Abstract:As a kind of facile tool, microfluidic paper-based analytical devices (μPADs) have been widely used in analytical and biomedical fields. However, because we lack the ability to control the continuous perfusion of these devices, they are not generally used in fields that require continuous flow, especially biofluidics fields such as cell culturing, drug screening, and organs on chips. In this study, we designed a novel, low-cost, and compact platform that can be used to control the continuous perfusion of μPADs. As most of the parts of this platform can be created using a three-dimensional (3D) desktop printer, our platform can be easily duplicated by other researchers. We demonstrate that with our system, μPADs can be promising paper-based biofluidic platforms for cell culturing and drug screening.
Fabrication of paper-based microfluidic devices by octadecyltrichlorosilane self-assembling and uv-patterning

Abstract:Paper-based microfluidic devices are emerging as a new technology for simple, low-cost, portable, and disposable diagnostic and monitoring platform. This article presents a novel method for fabricating microfluidic devices on paper by means of octadecyltrichlorosilane self-assembling and UV/O3-patterning. The hydrophilic filter paper was uniformly coated with a hydrophobic layer of octadecyltrichlorosilane (OTS) by dipping the paper into an OTS-hexane solution. The coated OTS layer was then treated with UV/O3 through a patterned quartz mask, generating hydrophobic barriers that define hydrophilic fluid channels, reservoirs and reaction zones. Contact angle measurements, XPS and ATR-FT-IR spectra confirmed that OTS self-assembled layer (OTS-SAL) was chemically immobilized on the filter paper surface and that the monolayer was degraded after UV/O3 treatment. The widths of the prepared hydrophilic channels and hydrophobic barriers could be as small as 80 μm and 130 μm, respectively. Colorimetric assays of nitrite ions were demonstrated with the developed paper-based microfluidic devices.

Abstract:Despite that microfluidic paper-based analytical devices (μPADs) provide effective analytical platforms for point-of-care diagnosis in resource-limited areas, it remains challenging to achieve simple and low-cost fabrication of μPADs. A novel method for fabrication of μPADs is developed in this study using a folded polydimethylsiloxane (PDMS)-coated paper mask with a specific pattern to form a sandwich structure with inserted chromatographic paper. PDMS penetrates the target paper from the front and the back sides, and then is cured in the target paper to form legible channels. This method for prototyping μPADs has many favorable merits including simple operation without the need of trained personnel, fast fabrication and low cost. We further investigated colorimetric detection of melamine in the μPADs, and it showed a remarkable measurement with a detection limit of 0.1 ppm in aqueous solutions and liquid milk discriminated by the naked eye, which meets the detection limit required by USA and China. The fabricating strategy developed in this study is very promising and attractive for the development of simple μPADs for point-of-care applications, including diagnostic testing, food safety control and environmental monitoring.

Abstract:When compared with conventional microfluidic chips made of glass and polymer substrates, paper-based microfluidic analysis devices (mu PADs) possess many unique advantages, including low-cost, easy-to-fabricate, strong capillary action and good biological compatibility. In recent years, mu PADs have attracted increased interest and attention, which has led to their rapid development. Thousands of literature reports regarding mu PADs have been published and a variety of mu PADs fabrication methods have been reported. This review focuses on the development of the fabrication methods of 2D and 3D mu PADs since 2007. A summary of the advantages and disadvantages of these methods is provided with particular attention paid to the resolution and cost of each method. Suitable applications of each method are discussed. Also, some trends of mu PADs are summarized.

