Abstract:Laser induced fluorescence (LIF) detection technique is one of the most sensitive detection method in micro total analytical systems (μTAS) as its good monochromaticity, strong collimation and high optical density. It is widely used in biochemical analysis systems and the detection limit could reach 10-9-10-12mol/L. Due to the important role in μTAS, different optical structure arrangements of LIF detection techniques were continuously developed. We reviewed the recent progress of the optical structure arrangement and the applications of the LIF detection techniques.

Abstract:Based on confocal configuration, a laser-induced fluorescence detector (LIFD) for high performance liquid chromatography (HPLC) is described and evaluated. A diode double-pumped solid-state laser emitting at 473 nm is used as excitation source. A detection cell of 7 microL physical volume and 0.1 nL effective optical volume was designed for conventional HPLC-LIFD. The angle between entrance and outlet channels of the cell was 120 degrees, without any change of internal diameters or right-angles along the channels. These measures insured the elimination of bubble accumulation or formation inside the cell, and additional band broadening effect. Riboflavin standard was used as test sample to evaluate the performance of the HPLC-LIFD. The analyses of real samples containing riboflavin were demonstrated.

Abstract:A laser induced fluorescence (LIF) detection system of transmission type optical path is described. To improve the performance of the detection system, confocal technique is used. For an ICE chip of certain size, some factors that are very important for the performance of the instrument are presented. The choice of the objective, the size of the pinhole and the intensity of the laser beam all influence SNR of the signal significantly. The diagrams, which show these relationships of these factors, are displayed.

Abstract:A highly sensitive in-column fiber-optic LIF detector for CE has been constructed and eval uated. In this detection system, a 457-nm diode-pumped solid-state blue laser was used as the excitation light source and an optical fiber (40 mu m od) was used to transmit the excitation light. One end of the optical fiber was inserted into the separation capillary and was in situ positioned at the detection window. The other end of the fiber was protruded from the capillary to capture the excitation light beam from the blue laser. Fluorescence emission was collected by a 40 x microscope objective, focused on a spatial filter, and passed through a yellow color filter before reaching the photomultiplier tube. The present CE-fluorescence detection is a simple and compact optical system. It reduces the laser scattering effect from the capillary and fiber as compared to the conventional LIF detection for CE. Its utility was successfully demonstrated by the separation and determination of D-penicillamine labeled with naphthalene-2,3-dicarboxaldehyde. The detection limit was 0.8 nM (S/N = 3). The present detection scheme has been proven to be attractive for sensitive fluorescence detection for CE.

Abstract:In this paper, we present a compact handheld laser-induced fluorescence (LIF) detector based on a 450nm laser diode and quasi-confocal optical configuration with a total size of 9.1×6.2×4.1cm3. Since there are few reports on the use of 450nm laser diode in LIF detection, especially in miniaturized LIF detector, we systematically investigated various optical arrangements suitable for the requirements of 450nm laser diode and system miniaturization, including focusing lens, filter combination, and pinhole, as well as Raman effect of water at 450nm excitation wavelength. As the result, the handheld LIF detector integrates the light source (450nm laser diode), optical circuit module (including a 450nm band-pass filter, a dichroic mirror, a collimating lens, a 525nm band-pass filter, and a 1.0mm aperture), optical detector (miniaturized photomultiplier tube), as well as electronic module (including signal recording, processing and displaying units). This detector is capable of working independently with a cost of ca. $2000 for the whole instrument. The detection limit of the instrument for sodium fluorescein solution is 0.42nM (S/N=3). The broad applicability of the present system was demonstrated in capillary electrophoresis separation of fluorescein isothiocyanate (FITC) labeled amino acids and in flow cytometry of tumor cells as an on-line LIF detector, as well as in droplet array chip analysis as a LIF scanner. We expect such a compact LIF detector could be applied in flow analysis systems as an on-line detector, and in field analysis and biosensor analysis as a portable universal LIF detector.

Abstract:Laser-induced fluorescence detector (LIFD) is one of the most sensitive detectors in analytical chemical files. Confocal optical configuration is widely used in LIFDs. Two effective approaches used to achieve the best signal to noise ratio (S/N) are increasing the confocal precision and minimizing the background noise. A novel three-dimensional adjustable confocal LIFD was developed, using a new three-dimensional adjustable supporter of reflector and modularized optical system. A detection limit (S/N = 3) of 1 x 10(-12) mol/L and a linear dynamic range of 3 orders of magnitude were obtained using fluorescein isothiocyanate (FITC) standard as the test sample. The noise level and drift levels were 8.0 x 10(-3) mV and 1.4 x 10(-3) mV/h, respectively, which were almost 10 times lower than before. And the stability of the LIFD was evaluated by five replicate injections of 5 x 10(-9) mol/L FITC, and the relative standard deviations (RSDs) of peak height and peak area were 0.38% and 0.41%, respectively. Further more, three biogenic amines, which were derivatized by FITC, were separated by high performance liquid chromatography (HPLC) and then detected by the novel LIFD. And the detection limits (S/N = 3) ranged 0.01 to 0.02 nmol/L, which were better than other methods. Therefore, the LIFD is highly sensitive, as well as shows a real low noise level and good reproducibility.

