Abstract:The palladium-catalyzed acyloxylation of 2-substituted 1,2,3-triazoles with acids via direct sp(2) CH bond activation is described. This reaction is applied to a wide variety of 2-substituted 1,2,3-triazoles and exhibits excellent functional group tolerance. The route is found to be efficient for the preparation of 2-substituted 1,2,3-triazole derivatives.

Abstract:A mild and efficient method for the direct alkenylation of 2-benzyl-1,2,3-triazoles via Pd-catalyzed C-H bond activation was developed. This protocol was compatible with various substrates and gave the corresponding products in good to excellent yields. Thus, the present study provides a novel and valuable method for the synthesis of 2-benzyl-1,2,3-triazole derivatives.

Abstract:A palladium-catalyzed ortho-acylation of 2-aryl-1,2,3-triazoles was developed in which low-toxicity, stable, commercially available toluene derivatives were used as acyl sources. A wide variety of aryl ketones containing 1,2,3-triazoles were obtained in moderate to excellent yields. A plausible mechanism is proposed.

Abstract:Palladium-catalyzed heteroarylation of 1-substituted 1,2,3-triazoles with furans and thiophenes has been developed in the presence of pyridine and Ag2CO3. The procedure is suitable for the regioselective preparation of 1,5-disubstituted 1,2,3-triazoles through conventional heating at reaction temperatures of 90-100 degrees C for 18 h.

Abstract:The palladium-catalyzed oxidative acyloxylation of 5-alkyl-4-aryl-thiazole-2-carboxylates via a direct ortho-Csp(2)-H bond activation has been described with a newly reported radical mechanism. Based on the late-stage functionalization of the highly substituted thiazoles, the reaction delivered multifarious valuable acyloxylated products with moderate-to-excellent yields. The transformation affords highly functionalized substrates, has broad scope of carboxylic acid coupling partners, excellent site-selectivity, rapid reaction rates, simple operation and mild conditions. The diverse acyloxylation reaction allows rapid access to a plethora of mono-acyloxylated products from the functionalized thiazoles.

Abstract:The reactivity of 2‐benzyl‐1,2,3‐triazole in palladium‐catalyzed direct arylation was studied. The reaction conditions for the selective synthesis of 2‐benzyl‐4‐aryl‐1,2,3‐triazoles in moderate to high yields using phosphine‐free Pd(OAc)2 catalyst and inexpensive KOAc base have been found. Then, from these 4‐aryl‐1,2,3‐triazoles, the palladium‐catalyzed C−H bond functionalization of the C5‐position allowed the synthesis of the corresponding 4,5‐diarylated 2‐benzyl‐1,2,3‐triazoles. This selective 4,5‐diarylation of 2‐benzyl‐1,2,3‐triazole was successfully applied for the straightforward building of the π‐extended polycyclic heteroaromatic structures phenanthro[9,10‐d][1,2,3]triazoles through Pd‐catalyzed C4‐ and C5‐intermolecular arylations followed by Pd‐catalyzed C−H intramolecular arylation.

Abstract:Pd-catalyzed direct arylation of 2-alkylthiazoles is a well-known reaction affording the corresponding 2-alkyl-5-arylthiazoles in very high yields. Conversely, the reactivity of 2-alkoxythiazoles has not been described yet. Herein, we report conditions for the Pd-catalyzed regioselective C5-arylation of 2-alkoxythiazoles. Moreover, we also found reaction conditions leading to 3-alkyl-5-arylthiazol-2(3H)-ones via a one-pot direct arylation with an O- to N-alkyl migratory rearrangement. The judicious choice of reaction temperature and time allows control over the selectivity of the reaction. In general, at 100 °C, 2-alkoxy-5-arylthiazoles were the major products, whereas, at 120 °C with a longer reaction time, 3-alkyl-5-arylthiazol-2(3H)-ones were obtained with good selectivities. The arylation reaction is promoted by a ligand-free air-stable palladium catalyst and a simple and inexpensive base, without oxidant or further additives, and tolerates a variety of useful substituents on the aryl bromide and also heteroaryl bromides.

Abstract:Heterocyclic compounds bearing 1,2,3-triazole scaffolds have found wide application both in medicinal chemistry and materials science. In this paper, rhodium(iii)-catalyzed triazole-directed alkylation reactions of arenes using diazo compounds as the alkylating agents are described. A number of polysubstituted arenes were provided from easily available materials in good yields under mild conditions. The reactions proceed via triazole-directed ortho C-H bond activation and subsequent carbene insertion originating from diazo compounds.