Abstract:As a pump-free and lightweight analytical tool, paper-based microfluidic analytical devices (μPADs) attract more and more interest. If the flow speed of μPAD can be programmed, the analytical sequences could be designed and they will be more popular. This reports presents a novel μPAD, driven by the capillary force of cellulose powder, printed by a desktop three-dimensional (3D) printer, which has some promising features, such as easy fabrication and programmable flow speed. First, a suitable size-scale substrate with open microchannels on its surface is printed. Next, the surface of the substrate is covered with a thin layer of polydimethylsiloxane (PDMS) to seal the micro gap caused by 3D printing. Then, the microchannels are filled with a mixture of cellulose powder and deionized water in an appropriate proportion. After drying in an oven at 60 °C for 30 min, it is ready for use. As the different channel depths can be easily printed, which can be used to achieve the programmable capillary flow speed of cellulose powder in the microchannels. A series of microfluidic analytical experiments, including quantitative analysis of nitrite ion and fabrication of T-sensor were used to demonstrate its capability. As the desktop 3D printer (D3DP) is very cheap and accessible, this device can be rapidly printed at the test field with a low cost and has a promising potential in the point-of-care (POC) system or as a lightweight platform for analytical chemistry.

Abstract:A novel and facile fabrication method of microfluidic paper-based analytical devices (mu PADs) with flash foam stamp lithography (FFSL) is presented in this paper. First, a flash foam(also called photosensitive seal) stamp with desired patterns is made by flash exposing. Next, the stamp is immersed in a hydrophobic solvent such as polydimethylsiloxane (PDMS) to absorb the ink. Finally, the hydrophobic solvent is stamped on filter paper to form hydrophobic barriers. After the hydrophobic solvent cures, the mu PAD is complete. Compared to common fabrication methods such as wax printing, inkjet printing, or direct writing, this paper will demonstrate that the FFSL method is convenient, quick, and cheap.

Abstract:Microfluidic paper-based chip analysis is a burgeoning microfluidic technique. It possesses great potentials for application in clinical diagnosis, food quality control as well as environmental monitoring due to its attractive features such as low-cost, easy-to-fabricate, easy-to-use and portable. Its importance and utility are widely acknowledged and extensive research has been conducted in the past several years. This paper mainly aims to review the developed techniques for fabrication of microfluidic paper-based chips, including UV photolithography, wax printing, plasma treatment, ink printing, ink jet etching, plotting, screen printing, flexography prinfing and laser treatment, and so on. The detection methods for the microfluidic paper-based chip analysis and applications of microfluidic paper-based chips are also reviewed.

Abstract:This work presents a novel and facile method for fabricating paper-based microfluidic devices by means of coupling of hydrophobic silane to paper fibers followed by deep UV-lithography. After filter paper being simply immersed in an octadecyltrichlorosilane (OTS) solution in n-hexane for 5 min, the hydrophilic paper became highly hydrophobic (water contact angle of about 125 degrees) due to the hydrophobic OTS molecules were coupled to paper's cellulose fibers. The hydrophobized paper was then exposed to deep UV-lights through a quartz mask that had the pattern of the to-be-prepared channel network Thus, the UV-exposed regions turned highly hydrophilic whereas the masked regions remained highly hydrophobic, generating hydrophilic channels, reservoirs and reaction zones that were well-defined by the hydrophobic regions. The resolution for hydrophilic channels was 233 +/- 30 mu m and that for between-channel hydrophobic barrier was 137 +/- 21 mu m. Contact angle measurement, X-ray photoelectron spectroscopy (XPS) and attenuated total reflectance Fourier transform-infrared (ATR-FT-IR) spectroscopy were employed to characterize the surface chemistry of the OTS-coated and UV/O-3-treated paper, and the related mechanism was discussed. Colorimetric assays of nitrite are demonstrated with the developed paper-based microfluidic devices.

