Abstract:Alfalfa is widely distributed worldwide and is one of the most important forage legumes.It is well-known as "the king of forage" because of its high yield,high quality,biological nitrogen fixation,etc.Alfalfa has a long history of having been planted in China and having maintained a high economic value.According to previous research,there are 46 species in the Medicago sativa L.genus.Studies on alfalfa breeding occurred later and have made a significant amount of progress in recent decades.By the end of 2014,a total of 77 alfalfa varieties had been registered,and 36 of them had been bred by crossbreeding technology.Alfalfa is not only an important high-protein type of forage but is also a soil and water conservation plant;thus,alfalfa has both important economic and ecological functions.Furthermore,the excellent germplasm resources of alfalfa have been discovered for breeding new alfalfa cultivars,which can not only increase the forage yield but can also accelerate the development of the dairy industry.Moreover,alfalfa can improve soil fertility as an important ecological protection for plants.Therefore,the development of the alfalfa industry plays an important role in the sustainable development of agriculture in China.This paper reviewed the distribution,germplasm resources,genetic breeding,and utilization of alfalfa on the basis of the present situation of alfalfa in China.It may provide some reference for future research of alfalfa.

Abstract:As a traditional fiber plant, kenaf also has tender branches and leaves which contain high and fine protein. Many domestic and international studies showed that kenaf has a good value as forage. With the speedy development of the stockbreeding and the forage industry, the forage with high and fine protein comes to be urgent goods. Further exploration and research on the feeding value of kenaf will be more helpful for the solution of the deficiency of the forage with high quality protein in South China. This paper summarizes the international research progress and prospect of the feeding value of kenaf and pro-vides references for further study.

Abstract:Currently,developed countries and multinational seed companies have made great progress in R&D of plant functional genomics,transgenic technology and genetically modified(GM) products.Production of GM crop is increasing rapidly and has become a new emerging industry in recent years.The Chinese government is highly concerned with the R&D and their industrialization of GM crops,and leads the developing countries in this respect, but there still has distance with the rapid development of international industry and can not meet the needs of our agricultural requirements.There exist several limitations in transgenic technology and product innovation, industrialization mechanisms and support conditions in China.Based on systematic comparison analysis,we propose to enhance our transgenic R&D capability building and transgenic based breeding,as well as to breakthrough transgenic core technology and develop GM products.With ever closer collaborations among research institutes,universities and companies,we aim to develop large enterprises with self-innovation capability and strong market competition.Meanwhile,we should promote public comprehension for GM crops and therefore establish an amenable social environment that propels rapid development of transgenic breeding-based emerging industry in China.

Abstract:Alfalfa is one of the most important legume forages in the world and contributes greatly to the improvement of ecosystems, nutrition, and food security. Diseases caused by pathogens and pests severely restrict the production of alfalfa. Breeding resistant varieties is the most economical and effective strategy for the control of alfalfa diseases and pests, and the key to breeding resistant varieties is to identify important resistance genes. Plant innate immunity is the theoretical basis for identifying resistant genes and breeding resistant varieties. In recent years, the framework of plant immunity theory has been gradually formed and improved, and considerable progress has been made in the identification of alfalfa resistance genes and the revelation of the related mechanisms. In this review, we summarize the basic theory of plant immunity and identify alfalfa resistance genes to different pathogens and insects and resistance mechanisms. The current situation, problems, and future prospects of alfalfa resistance research are also discussed. Breeding resistant cultivars with effective resistance genes, together with other novel plant protection technologies, will greatly improve alfalfa production.

Abstract:Alfalfa (Medicago sativa) is one of the most important forage crops in the world; however, its molecular genetics and breeding research are hindered due to the lack of a high-quality reference genome. Here, we report a de novo assembled 816-Mb high-quality, chromosome-level haploid genome sequence for 'Zhongmu No.1' alfalfa, a heterozygous autotetraploid. The contig N50 is 3.92 Mb, and 49 165 genes are annotated in the genome. The alfalfa genome is estimated to have diverged from M. truncatula approximately 8 million years ago. Genomic population analysis of 162 alfalfa accessions revealed high genetic diversity, weak population structure, and extensive gene flow from wild to cultivated alfalfa. Genome-wide association studies identified many candidate genes associated with important agronomic traits. Furthermore, we showed that MsFTa2, a Flowering Locus T homolog, whose expression is upregulated in salt-resistant germplasms, may be associated with fall dormancy and salt resistance. Taken together, these genomic resources will facilitate alfalfa genetic research and agronomic improvement.

