Abstract:A direct catalytic asymmetric aldol-type reaction of 3-substituted-2-oxindoles with glyoxal derivatives and ethyl trifluoropyruvate, catalyzed by a chiral N,N'-dioxide-Sc(OTf)(3) (Tf = trifluoromethanesulfonyl) complex, has been developed that tolerates a wide range of substrates. The reaction proceeds in good yields and excellent enantioselectivities (up to 93% yield, 99:1 diastereomeric ratio (dr), and >99% enantiomeric excess (ee)) under mild conditions, to deliver 3-(alpha-hydroxy-beta-carbonyl) oxindoles with vicinal quaternary-tertiary or quaternary-quaternary stereocenters. Even with 1 mol % catalyst loading or on scaleup (10 mmol of starting material), maintenance of ee was observed, which showed the potential value of the catalyst system. In studies probing the reaction mechanism, a positive nonlinear effect was observed and Sc(III)-based enolate intermediates were detected by using ESIMS. On the basis of the experimental results and previous reports, a possible catalytic cycle was assumed.

Abstract:A palladium-catalyzed decarboxylative asymmetric [4 + 2] annulation of methyleneindolinones with a zwitterionic oxo-1,4-dipole intermediate was successfully developed to access spirocyclic oxindoles bearing two vicinal stereocenters in good yields with highly diastereoselectivities and enantioselectivities. This strategy is featured with a broad substrate scope (28 examples), allowed for efficient scale-up. Further selective transformation of the product and preliminary mechanistic studies were well conducted.

Abstract:The first Lewis acid catalyzed asymmetric Friedel-Crafts alkylation reaction of ortho-hydroxybenzyl alcohols with C3-substituted indoles is described. A chiral N,N'-dioxide Sc(OTf)3 complex served not only to promote formation of ortho-quinone methides (o-QMs) in situ but also induced the asymmetry of the reaction. This methodology enables a novel activation of ortho-hydroxybenzyl alcohols, thus affording the desired chiral diarylindol-2-ylmethanes in up to 99 % yield and 99 % ee. A range of functional groups were also tolerated under the mild reaction conditions. Moreover, this strategy gives concise access to enantioenriched indole-fused benzoxocines.

Abstract:A palladium-catalyzed asymmetric chloroenolate allylation with vinyl benzoxazinanones under mild reaction conditions has been developed, affording a series of optically active 3,3-disubstituted oxindoles exhibiting a chloro-group and a linear aryl amino side chain in good yields with up to 96% ee. Versatile functional group tolerance on the benzene ring has been demonstrated, and the utility of this method was probed by a scale-up synthesis and highlighted by product derivatizations.

Abstract:Herein, we report an efficient scandium‐catalytic reaction protocol for the classical Fischer indole synthesis, which can tolerate a wide variety of polycyclic substrates and allow us to easily construct complex indole structures in high yields for most cases. Moreover, in order to demonstrate the reaction efficacy, we also synthesized two quinazolinone‐containing natural alkaloids, their analog, as well as an anticancer indenoindolone compound.

Abstract:Asymmetric synthesis of enantioenriched C3-arylated oxindoles bearing tetrasubstituted stereocenter has been achieved using chiral N,N′-dioxide/transition metal complex which promote Friedel–Crafts alkylation of 3-bromo-3-substituted oxindoles with arenes and heteroarenes i.e., anisole derivatives, furans, pyrroles, 2-methoxythiophene, anilines, and phenols. A series of 3-aryl-indolinones with quaternary stereocenters have been obtained up to 99% yield, 99% ee. In addition, the rationale of the reactivity is based on the nucleophilicity parameters of arenes and heteroarenes were predicted from Mayr's Database. Also, a gram-scale synthesis has been achieved and this method has been applied to synthesis of bioactive compounds. Additionally, a plausible catalytic cycle has been proposed.

Abstract:An efficient approach to access functionalized indole derivatives has been developed through Cu(OTf)2-catalyzed C3 aza-Friedel-Crafts alkylation of substituted indoles 5a-5m, N-methyl-pyrrole with linear N,O-acetals 4a-4l. As a result, a series of C3 amide aza-alkylated indole derivatives 6a-6ag and 7 were synthesized in moderate to excellent yields.

Abstract:An efficient formation of fused tricyclic [1,3]oxazino[3,4‐a]indol‐1‐ones and dihydropyrimido[1,6‐a]indol‐1(2H)‐ones via Rh(III)‐catalyzed [3+3] or [4+2] annulation of N‐methoxy‐1H‐indole‐1‐carboxamides with sulfoxonium ylides has been developed. These selective annulation reactions were carried out by switching the additives and notable features of this protocol were low catalyst loading and a broad substrate scope providing the corresponding products in up to 99% yields.

Abstract:A novel palladium-catalyzed [4+3]-annulation of oxotryptamines with allyl dicarbonates has been developed. The reaction furnishes various spiro[azepane-4,3'-oxindoles] in moderate to excellent yields (30%-96%) with up to >20:1 Z/E ratio through a tandem allylic substitution strategy. This protocol enables the synthesis of a wide range of seven-membered spirocyclic oxindoles in high Z-selectivity. In addition, the gram-scale reaction and further derivatizations were also investigated, demonstrating the synthetic utility of this approach.

Abstract:A novel palladium-catalyzed [2 + 2 + 1] annulation of alkyne-tethered aryl iodides with diaziridinone was developed, leading to the formation of 3,4-fused tricyclic indoles. From a mechanistic standpoint, the formation of fused tricyclic indole scaffolds involved C,C-palladacycles, which were synthesized through the intramolecular reaction of aryl halides and alkynes. The cascade reaction described herein could be carried out with a broad range of substrates and provided various 3,4-fused tricyclic indoles with yields up to 98%.

Abstract:Here we report a general rhodium-catalyzed asymmetric intermolecular dearomative cyclopropanation of indoles using trifluoromethyl N-triftosylhydrazones as carbene precursors. The reaction enables the rapid construction of diverse cyclopropane fused indolines bearing a trifluoromethylated quaternary stereocenter in high enantioselectivities (up to 99% ee). This mild method exhibits a broad substrate scope, is compatible with various functional groups, and can even be utilized for the late-stage diversification of complex bioactive molecules. DFT calculations suggest that the formation of a key zwitterionic intermediate is responsible for the chiral induction. Overall, this approach opens up new possibilities for asymmetric cyclopropanation of challenging aromatic heterocyclic compounds.

Abstract:The rhodium-catalyzed diastereo- and enantioselective Michael addition of arylboronic acids to 3-alkylenyloxindoles has been developed with (R)-binap as a ligand. Abstract:Palladium‐catalyzed thioesterifications of (aza)indole‐tethered aryl iodides, Mo(CO)6, and sulfonyl chlorides are reported. A series of C2 thioester‐group substituted indoline‐fused ring compounds are efficiently constructed using intramolecular Heck‐cyclization and carbonylation chemistry involving N‐(2‐iodobenzoyl)indole. The salient features of this three‐component cascade sequence include the use of solid Mo(CO)6 as the carbon monoxide source and good functional group tolerance.

Abstract:A Rh(III)-catalyzed C-H bond activation for the synthesis of fused 2H-isoindole scaffolds from oxadiazolones with diazo compounds was developed. The reaction proceeded through C-H activation of oxadiazolones/[4 + 1] annulation, intramolecular cyclization, and an unusual acyl migration cascade to afford target scaffolds with good yields. These 2H-isoindole derivatives could be further transformed into intriguing drug privileged scaffolds.
