Abstract:Exploiting the variances in light absorption among different components of biological tissues and utilizing the light absorption coefficient as an imaging contrast, photoacoustic imaging achieves remarkable imaging resolution. Within the laser wavelength range commonly employed in photoacoustic imaging, biological tissue components like hemoglobin and melanin exhibit higher light absorption coefficients compared to other components. Consequently, these components generate stronger photoacoustic signals during the imaging process. However, this heightened signal strength obscures components of biological tissues with low light absorption coefficients, making it challenging to clearly visualize them in the resulting images. To address this limitation and enhance the visibility of low absorption coefficient components against a background of intense photoacoustic signals produced by high absorption coefficient components, we propose a method that utilizes lowintensity unfocused ultrasound to induce the temperature difference (TD) effect in biological tissues during photoacoustic imaging. By analyzing the disparities between the photoacoustic signals collected before and after introducing the TD effect, we establish a novel imaging parameter that improves the imaging contrast. To validate the feasibility and effectiveness of inducing photoacoustic effects in biological tissues, we conducted simulations using the finite element method, which yielded positive results. Subsequent simulations of photoacoustic imaging incorporating the TD effect demonstrated enhanced visualization of components with low light absorption coefficients in biological tissue. The method proposed in this paper is expected to provide a research idea for improving the photoacoustic imaging contrast of biological tissues.

Abstract:Non-invasive laser-induced photoacoustic tomography is attracting more and more attentions in the biomedical optical imaging field. This imaging modality takes the advantages in that the tomography image has the optical contrast similar to the optical techniques while enjoying the high spatial resolution comparable to the ultrasound. Currently, its biomedical applications are mainly focused on breast cancer diagnosis and small animal imaging. In this paper, we report in detail a photoacoustic tomography experiment system constructed in our laboratory. In our system, a Q-switched ND: YAG pulse laser operated at 532nm with a 10ns pulse width is employed to generate photoacoustic signal. A tissue-mimicking phantom was built to test the system. When imaged, the phantom and detectors were immersed in a water tank to facilitate the acoustic detection. Based on filtered back-projection process of photoacoustic imaging, the two-dimension distribution of optical absorption in tissue phantom was reconstructed.

Abstract:Photoacoustic imaging (PAI) and thermoacoustic imaging (TAI) are two emerging biomedical imaging techniques that both utilize ultrasonic signals as an information carrier. Unique advantages of PAI and TAI are their abilities to provide high resolution functional information such as hemoglobin and blood oxygenation and tissue dielectric properties relevant to physiology and pathology. These two methods, however, may have a limited detection depth and lack of endogenous contrast. An exogenous contrast agent is often needed to effectively resolve these problems. Such agents are able to greatly enhance the imaging contrast and potentially break through the imaging depth limit. Furthermore, a receptor-targeted contrast agent could trace the molecular and cellular biological processes in tissues. Thus, photoacoustic and thermoacoustic molecular imaging can be outstanding tools for early diagnosis, precise lesion localization, and molecular typing of various diseases. The agents also could be used for therapy in conjugation with drugs or in photothermal therapy, where it functions as an enhancer for the integration of diagnosis and therapy. In this article, we present a detailed review about various exogenous contrast agents for photoacoustic and thermoacoustic molecular imaging. In addition, challenges and future directions of photoacoustic and thermoacoustic molecular imaging in the field of translational medicine are also discussed.

Enhancement of Photoacoustic Tomography in the Tissue with Speed-of-sound Variance Using Ultrasound Computed Tomography

Abstract:The speed-of-sound variance will decrease the imaging quality of photoacoustic tomography in acoustically inhomogeneous tissue. In this study, ultrasound computed tomography is combined with photoacoustic tomography to enhance the photoacoustic tomography in this situation. The speed-of-sound information is recovered by ultrasound computed tomography. Then, an improved delay-and-sum method is used to reconstruct the image from the photoacoustic signals. The simulation results validate that the proposed method can obtain a better photoacoustic tomography than the conventional method when the speed-of-sound variance is increased. In addition, the influences of the speed-of-sound variance and the fan-angle on the image quality are quantitatively explored to optimize the image scheme. The proposed method has a good performance even when the speed-of-sound variance reaches 14.2%. Furthermore, an optimized fan angle is revealed,which can keep the good image quality with a low cost of hardware. This study has a potential value in extending the biomedical application of photoacoustic tomography.

