Abstract:Coumarins are a specific type of secondary metabolite that can be found in many plants. These compounds are predominantly produced through the phenylpropanoid pathway. Coumarins have been proven to possess a range of biological activities, including antimicrobial properties and antioxidant functions that aid in plant disease resistance response. The antimicrobial effect of coumarins is achieved through various mechanisms. They disrupt the cell membranes of pathogens, inhibit enzymatic activity, and hinder nucleic acid synthesis. Additionally, coumarins stimulate plant defense responses by triggering the production of reactive oxygen species (ROS) and activating the expression of immunity-related genes and signaling pathways such as the salicylic acid-dependent pathway. Due to their crucial role in defense mechanisms, coumarins can be effectively used in sustainable agriculture practices that emphasize environmentally friendly integrated pest management strategies. By providing a comprehensive overview of the biosynthetic pathways, mode of action, and application of coumarins in plant defense, this review aims to highlight the potential importance of coumarins in developing safe and sustainable crop protection strategies.

Abstract:Abstract Coumarins are a family of plant-derived secondary metabolites that are produced via the phenylpropanoid pathway. In the past decade, coumarins have emerged as iron-mobilizing compounds that are secreted by plant roots and aid in iron uptake from iron-deprived soils. Members of the coumarin family are found in many plant species. Besides their role in iron uptake, coumarins have been extensively studied for their potential to fight infections in both plants and animals. Coumarin activities range from antimicrobial and antiviral to anticoagulant and anticancer. In recent years, studies in the model plant species tobacco and Arabidopsis have significantly increased our understanding of coumarin biosynthesis, accumulation, secretion, chemical modification and their modes of action against plant pathogens. Here, we review current knowledge on coumarins in different plant species. We focus on simple coumarins and provide an overview on their biosynthesis and role in environmental stress responses, with special attention for the recently discovered semiochemical role of coumarins in aboveground and belowground plant–microbe interactions and the assembly of the root microbiome.

Abstract:Coumarins are natural products with benzopyran ring as the parent nucleus. Numerous coumarin derivatives exhibit a variety of pharmacological activities, including antibacterial, anti-inflammatory, antitumor, anti-coagulant, anti-osteoporotic, and insecticidal activities. Therefore, they play an important role in both medicine and agriculture. The development and utilization of coumarin derivatives have attracted increasing attention. The advancement of gene sequencing technology and the rapid progress in synthetic bio-logy have led to significant advancement in the biosynthesis of coumarin derivatives, and has received increasing attention from global researchers. This paper presents a comprehensive overview of the key biosynthesis-related enzymes of coumarin derivatives, such as cytochrome P450 enzyme(CYP450), prenyltransferase(PT), UDP-glucosyltransferase(UGT). Additionally, the pharmacological activities of these enzymes, including anti-tumor, anti-inflammatory, antioxidant, and antibacterial activities, are systematically summarized. This review aims to provide a valuable reference for the biosynthesis of coumarin derivatives and further exploration of their medicinal potential.

Abstract:Coumarins are naturally derived compounds fused benzene and α-pyrone rings which are found in large range of plant sources.Recently,several studies have been conducted on the biological activity of coumarins,such as anticancers,anti-inflammatory,anticoagulants,antibacterial and antifungals.In this study,the advances on the antibacterial activity of coumarins were summarized.The main target in medicine is Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa,β-hemolytic streptococcus,Haemophilus infuenzae and Vibrio cholera,etc.,in the agricultural areas are concentrated in Ralstonia solanacearum,Phytophthora capsici,Powdery mildew,Botrytis cinerea and Fusarium graminearum,etc.Indeed,the structure-activity relationship and mechanism action of coumarins were analyzed.Coumarins have multiple substitution sites and can used as a kind of lead compound to synthesize more effective coumarin derivatives with widely application values.
Effects of Coumarin on Rhizosphere Microbiome and Metabolome of Lolium multiflorum

