman) is distinct from traditional techniques in its material-specific thermal sensing capability and integral way of ultrafast thermal sensing. Detailed physics discussions are provided for all these techniques, including their advantages and limitations. Still, large development rooms exist for the relatively new techniques, including TPET, PLTR, and ET-Raman. These include measurements under extreme situations, sensitivity improvement, and distinguishing conjugated physical processes.

Abstract:Photoacoustic tomography is a potential and noninvasive medical imaging technology. It combines the advantages of pure optic imaging and pure ultrasound imaging. Photoacoustic signals induced by a short pulse laser cover a wide spectral range. We have explored the influences of attenuation of photoacoustic signals, which vary according to frequencies, to the quality of reconstructed photoacoustic images. It reveals that the attenuation of low frequent components are less than that of high frequencies, and the latter is more important for photoacoustic imaging with high resolution. Based on the ultrasonic attenuation theory, the photoacoustic imaging with rectification of the attenuation of different frequent component was performed. The experiments results show that this method improves the resolution of reconstructed images, which improves from 0.3mm to 0.2mm. A Q-switched Nd:YAG laser operating at 1064nm was used as light source. The laser had a pulse width of 6ns and a repetition frequency of 20Hz. A needle PVDF hydrophone with diameter of 1mm was used to detect photoacoustic signals.

Abstract:The dual-pulse nonlinear photoacoustic technique is a recently developed technology based on temperature dependence of the Grüneisen parameter and involves consecutive excitations of biological tissue using two laser pulses with a short time delay. Here we review the principle of the technique and give a discussion about its technical aspects, including selection and combination of excitation laser wavelengths, determination of laser fluence, estimation of thermal relaxation function and probability of photoablation or cavitation. Comparisons between the dual-pulse technique and conventional photoacoustics as well as thermal photoacoustics are also presented. These investigations are supported by experimental results and will give a practical reference and guide for further developments of the technique.

Abstract:SignificanceIn the photoacoustic (PA) technique, the laser irradiation in the time domain (i.e., laser pulse duration) governs the characteristics of PA imaging—it plays a crucial role in the optical-acoustic interaction, the generation of PA signals, and the PA imaging performance.AimWe aim to provide a comprehensive analysis of the impact of laser pulse duration on various aspects of PA imaging, encompassing the signal-to-noise ratio, the spatial resolution of PA imaging, the acoustic frequency spectrum of the acoustic wave, the initiation of specific physical phenomena, and the photothermal–PA (PT-PA) interaction/conversion.ApproachBy surveying and reviewing the state-of-the-art investigations, we discuss the effects of laser pulse duration on the generation of PA signals in the context of biomedical PA imaging with respect to the aforementioned aspects.ResultsFirst, we discuss the impact of laser pulse duration on the PA signal amplitude and its correlation with the lateral resolution of PA imaging. Subsequently, the relationship between the axial resolution of PA imaging and the laser pulse duration is analyzed with consideration of the acoustic frequency spectrum. Furthermore, we examine the manipulation of the pulse duration to trigger physical phenomena and its relevant applications. In addition, we elaborate on the tuning of the pulse duration to manipulate the conversion process and ratio from the PT to PA effect.ConclusionsWe contribute to the understanding of the physical mechanisms governing pulse-width-dependent PA techniques. Abstract:Photoacoustic sensing and imaging techniques have been studied widely to explore optical absorption contrast based on nanosecond laser illumination. In this paper, we report a long laser pulse induced dual photoacoustic (LDPA) nonlinear effect, which originates from unsatisfied stress and thermal confinements. Being different from conventional short laser pulse illumination, the proposed method utilizes a long square-profile laser pulse to induce dual photoacoustic signals. Without satisfying the stress confinement, the dual photoacoustic signals are generated following the positive and negative edges of the long laser pulse. More interestingly, the first expansion-induced photoacoustic signal exhibits positive waveform due to the initial sharp rising of temperature. On the contrary, the second contraction-induced photoacoustic signal exhibits exactly negative waveform due to the falling of temperature, as well as pulse-width-dependent signal amplitude. An analytical model is derived to describe the generation of the dual photoacoustic pulses, incorporating Gruneisen saturation and thermal diffusion effect, which is experimentally proved in vivo.
Photoacoustic Response Optimization of Gold Nanorods in the Near-Infrared Region

Abstract:Photoacoustic imaging (PAI) combines the advantage of optical and ultrasonic imaging, which has a high signal-to-noise ratio, and spatial resolution. To further improve the performance of PAI, gold nanorods can be utilized as exogenous contrast agents, and their size can be controlled. The size change of gold nanorods will change their absorption and heat transfer characteristics, and then affect their photoacoustic characteristics. Therefore, in the present work, the influences of absorption characteristics (absorption cross section and volume absorption coefficient) and heat transfer characteristics (specific surface area) of gold nanorods on photoacoustic response are studied by FDTD (FDTD Solutions, Lumerical) and FEM (Comsol Multiphysics). Results show that the increase of specific surface area will enhance the thermal coupling between gold nanorods and water, so as to improve the heat transfer and produce different photothermal responses without significantly affecting the photoacoustic quantum yield (As the specific surface area increases by about 10 times, the temperature increases by about 12 times, while the photoacoustic quantum yield increases by about 24%). On the contrary, absorption characteristics are decisive factors for both photothermal and photoacoustic responses. Therefore, in terms of improving photoacoustic quantum yield, when the size of gold nanorods changes, more attention should be paid to the improvement of the absorption characteristics rather than the specific surface area. In addition, increasing the specific surface area can significantly improve the photostability of gold nanorods, which is very important for practical applications.

Abstract:Recent developments in time-resolved photoacoustic calorimetry (PAC) are discussed. An equation is derived relating the amplitude of the maximum photoacoustic signal to parameters of a photoacoustic cell, to physical properties of the solvents, and to the energy deposited as heat. The equation is validated by a series of scaling experiments: it correctly correlates the dependence of the PAC signal on cell thickness, on energy deposition, and on solvent properties. The dominant sources of background signal have been determined and the background signal reduced substantially relative to previous work. It is now possible to measure energies and lifetimes of transients in solutions with optical densities as low as ∼5×10−4. Data are presented for the energy of 2-cyclopentenone triplet, a transient for which interpretable PAC measurements were previously not possible. Its triplet energy is 73.1±1.1 kcal/mole. With reduced background and a faster digital storage oscilloscope, a quite short transient lifetime, 7.53±0.2 ns, and a triplet energy of 51.0±2.1 kcal/mol have been measured for 1,1-bis-(p-bromophenyl)ethene. The lifetime agrees very well with that determined by kinetic absorption spectroscopy and the triplet energy agrees with expectation based on the unsubstituted 1,1-diphenylethene.

Abstract:Acoustic-resolution microscopy is an important imaging method in studying biological tissues with deep penetration. Using high-frequency transducer could reduce the size of the acoustic focal spot, thereby improving the resolution of microscopy. However, high-frequency photoacoustic signals usually suffer from acoustic attenuation which weakens their energy and distorts the image. In this paper, an attenuation compensation for high-frequency acoustic-resolution photoacoustic imaging is proposed. This technique upgrades the wavenumber term by considering acoustic attenuation and dispersion during wavefield extrapolation, which is a Fourier-domain image reconstruction. It is able to deal with the space-variant attenuation effect and inherits the high computational efficiency of wavefield extrapolation methods. According to the results of simulations and experiments, attenuation compensation successfully eliminates the dispersion induced reconstruction errors, obviously improves the resolution of the image and clearly presents the edges of targets.