Abstract:Photoacoustics has been broadly studied in biomedicine, for both human and small animal tissues. Photoacoustics uniquely combines the absorption contrast of light or radio frequency waves with ultrasound resolution. Moreover, it is non-ionizing and non-invasive, and is the fastest growing new biomedical method, with clinical applications on the way. This review provides a brief recap of recent developments in photoacoustics in biomedicine, from basic principles to applications. The emphasized areas include the new imaging modalities, hybrid detection methods, photoacoustic contrast agents and the photoacoustic Doppler effect, as well as translational research topics.

Abstract:Photoacoustic (PA) imaging is an emerging technology that could map the functional contrasts in deep biological tissues in high resolution by "listening" to the laser induced thermoelastic waves. Almost all of the current studies in PA imaging are focused on the intensity of the PA signals as an indication of the optical absorbance of the biological tissues. Our group has for the first time demonstrated that the frequency domain power distribution of the broadband PA signals encode the texture information within the regions-of-interest (ROI). Following the similar method of ultrasound spectral analysis (USSA), photoacoustic spectrum analysis (PASA) could evaluate the relative concentrations and, more importantly, the dimensions of microstructures of the optically absorbing materials in biological tissues, including lipid, collagen, water and hemoglobin. By providing valuable insights into tissue pathology, PASA should benefit basic research and clinical management of many diseases, and may help achieve eventual "noninvasive biopsy". In this work, taking advantage of the optical absorption contrasts contributed by lipid and hemoglobin at 1200-nm and 532-nm wavelengths respectively, we investigated the capability of PASA in identifying histological changes corresponding to fat accumulation livers through the study on ex vivo and in situ mouse models. The PA signals from the mouse livers were acquired using our PA and US dual-modality imaging system, and analyzed in the frequency domain. After quantifying the power spectrum by fitting it to a first order model, three spectral parameters, including the intercept, the midband fit and the slope, were extracted and used to differentiate fatty livers from normal livers. The comparison between the PASA parameters from the normal and the fatty livers supports our hypotheses that PASA can quantitatively identify the microstructure changes in liver tissues for differentiating normal and fatty livers.

Abstract:Photoacoustic tomography is a developing, promising, non-invasive imaging method in the medical clinic diagnosis. It is an ultrasound-mediated biophotonic imaging method based on the intrinsic optical absorption properties of tissue and ultrasonic detection, and combines the merits of both high contrast advantage of pure optical imaging and high resolution advantage of pure ultrasound imaging. Photoacoustic tomography can be performed by detecting photoacoustic waves instead of detecting photons. In photoacoustic tomography, imaging contrast is based primarily on the optical properties of biological tissues, and imaging resolution is based primarily on the ultrasonic waves. It can avoid the influence of optical scattering on imaging resolution in principle, and can provide tomography of tissues with high contrast and high spatial resolution at medium depths. Photoacoustic tomography can provide an effective approach to studying the structures, physiological properties, metabolisms, pathological properties of biological tissues. It has important potential clinical applications in the early non-invasive detection of cancers, structural and functional in vivo imaging. A brief introduction of photoacoustic imaging mechanisms is gives, and the imaging methods, the image reconstruction algorithm and the potential biomedical applications of photoacoustic tomography are reviewed.