Abstract:Rhizosphere microorganisms can help plants absorb nutrients, coordinate their growth, and improve their environmental adaptability. Coumarin can act as a signaling molecule that regulates the interaction between commensals, pathogens, and plants. In this study, we elucidate the effect of coumarin on plant root microorganisms. To provide a theoretical basis for the development of coumarin-derived compounds as biological pesticides, we determined the effect of coumarin on the root secondary metabolism and rhizosphere microbial community of annual ryegrass (Lolium multiflorum Lam.). We observed that a 200 mg/kg coumarin treatment had a negligible effect on the rhizosphere soil bacterial species of the annual ryegrass rhizosphere, though it exhibited a significant effect on the abundance of bacteria in the rhizospheric microbial community. Under coumarin-induced allelopathic stress, annual ryegrass can stimulate the colonization of beneficial flora in the root rhizosphere; however, certain pathogenic bacteria, such as Aquicella species, also multiply in large numbers in such conditions, which may be one of the main reasons for a sharp decline in the annual ryegrass biomass production. Further, metabolomics analysis revealed that the 200 mg/kg coumarin treatment triggered the accumulation of a total of 351 metabolites, of which 284 were found to be significantly upregulated, while 67 metabolites were significantly downregulated in the T200 group (treated with 200 mg/kg coumarin) compared to the CK group (control group) (p < 0.05). Further, the differentially expressed metabolites were primarily associated with 20 metabolic pathways, including phenylpropanoid biosynthesis, flavonoid biosynthesis, glutathione metabolism, etc. We found significant alterations in the phenylpropanoid biosynthesis and purine metabolism pathways (p < 0.05). In addition, there were significant differences between the rhizosphere soil bacterial community and root metabolites. Furthermore, changes in the bacterial abundance disrupted the balance of the rhizosphere micro-ecosystem and indirectly regulated the level of root metabolites. The current study paves the way towards comprehensively understanding the specific relationship between the root metabolite levels and the abundance of the rhizosphere microbial community.

Abstract:Coumarins are an important class of natural compounds found in different species of plants in nature.Experimental studies have found that coumarins have multi-biological activities such as anti-HIV,anti-tumor,anti-oxidation,anti-flammatory and other pharmacological activities,which are widely applied to anti-coagulation and lymphatic edema in clinic.In recent years,studies have found that coumarins have clear toxic effects with species and site-specific in rodents,which is related to metabolic pathways and CYP2A6 polymorphisms.In addition,toxic effects are closely related to dose and route of administration,and oral and high dose are more prone to toxicity.This paper reviews the advances in pharmacology and toxicology of coumarin and its derivatives,with a view to the development of coumarins and clinical applications.

Abstract:Coumarin (2H-1-benzopyran-2-one) is a phenolic compound derived from the shikimate pathway and synthesized by various medicinal and aromatic plants as parent molecule of a large group of secondary metabolites, namely coumarins. Its main utilization is as fixative in perfumes and flavour enhancer. Given its role as phytoalexin and phagodepression activity, herein we evaluated for the first time its efficacy against several insect species: the green peach aphid, Myzus persicae, the moth Spodoptera littoralis, the housefly, Musca domestica and the filariasis vector Culex quinquefasciatus. Two non-target species were also included in our toxicity evaluation experiments: the ladybug Harmonia axyridis and the earthworm Eisenia fetida. Results highlighted remarkable selectivity of coumarin, being highly toxic to M. persicae aphids (LC50(90) values of 1.3(1.9) mg L−1) and friendly to natural enemies of aphids as well as soil invertebrates.

Abstract:Quorum sensing (QS) is a bacterial cell-to-cell communication mechanism that plays an essential role in bacterial pathogenesis. QS governs bacterial behavior and controls biofilm formation, which in turn contributes to antibiotic resistance. Therefore, identifying and synthesizing novel compounds to overcome QS and inhibit biofilm formation are essential. Coumarins are important plant-derived natural products with wide-ranging bioactivities and extensive applications, including antibacterial, antifungal, anticoagulant, antioxidant, anticancer, and anti-inflammatory properties. Additionally, coumarins are capable of QS rewiring and biofilm formation inhibition, leading to higher susceptibility to antimicrobial agents and less antibiotic resistance. Therefore, in this review, we aim to provide an overview of QS and biofilm formation. This review also discusses the role of natural and synthesized coumarins in controlling QS, inhibiting biofilm formation, and inducing synergy in antibiotic–coumarin combinations. Hence, this review emphasizes the potential of coumarin compounds to act as antibacterial agents and demonstrates their ability to alleviate antibiotic resistance.

