Abstract:Many [1, 2, 4] triazolobenzoheteroazepine derivatives show interesting biological activities. In this paper, the geminal arylazo isocyanato compounds 3 were prepared from chroman-4-ones in three steps, i.e. condensation with arylhydrazine, cycloaddition of the resulting hydrazone with HNCO, and oxidative ring cleavage by KMnO4. Compounds 3 were allowed to react with HBF4, producing the respective 3-spiro substituted 1-aryl-4, 5-dihydro-5-oxo-3H-1, 2, 4-triazolium salts 4. The concurrent rearrangement of compound 4 with insertion of the nitrogen atom into the carbon skeleton provided, after the basic work-up, 2-oxo-[1, 2, 4] triazolo [3, 2-d] [1, 5] benzoxazepines 5a-5h. An unambigous structutal proof was obtained from the X-ray diffraction analysis for compound 5h.

Abstract:A series of novel tricyclic O,N-heterocycles, [1,2,4]triazolo[3,2-d][1,5]benzoxazepin-2-thiones 7 were achieved via acid-induced ring closure of the geminal arylazo isothiocyanate compounds 5 which were derived from substituted chroman-4-ones, followed by feasible ring expansion with simultaneous insertion of the nitrogen atom into the carbon skeleton. The X-ray crystal structure of 7d was also described.

Abstract:The synthesis of a series of unprecedented tricyclic heterocycles, 1,2,4-triazolo[3,2-d][1,5]benzothiazepine 6-oxides (5a-i), was accomplished via cycloaddition of the allene-like cations 3, derived from thiochroman-4-one arylhydrazones, to the triple bond of nitriles and ensuing rings expansion annulation. The structure of one product (5h) has been unambiguously determined by X-ray diffraction analysis.

Abstract:Several new 6-(1-aryl-5-methyl-1, 2, 3-triazol-4-yl)-3-(4-pyridyl)-s-triazolo[3,4-b]-1, 3, 4-thiadiazoles 3a-j have been synthesized by the condensation of 4-amino-5-mercapto-3-(4-pyridyl)-1, 2, 4-triazole 2 with 1-aryl-4-carboxy-5-methyl-1, 2, 3-triazoles 1a-j in the presence of phosphorus oxychloride. The structures of the products obtained are characterized by elemental analyses and spectral data and the fragmentation pattern of the mass spectra of 3a-j is discussed. 3a-j have been screened for their antibacterial activity against B. subtilis and E. coli.

Abstract:Two new series of tricyclic heterocycles, namely 5,6-dihydro-4H-benzo[b][1,2,4]triazolo[1,5-d][1,4]diazepinium salts 10 and the related neutral, free bases 13 were synthesized from 4-acetoxy-1-acetyl-4-phenylazo-1,2,3,4-tetrahydroquinolines 8 and nitriles 9 in the presence of aluminium chloride by the [3+ + 2]-cycloaddition reaction of the in situ generated azocarbenium intermediates 14 followed by a ring-expansion rearrangement. In the rearrangement reaction, the phenyl substituent in the initially formed spiro-triazolium adducts 16 underwent a [1,2]-migration from C(3) to the electron-deficient N(2). This led to the ring expansion from 6-membered piperidine to 7-membered diazepine furnishing the tricyclic 1,2,4-triazole-fused 1,4-benzodiazepines.

Abstract:A synthetic approach to the previously unknown thieno[2,3-f][1,2,4]triazolo[1,5-a]azepine derivatives was efficiently accomplished by cycloaddition of the bicyclic 1-aza-2-azoniaallene salts 6, derived from 6,7-dihydrobenzo[b]thiophen-4(5H)-one hydrazones 4, to the triple bond of nitriles, followed by ring enlargement. The X-ray crystal diffraction analysis of the picrate 10a provided affirmative proof for the structural assignment made.