Abstract:One of the major challenges for photoacoustic tomography is the variance of the speed of sound (SOS) in realistic tissue, which could lead to defocusing in image reconstruction and degrade the reconstructed image. In this study, we propose a method to optimize the SOS used for image reconstruction based on a memory effect of photoacoustic signal. We reveal that the photoacoustic signals received by two adjacent transducers have a high degree of similarity in waveform, while a time delay exists between them. The time delay is related to the SOS. Based on this physical phenomenon, an iterative operation is implemented to estimate the SOS used for image reconstruction. Both simulations and experiments confirm that the method significantly enhances the reconstructed image in inhomogeneous tissue. This study may have potential value in improving the performance of photoacoustic tomography in biomedical applications. Abstract:Photoacoustic imaging is a novel imaging modality that utilizes pulsed laser to illuminate samples and ultrasonic transducers for detection of the laser absorption generated ultrasonic/photoacoustic waves. Benefited from its high optical contrast and acoustic resolution at depth, as well as the capability of acquiring optical spectral information, photoacoustic imaging allows for structural and functional imaging of the biological relevant information such as hemoglobin concentration, blood oxygen metabolism, and lipid with high specificity, and is able to capture the corresponding physiological feature dynamics with high sensitivity. In terms of the imaging system, photoacoustic imaging manifests a variety of embodiments catering the multi-scale biomedical imaging needs. The above mentioned features and capabilities enable extensive biomedical application potentials of this imaging modality in both clinical diagnosis and pathological analysis, such as malignant tumor detection, atherosclerotic plaque assessment, and microcirculation abnormality diagnosis. Therefore, it is attracting more and more attention and interests in a wide range of biomedical research areas. This article presented the recent progress of photoacoustic technology development and application studies in our group in the fields of microvascular imaging, intravascular imaging, gastrointestinal endoscopic imaging, and sentinel lymph node imaging.
Listening to Light by Fast Photoacoustic Tomography Based on a Digital Phased Array System

Abstract:Photoacoustic imaging technique, which provided high ultrasonic resolution and high optical contrast tissue images, can overcome the disadvantages of pure optical imaging by measurement of laser-induced sound waves. The waves produced by tissue are high-frequency ultrasounds, meaning that they cannot be heard by human ear. However, it can be picked up with ultrasonic transducer and analyze them with a computer. The laser-induced ultrasonic signals from a biological sample can be used to reveal the tissues structure based on optical contrast. In current experiment system, an integrity multi-element synthetic aperture focusing (M-SAF) photoacoustic imaging system using real-time digital beamformer is developed. This system relies on pumping laser source to irradiate the biological tissue to produce photoacoustic signal, a linear ultrasonic transducer array is connected to a multichannel signal acquisition and real-time digital beam-formation system providing techniques of real-time dynamic receiving focus and dynamic receiving apodization to process the photoacoustic signal. Each element of the transducer array has a thin cylinder ultrasonic lens to select 2D image plane and suppress the out-of-plane signals to realize photoacoustic computed tomography. This method and system can provide a fast and reliable photoacoustic tomography approach that could be applied to noninvasive imaging and clinic diagnosis.

Abstract:Photoacoustic (PA) tomography (PAT), also referred to as optoacoustic tomography, is an emerging biomedical imaging method based on the PA eect (Xu and Wang 2006, Li and Wang 2009, Oraevsky and Karabutov 2003). e PA eect was reported by Alexander Graham Bell in 1880 (Bell 1880), and refers to the generation of acoustic waves by substances being illuminated by electromagnetic (EM) waves of varying power. e PA eect results from the variational thermal expansion within the material as it absorbs the EM radiative energy. Since the ability of EM absorption re¦ects a material’s characteristics, much physical and chemical information can be revealed by studying PA signals. Historically, the PA eect was of little research interest for a long time, but the invention of laser, an adequate PA illumination source, changed matters drastically. Now, variousdetection methods based on PA eect have been widely implemented in physics, chemistry, biology, engineering, and medicine (Rosencwaig 1980, Gusev et al. 1993).

Abstract:Photoacoustic tomography has the potential to image the configuration or function of animal or human organs with simultaneous high contrast and high spatial resolution. This article provides an overview of photoacoustic tomography with an acoustic lens. And a new fast photoacoustic tomography technique is introduced. An acoustic lens is able to map the initial photoacoustic pressure distribution in an object in real time without the need of complicated computational reconstruction. Further more, this method produces no artifacts in reconstructed images. Finally, a new fast photoacoustic tomography system based on acoustic is discussed in detail in this paper.

