Abstract:Coumarins are an important class of natural compounds found in different species of plants in nature.Experimental studies have found that coumarins have multi-biological activities such as anti-HIV,anti-tumor,anti-oxidation,anti-flammatory and other pharmacological activities,which are widely applied to anti-coagulation and lymphatic edema in clinic.In recent years,studies have found that coumarins have clear toxic effects with species and site-specific in rodents,which is related to metabolic pathways and CYP2A6 polymorphisms.In addition,toxic effects are closely related to dose and route of administration,and oral and high dose are more prone to toxicity.This paper reviews the advances in pharmacology and toxicology of coumarin and its derivatives,with a view to the development of coumarins and clinical applications.

Abstract:Coumarin compounds represent an important type of naturally occurring and synthetic oxygen-containing heterocycles with typical benzopyrone framework. This type of special benzopyrone structure enables its derivatives readily interact with a diversity of enzymes and receptors in organisms through weak bond interactions, thereby exhibit wide potentiality as medicinal drugs. So far, some coumarin-based drugs such as anticoagulant and antineurodegenerative agents have been extensively used in clinic. Coumarin-containing supramolecular medicinal agents as a new increasing expansion of supramolecular chemistry in pharmaceutical science have also been actively investigated in recent years. Coumarin-derived artificial ion receptors, fluorescent probes and biological stains are growing quickly and have a variety of potential applications in monitoring timely enzyme activity, complex biological events as well as accurate pharmacological and pharmacokinetic properties. This review provides a systematic summary and insight of the whole range of medicinal chemistry in the current developments of coumarin compounds as anticoagulant, antineurodegenerative, anticancer, antioxidative, antibacterial, antifungal, antiviral, antiparasitic, antiinflammatory and analgesic, antidiabetic, antidepressive and other bioactive agents as well as supramolecular medicinal drugs, diagnostic agents and pathologic probes, and biological stains. Some rational design strategies, structure-activity relationships and action mechanisms are discussed. The perspectives of the future development of coumarinbased medicinal chemistry are also presented.

Abstract:Plants produce and release a vast variety of secondary metabolites with diversified functions, and possess ecological, toxicological and biological effects that mimic the activities of synthetic chemicals. Coumarins extracted from bacteria, fungi and many edible plants are widely used for their antibacterial, antifungal, antiviral, anti-HIV and anticancer activities. This review presents a concise summary based on the latest knowledge of the biological and pharmaceutical uses of coumarin and its derivatives, including an evaluation of future therapeutic potential. The presence of coumarins in various plant organs like fruits, seeds, roots, leaves and latex supplement recent work reported in scientific literature related to these compounds and their development. Due to low production in plants, the upscaling and industrial scale production, commercialization and industry demand of coumarins has faced hurdles. We searched Google Scholar and Web of Science for relevant literature upto 2018 using the keywords pharmaceutical, biological activities and coumarins. This review has thoroughly overviewed the related facts and figures about coumarins and its derivatives, especially in terms of biological and pharmaceutical properties including anti-microbial, anti-viral, anti-diabetic, anticoagulant, estrogenic, dermal photosensitizing, vasodilator, molluscacidal, antithelmintic, sedative and hypnotic, analgesic, hypothermic, anti-cancer activity, anti-oxidant, anti-parasitic, antihelmintic, anti-proliferative, anti-convulsant, anti-inflammatory, and anti-hypertensive activities. The pharmaceutical impact of coumarins on public health is a complex phenomenon, with several questions in relation to safety during medical therapies and consumption through diet. The present review focuses on coumarin clinical studies in the treatment of various chronic diseases such as cancer, diabetes, depression, Alzheimer's, Parkinson's and HIV. However, further research and review are necessary to broaden the therapeutic effectiveness of coumarin in patients suffering from such ailments.

Abstract:Many naturally occurring substances, traditionally used in popular medicines around the world, contain the coumarin moiety. Coumarin represents a privileged scaffold for medicinal chemists, because of its peculiar physicochemical features, and the versatile and easy synthetic transformation into a large variety of functionalized coumarins. As a consequence, a huge number of coumarin derivatives have been designed, synthesized, and tested to address many pharmacological targets in a selective way, e.g., selective enzyme inhibitors, and more recently, a number of selected targets (multitarget ligands) involved in multifactorial diseases, such as Alzheimer’s and Parkinson’s diseases. In this review an overview of the most recent synthetic pathways leading to mono- and polyfunctionalized coumarins will be presented, along with the main biological pathways of their biosynthesis and metabolic transformations. The many existing and recent reviews in the field prompted us to make some drastic selections, and therefore, the review is focused on monoamine oxidase, cholinesterase, and aromatase inhibitors, and on multitarget coumarins acting on selected targets of neurodegenerative diseases.

