Abstract:2-Chloro-5-methylthiophene (1) is a versatile intermediate for the synthesis of pharmaceuticals and agrochemicals.A facile and low-cost route for the preparation of 1 was reported through the reaction of 2 methylthiophene with sulfuryl chloride.The process produced the product in 73.96% yield.

Abstract:The chloromethylation reaction of 2-chloro-thiophene via phase transfer catalysis was carried out to synthesize 2-chloro-3-chloromethyl-thiophene, where the catalyst was tetrabutylammonium bromide (TBAB). The chloromethylation reagents used were paraformaldehyde and 37% concentrated hydrochloric acid. The structure of 2-chloro-3-chloromethyl-thiophene was confirmed by means of GC-MAS, IR and ^1H NMR spectrometries. The yield was 77.6%. The O-alkylation reactions between 1-(2,4-dichloro-phenyl)-2-(1-imidazol)-ethanol and 2-chloro-3-chloromethyl-thiophene were performed in a two -phase system of organic solvent/solid NaOH under phase transfer catalytic condition. At the end, the tioconazole nitrate was formed by the addition of nitric acid. The structure of tioconazole was confirmed with IR and ^1H NMR spectra. The yield of tioconazole was 53.0%.

Abstract:Dithieno[3,2-b:2',3'-d]thiophene (DTT) derivative contains three annulated thiophene rings, which have attracted great attention in the field of materials due to its planar and conjugated molecular, structural rigidity, and increased solubility. Herein, the one-step modular construction of a series of structurally well-defined 2,6-disubstituted dithieno[3,2-b:2',3'-d]thiophenes (2,6-DTT) involves Pd/Cu(I)-catalyzed Sonogashira coupling and LiNH2-mediated ring-closing reaction with element sulfur was reported. The method allows the selective and straightforward introduction of different aryl substitution into DTT without otherwise tedious separation and purification processes. The preliminary study of the thermal and photophysical properties demonstrated that the introduction of substituent in phenyl has great impact for the parent molecule, which enable this protocol to synthesis high-performance 2,6-DTT through the different alkynes.

Abstract:A mild and efficient method for the synthesis of 2,5-disubstituted chalcogenophenes is reported. This method involves two steps of reactions via one-pot approach by employing commercially available and cost-effective alkynes and element chalcogen. Further, by using LiNH2 as the promoter and DMSO/EtOH as the solvent, Cu-catalyzed Glaser coupling-cycloaddition processes are dramatically enhanced that a diverse range of chalcogenophenes can be obtained in good to excellent yields under mild conditions. Controlled experiments show that trisulfur radical anion (S3 & sdot;-) is the key intermediate and DMSO/EtOH serves as the hydrogen source, while alcohol may facilitate the transition state. The mild and highly efficient constructions of 2,5-disubstitued thiophenes, selenophenes and tellurophenes are achieved by using a one-pot, two-step protocol that combine Cu-catalyzed Glaser coupling with LiNH2-mediated cycloaddition. And the DMSO/EtOH mixture solvent is found to short the reaction time and improve the reaction yield by promoting the production of S3 & sdot;- and stabilizing the transition state.image

Abstract:An improved procedure for the synthesis of 3,4-ethylenedioxythiophene is reported starting from ethyl chloroacetate. Reaction with sodium sulfide gave diethyl thiodiglycolate which was then reacted with diethyl oxalate, and then 1,2-dibromoethane to give 2,5-dicarbethoxy-3,4-ethylenedioxythiophene. Hydrolysis, and decarboxylation gave 3,4-ethylenedioxythiophene in 16% overall yield. The structures of the key intermediates in synthetic routes were confirmed, and every step was optimised, to give a procedure suitable for large-scale industrial production.

Abstract:A convenient and efficient synthesis of 2-substituted benzo[b]thiophenes is described. This one-step reaction of alkylthiobenzaldehyde and activated electrophiles proceeded smoothly in toluene at 120 degrees C with magnesium oxide as a catalyst. The target 2-substituted benzo[b]thiophenes are obtained in moderate yields and the side reaction is inhibited via azeotropic distillation. According to the experiments, a plausible mechanism is presented.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:A new route for the synthesis of 6-bromo-4,4-dimethylthiochroman was proposed and the optimized conditions for each step were obtained.Bromobenzene was selected as starting material and 6-bromo-4,4-dimethylthiochroman was prepared via chlorosulfonation,reduction,thioetherification and cyclization with the yield of 88.5%.Except the reduction,which had to be performed at a higher temperature,the other three steps proceeded at temperatures lower than 50oC.Thus both the energy cost and byproducts were decreased.Compared to previous routes,the proposed one in this work is advanced for its lower cost and less contamination,so it is suitable to be industrially amplified.

