Abstract:Asymmetric expansion: A catalytic asymmetric ring-expansion reaction of the title compounds occurs in the presence of a Sc(OTf)(3) catalyst bearing an N,N'-dioxide-based ligand. Highly functionalized 2-quinolone derivatives containing a chiral C4-quaternary stereocenter were obtained in high yields and high levels of selectivity under mild reaction conditions (see scheme; Tf=trifluoromethanesulfonyl).

Abstract:We describe here a Cu-catalyzed and 4-OH-TEMPO-mediated sequential dehydrogenation/aza-Michael addition/annulation cascade reaction for the construction of N-alkyl 2-arylindoles from facilely available saturated ketones and 2-arylethynylanilines. This reaction shows high regioselectivity and tolerates a variety of functional groups. Moreover, 3-alkyl-substituted indoles can also be achieved when using 2-alkylethynylanilines as starting materials.

Abstract:Palladium-catalyzed [4+2] annulation of aryl and alkenyl carboxamides with 1,3-dienes via C–H functionalization is developed using air as the terminal oxidant. The method demonstrates good functional group tolerance and high stereoselectivity, affording a series of 3,4-dihydroisoquinolones and 5,6-dihydropyridinones in yields of up to 99%.

Abstract:We report a palladium-catalyzed asymmetric [4+4] cycloaddition reaction between 2-alkylidenetrimethylene carbonate and electron-deficient indole-2,3-quinodimethanes (IQDMs). This reaction features exclusive regioselectivity, high yield (up to 98%), excellent enantioselectivity (up to 95% ee), and easy scale-up without any loss of efficiency, making it valuable for the synthesis of indole-fused eight-membered oxa-rings. The Pd-catalyzed asymmetric [4+4] cycloaddition reaction between 2-alkylidenetrimethylene carbonate and electron-deficient indole-2,3-quinodimethanes (IQDMs) results in the formation of indole-fused eight-membered oxa-rings with high enantioselectivity and excellent yield (up to 98% yield, 95% ee). Abstract:A base-promoted selective C2−N1 ring-expansion reaction of indolones toward substituted quinolines under mild conditions has been developed. This is the first example of transition-metal free ring-expansion reaction of indolones via the selective insertion of alkynoates into C2-N1 bond. The process achieved the cleavage of one C-N bond and three C-C bonds, and the formation of one C-N and three C-C bonds in one step.

Abstract:A novel Brønsted-acid-promoted selective C2-N1 ring-expansion reaction of indoles has been developed that provides a rapid and efficient protocol for the preparation of fused quinolines. A variety of corresponding quinolines were obtained in high yields. Controlled experiments revealed that C2-spiroindolenines might be intermediates of this C2-N1 ring-expansion reaction. The notable advantages of this process include excellent yields, good functional group tolerance, and operational simplicity.

Abstract:A Pd(II) catalyzed tandem reaction of o-alkynylbenzaldehydes or o-alkynylbenzaldimines with substituted indoles initiated by the intermolecular addition of indoles to the carbonyl or imine group followed by the nucleopalladation of an intramolecular alkyne and quenching the carbon-palladium bond by protonolysis to regenerate the Pd(II) species was developed. The reaction can be carried out under mild conditions without the necessity of a redox system.

Abstract:A palladium-catalyzed aromatic dual C-H acylations followed with intramolecular cyclizations have been developed by the assistance of bidentate N-(quinolin-8-yl)benzamide. This tandem process involves the formation of three new chemical bonds, providing access to novel quinoline-substituted hydroxyl isoindolones skeleton under simple reaction conditions. The deuterium-labeled competition reaction has revealed that C-H bond cleavage is the turnover limiting step.

Abstract:A novel method for the construction of a cyclopenta[c]quinoline ring via cyclization of 3-bromoindoles with internal alkynes in the presence of palladium is described. The formation of the cyclopenta[c]quinoline ring is proposed from a double [1,5] carbon sigmatropic rearrangement of the spirocyclic cyclopentadiene intermediate, which is generated in situ from the cyclization of 3-bromoindoles with internal alkynes involving a sequential double alkyne insertion into the carbon-palladium bond and dearomatization of indole. The present studies have developed a novel ring-expansion reaction of the pyrrole ring to pyridine via one carbon insertion into the C2-C3 bond of indoles and provided a simple and distinct route for the construction of tricyclic fused-quinoline derivatives that are not easy to access with conventional methods.

Abstract:A cobalt‐catalyzed annulation of o‐phenylenediamines and internal alkynes for the synthesis of quinoxalines is developed. This method provides a variety of quinoxalines in good to high yields under a simple and mild reaction conditions with molecular O2 as a terminal oxidant. Preliminary mechanistic studies show that a novel reaction mechanism is involved in the current reaction, which is different from the previous reported dicarbonyl compound mechanism via the oxidation of the alkyne.

Abstract:We describe a selective preparation of a variety of functionalized pyrrolo[2,3‐c]tetrahydroquinolines and indolo[3,2‐c]tetrahydroquinolines in 40‐82% and 40‐80% yields from alkynyl‐tethered oximes with diaryliodonium triflates, respectively. The one‐pot reaction underwent N‐arylation, intramolecular [3+2] cycloaddition, selective [1.3] or [3,3]‐rearrangement of N‐O bond in total three steps. Experimental studies revealed that EtOAc solvent and copper(II)‐catalyst played crucial roles on the formation of these two tetrahydroquinoline scaffolds. Moreover, these two tetrahydroquinoline scaffolds could be converted into various tetrahydroquinoline building blocks by further transformations.

Abstract:An efficient indium-mediated cascade annulation reaction of 2-azidoaryl aldehydes with propargyl bromides is reported. The aromatic 5/6/6-fused heterocycles, [1,2,3]triazolo[1,5-a]quinoline derivatives, could be constructed in one pot in moderate yields with a broad substrate scope. Mechanistic studies indicated that the reaction proceeded through allenol formation, azide-allene [3 + 2] cycloaddition, and dehydration. The synthetic potential of the products including the denitrogenative functionalization and the Pd-catalyzed coupling reactions has also been explored.

Abstract:An efficient oxidative cross-coupling reaction between 1,3-diarylpropenes and indoles in the presence of palladium chloride was achieved with 2,3-dichloro-5,6-dicyanoquinone (DDQ) as oxidant. The reaction afforded 1,3-diphenylallylindoles in moderate to high yields under mild conditions, thus providing a novel methodology to synthesize the respective products.

Abstract:An attractive palladium-catalyzed three-component reaction of <i>ortho</i>-amino aryl diazo esters, allyl carboxylates, and carbon monoxide (CO) has been developed. This catalytic system rendered domino carbene migratory insertion and carbonylation. Remarkably, 2-indolones <b>3</b> with a C3 all-carbon quaternary center can be selectively obtained in good to excellent yields via one-pot synthesis, in which two different C-C bonds and one C-N bond were formed in a straightforward manner.

Abstract:We disclosed an efficient protocol for regioselective C6 C‐H/N‐H activation/annulation reaction of indole‐7‐carboxamides with alkynes to synthesize highly substituted pyrrolo[3,2‐h]isoquinolin‐9‐one derivatives. Under optimized reaction conditions, electron‐deficient and electron‐rich internal alkynes reacted efficiently with various indole‐7‐carboxamides to deliver desired products in good to excellent yields. The synthetic utility of the product is demonstrated by its selective oxidation to the corresponding isatin derivative. Deuterium insertion and intermolecular competition experiments were also conducted to gain mechanistic insights.