Abstract:Abstract State-of-the-art microfluidic systems rely on relatively expensive and bulky off-chip infrastructures. The core of a system—the microfluidic chip—requires a clean room and dedicated skills to be fabricated. Thus, state-of-the-art microfluidic systems are barely accessible, especially for the do-it-yourself (DIY) community or enthusiasts. Recent emerging technology—3D-printing—has shown promise to fabricate microfluidic chips more simply, but the resulting chip is mainly hardened and single-layered and can hardly replace the state-of-the-art Polydimethylsiloxane (PDMS) chip. There exists no convenient fluidic control mechanism yet suitable for the hardened single-layered chip, and particularly, the hardened single-layered chip cannot replicate the pneumatic valve—an essential actuator for automatically controlled microfluidics. Instead, 3D-printable non-pneumatic or manually actuated valve designs are reported, but their application is limited. Here, we present a low-cost accessible all-in-one portable microfluidic system, which uses an easy-to-print single-layered 3D-printed microfluidic chip along with a novel active control mechanism for fluids to enable more applications. This active control mechanism is based on air or gas interception and can, e.g., block, direct, and transport fluid. As a demonstration, we show the system can automatically control the fluid in microfluidic chips, which we designed and printed with a consumer-grade 3D-printer. The system is comparably compact and can automatically perform user-programmed experiments. All operations can be done directly on the system with no additional host device required. This work could support the spread of low budget accessible microfluidic systems as portable, usable on-the-go devices and increase the application field of 3D-printed microfluidic devices.

Microfluidic Time-Delay Valve Mechanism on Paper-Based Devices for Automated Competitive ELISA

Abstract:Paper-based technologies have been drawing increasing attentions in the biosensor field due to their economical, ecofriendly, and easy-to-fabricate features. In this paper, we present a time-delay valve mechanism to automate a series of procedures for conducting competitive enzyme-linked immunosorbent assay (ELISA) on a paper-based device. The mechanism employs a controllable time-delay valve, which has surfactants to dissolve the hydrophobic barriers, in a fluid pathway. The valves can regulate the liquid and sequentially deliver the sample flow for automating ELISA procedures in microchannels. Competitive ELISA is achieved in a single step once the sample, or small molecule pesticide (e.g., Imidacloprid), is applied onto the paper-based device with a comparable sensitivity to plate-based competitive ELISA. The results further demonstrate the appositeness of using paper-based devices with the valve designs for on-the-go ELISA detection in agriculture and biomedical applications.

Abstract:Since the monumental work conducted by Whitesides et al. in 2007, research and development of paper-based microfluidics has been widely carried out, with its applications ranging from chemical and biological detection and analysis, to environmental monitoring and food-safety inspection. Paper-based microfluidics possesses several competitive advantages over other substrate materials, such as being simple, inexpensive, power-free for fluid transport, lightweight, biodegradable, biocompatible, good for colorimetric tests, flammable for easy disposal of used paper-based diagnostic devices by incineration, and being chemically modifiable. Myriad methods have been demonstrated to fabricate paper-based microfluidics, such as solid wax printing, cutting, photolithography, microembossing, etc. In this study, fabrication of paper-based microfluidics was demonstrated by spray on the printed paper. Different from the normally used filter papers, printing paper, which is much more accessible and cheaper, was utilized as the substrate material. The toner was intended to serve as the mask and the patterned hydrophobic barrier was formed after spray and heating. The processing parameters such as toner coverage on the printing paper, properties of the hydrophobic spray, surface properties of the paper, and curing temperature and time were systematically investigated. It was found that, after repetitive printing four times, the toner was able to prevent the hydrophobic spray (the mixture of PDMS and ethyl acetate) from wicking through the printing paper. The overall processing time for fabrication of paper-based microfluidic chips was less than 10 min and the technique is potentially scalable. Glucose detection was conducted using the microfluidic paper-based analytical devices (μPADs) as fabricated and a linear relationship was obtained between 1 and 10 mM.