Abstract:Coumarin is a natural product known for its diverse biological activities. While its antifungal properties in agricultural chemistry have been extensively studied, there is limited research on its antibacterial potential. In this study, we developed several novel coumarin derivatives by combining coumarin with pyridinium salt through molecular hybridization and chemical synthesis. Our findings reveal that most of these derivatives exhibit promising antibacterial activity. Among them, derivative A25 has been identified as the most effective compound based on three-dimensional quantitative structure-activity relationships. It demonstrates significant in vitro and in vivo activity against Xanthomonas oryzae pv. oryzae (Xoo), Xanthomonas oryzae pv. oryzicola (Xoc), and Xanthomonas campestris pv. citri (Xac), outperforming the commercially available thiediazole copper. Initial investigations into its mechanism of action suggest that A25 disrupts the cell membranes of Xoc and Xoo, thereby inhibiting bacterial growth. Additionally, A25 enhances the activity of defense enzymes in rice and modulates the expression of proteins related to the pyruvate metabolism pathway. This dual action contributes to rice's resistance against bacterial infestation. We anticipate that this study will serve as a foundation for the development of coumarin-based bactericides.

Abstract:Coumarins are widely used due to their wide range of biological activities, but the long-term or excessive use of coumarin flavors can pose serious health hazards. Therefore, sensitive and specific methods for the quantification of these compounds in different matrices have been developed. In this review, an updated overview of the latest trends in sample preparation techniques and methods used to detect coumarins from March 2012 to April 2019 is provided. This study reviews different analytical methods (such as liquid chromatography coupled with different detectors, electrochemical sensors, capillary electrophoresis, etc.) and different pretreatment methods (such as liquid-liquid extraction, solid-phase extraction, dispersive liquid-liquid microextraction, etc.). Different methods for the pretreatment and determination of coumarins in plant, food, environmental, pharmaceutical and biological samples are summarized, discussed and compared.

Abstract:BACKGROUND Osthol is a natural coumarin and lead compound that has been developed into commercial fungicides in China. Natural coumarins comprise five major subtypes: simple coumarins, linear furanocoumarins, angular furanocoumarins, linear pyranocoumarins and angular pyranocoumarins. Studies pertaining to the antifungal activities of linear pyranocoumarins are few, and no reports exist for the antifungal activities of angular pyranocoumarins. In order to discover more antifungal natural coumarins, we synthesised a series of simple natural coumarins and isolated several plant-based furanocoumarins and pyranocoumarins using previously described methods. The compounds were biologically evaluated against some plant fungal pathogens. RESULTS Several of the 35 coumarins evaluated here exhibited strong activities against specific fungal species, including compound 25 (Pd-D-V, a linear pyranocoumarin), compound 26 (libanorin, an angular furanocoumarin) and compound 34 (disenecioyl khellactone, an angular pyranocoumarin). Compound 25 exhibited a high activity against Sclerotinia sclerotiorum (EC50 = 13.2 µg mL-1 ); compound 34 displayed a strong antifungal activity against Botrytis cinerea (EC50 = 11.0 µg mL-1 ). CONCLUSION This study demonstrates that several natural coumarins (one linear pyranocoumarin and one angular pyranocoumarin in particular) exhibit strong antifungal activities. These results call for further studies, where these coumarins can be examined as potential lead compounds for developing novel antifungal agents. © 2016 Society of Chemical Industry.

Abstract:This paper reviewed the advances on effective constituents and biological activities of coumarins in recent ten years. Coumarins are a group of important natural compounds, and have been found to have multi-biological activities such as anti-HIV, anti-tumor, anti-hypertension, anti-arrhythmia, anti-osteoporosis, assuaging pain, preventing asthma and antisepsis. Therefore, further investigation should emphasize on improving techniques for extraction and separation, searching the effective precursory compound, and synthesizing and screening out courmarin derivatives with high activity and low toxicity.
New Insights into the Antibacterial Activity of Hydroxycoumarins against Ralstonia solanacearum