Abstract:The novel tricyclic heterocycles of 4,5-dihydro[ 1,2,4]triazolo[3,2-d][1,5]benzoxazepine derivatives were prepared by the cycloaddition of the corresponding bicyclic cationic 1,3-dipoles, which were easily generated from the azoacetates by reaction with AlCl3 as a Lewis acid, to the triple bond of nitriles along with the consecutive ring expansion. The formation of products deviating from the normal reaction aptitudes was observed and the mechanistic aspects are discussed herein. Crystal diffraction analysis for two of the products unequivocally established the respective structures proposed.

Abstract:This paper reports our recent efforts to develop novel tricycles based on 4H-benzo[1,4]thiazin-3-one (2) and 1,1-dioxo-1,4-dihydro-2H- 1 lambda(6)-benzo[1,4]thiazin-3-one (3) using 1,5-difluoro-2,4-dinitrobenzene (1). All of these tricycles integrate two privileged structures into one skeleton, including 3,8-dihydro-5-thia-1,3,8tri aza-cyclopenta [b] naphthalene-7 -one (4, 10, 12), 5,5-dioxo-3,5,6,8-tetrahydro-5 lambda(6) -thia- 1,3,8-triaza-cyclopenta [b] naphthalene-7 -one (5, 11),, 8 7 dihydro-5 -thia- 1,2,3,8 -tetrqaza-cyclopenta[b] naphthalen-7 -one (6), 5,5-dioxo-3,5,6,8-tetrahydro-5 lambda(6) -thia- 1,2,3,8 -tetraaza-cyclopenta[b] naphthalene-7 -one (7), 3,8-dihydro-1H- lambda 6-thi, 5-thia- 1,3,8-triaza-cyclopenta[b] naphthalene 2,7-dione (8), and 5,5-dioxo-3,5,6,8-tetrahydro-1H-5 lambda(6) -thia-1,3,8-triaza-cyclopenta[b]naphthalene-2,7-dione (9). A typical library of scaffold 5 was synthesized in a parallel solution-phase manner and analyzed by HPLC-UV-MS or HPLC-UV-ELSD method.

Abstract:A transition-metal-free oxidative N-N bond formation strategy was developed to generate various structurally interesting [1,2,4]triazolo[1,5-a]benzazoles efficiently. The mechanism of the key oxidative N-N bond formation was investigated by using an intramolecular competition reaction. Notably, the first single crystal structure was also obtained to confirm the structure of 2-aryl[1,2,4]triazolo[1,5-a]benzimidazole.

Abstract:This paper describes our recent efforts to synthesize novel compound scaffolds integrating 2-quinoxalinol with privileged structures of 1,3-dihydro-benzoimidazol-2-one, 1,3-dihydro-benzoimidazole-2-thione, 3-hydroxy-1H-quinoxalin-2-one, 2H-benzo[1,4]oxazin-3-ol, 2H-benzo[1,4]thiazin-3-ol, and 1,3,4,5-tetrahydro-benzo[1,4]diazepin-2-one, respectively. Eight novel benzofused tricycles and their substituent diversity points were developed. These include pyrazino[2,3-g]quinoxaline-2,8-diol (I), 3-hydroxy-6,8,9,10-tetrahydro-1,4,6,10-tetraaza-cyclohepta[b]naphthalen-7-one (II), 6-hydroxy-4H-1-oxa-4,5,8-triaza-anthracen-3-one (III), 6-hydroxy-4H-1-thia-4,5,8-triaza-anthracen-3-one (IV), 6-hydroxy-1,1-dioxo-1,4-dihydro-2H-1 lambda(6)-thia-4,5,8-triaza-anthracen-3-one (V), 6-hydroxy-1,3-dihydro-imidazo[4,5-g]quinoxalin-2-one (VI), 6-hydroxy-1,3-dihydro-imidazo[4,5-g]quinoxaline-2-thione (VII) and 7-hydroxy-1,4-dihydro-pyrazino[2,3-g]quinoxaline-2,3-dione (VIII). This strategy of integrating two benzofused privileged structures into one molecule may provide a greater chance for the discovery of novel lead compounds.