Abstract:Photoacoustic (optoacoustic) tomography, combining optical absorption contrast and highly scalable spatial resolution (from micrometer optical resolution to millimeter acoustic resolution), has broken through the fundamental penetration limit of optical ballistic imaging modalities-including confocal microscopy, two-photon microscopy, and optical coherence tomography-and has achieved high spatial resolution at depths down to the diffusive regime. Optical absorption contrast is highly desirable for microvascular imaging and characterization because of the presence of endogenous strongly light-absorbing hemoglobin. We focus on the current state of microvascular imaging and characterization based on photoacoustics. We first review the three major embodiments of photoacoustic tomography: microscopy, computed tomography, and endoscopy. We then discuss the methods used to characterize important functional parameters, such as total hemoglobin concentration, hemoglobin oxygen saturation, and blood flow. Next, we highlight a few representative applications in microvascular-related physiological and pathophysiological research, including hemodynamic monitoring, chronic imaging, tumor-vascular interaction, and neurovascular coupling. Finally, several potential technical advances toward clinical applications are suggested, and a few technical challenges in contrast enhancement and fluence compensation are summarized.

Abstract:In recent years, photoacoustics has attracted intensive research for both anatomical and functional biomedical imaging. However, the physical interaction between photoacoustic generated endogenous waves and an exogenous ultrasound wave is a largely unexplored area. Here, we report the initial results about the interaction of photoacoustic and external ultrasound waves leading to a micro-Doppler photoacoustic (mDPA) effect, which is experimentally observed and consistently modelled. It is based on a simultaneous excitation on the target with a pulsed laser and continuous wave (CW) ultrasound. The thermoelastically induced expansion will modulate the CW ultrasound and leads to transient Doppler frequency shift. The reported mDPA effect can be described as frequency modulation of the intense CW ultrasound carrier through photoacoustic vibrations. This technique may open the possibility to sensitively detect the photoacoustic vibration in deep optically and acoustically scattering medium, avoiding acoustic distortion that exists in state-of-the-art pulsed photoacoustic imaging systems.

Abstract:Lights, Sound, Images Optical microscopy can readily image thin samples such as cells, but thicker samples, such as tissue, are more difficult to image directly, because of the multiple scattering of light. Wang and Hu (p. 1458 ) review methods for imaging biological samples on length scales ranging from organelles to whole organs that rely on the photoacoustic effect—the excitation of ultrasonic pressure waves when light is absorbed by molecules in solution. The incident light can be focused and scanned across a sample in a microscopy mode to create ultrasound images, or the entire region of interest can be illuminated and the ultrasound waves analyzed with a computer algorithm in a tomography mode. Imaging studies can reveal changes in oxygen metabolism and gene expression and in image biomarkers and vasculature.

High Quality Photoacoustic Tomography in Scattering Biological Tissue

Abstract:Photoacoustic tomography is an emerging biomedical imaging method, which has a high image contrast and good spatial resolution in deep tissues. Because of these merits, PAT has broad applications in biomedical imaging. However, acoustic scattering is unavoidable in tissue with inhomogeneous acoustic properties. Acoustic scattering brings a challenge for the applications of PAT. A new PAT method is presented to obtain the photoacoustic images in scattering tissue. This method is based on the time-reversal invariance of acoustic wave propagation during the acoustic scattering process. Both numerical simulations and experiments are used to validate this method. The results show that the proposed method can provide better PAT images in scattering tissues than the traditional photoacoustic tomography. Therefore, this method could serve as an alternative for imaging inhomogeneous biological tissues with acoustic scattering.

Abstract:Photoacoustic microscopy is a fast-developing imaging technology, which depends on the optical absorption differences and inherits high contrast of optical imaging and deeper depth of ultrasound imaging. Thus, photoacoustic microscopy presents more merit than optical microscopy. Photoacoustic microscopy realizes multiscale imaging from acoustical resolution to the optical resolution, which provides multi-parameters imaging from simple absorption structure to its function, multi-contrast imaging from endogenous contrast agents to exogenous contrast agent, and imaging from ultrasound-transducer-based to all-optical excitation and detection, and multimodality imaging combined with optical coherence tomography, fluorescence imaging, two-photon imaging, second harmonic imaging, etc. Photoacoustic microscopy is widely applied in the field of biomedicine, such as vascular biology, oncology, neurology, ophthalmology, dermatology, and so on.