Abstract:Coumarins are important plant-derived natural products with wide-ranging bioactivities and extensive applications. In this study, we evaluated for the first time the antibacterial activity and mechanisms of action of coumarins against the phytopathogen Ralstonia solanacearum, and investigated the effect of functional group substitution. We first tested the antibacterial activity of 18 plant-derived coumarins with different substitution patterns, and found that daphnetin, esculetin, xanthotol, and umbelliferone significantly inhibited the growth of R. solanacearum. Daphnetin showed the strongest antibacterial activity, followed by esculetin and umbelliferone, with MICs of 64, 192, and 256 mg/L, respectively, better than the archetypal coumarin with 384 mg/L. We further demonstrated that the hydroxylation of coumarins at the C-6, C-7 or C-8 position significantly enhanced the antibacterial activity against R. solanacearum. Transmission electron microscope (TEM) and fluorescence microscopy images showed that hydroxycoumarins may interact with the pathogen by mechanically destroying the cell membrane and inhibiting biofilm formation. The antibiofilm effect of hydroxycoumarins may relate to the repression of flagellar genes fliA and flhC. These physiological changes in R. solanacearum caused by hydroxycoumarins can provide information for integral pathogen control. The present findings demonstrated that hydroxycoumarins have superior antibacterial activity against the phytopathogen R. solanacearum, and thus have the potential to be applied for controlling plant bacterial wilt.

Abstract:The increasing resistance of plant diseases caused by phytopathogenic fungi highlights the need for highly effective and environmentally benign agents. The antifungal activities of Cnidium monnieri fruit extracts and five isolated compounds as well as structurally related coumarins against five plant pathogenic fungi were evaluated. The acetone extract, which contained the highest amount of five coumarins, showed strongest antifungal activity. Among the coumarin compounds, we found that 4-methoxycoumarin exhibited stronger and broader antifungal activity against five phytopathogenic fungi, and was more potent than osthol. Especially, it could significantly inhibit the growth of Rhizoctonia solani mycelium with an EC 50 value of 21 µg mL -1 . Further studies showed that 4-methoxycoumarin affected the structure and function of peroxisomes, inhibited the β-oxidation of fatty acids, decreased the production of ATP and acetyl coenzyme A, and then accumulated ROS by damaging MMP and the mitochondrial function to cause the cell death of R. solani mycelia. 4-Methoxycoumarin presented antifungal efficacy in a concentration- dependent manner in vivo and could be used to prevent the potato black scurf. This study laid the foundation for the future development of 4-methoxycournamin as an alternative and friendly biofungicide.

Abstract:Plants produce and release a vast variety of secondary metabolites with diversified functions, and possess ecological, toxicological and biological effects that mimic the activities of synthetic chemicals. Coumarins extracted from bacteria, fungi and many edible plants are widely used for their antibacterial, antifungal, antiviral, anti-HIV and anticancer activities. This review presents a concise summary based on the latest knowledge of the biological and pharmaceutical uses of coumarin and its derivatives, including an evaluation of future therapeutic potential. The presence of coumarins in various plant organs like fruits, seeds, roots, leaves and latex supplement recent work reported in scientific literature related to these compounds and their development. Due to low production in plants, the upscaling and industrial scale production, commercialization and industry demand of coumarins has faced hurdles. We searched Google Scholar and Web of Science for relevant literature upto 2018 using the keywords pharmaceutical, biological activities and coumarins. This review has thoroughly overviewed the related facts and figures about coumarins and its derivatives, especially in terms of biological and pharmaceutical properties including anti-microbial, anti-viral, anti-diabetic, anticoagulant, estrogenic, dermal photosensitizing, vasodilator, molluscacidal, antithelmintic, sedative and hypnotic, analgesic, hypothermic, anti-cancer activity, anti-oxidant, anti-parasitic, antihelmintic, anti-proliferative, anti-convulsant, anti-inflammatory, and anti-hypertensive activities. The pharmaceutical impact of coumarins on public health is a complex phenomenon, with several questions in relation to safety during medical therapies and consumption through diet. The present review focuses on coumarin clinical studies in the treatment of various chronic diseases such as cancer, diabetes, depression, Alzheimer's, Parkinson's and HIV. However, further research and review are necessary to broaden the therapeutic effectiveness of coumarin in patients suffering from such ailments.