Abstract:Alfalfa ( Medicago sativa L .) is one of the most important forage crops in the world. Drought is recognized as a major challenge limiting alfalfa production and threatening food security. Although some literature reviews have been conducted in this area, bibliometric reviews based on large amounts of published data are still lacking. In this paper, a bibliometric analysis of alfalfa drought stress from 1998–2023 was conducted using the Web of Science Core Collection database in order to assess global trends in alfalfa drought stress research and to provide new directions for future research. The results showed that the annual publication output maintained an increase in most years, with China and the United States contributing significantly to the field. Most of the journals published are specialized journals in botany, environmental science, soil science and crop science, as well as related agribusiness journals. "plant growth" and "yield" were the most frequently used keywords, reflecting the important purpose of research in this field. And two main research directions were identified: research on drought response mechanism of alfalfa and exploration of drought-resistant technology. In addition, physiological, biochemical, and molecular responses of drought tolerance and high yield in alfalfa, transgenics, and microbial fertilizer research have been hot research topics in recent years and may continue in the future. The ultimate goal of this paper is to provide a foundational reference for future research on alfalfa's drought resistance and yield optimization mechanisms, thereby enhancing the crop's application in agricultural production.

Abstract:Abstract Artificially improving traits of cultivated alfalfa ( Medicago sativa L.), one of the most important forage crops, is challenging due to the lack of a reference genome and an efficient genome editing protocol, which mainly result from its autotetraploidy and self-incompatibility. Here, we generate an allele-aware chromosome-level genome assembly for the cultivated alfalfa consisting of 32 allelic chromosomes by integrating high-fidelity single-molecule sequencing and Hi-C data. We further establish an efficient CRISPR/Cas9-based genome editing protocol on the basis of this genome assembly and precisely introduce tetra-allelic mutations into null mutants that display obvious phenotype changes. The mutated alleles and phenotypes of null mutants can be stably inherited in generations in a transgene-free manner by cross pollination, which may help in bypassing the debate about transgenic plants. The presented genome and CRISPR/Cas9-based transgene-free genome editing protocol provide key foundations for accelerating research and molecular breeding of this important forage crop.

Urgency of Further Developing Alfalfa Industry in China

Abstract:The rapid development of animal husbandry in China, especially the prosperous development of dairy farming, greatly increases the need for herbage of high quality, particularly alfalfa. This study pointed out that the alfalfa yield was low, and its quality was poor, and the supply of alfalfa did not meet its market demand in China. Consequently, the total quantity of imported alfalfa showed a increasing trend and came to 227 200 t in 2010, threatening the development of alfalfa industry in China. This study pro- posed that it is urgent to develop the alfalfa industry because alfalfa industry was regulated by many fac- tors. Finally, this study proposed some suggestions for further developing the alfalfa industry. Firstly, it was urgent to make development plan of alfalfa industry; secondly, the compensation package needed to be improved for alfalfa producer; thirdly, it was effort to innovate the utilization technology and support the leading enterprise of alfalfa industry; fourthly, establishing the demonstration base with modern technology was necessary by integrating the alfalfa resources, which would benefit the establishment of alfalfa industry belt and increased the comprehensively productive ability and supply ability by optimizing regional distribution, planting scale, exercising mechanization and rationally managing with standard.

