Abstract:Chemical examination of a Chinese sponge Axinyssa sp. resulted in the isolation of 12 new formamidobisabolene-based analogues named axinyssines A–L (1–12), along with a new natural product (13) and three known formamidobisabolenes (14–16). Their structures were determined on the basis of extensive NMR and mass spectroscopic analyses in association with ICD data, Mosher reaction and chemical conversion for the assignment of absolute configurations. All compounds were evaluated for antitumor and antimicrobial activities.

Abstract:Chemical examination of the Chinese marine sponge Xestospongia testudinaria resulted in the isolation and characterization of twelve new brominated acetylenic acids, namely xestospongienols A-L (1a-1d, 2a-2d, and 3a-3d), together with the known analog 4. The structures of xestospongienols A-L were determined on the basis of 1D- and 2D-NMR, IR, and MS analysis in association with the modified Mosher method. The structural patterns are characteristic of brominated unbranched C(16)-alkyl chains containing polyene and acetylene moieties.

Abstract:The precursor ion scanning-supercritical fluid chromatography (PI-SFC) method was applied to explore new methionine sulfoxide-containing cycloheptapeptides, axinellasins A-D (1-4), from the marine sponge Axinella sp. Their structures, including absolute configurations, were elucidated by detailed spectroscopic analyses and X-ray crystallography. The total synthesis of 4 was completed via an Fmoc solid/solution-phase synthesis. Compounds 1-4 exhibited immunosuppressive effects via inhibition of T and B cell proliferation, and 1 and 4 showed better inhibitory activities than their corresponding diastereomers.

Abstract:A new scalarane sesterterpene, phyllofolactone M (1), and a new sterol, (24E)-5 alpha,6 alpha-epoxystigmasta-7,24(28)-dien-3 beta-ol (3), together with a known sesterterpene, phyllofolactone B (2), were isolated from the South China Sea sponge Phyllospongia foliascens. Their structures were elucidated by spectroscopic analysis and comparison with known compounds. In addition, previous NMR data assignments for the known sesterterpene phyllofolactone B (2) were revised.

Abstract:One new naphthalene derivative, 1,8-dihydroxynaphthol-1-O-α-l-rhamnopyranoside (1), together with one known α-dibenzopyrone, alternariol (2), and five xanthones (3-7) were isolated from the sponge-derived fungus Arthrinium sp. ZSDS1-F3. All the isolated compounds (1-7) were established by comprehensive analysis of the spectral data, especially 1D and 2D NMR (HMQC and HMBC) spectra. In the primary bioassays, compound 3 exhibited moderate COX-2 inhibition, with IC50 values of 12.2 μM.

Abstract:Seven new formamido-diterpenes, cavernenes A–D (1–4), kalihinenes E and F (5–6), and kalihipyran C (7), together with five known compounds (8–12), were isolated from the South China Sea sponge Acanthella cavernosa. Structures were established using IR, HRESIMS, 1D and 2D NMR, and single X-ray diffraction techniques. The isolated compounds were assessed for their cytotoxicity against a small panel of human cancer cell lines (HCT-116, A549, HeLa, QGY-7701, and MDA-MB-231) with IC50 values in the range of 6–18 μM. In addition, compound 9 showed weak antifungal activity against Trichophyton rubrum and Microsporum gypseum with MIC values of 8 and 32 μg/mL, respectively, compound 10 displayed weak antifungal activity against fungi Candida albicans, Cryptococcus neoformans, T. rubrum, and M. gypseum with MIC values of 8, 8, 4, and 8 μg/mL, respectively.

Abstract:Four new bisabolane-type sesquiterpenoids, aspergiterpenoid A (1), (-)-sydonol (2), (-)-sydonic acid (3), and (-)-5-(hydroxymethyl)-2-(2',6',6'-trimethyltetrahydro-2H- pyran-2-yl)phenol (4) together with one known fungal metabolite (5) were isolated from the fermentation broth of a marine-derived fungus Aspergillus sp., which was isolated from the sponge Xestospongia testudinaria collected from the South China Sea. Four of them (1-4) are optically active compounds. Their structures and absolute configurations were elucidated by using NMR spectroscopic techniques and mass spectrometric analysis, and by comparing their optical rotations with those related known analogues. Compounds 1-5 showed selective antibacterial activity against eight bacterial strains with the MIC (minimum inhibiting concentrations) values between 1.25 and 20.0 µM. The cytotoxic, antifouling, and acetylcholinesterase inhibitory activities of these compounds were also examined.

Abstract:One new aromadendrane-type sesquiterpenoid, namely ximaocavernosin P [(+)-1], and three new cadinane-type sesquiterpenoids, namely (+)-maninsigin D [(+)-4], (+)- and (-)-ximaocavernosin Q [(+)- and (-)-5], together with five related known ones [2, 3, (-)-4, 6, and 7], were isolated from the Hainan sponge Acanthella cavernosa. Compounds 4 and 5 were isolated as racemic forms, which were further separated to the corresponding enantiomers [(+)-4/(-)-4 and (+)-5/(-)-5], respectively, by using chiral-phase HPLC. The structures of new compounds were elucidated by extensive spectroscopic analysis and comparison with the reported data. In addition, the absolute configuration of optically pure (+)-1 and 2 were determined by time-dependent density functional theory/electronic circular dichroism (TDDFT-ECD) calculations or X-ray diffraction analysis. A plausible biosynthetic pathway of these sesquiterpenoids and their internal correlation were proposed and discussed. In an in vitro bioassay, (+)-aristolone (3) exhibited promising anti-inflammatory activity by the inhibition of LPS-induced TNF-α and CCL2 release in RAW 264.7 macrophages.

