Abstract:An efficient synthesis of novel fused heterocycles has been established via microwave-assisted palladium-catalyzed intramolecular direct arylation. The acyclic aryl bromides are readily available from microwave-assisted one-pot annulation of N-(2-bromobenzyl)-2-aminophenois and ethyl 2-bromoalkanoates. The intramolecular direct arylation is then performed in the presence of palladium(II) acetate and dppf (10 mol% each) in toluene using potassium carbonate (2 equiv) as the base under microwave heating (150 degrees C, 1 h) to afford the products in 43-99% yields. Steric effect is observed for Ar-Ar bond formation, giving a substantial amount of the debromination byproduct.

Abstract:A sterically congested all-carbon quaternary center was installed for the first time via a Pd-catalyzed cascade diarylation with aryl bromides and acyclic 1,3-diketones. This method was used as a key step in the total synthesis of (±)-spiroaxillarone A. Computational experimental results indicated that the selective diarylation is accelerated by the higher free-energy barriers of the endothermic transmetalation and reductive elimination in the first arylation step.

Abstract:A general and convenient selective direct arylation of the 3‐position of triimidazo[1,2‐a:1',2'‐c:1'',2''‐e][1,3,5]triazine (1) with (hetero)aryl halides in DMA was successfully achieved in the presence of K2CO3 as the base and a catalyst precursor consisting of Pd(OAc)2 and P(2‐furyl)3. Electron‐poor and ‐rich (hetero)aryl moieties, including the strongly deactivated and sterically encumbered 2,4,6‐trimethoxyphenyl unit, are well tolerated in the electrophilic partner. The data obtained in this synthetic study support a reaction mechanism involving an electrophilic attack of an arylpalladium‐(II) halide species onto the triazine ring.

Abstract:Recently, we realized the highly selective one-pot synthesis of 2,6-diarylpyridines by using a Pd-catalyzed direct C-H arylation approach via a transient activator strategy. Although methylation reagent as a transient activator and Cu(I) salt or oxide were found to be prerequisites, details regarding the mechanism remained unclear. In this paper, DFT calculations combined with experimental investigations were carried out to elucidate the principle features of this transformation. The results reveal (1) the origin of the exquisite diarylating selectivity of the pyridine under the transient strategy; (2) the possible demethylating reagent as the counteranion of the pyridinium salt; (3) the reason why Cu2O is a better Cu(I) resource than others.

Abstract:A cooperative Rh(II)/Pd(0) dual-catalysis strategy that enabled divergent reactions of α-diazo 1,3-dicarbonyl compounds with allylic carbonates involving ketene versus carbene intermediates is described. The efficient synthesis of α-quaternary allylated β-keto-esters was accomplished by the Rh(II)/Pd(0) dual-catalysis allylic alkylation of α-diazo 1,3-dicarbonyl compounds. Alternatively, an unprecedented (1+4) annulation of α-diazo 1,3-dicarbonyl compounds with 2-(hydroxymethyl)allyl carbonates via Rh(II)/Pd(0) dual catalysis was also successfully developed, affording a wide variety of α-quaternary tetrahydrofurans in good to high yields.

Abstract:A C-H arylation of thiopyran derivatives with aryl halides has been developed. Under the catalysis of Pd(OAc)2/Ag2CO3, the C-H arylation takes place at the α-position of the thiopyran ring. When dibromo-substituted compounds are used as reactants, double C-H arylations may occur on the same thiopyran ring at its α- and β-positions.

Abstract:The reactivity of 2‐benzyl‐1,2,3‐triazole in palladium‐catalyzed direct arylation was studied. The reaction conditions for the selective synthesis of 2‐benzyl‐4‐aryl‐1,2,3‐triazoles in moderate to high yields using phosphine‐free Pd(OAc)2 catalyst and inexpensive KOAc base have been found. Then, from these 4‐aryl‐1,2,3‐triazoles, the palladium‐catalyzed C−H bond functionalization of the C5‐position allowed the synthesis of the corresponding 4,5‐diarylated 2‐benzyl‐1,2,3‐triazoles. This selective 4,5‐diarylation of 2‐benzyl‐1,2,3‐triazole was successfully applied for the straightforward building of the π‐extended polycyclic heteroaromatic structures phenanthro[9,10‐d][1,2,3]triazoles through Pd‐catalyzed C4‐ and C5‐intermolecular arylations followed by Pd‐catalyzed C−H intramolecular arylation.

Abstract:Pd-catalyzed direct arylation of 2-alkylthiazoles is a well-known reaction affording the corresponding 2-alkyl-5-arylthiazoles in very high yields. Conversely, the reactivity of 2-alkoxythiazoles has not been described yet. Herein, we report conditions for the Pd-catalyzed regioselective C5-arylation of 2-alkoxythiazoles. Moreover, we also found reaction conditions leading to 3-alkyl-5-arylthiazol-2(3H)-ones via a one-pot direct arylation with an O- to N-alkyl migratory rearrangement. The judicious choice of reaction temperature and time allows control over the selectivity of the reaction. In general, at 100 °C, 2-alkoxy-5-arylthiazoles were the major products, whereas, at 120 °C with a longer reaction time, 3-alkyl-5-arylthiazol-2(3H)-ones were obtained with good selectivities. The arylation reaction is promoted by a ligand-free air-stable palladium catalyst and a simple and inexpensive base, without oxidant or further additives, and tolerates a variety of useful substituents on the aryl bromide and also heteroaryl bromides.

Abstract:With the combination of the N-heterocyclic carbene-PdCl(2)-1-methylimidazole complex and Cu(2)O, we succeeded in the first example of double direct C-H bond arylation reactions between thiophenes and aryl chlorides, giving the desired 2,5-diarylated thiophenes in moderate to high yields under suitable conditions, consistent with the density functional theory calculations.