Abstract:5-[2-(ethylthio)propyl]-1,3-cyclohexanedione, the title compound of this study, is an important intermediate for preparation of 3-hydroxy-cyclohex-2-enones herbicides, such as clethodim, sethoxydim, cloproxydim, etc.. The optimization of the synthesis route and the preparation method of the title compound were studied and the results were described concisely as follows. Firstly, 3-ethylthiobutanal was prepared with high yield by reacting crotonaldehyde with ethanethiol in triethylamine. Secondly, methyl acetoacetate was converted to 6-ethylthiohept-3-en-2-one via three steps, hydrolysis, condensation with 3-ethylthiobutanal and dehydration, then the 6-ethylthiohept-3-en-2-one was cyclocondensed with di-Me malonate by Michael addition and Clasisen condensation, and further via saponification and decarboxylation , the title compound was obtained. The products of each steps and the final title compound obtained were compared with each its own standerd samples and were confirmed by LC/MS.

Abstract:As an intermediate for COX-2 inhibiters,4-(methylthio)benzeneacetic acid(Ⅳ) was prepared via Friedel-Crafts and Willgerodt-Kindler reaction from thioanisole(Ⅰ).When the molar ratio of Ⅰ to acetyl chloride and AlCl_3 was 1∶1.1∶1.3 and CS_2 was used as solvent,after reacting at(0°C) for 6 h the yield of 4-(methylthio) acetophenone(Ⅱ) was 75.9%.Ⅱ was refluxed 5 h with morpholine and sulfur to give thioacetmorpholide(Ⅲ),then Ⅲ was refluxed in 10% NaOH for 10 h to give Ⅳ with an overall yield of 63.5% and purity of 99.0%(by HPLC).The structure was confirmed by IR and()~1H NMR.

Abstract:p-Trifluoromethylthiophenol was synthesized from p-hydroxylphenylthiol in DMF initiated by ·CF_3 which was formed from CF_3Br using Na_2S_2O_4 as a stimulant. The purity of the product was 98% and the yield was 81.5%.

Abstract:A new method for the 100 g scale synthesis of N-difluoromethylthiophthalimide 1 from cheap commodity chemical benzyl mercaptan and HCF2Cl or other difluorocarbene precursors was described. Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:sulfomethyltetrazole 5 thiol disodium salt, a side chain of cefonicid,was synthesized from sodium bisulfite,ammonium hydroxide and formaldehyde through five steps of reactions. The yield of product was 10.8% in the optimum condition. The process was more economic in which the iodomethane is substituted to the bromoethane in the second reaction.

Abstract:1,4-Bis(tert-butylthio)-3,6-diphenyl- and 3,6-di(2-thienyl)thieno[3,4-c]thiophenes (2a,b)were synthesized from 2-(tert-butylthio)-3-phenyl- and 3-(2-thienyl)cyclopropenethiones (1a,b) and triphenylphosphine in dry benzene at 50 degrees C, although similar treatment of 2,4,6-triisopropylphenyl, N,N-diethylamino, pyrrolidino, and diphenyl-substituted cyclopropenethiones (1c-h) did not result in the production of the corresponding thieno[3,4-c]thiophene derivatives. The possible reaction pathway for the formation of 2a,b is described. The protonation of 2a with trifluoroacetic acid (TFA) gave 4-(tert-butylthio)-3,6-diphenylthieno[3,4-c]thiophene-1(3H)-thione (13a), the treatment of which with sodium hydride and then isopropyl iodide led to 4-(tert-butylthio)-3,6-diphenyl-1-(isopropylthio)thieno[3,4-c]thiophene (16) by the regeneration of the thieno[3,4-c]thiophene ring system, this making possible the synthesis of other alkylthio-substituted thieno[3,4-c]thiophene derivatives. The reactions of 2a,b with N-phenylmaleimide (NPM) gave predominantly the endo-cycloadducts (17a,b) at the 1- and 3-positions, and that of 2a with dimethyl acetylenedicarboxylate (DMAD) led to the benzo[c]thiophene derivative (19) by desulfurization.