Abstract:A laser induced fluorescence detector, with a novel Z-type reflective flow cell and solid-state diode la-ser emitting at 473nm, was invented. The performance of the detector, including the sensitivity and linear dynamic range, was evaluated by FITC standards and five pre-derivatizated amines as test samples. The detection limit of FITC was 1×10-12mol/L (S/N=15) and the linear dynamic range was over 4 degree. The detection limit of amines achieved pg level and good reproducibility was observed. It was concluded that the LIFD had highly sensitivity, a wide linear dynamic range and good reproducibility.

Abstract:In many research fields, the demand for miniaturized laser-induced fluorescence (LIF) detection systems has been increasing. This work has developed a compact LIF detector, employing a laser diode as the excitation source and a photodiode as the photodetector with an adjustable laser focal spot, to meet the diverse requirements of various observation targets, such as capillaries, PCR tubes, and microfluidic chips. It features the functionalities of background fluorescence correction, the adaptive adjustment of the dynamic range, and constant power control for the laser. The influence of the excitation power on the detection limit was studied through experiments, and the configuration results for LED/LD as light sources and 487/450 nm wavelengths were compared and optimized. A fully integrated, compact, modular epifluorescence LIF detector was subsequently constructed, measuring 40 × 22 × 38 mm3 in total size, with a cost of USD 320, and achieving a detection limit of 0.4 nM for fluorescein sodium. Finally, the detector was integrated into a nucleic acid detection system with a microfluidic chip on the Chinese Space Station (CSS) and was also tested with PCR tubes and capillaries, proving its broad practicality and adaptability to various analytical systems.

Abstract:The invention discloses a laser induction fluorescence detecting machine, which is mainly used for the micro sample inspecting in the field of clinical medicine and life science. The invention comprises an excitation light path (10), a separating micro column (20) and a fluorescence light collecting path (30). The axis shaft line of the excitation light path (10) is vertical with the axis shaft line of the fluorescence light collecting path (30), and the excitation light path (10) has the same axis with the separating micro column (20), an optical fiber (40) is equipped between the excitation light path (10) and the separating micro column (20); the front end of the optical fiber (40) is coupled with the excitation light path (10), and the rear end extends to the inspecting window (21) of the separating micro column (20). The invention has the advantage that the invention has high fluorescence stimulation efficiency, The optical structure is simple, the volume is small, the fluorescence stimulation of the sample can be directly realized, the sensitivity and accuracy can be enhanced; and the invention can be used together with the micro column separating systems like capillary tube electrophoresis, capillary tube liquid chromatography and flowing injection, etc.

Abstract:There is a growing urgent requirement for miniaturized laser-induced fluorescence (LIF) detection systems in many research fields. In this work, miniaturized LIF detectors with three different optical configurations of orthogonal, confocal, and oblique were developed, using a laser diode as the excitation source and a photodiode as the photodetector. The computer simulation and experimental methods were used to investigate the distributions of laser scattered light and fluorescent light near the detection window. Other conditions including the solution preparation, sample flow rate, alignment method and filter model were also optimized. Under the optimized conditions, the detection limits of sodium fluorescein for orthogonal and confocal LIF detectors were 40 pM and 50 pM, respectively, while the limit of detection (LOD) for oblique LIF detector were 1 nM (45 degrees) and 7 nM (67.5 degrees). We further built a fully integrated handheld orthogonal LIF detector with a total size of 50 x 20 x 46 mm(3), a cost of $380, and a detection limit of 10 pM for sodium fluorescein. It is expected that such a LIF detector could be applied in field analysis as a portable instrument or in other analysis systems as a detection module.

The detector can incorporate excitation sources of different wavelength, and an on-column or a flow cell detection modes, which can meet with ahnost all flow analysis system. Abstract:In this work, a simple LIF detection system based on an orthogonal optical arrangement for microfluidic chips was developed. Highly sensitive detection was achieved by detecting the fluorescence light emitted in the microchannel through the sidewall of the chip to reduce scattered light interference from the laser source. A special crossed-channel configuration, with a 1.5-mm distance from the separation channel to the sidewall of the glass chip, was designed in order to facilitate collection of emitted fluorescence light through the sidewall. The significant difference in intensity distribution of scattered laser light on the chip plane observed in this study was fully exploited to optimize S/N ratio of detected signals by rejection of scattered light, both through systematic measurements and employing ray-tracing simulation. A fluorescence collection angle of 45 degrees in the chip plane gave the best result, with a scattered light intensity 1/38 of that obtained at an angle of 90 degrees. Sodium fluorescein and fluorescein isothiocyanate-labeled amino acids were used as model samples to demonstrate the performance of the LIF system. A detection limit (S/N = 3) of 1.1 pM fluorescein was obtained, which is comparable to that of optimized confocal LIF systems for chip-based capillary electrophoresis. Apart from the high detection power, the system also has the advantages of simple optical structure, compactness, and ease in building.