Abstract:Alfalfa (Medicago sativa L.) is the most valuable perennial forage crop, and assessing its yield and quality properties is essential to evaluate forage value of livestock production. However, regional patterns and con-trolling factors of the yield and quality of alfalfa have not been well documented. In this study, using a dataset of 7166 observations from 301 sites across the country collected from 334 publications, we explored the re-lationships between the yield and quality properties of alfalfa in China and the effects of environmental factors and management practices on them. The average dry matter yield of alfalfa was 11.18 ?? 6.69 Mg ha-1, with average crude protein, ether extract, crude fiber, acid detergent fiber, neutral detergent fiber, crude ash, nitrogen-free extract, calcium, phosphorus and relative feed values of 19.05 ?? 2.87%, 2.62 ?? 0.97%, 27.16 ?? 5.21%, 31.29 ?? 5.58%, 40.48 ?? 6.34%, 9.49 ?? 1.59%, 38.67 ?? 7.49%, 1.68 ?? 0.55%, 0.25 ?? 0.12%, and 151 ?? 31.94, respectively. Alfalfa quality decreased with increasing yield. Geographically, alfalfa had higher yield and crude protein content in South China than in other regions. Quality was positively correlated with both precipitation and temperature, while yield showed a positive relationship with temperature but no significant relationship with precipitation. High soil nutrient availability (e. g., soil available nitrogen, SAN; soil available phosphorus, SAP; soil available potassium, SAK) improved the yield, while high SAN and SAP contents increased the crude protein content. The alfalfa yield increased and the quality decreased with advancing maturity and age, whereas increasing the number of harvests may benefit the quality. This study documents comprehensive in-formation on the yield and quality of alfalfa and provides insights for policy makers for the effective management and sustainable development of alfalfa production.

Abstract:Alfalfa (Medicago sativa L.) is the most important legume forage crop worldwide with high nutritional value and yield. For a long time, the breeding of alfalfa was hampered by lacking reliable information on the autotetraploid genome and molecular markers linked to important agro-nomic traits. We herein reported the de novo assembly of the allele-aware chromosome-level genome of Zhongmu-4, a cultivar widely cultivated in China, and a comprehensive database of genomic variations based on resequencing of 220 germplasms. Approximate 2.74 Gb contigs (N50 of 2.06 Mb), accounting for 88.39% of the estimated genome, were assembled, and 2.56 Gb contigs were anchored to 32 pseudo-chromosomes. A total of 34,922 allelic genes were identified from the allele-aware genome. We observed the expansion of gene families, especially those related to the nitrogen metabolism, and the increase of repetitive elements including transposable elements, which probably resulted in the increase of Zhongmu-4 genome compared with Medicago truncatula. Population structure analysis revealed that the accessions from Asia and South America had rela-tively lower genetic diversity than those from Europe, suggesting that geography may influence alfalfa genetic divergence during local adaption. Genome-wide association studies identified 101 sin-gle nucleotide polymorphisms (SNPs) associated with 27 agronomic traits. Two candidate genes were predicted to be correlated with fall dormancy and salt response. We believe that the allele -aware chromosome-level genome sequence of Zhongmu-4 combined with the resequencing data of the diverse alfalfa germplasms will facilitate genetic research and genomics-assisted breeding in variety improvement of alfalfa.

Abstract:Alfalfa (Medicago sativa L.) is an important forage crop worldwide. However, little is known about the effects of breeding status and different geographical populations on alfalfa improvement. Here, we sequenced 220 alfalfa core germplasms and determined that Chinese alfalfa cultivars form an independent group, as evidenced by comparisons of FST values between different subgroups, suggesting that the geographical origin plays an important role in group differentiation. By tracing the influence of geographical regions on the genetic diversity of alfalfa varieties in China, we identified 350 common candidate genetic regions and 686 genes under selection. We also defined 165 loci associated with 24 important traits from genome-wide association studies (GWASs). Of those, 17 genomic regions closely associated with a given phenotype were under selection, with the underlying haplotypes showing significant differences between subgroups of distinct geographical origins. Based on results from expression analysis and association mapping, we propose that 6-phosphogluconolactonase (MsPGL) and a gene encoding a protein with NHL domains (MsNHL) are critical candidate genes for root growth. Critical Reviews in Biotechnology

Abstract:Current research needs to be more focused on agronomical plants to effectively utilize the knowledge obtained from model plant species. Efforts to improve legumes have long employed common breeding tools. Recently, biotechnological approaches facilitated the development of improved legumes with new traits, allowing them to withstand climatic changes and biotic stress. Owing to its multiple uses and profits, alfalfa (Medicago sativa L.) has become a prominent forage crop worldwide. This review provides a comprehensive research summary of tissue culture-based genetic transformation methods, which could be exploited for the development of transgenic alfalfa with agronomically desirable traits. Moreover, advanced bio-imaging approaches, including cutting-edge microscopy and phenotyping, are outlined here. Finally, characterization and the employment of beneficial microbes should help to produce biotechnologically improved and sustainable alfalfa cultivars.