Abstract:A chemical investigation of marine sponge Jaspis stellifera, collected from South China Sea, led to the isolation of two new compounds, Jaspiferin A and B (1-2), and six known compounds, gibepyrone F (3), p-hydroxy benzaldehyde (4), 3-Indole-3-aldehyde (5), Thymine (6), 24(28)-dehydroaplysterol (7), (25s)-26-methylene-cholest-4-en-3-one (8). Their structures were determined by extensive spectroscopic analysis in association with physical and chemical properties, as well as comparison of their spectral data with these reported in literatures. The biogenetic transformation of compound 2 was also speculated.

Abstract:Two new xanthone derivatives, 12-dimethoxypinselin (1) and 12-O-acetyl-AGI-B4 (2), as well as two new phenolic bisabolane-type sesquiterpenes, 11,12-dihydroxysydonic acid (15) and 1-hydroxyboivinianic acid (16), together with one new alkaloid, scopulamide (21) and one new α-pyrone derivative, scopupyrone (26), in addition to twenty-three known compounds (3–14, 17–20, 22–25, 27–29) were isolated from solid rice cultures of the marine-derived fungus Scopulariopsis sp. obtained from the Red Sea hard coral Stylophora sp. All compounds were unambiguously identified through extensive NMR spectroscopic analyses, and by comparison with the literature. Marfey's reaction was performed to determine the absolute configuration of scopulamide (21) and TDDFT-ECD calculations were used to assign the configuration of AGI-B4 (3) and scopupyrone (26). All isolated compounds were evaluated for their cytotoxicity against L5178Y mouse lymphoma cells and the structure-activity relationships were discussed.

Abstract:Three new scalarane sesterterpenes, phyllofolactone L(1), phyllofenone D (2) and phyllofenone E (3), were isolated from the acetone extract of the South China Sea sponge Phyllospongia foliascens. Their structures were elucidated on the basis of spectroscopic analysis. Phyllofenone D (2) was cytotoxic against the P388 leukemia cell line with an IC50 value of 6.5 mu/ml.

Abstract:The chemical screening of an algae-containing marine sponge identified as Lendenfeldia species has isolated two 24-homoscalarane sesterterpenoids, including a known compound, felixin A ( 1 ) and a new analogue, lendenfeldarane V ( 2 ). The structure of 1 , obtained in a previous study, was cited for the first time in this study via single-crystal X-ray diffraction analysis. The structure of 2 was ascertained via 2D NMR experiments and a literature review. The absolute configurations of 1 and 2 were delineated with DP4+ computation and specific optical rotation. Homoscalarane 1 exhibited cytotoxicity towards MG63 human osteosarcoma cells.

Their structures were deter-mined by analyses of extensive NMR data and HRESI-MS. Abstract:Chemical investigation of the marine sponge Agelas sp. collected from the South China Sea resulted in the isolation of four new dimeric bromopyrrole alkaloids, including hexazosceptrin (1) with a rare cyclohexane-fused-cyclobutane skeleton, agelestes A and B (2–3), the sceptrin analogues without any aminoimidazole substitution, and (9S, 10R, 9′S, 10′R)-nakamuric acid (4), of which the absolute configuration was confirmed for the first time. The structure elucidation and absolute configuration assignments were unambiguously determined via extensive spectroscopic analyses and the comparison of experimental and calculated electronic circular dichroic spectra. Compounds 1 and 4 exhibited moderate antimicrobial activity.

Abstract:Chemical examination of a marine sponge Xestospongia sp. resulted in the isolation of 20 sterol derivatives (1 – 20), including eight new sterols namely aragusterols J – L (1 – 3), (5α,7α,12β,22E)‐7,12,18‐trihydroxystigmast‐22‐en‐3‐one (4), (5α,7α,12β,24R)‐ and (5α,7α,12β,24S)‐7,12,20‐trihydroxystigmastan‐3‐one (5/6), and (5α,7α,12β,22E,24R)‐ and (5α,7α,12β,22E,24S)‐7,12,20‐trihydroxyergost‐22‐en‐3‐one (7/8). The structures of new compounds were determined through extensive spectroscopic analyses and chemical conversion. The sterol diversity was mainly characterized by the presence of a cyclopropane unit at side chain, while compound 4 with 18‐hydroxymethyl group was found in stigmasterol family for the first time. Cytotoxic test revealed the inhibitory effects of compounds 1, 4, and 17 against human leukemia cell line K562 with IC50 values of 18.3, 24.1, and 34.3 μm, respectively.
