Abstract:Coumarin is bioactive scaffold found both in natural and synthetic compounds including drugs such as warfarin, hymecromone, and novobiocin. Starting from 2‐(7‐hydroxy‐2‐oxo‐2H‐chromen‐4‐yl)acetic acid, a derivative of hymecromone, we synthesized new coumarin derivatives through amide linkage with various substituted anilines and 2‐aminopyridines. Among these, the para bromo substituted 6b inhibited strongly MCF‐7 3D tumor sphere formation thus represents attractive lead for further optimization and mechanistic study. Nine new coumarin derivatives of 2‐(7‐hydroxy‐2‐oxo‐2H‐chromen‐4‐yl)acetic acid 6a–i have been successfully synthesized through amide coupling with various substituted anilines and 2‐aminopyridines. The synthesized derivatives were screened for anticancer activity using MTT assay on MCF‐7 cell line. The results showed that compound 6b inhibited MCF‐7 cell growth in a dose‐dependent manner and reached 47% inhibition at 40 μM, being better than starting material 5 and references 3 and 5FU drug. Moreover, 6b inhibited significantly 3D tumorsphere formation. The para‐Br substitution of 6b seems to be important for activity. Docking study suggests estrogen receptor and/or 3a‐HSD type 3 protein could be the target(s) for anticancer activity of this class of compounds. Further optimization of compound 6b should lead to more potent compound.

Abstract:This paper reviewed the advances on effective constituents and biological activities of coumarins in recent ten years. Coumarins are a group of important natural compounds, and have been found to have multi-biological activities such as anti-HIV, anti-tumor, anti-hypertension, anti-arrhythmia, anti-osteoporosis, assuaging pain, preventing asthma and antisepsis. Therefore, further investigation should emphasize on improving techniques for extraction and separation, searching the effective precursory compound, and synthesizing and screening out courmarin derivatives with high activity and low toxicity.

Abstract:Based on the pharmaceutical potentials of coumarins, which have antitumor activity, we synthesized new coumarin derivatives and evaluated their biological activities. The new coumarin derivatives were chemically synthesized from 4-hydroxycoumarin, and their structures were confirmed by nuclear magnetic resonance data. Ten of the synthesized compounds were investigated for antimetastatic activity against lung carcinoma cells. Several of the tested compounds showed good to mild inhibitory effects on lung cancer cell motility. There were no cytotoxic effects related to the use of these compounds. 4-Hydroxycoumarin derivatives, 4h and 4i, elicited the significant inhibitory effect on lung cancer cell motility by suppressing expression of the epithelial-mesenchymal transition markers N-cadherin, Snail, and Twist.

Abstract:Many organic and medicinal chemists are interested in the coumarin compounds due to their potential for use in pharmaceuticals and other industries. Coumarin compounds are a significant class of biologically active chemicals. Here, we outline the steps forproducing coumarin derivatives quickly and effectively from commercially accessible substrates via multicomponent condensation processes involving pyrone and benzene derivatives. These substances were examined for their potential anti-tumor effects, including their ability to block carbonic anhydrase, activate the cell apoptosis protein, and coumarin and its derivatives are the primary oral anticoagulants. Candida albicans, Aspergillus fumigatus, and Fusarium solani are three distinct fungus species that are resistant to the antifungal properties of synthetic coumarins. Long known are the coumarins' antibacterial due to its biological activities against some kinds of bacteria such as Zeitschrift für Naturforschung C, as well as their antifilarial, antiulcerogenic, anti-inflammatory, and antioxidant effects for its capacity to inhibit their growth activities. Coumarin has long been valued as a major raw element in the fragrance sector due to its distinct sweet aroma and stability. When mixed with organic essential oils like lavender, citrus, rosemary, and oak moss, it is used as an odour-enhancer to produce a long-lasting impact. It has been utilized as a bleaching or brightening agent in detergents when it revealed a remarkable brightening effect with excellent overall stability on fibers of synthetic origin. Materials such as liquid crystalline, organic/inorganic composites, as well as light harvesting, energy transferring compounds, and electro-optics materials are all employed as a dye in organic photo-redox catalysis and as potent photo-sensitizers reagent. These materials polymerize epoxy-silicones, under near-UV and visible light. Coumarin has also found useful as metal-free sensitizers for solar cells.

Abstract:A series of coumarin derivatives (A01-A13) have been designed and synthesized,and their biological activities were evaluated in vitro in 5 different cell lines:Liver hepatocellular cells (HepG2),breast cancer cells (MCF-7),human malignant melanoma cells (A375),human colonic carcinoma (HT-29) and non-small cell lung cancer cells (A549).Furthermore,A05 was screened in kinase sets and the results showed that the possible targets of it were MST3/STK24 and TNIK.Compounds A01 and A05 displayed the most potent antiproliferative activities in vitro,which suggested that the picolinamide moiety was important for antiproliferative activities.Compound A05 was screened against a set of kinases and the results indicated the kinases MST3/STK24 and TNIK as possible targets.
The angiogenesis-modulating effects of coumarin-derivatives

Abstract:Coumarin is a natural compound that is rich in plants. Coumarin and its derivates were reported to have many biological activities, such as anti-bacterial, anti-tumor, and anti-coagulation. In this study, we examined the angiogenic modulating activities of six previously synthesized coumarin derivatives (Compound #1-#6) in zebrafish embryos and further confirmed them in a chick model. According to the survival rate in a zebrafish model, Compound #1 (100 %), #2 (82.5-100 %), and #4 (100 %) showed much less toxicity than Compound #3 (19.2-100 %), #5 (0-100 %), and #6 (0-100 %). Using a green blood vessel fluorescent transgenic fish Tg(fli1:egfp) to record the angiogenesis-modulating effects of Compound #1, #2, and #4, we found that Compound #2 had the highest effects in interfering intersegmental vessel growth, subintestinal vein growth, and caudal vein plexus remodeling. Chick chorioallantoic membrane (CAM) assay also showed that Compound #2 exposure led to a reduction of blood vessel growth. Real-time PCR experiments revealed that Compound #2 significantly changed the expression of vascular growth-related genes flt1, cdh5, and nrp1a in zebrafish. Based on our data from zebrafish and chick models, a new coumarin-derivative (Compound #2) possesses anti-angiogenic activity with low toxicity, but further investigation in mammal models is asked to confirm our findings.

