Abstract:The first catalytic enantioselective 1,2-reduction of enones with 0.45 mol equiv potassium borohydride solution catalyzed by a chiral N,N'-dioxide-Sc(III) complex catalyst was accomplished under mild reaction conditions. A number of optically active allylic alcohols were obtained in good to excellent enantioselectivities (up to 95% ee) with nearly quantitative yields.

Abstract:The first Lewis acid catalyzed asymmetric Friedel-Crafts alkylation reaction of ortho-hydroxybenzyl alcohols with C3-substituted indoles is described. A chiral N,N'-dioxide Sc(OTf)3 complex served not only to promote formation of ortho-quinone methides (o-QMs) in situ but also induced the asymmetry of the reaction. This methodology enables a novel activation of ortho-hydroxybenzyl alcohols, thus affording the desired chiral diarylindol-2-ylmethanes in up to 99 % yield and 99 % ee. A range of functional groups were also tolerated under the mild reaction conditions. Moreover, this strategy gives concise access to enantioenriched indole-fused benzoxocines.

Abstract:A facile technique involving a novel Lewis-acid-catalyzed tandem selective ring-opening of oxirane and Friedel-Crafts alkylation of 1H-indol-1-yl derived oxiranes was developed. The developed protocol affords functionalized [1,4]oxazino[4,3-a]indoles and 3,4-dihydro-1H-pyrrolo[2,1-c][1,4]oxazines in up to 95% yield.

Abstract:A cooperative catalytic system established by the combination of an iron salt and a chiral Bronsted acid has proven to be effective in the asymmetric Friedel-Crafts alkylation of indoles with beta-aryl alpha'-hydroxy enones. Good to excellent yields and enatioselectivities were observed for a variety of alpha'-hydroxy enones and indoles, particularly for the beta-aryl alpha'-hydroxy enones bearing an electron-with-drawing group at the para position of the phenyl ring (up to 90% yield and 91% ee). The proton of the chiral Bronsted acid, the Lewis acid activation site, as well as the inherent basic site for the hydrogen-bonding interaction of the Bronsted acid are responsible for the high catalytic activities and enantioselectivities of the title reaction. A possible reaction mechanism was proposed. The key catalytic species in the catalytic system, the phosphate salt of Fe-III, which was thought to be responsible for the high activity and good enantioselectivity, was then confirmed by ESIMS studies.

Abstract:Highly diastereoselective hydroxyalkylation of phenols with chiral glyoxylates was performed. Hydrolysis or reduction of the esters 3 afforded 2-hydroxymandelic acids 6 and diols 5 in high yields and enantiomeric pure form. Abstract:Chiral phosphoric acid-catalyzed enantioselective aza-Friedel-Crafts reaction of trifluoromethyl benzoxazinones with pyrroles is reported. Under mild conditions, a range of enantioenriched dihydrobenzoxazinones bearing trifluoromethylated quaternary stereocenters could be obtained in good to excellent yield and ee. A remarkable fluorine effect is observed, and preliminary mechanistic studies combined with theory calculations suggest that triple-hydrogen-bonding interactions hold the transition structure rigidly and allow the bulky substituents of the catalyst to influence the enantioselectivity.

Abstract:This paper describes an efficient scandium triflate- and triflic acid-catalyzed benzannulalion of 2-(2-alkynylarylidene)-1,3-dicarbonyl compounds to afford polysubstituted naphthalenes and benzo[a]fluorenols. The product selectivity could be tuned by subtle choice of the catalyst. An unprecedented process between alkynes and ketones is also explored.

Abstract:A series of pyrazolone derivatives bearing a tetrasubstituted chiral center were prepared by virtue of a Lewis acid-catalyzed Friedel-Crafts reaction, in which a chiral copper complex was employed as the catalyst. This reaction can be carried out smoothly under mild condition to afford the pyrazolone derivatives with high yields (up to 85%) and excellent enantioselectivities (up to 99%). In addition, the gram scale synthesis proved the practicality of this reaction.

Abstract:A novel class of chiral spiro-fused bisoxazoline ligands possessing a deep chiral pocket was prepared. The developed ligands have been employed in the nickel-catalyzed highly enantioselective Michael-type Friedel-Crafts reaction, affording the products bearing a trifluoromethylated all-carbon quaternary stereocenter with moderate to excellent yields (up to 99%) and good to excellent enantioselectivies (up to > 99.9% ee). Moreover, a proposed model of chiral pocket revealed that the attack of indole from the Re-face of β-CF3-β-disubstituted nitroalkene was favorable.