Abstract:Many organic and medicinal chemists are interested in the coumarin compounds due to their potential for use in pharmaceuticals and other industries. Coumarin compounds are a significant class of biologically active chemicals. Here, we outline the steps forproducing coumarin derivatives quickly and effectively from commercially accessible substrates via multicomponent condensation processes involving pyrone and benzene derivatives. These substances were examined for their potential anti-tumor effects, including their ability to block carbonic anhydrase, activate the cell apoptosis protein, and coumarin and its derivatives are the primary oral anticoagulants. Candida albicans, Aspergillus fumigatus, and Fusarium solani are three distinct fungus species that are resistant to the antifungal properties of synthetic coumarins. Long known are the coumarins' antibacterial due to its biological activities against some kinds of bacteria such as Zeitschrift für Naturforschung C, as well as their antifilarial, antiulcerogenic, anti-inflammatory, and antioxidant effects for its capacity to inhibit their growth activities. Coumarin has long been valued as a major raw element in the fragrance sector due to its distinct sweet aroma and stability. When mixed with organic essential oils like lavender, citrus, rosemary, and oak moss, it is used as an odour-enhancer to produce a long-lasting impact. It has been utilized as a bleaching or brightening agent in detergents when it revealed a remarkable brightening effect with excellent overall stability on fibers of synthetic origin. Materials such as liquid crystalline, organic/inorganic composites, as well as light harvesting, energy transferring compounds, and electro-optics materials are all employed as a dye in organic photo-redox catalysis and as potent photo-sensitizers reagent. These materials polymerize epoxy-silicones, under near-UV and visible light. Coumarin has also found useful as metal-free sensitizers for solar cells.

Abstract:Introduction: Coumarins are naturally occuring metabolites from plants and a few micro-organisms. They have been widely used in the food and drug industry in their natural or synthetic forms. Numerous coumarins possess several biological activities such as anti-inflammatory, anti-ulcers, anti-tumour, anti-microbial, anti-coagulant. The aim of this study was to assess the bioactivity, and toxicity of coumarins from African medicinal plants. Methods: We searched online databases and search engines such as PubMed, Google Scholar and Web of Science for key terms such as coumarins, toxicity, bioavailability, bioactivity with appropriate Boolean operators. Only full-length research articles published in English between 1956 to 2023 were reviewed. Results: We recorded 22 coumarins from 15 plant species from Africa. Most of the plant species (33%) were from North Africa. These were followed by East Africa at 21%, then West, and Central Africa at 18.2% each. Most of the coumarins (21.3%) were isolated from the entire plant and the leaves (19.1%) and most of them (46.7%) had some antimicrobial activity. Five coumarins viz osthole, pseudocordatolide C & calanolide, chartreusin and esculetin had either antitumor or anticancer activity. Six coumarins had varying levels and types of toxicity ranging from inhibiting blood clotting as anticoagulants, to cytotoxic effects, causing hyperventilation, tremor, & photophobia, pulmonary haemorrhage, carcinogenic activity, severe neurotoxicity, hepato- and phototoxicity. Conclusion: Several African medicinal plants are sources of various coumarins that possess several biological activities as well as toxicities. This calls for more research into their safety and efficacy because of their wide spread applications as therapeutic agents.

Abstract:BACKGROUND:As an important plant allelochemical, coumarin can effectively inhibit the germination of various seeds. However, little is known about the inhibition mechanism of coumarin on weed seed germination. Moreover, the herbicidal activity of coumarin is needed to be improved as a natural pesticide.RESULTS:Coumarin had the highest inhibition effect on the ryegrass (Lolium perenne) seed, where coumarin disturbed the hormone pathway by decreasing the content of gibberellic acid 3, resulting in the reduction of amylase activity and consumption of starch during the germination process of ryegrass seed. Moreover, coumarin induced decreased activity of catalase and subsequently led to the accumulation of hydrogen peroxide and malondialdehyde, causing oxidative stress during the germination process of ryegrass seed. Furthermore, to enhance the herbicidal activity of coumarin, carbon dots (CDs) modified with polyetherimide were prepared, characterized, and then combined with coumarin to form coumarin-carbon dots (Cm-CDs) nanocomposites. Compared with coumarin, Cm-CDs nanocomposites significantly increased the herbicidal activity of coumarin on ryegrass, which implies that Cm-CDs nanocomposites could be used as a potential formulation to improve the herbicidal activity of coumarin.CONCLUSION:This study not only reveals the mechanism of coumarin on ryegrass germination, but also develop Cm-CDs nanocomposites to enhance the herbicidal activity of coumarin. Our findings will stimulate the application of Cm-CDs nanomaterials as an effective and environmentally friendly formulation in agriculture. © 2023 Society of Chemical Industry.