Abstract:Background: Coumarins, abundantly distributed in a plethora of biologically active compounds, serve as a fundamental motif in numerous natural products, drugs, and therapeutic leads. Despite their small size, they exhibit a diverse range of biological activities, intriguing researchers with their immense pharmacological potential. Purpose: This study consolidates the evidence regarding the essential role of coumarins in modern drug discovery, exploring their broad-spectrum pharmaceutical effects, structural versatility, and mechanisms of action across various domains. Methods: For literature search, we utilized PubMed, Google scholar, and SciFinder databases. Keyword and keyword combinations such as "coumarins", "natural coumarins", "specific natural coumarins for particular diseases", and "therapeutic effects" were employed to retrieve relevant studies. The search encompassed articles published between 2005 and 2023. Selection criteria included studies reporting on the pharmacological activities of natural coumarins against various diseases. Results: The results highlight the therapeutic potential of natural coumarins against various diseases, demonstrating anti-cancer, anti-oxidant, and anti-inflammatory activities. They also act as monoamine oxidase inhibitors and phosphodiesterase inhibitors, and as anti-thrombotic, anti-diabetic, and hepatoprotective agents. They also show efficacy against diabetic nephropathy, neurodegenerative diseases, microbial infections and many other diseases. Conclusion: This review underscores the significant role of natural coumarins in medicinal chemistry and drug discovery. Their diverse biological activities and structural versatility make them promising therapeutic agents. This study serves as a catalyst for further research in the field, aiming to address emerging challenges and opportunities in drug development.

Abstract:This comprehensive review, covering 2021–2023, explores the multifaceted chemical and pharmacological potential of coumarins, emphasizing their significance as versatile natural derivatives in medicinal chemistry. The synthesis and functionalization of coumarins have advanced with innovative strategies. This enabled the incorporation of diverse functional fragments or the construction of supplementary cyclic architectures, thereby the biological and physico-chemical properties of the compounds obtained were enhanced. The unique chemical structure of coumarine facilitates binding to various targets through hydrophobic interactions pi-stacking, hydrogen bonding, and dipole-dipole interactions. Therefore, this important scaffold exhibits promising applications in uncountable fields of medicinal chemistry (e.g., neurodegenerative diseases, cancer, inflammation).

Abstract:Many organic and medicinal chemists are interested in the coumarin compounds due to their potential for use in pharmaceuticals and other industries. Coumarin compounds are a significant class of biologically active chemicals. Here, we outline the steps forproducing coumarin derivatives quickly and effectively from commercially accessible substrates via multicomponent condensation processes involving pyrone and benzene derivatives. These substances were examined for their potential anti-tumor effects, including their ability to block carbonic anhydrase, activate the cell apoptosis protein, and coumarin and its derivatives are the primary oral anticoagulants. Candida albicans, Aspergillus fumigatus, and Fusarium solani are three distinct fungus species that are resistant to the antifungal properties of synthetic coumarins. Long known are the coumarins' antibacterial due to its biological activities against some kinds of bacteria such as Zeitschrift für Naturforschung C, as well as their antifilarial, antiulcerogenic, anti-inflammatory, and antioxidant effects for its capacity to inhibit their growth activities. Coumarin has long been valued as a major raw element in the fragrance sector due to its distinct sweet aroma and stability. When mixed with organic essential oils like lavender, citrus, rosemary, and oak moss, it is used as an odour-enhancer to produce a long-lasting impact. It has been utilized as a bleaching or brightening agent in detergents when it revealed a remarkable brightening effect with excellent overall stability on fibers of synthetic origin. Materials such as liquid crystalline, organic/inorganic composites, as well as light harvesting, energy transferring compounds, and electro-optics materials are all employed as a dye in organic photo-redox catalysis and as potent photo-sensitizers reagent. Abstract:Coumarin (2H-1-benzopyran-2-one) presents the fundamental structure of an enormous class of biologically active compounds of natural, semi-synthetic, and synthetic origin. Extensive efforts are continually being put into the research and development of coumarin derivatives with medicinal properties by the broad scientific community. Transition metal coordination compounds with potential biological activity are a "hot topic" in the modern search for novel drugs. Complexation with transition metals can enhance the physiological effect of a molecule, modify its safety profile, and even imbue it with novel attributes of interest in the fields of medicine and pharmacy. The present review aims to inform the reader of the latest developments in the search for coumarin transition metal complexes with biological activity, their potential applications, and structure-activity relationships, where such can be elucidated. Abstract:Introduction: Coumarins are naturally occuring metabolites from plants and a few micro-organisms. They have been widely used in the food and drug industry in their natural or synthetic forms. Numerous coumarins possess several biological activities such as anti-inflammatory, anti-ulcers, anti-tumour, anti-microbial, anti-coagulant. The aim of this study was to assess the bioactivity, and toxicity of coumarins from African medicinal plants. Methods: We searched online databases and search engines such as PubMed, Google Scholar and Web of Science for key terms such as coumarins, toxicity, bioavailability, bioactivity with appropriate Boolean operators. Only full-length research articles published in English between 1956 to 2023 were reviewed. Results: We recorded 22 coumarins from 15 plant species from Africa. Most of the plant species (33%) were from North Africa. These were followed by East Africa at 21%, then West, and Central Africa at 18.2% each. Most of the coumarins (21.3%) were isolated from the entire plant and the leaves (19.1%) and most of them (46.7%) had some antimicrobial activity. Five coumarins viz osthole, pseudocordatolide C & calanolide, chartreusin and esculetin had either antitumor or anticancer activity. Six coumarins had varying levels and types of toxicity ranging from inhibiting blood clotting as anticoagulants, to cytotoxic effects, causing hyperventilation, tremor, & photophobia, pulmonary haemorrhage, carcinogenic activity, severe neurotoxicity, hepato- and phototoxicity. Conclusion: Several African medicinal plants are sources of various coumarins that possess several biological activities as well as toxicities. This calls for more research into their safety and efficacy because of their wide spread applications as therapeutic agents.