Abstract:A wide range of applications are possible with paper-based analytical devices, which are low priced, easy to fabricate and operate, and require no specialized equipment. Paper-based microfluidics offers the design of miniaturized POC devices to be applied in the health, environment, food, and energy sector employing the ASSURED (Affordable, Sensitive, Specific, User-friendly, Rapid and Robust, Equipment free and Deliverable to end users) principle of WHO. Therefore, this field is growing very rapidly and ample research is being done. This review focuses on fabrication and detection techniques reported to date. Additionally, this review emphasises on the application of this technology in the area of medical diagnosis, energy generation, environmental monitoring, and food quality control. This review also presents the theoretical analysis of fluid flow in porous media for the efficient handling and control of fluids. The limitations of PAD have also been discussed with an emphasis to concern on the transformation of such devices from laboratory to the consumer.

With customized circuits, the platform takes the advantages of portability and easy-application with a friendly user interface. It can manipulate droplet moving, merging and mixing through an application (App) on android smartphone. Abstract:In this report, we describe a simple, low-cost, straight forward and highly reproducible fabrication method of microfluidic systems. This system was cut on a glass fiber membrane by a common cutter without using any other sophisticated equipment or organic solvents. This format represents a novel type of paper-based microfluidics with high resolution of the microchannel down to similar to 137 mu m, comparable to those made by conventional photolithography. We successfully applied this method to microfluidics to create a star micro-array format of multiplexed urine tests in this study.

Abstract:Background: The microfluidic paper-based analytical devices (μPADs) have been highly regarded as effective tools that offer a cost-effective and portable solution for point-of-care testing (POCT) and on-site detection. Utilizing paper substrates such as cellulose and nitrocellulose membranes, μPADs have proven beneficial for a range of applications from medical diagnostics to environmental monitoring. Despite their advantages, the fabrication of μPADs often requires sophisticated techniques and equipment, posing challenges for widespread adoption, especially in resource-limited settings. This study addresses the need for a simplified, low-cost method for fabricating μPADs that is accessible without specialized training or equipment. Results: This research introduces a novel, efficient method for producing μPADs using 3D-printed slidable chambers and super glue vapor, bypassing traditional, more complex fabrication processes. The method utilizes super glue (ethyl-cyanoacrylate) vapor to create hydrophobic barriers on paper substrates. By optimizing the exposure sequence to super glue and water vapors and the heating conditions, we achieved rapid hydrophobization within 5 min, creating effective hydrophobic barriers and hydrophilic channels on paper substrates. The technique's simplicity allows for use by individuals without specialized training. The practical application of the fabrication method is demonstrated by the fabrication of μPADs that can detect multiple target analytes. We perform the simultaneous detection of glucose, proteins, and also the simultaneous detection of heavy metal ions nickel (Ni2+) and copper (Cu2+), highlighting its potential for broad applications in point-of-care diagnostics. Significance: This study is the first to report a method for selective exposure of ethyl-cyanoacrylate vapor for the fabrication of μPADs. This method significantly reduces the complexity, time, and fabrication cost, making it feasible for use in various settings. It also eliminates the need for specialized equipment and can be executed by individuals without specialized training. We believe that the proposed fabrication method contributes to the wider adoption and deployment of μPADs across various sectors.

Abstract:Controlling the spontaneous directional transport of droplets plays an important role in the application of microchemical reactions and microdroplet detection. Although the relevant technologies have been widely studied, the existing spontaneous droplet transport strategies still face problems of complex structure, single function, and poor flexibility. Inspired by the spontaneous droplet transport strategy in nature, an asymmetric wettability surface with microcone channels (AWS-MC) is prepared on a flexible fabric by combining surface modification and femtosecond laser manufacturing technology. On this surface, the capillary force and Laplace pressure induced by the wettability gradient and the geometric structure gradient drive the droplet transport from the hydrophobic surface to the hydrophilic surface. Notably, droplets in adjacent hydrophilic regions do not exchange substances even if the gap in the hydrophilic region is only 1 mm, which provides an ideal platform for numerous detections by a single drop. The droplet transport strategy does not require external energy and can adapt to the manipulation of various droplet types. Application of this surface in the blood of organisms is demonstrated. This work provides an effective method for microdroplet-directed self-transport and microdroplet detection.
