Abstract:A chemical investigation of Jaspis sp., the marine sponge collected from the South China Sea led to the isolation of six new isomalabaricane-type compounds, jaspolides A-F (1-6). The structures of those compounds were elucidated by extensive spectroscopic methods. The structure-types of 1 to 6 could be classified into triter-penes (1, 2), sesterterpene (6), diterpenes (3, 4), and nortriterpene (5). The biogenetic transformation of the isolated compounds was also speculated.

Abstract:A chemical investigation of marine sponge Jaspis stellifera, collected from the South China Sea, led to the isolation of four new isomalabaricane-type triterpenoids, jaspiferins C–F (1–4). The structures of those compounds were elucidated by extensive spectroscopic methods. Jaspiferin C (1), which has the six-membered carbon ring at the side chain, was discovered for the first time from the isomalabaricane-type triterpenoids. The hypothesis of a biogenetic pathway to generate jaspiferin C (1) was depicted.

Abstract:A chemical investigation of marine sponge Jaspis stellifera, collected from South China Sea, led to the isolation of two new compounds, Jaspiferin A and B (1-2), and six known compounds, gibepyrone F (3), p-hydroxy benzaldehyde (4), 3-Indole-3-aldehyde (5), Thymine (6), 24(28)-dehydroaplysterol (7), (25s)-26-methylene-cholest-4-en-3-one (8). Their structures were determined by extensive spectroscopic analysis in association with physical and chemical properties, as well as comparison of their spectral data with these reported in literatures. The biogenetic transformation of compound 2 was also speculated.

Abstract:Two new jaspamide derivatives 2 and 3, together with the parent compound jaspamide (1) have been isolated from the marine sponge Jaspis splendens collected in Kalimantan (Indonesia). The structures of the new compounds were unambiguously elucidated based on 1D and 2D NMR spectral data, mass spectrometry and comparison with jaspamide (1). The new derivatives inhibited the growth of mouse lymphoma (L5178Y) cell line in vitro with IC50 values of < 0.1 mu g/mL.

Abstract:A new acylic jasplakinolide congener (2), another acyclic derivative requiring revision (4), together with two jasplakinolide derivatives including the parent compound jasplakinolide (1) were isolated from the Indonesian marine sponge Jaspis splendens. Abstract:Chemical examination of the Chinese marine sponge Xestospongia testudinaria led to the isolation of 39 new brominated polyunsaturated compounds, which were designated with the trivial names xestospongienes A–Z and Z1–Z13. The structures of these compounds were elucidated by analyzing extensive spectroscopic (IR, MS, 1D, and 2D NMR) data and the results from CD spectral assignment, chemical conversion, and Mosher reaction.

Abstract:Jaspolide B is a novel isomalabaricane-type triterpene isolated from sponge Jaspis sp. We investigated its effects on human hepatoma cells in this study. After 48 h of incubation, jaspolide B inhibited the growth of Bel-7402 and HepG2 cells with IC50 values of 29.1 and 29.5 μM, respectively. Incubation with 0.5 μM of jaspolide B caused time-dependent induction of apoptosis in up to 66.8% of Bel-7402 cells for 48 h, and the induction of apoptosis was confirmed by the enhancement of mitochondrial masses and cell membrane permeability, and nuclear condensation in Bel-7402 and HepG2 cells. Moreover, jaspolide B arrested cell cycle progression at G1 phase of human hepatoma cells in a dose- and time-dependent manner. In addition, treatment of the compound caused dose-dependent disassembly of microtubule cytoskeleton in Bel-7402 cells at indicated concentrations, and this effect being similar but weaker than that of colchicine, a well-known microtubule-disassembly agent. We conclude that the anti-cancer effect of jaspolide B relates to the apoptosis induction, cell cycle arrest and microtubule disassembly, and these multiple mechanisms of jaspolide B, especially the induction of apoptosis, open interesting perspectives for further exploration of the isomalabaricane-type terpenes and compounds of marine sponge origin as potential anticancer agents.