Abstract:The accumulation of pyranocoumarins, the main active ingredient of Peucedanum praeruptorum Dunn, is strictly regulated by key enzyme genes in the coumarin biosynthesis pathway. However, little is understood about the correlation between the expression of key enzyme genes and the accumulation of coumarin content at various growth and development phases. In this work, a high-performance liquid chromatographic method and the real-time quantitative PCR system were prepared to establish the pyranocoumarins contents and the key enzyme expression in the roots, stems, and leaves of Peucedanum praeruptorum Dunn. The results showed that the coumarin content in the roots was the highest before blotting and was the lowest after flowering. Particularly, the content of Praeruptorin A, Praeruptorin B and Praeruptorin E was reduced to the lowest in roots after flowering. Consistent with the variation trend of the coumarin content in the stem, the relative expression of most key enzyme genes in the coumarin synthesis pathway was also significantly decreased. Especially for PpBMT gene, the relative expression in roots were extremely significantly decreased up to 90%. However, the variation trend of the coumarin content and the key enzyme genes expression was opposite in the roots and leaves. These results contribute to our knowledge of the process by which important enzymes control the production and accumulation of coumarin.

Abstract:<p>Infections caused by Gram‐positive and ‐negative bacteria are one of the foremost causes of morbidity and mortality globally. Antibiotics are the mainstay of therapy for bacterial infections, but the emergence and wide spread of drug‐resistant pathogens have already become a huge issue for public healthcare systems. The coumarin moiety, which is ubiquitous in nature, could bind to the B subunit of DNA gyrase in bacteria and inhibit DNA supercoiling by blocking the ATPase activity; hence, coumarin derivatives possess potential antibacterial activity. Several coumarin‐containing hybrids such as coumermycin A1, clorobiocin, and novobiocin have already been used in clinical practice for the treatment of various bacterial infections; thus, it is conceivable that hybridization of the coumarin moiety with other antibacterial pharmacophores may provide opportunities for the development of novel antibiotics. This review outlines the advances in coumarin‐containing hybrids with antibacterial potential in the recent 5 years and the structure–activity relationships are also discussed.</p>

Abstract:Phytocoumarins exhibit a range of biological activities, including antibacterial, anti-inflammatory, antiradical, oncolytic, and reduction of monoamine oxidase B effects. These compounds are frequently used by researchers to develop novel, entirely or partially laboratory-made pharmaceutical medicines derived from coumarin. Several of these medications are hybrid chemicals with different pharmacological effects that were purposefully created by applying the molecular hybridization concept. The combinations of compounds exhibit multifunctional characteristics, making them attractive options for developing therapies against complex diseases like cancer, Alzheimer's disease, dysmetabolic syndrome, AIDS, plasmodium infection, and cardiovascular disorders. This study summarizes findings on the development of various coumarin hybrids, groups them based on prospective therapeutic uses, and makes recommendations about possible structure–activity relationships. We searched a number of database servers, including PubMed, Scopus, Google Scholar, Web of Science, and others. Following a sieve, we eventually found and incorporated 45 relevant papers that were released between 2004 and the beginning of 2024. In order to help medicinal chemists cure a range of human ailments, the authors set out to create and produce highly efficient, tailored coumarin hybrid molecules.