Abstract:A wall-free detection method based on liquid junction in a capillary gap was proposed for laser-induced fluorescence (LIF) of capillary electrophoresis (CE). The capillary gap of the wall-free cell was fabricated by etching a 10-mm x 50-microm I.D. fused-silica capillary to obtain a polyimide coating sleeve, decoating about 6mm at one end of both 50 microm I.D. separation and liquid junction capillary, inserting the treated capillary ends into the coating sleeve oppositely, fixing the capillaries with a gap distance of 140 microm by epoxy glue and removing the coating sleeve by burning. The theoretical model, experimental results and wall-free cell images indicated that the gap distance and applied voltage were main influence factors on the wall-free detection. Since the wall-free cell increased the absorption light path and avoided the stray light from the capillary wall, it improved the ratio of signal to noise and limit of detection (LOD) of CE-LIF. Three flavin compounds of riboflavin (RF), flavin mononucleotide sodium (FMN) and flavin adenine dinucleotide disodium (FAD) were used to evaluate the wall-free detection method. Compared with on-column cell, the LODs of the wall-free cell were improved 15-, 6- and 9-fold for RF, FMN and FAD, respectively. The linear calibration concentrations of the flavins ranged from 0.005 to 5.0 micromol/L. The column efficiency was in the range from 1.0 x 10(5) to 2.5 x 10(5) plates. The wall-free detection of CE-LIF was applied to the analysis of the flavins in spinach and lettuce leaves.

Abstract:A 64-channel detection system for laser-induced fluorescence (LIF) detection at the cell level is established and applied to single event counting. Generally, fluorescence detection at the cellular level requires a dyeing label to enhance the scattered light intensity for the photodetector. However, the dyeing labels, such as fluorophores, probes, and dyes, complicate sample preparation and increase cytotoxicity in the process. Therefore, label-free detection becomes essential for in vivo research. The presented 64-channel detection system is designed for label-free detection with the ability to record feeble emissions. Two linear photodetector devices are included in the system, extending the wavelength detection range to 366-680 nm with an improved spectral resolution at an average of 4.9 nm. The performance of the system was validated by detecting unlabeled human hepatocytes (L-02) and other cell-level biologic samples. In addition, the 64-channel detection system was also used for particle counting with a quartz microfluidic chip. The counting method is based on fluorescence spectra differs from those of other devices (i.e., flow cytometry and cell-sorting equipment), which use isolated irradiance intensities.

Abstract:For laser-induced fluorescence (LIF), the SNR and related concentration detectability was severely limited by background signal (noise). Herein, an axial-excitation LIF based on pinhole metal-capillary (PMC) and direct laser-diode excitation was proposed. An overall ~480 fold improvement on SNR was realized, because the laser-sidewall interaction (LSI) and related background signal (noise) can be effectively suppressed in PMC by avoidance of laser leakage and laser contact with capillary sidewall. For rhodamine B detection without sample enrichment, a detection limit of concentration (DLC) of 0.4 pM was obtained, which is 2.5 fold lower than the DLC (1 pM) of commercial LIFs equipped with bulky and expensive Ar+ laser. For selective detection of Cu2+, a DLC of 5 pM was realized, which is more than 40 fold lower than that of previous fluorimetry detection with similar quantum-dot probe. The PMC-LIF features compact (16 × 4 × 4 cm3), low-cost and high concentration detectability.

Abstract:A novel miniaturized, integrated whole- column imaging detection (WCID) system on a microchip is presented. In this system, a program controlled organic light emitting diode (OLED) array was used as a spatial-scanning light source, to achieve imaging by the time sequence of the excited fluorescence. By this mechanism, a photomultiplier tube (PMT) instead of a charge coupled detector (CCD) can be applied to the imaging. Unlike conventional systems, no lenses, fibers or any mechanical components are required either. The novel flat light source provides uniform excitation light without size limitations and outputs a stronger power by pulse driving. The scanning mode greatly reduced the power consumption of the light source, which is valuable for a portable system. Meanwhile, this novel simplified system has a broader linear range, higher sensitivity and higher efficiency in data collection. Isoelectric focusing of R- phycoerythrin (PE) and monitoring of the overall process with WCID were performed on this system. The limit of detection (LOD) was 38 ng mL(-1) or 3.2 pg at 85 nL per column injection of PE. The system provides a technique for WCID capillary isoelectric focusing (cIEF) on chip and can be used for throughput analysis.