Abstract:New pyranocoumarin and coumarin-sulfonamide derivatives were prepared and evaluated for their antioxidant, antimicrobial, and/or anti-inflammatory activities. Coumarin-sulfonamide compounds 8a–d demonstrated significant antioxidant activity, while 7c,d, 8c,d, and 9c,d exhibited antimicrobial activity equal to or higher than the standard antimicrobials against at least one tested microorganism. Regarding the anti-inflammatory testing, pyranocoumarins 2b, 3a,b and 5c and coumarin-sulfonamide compound 9a showed more potent antiproteinase activity than aspirin in vitro; however, five compounds were as potent as aspirin. The anti-inflammatory activity of the promising compounds was further assessed pharmacologically on formaldehyde-induced rat paw oedema and showed significant inhibition of oedema. For in vitro COX-inhibitory activity of coumarin derivatives, pyranocoumarin derivative 5a was the most selective (SI = 152) and coumarin-sulfonamide derivative 8d was most active toward COX-2 isozyme. The most active derivatives met the in silico criteria for orally active drugs; thus, they may serve as promising candidates to develop more potent and highly efficient antioxidant, antimicrobial, and/or anti-inflammatory agents.

Abstract:In this study, several novel coumarin derivatives, 7-hydroxy-2-oxo-2H-chromene-3-carboxyl-Trp-Trp-AA-OBzl compounds, were designed and synthesized as potential anticancer agents. Their in vitro cytotoxic activities were evaluated using methylthiazoltetrazolium (MTT) assay. The anti-tumor activity of the newly coumarin derivatives was determined in a S180 bearing mouse model and some of the compounds demonstrated tumor growth inhibition similar to the positive control, doxorubicin. Compared to doxorubicin, most of the compounds exhibited enhanced immunologic function suggesting a relatively minor toxic effect. The intercalation of the coumarin derivatives synthesized with calf thymus (CT) DNA was also studied.

Abstract:Coumarin derivatives are an important class of C6-C3 plant metabolites that show a variety of bioactivities. Currently, most clinical anticoagulant agents are coumarins, such as warfarin, dicoumarol and acenocoumarol, and patients taking these drugs must be monitored for adverse reactions. In a search for safe and effective anticoagulant compounds from Chinese herbal medicine, a screening procedure on the whole plant of Ainsliaea fragrans was performed. The phytochemical investigation of this plant afforded five new coumarin derivatives, including a pair of natural 4-hydroxycoumarin enantiomers (1), a pair of coumarin enantiomers with a rare polycyclic pyrano[3-2c] carbon skeleton (2) and a 7-hydroxycoumarin derivative (3), together with 5 known biogenetically related compounds (4-8). Enantioseparation of 1 and 2 produced optically pure compounds 1a, 1b, 2a and 2b. The absolute configurations of the new compounds were confirmed by single-crystal X-ray diffraction analysis. In addition, we evaluated the anticoagulant activity of all isolates via activated partial thromboplastin time (APTT), thrombin time (TT) and prothrombin time (PT) assays in vitro and in vivo. Of note, compound 3 displayed potent anticoagulant activity and no significant hepatic or renal toxicity, which could make it a promising agent for further preclinical evaluation for preventing abnormal blood clotting.

Abstract:The structural motif of coumarins is related with various biological activities and pharmacological properties. Both natural coumarin extracted from various plants or a new coumarin derivative synthesized by modification of the basic structure of coumarin, in vitro experiments showed that coumarins are a promising class of anti-tumor drugs with high selectivity. Cancer is a complex and multifaceted group of diseases characterized by the uncontrolled and abnormal growth of cells in the body. This review focuses on the anticancer mechanism of various coumarins synthesized and isolated in more than a decade. Isopentenyloxycoumarins inhibit angiogenesis by reducing CCl2 chemokine levels. Ferulin C is a potent colchicine-binding agent that destabilizes microtubules, exhibiting antiproliferative and anti-metastatic effects in breast cancer cells through PAK1 and PAK2-mediated signaling. Trimers of triphenylethylene-coumarin hybrids demonstrate significant proliferation inhibition in Hela, A549, K562, and MCF-7 cell lines. Platinum(IV) complexes with 4-hydroxycoumarin have the potential for high genotoxicity against tumor cells, inducing apoptosis in SKOV-3 cells by up-regulating caspase3 and caspase9 expression. Derivatives of 3-benzyl coumarin seco-B-ring induce apoptosis, mediated through the PI3K/Akt/mTOR signaling pathway. Sesquiterpene coumarins inhibit the efflux pump of multidrug resistance-associated protein. Coumarin imidazolyl derivatives inhibit the aromatase enzyme, a major contributor to estrogen overproduction in estrogen-dependent breast cancer.
