Abstract:Several new 6-(1-aryl-5-methyl-1, 2, 3-triazol-4-yl)-3-(4-pyridyl)-s-triazolo[3,4-b]-1, 3, 4-thiadiazoles 3a-j have been synthesized by the condensation of 4-amino-5-mercapto-3-(4-pyridyl)-1, 2, 4-triazole 2 with 1-aryl-4-carboxy-5-methyl-1, 2, 3-triazoles 1a-j in the presence of phosphorus oxychloride. The structures of the products obtained are characterized by elemental analyses and spectral data and the fragmentation pattern of the mass spectra of 3a-j is discussed. 3a-j have been screened for their antibacterial activity against B. subtilis and E. coli.

Abstract:A series of novel 6-aryl-3-(1-p-chlorophenyl-5-methyl-1,2,3-triazol-4-yl)-s-triazolo-[3,4-b]-1,3,4-thiadiazoles 6a-m have been synthesized in 49-64% yields by the cyclocondensation of 4-amino-3-(1-p-chlorophenyl-5-methyl-1,2,3-triazol-4-yl)-5-mercapto-1,2,4-triazole 5 with various aromatic acids in the presence of POCl3. Biological activity of 6a-m was studied.

Abstract:In the present study, we have synthesized a series of novel glucosyl triazoles for the first time. The glucosyl triazoles 4a-e were synthesized by reaction of some azidoglycosides with various terminal alkynes via a copper-catalyzed [3+2] cycloaddition (oclick chemistryo) and were deprotected to afford the corresponding glucosyl triazoles 5a-e in good yields. The structures of the new compounds were determined by IR, NMR spectroscopy, and mass spectrometry. The antitumor (human cervical cancer cell) activity was evaluated for the target compounds.

Abstract:Fifteen new 3-[(4-hydroxy-6-methly-2(1H)-pyridinones)-3-yl]-4-substituted- (1H)-1,2,4-triazole-5-thione compounds were synthesized from methyl 4-hydroxy-6- methyl-2-oxo-1,2-dihydropyridine-3-carboxylate in good overall yields by four step reactions. Molecular structure of these compounds was confirmed by IR, NMR, MS, elemental analysis and X-ray diffraction. Their antitumor activities were evaluated in vitro by the SRB and MTT methods. The results show that there are two compounds which are highly effective against KB cells and one compound which is highly effective against Bel-7402 cells.

Abstract:Ten novel benzoyl ureas containing a 1,3,4-thiadiazole group were synthesized by the reaction of 2-amino-5-substituted phenyl-1,3,4-thiadiazole and 2,6-difluorobenzoyl isocyanate, and structurally confirmed by H-1 NMR, IR, ESI-MS techniques and elemental analysis. Preliminary bioassay test indicated that some compounds showed insecticidal activity. Compounds 3d (3,5-(CH3)(2)) and 3g (4-C4H9) exhibit good insecticidal activity against Aphis craccivora with higer than 90% mortality.

Abstract:A one-pot reaction of 6-methyl-3-thioxo-3,4-dihydro-[1,2,4]triazin-5-one 1 with selected aldehydes 2a-d and chloroacetic acid afforded the respective 2-arylidene-6-methyl-thiazolo[3,2-b][1,2,4]triazine-3,7-diones 4a-d. Compunds 4a-d could be also obtained via the reaction of 1 with chloroacetic acid in refluxing acetic acid to give 6-methyl-thiazolo[3,2-b][1,2,4]triazine-3,7-dione 3 then, Knoevenagel condensation of 3 with aldehydes 2a-d gave compounds 4a-d. Heterocyclization of 4a-c with hydrazine hydrate and phenylhy- drazine gave the corresponding pyrazolines 5a-c and 6a-c, respectively. Moreover, 7-amino-9-(aryl)-3-methyl-2-oxo-2H-pyrido[2',3':4,5][1,3]thiazolo[3,2-b][1,2,4]triazine-8-carbonitrles 7a-c were synthesized by the reaction of 4a-c with malononitrile in the presence of ammonium acetate. The structures of newly synthesized compounds were confirmed by analytical and spectroscopic measurements. Some selected new compounds were screened for their cytotoxic activities against three human cancer cell lines (HepG2, MCF-7 and A549) using SRB assay and the structure-activity relationship (SAR) was discussed. The biochemical assays including antioxidant enzyme, oxidative stress and estimation of nucleic acids and proteins have been discussed for some selected compounds. The molecular docking of 4c and 7b has been also studied.

Abstract:The synthesis of a series of unprecedented tricyclic heterocycles, 1,2,4-triazolo[3,2-d][1,5]benzothiazepine 6-oxides (5a-i), was accomplished via cycloaddition of the allene-like cations 3, derived from thiochroman-4-one arylhydrazones, to the triple bond of nitriles and ensuing rings expansion annulation. The structure of one product (5h) has been unambiguously determined by X-ray diffraction analysis.

Abstract:The title compound C18H16N4O2S has been synthesized by the reaction of 4-amino-3-(4-ethoxyphenyl)-5-mercapto-1,2,4-triazole with phenoxyacetic acid in phospho- rus oxychloride, and characterized by IR, NMR spectra and elemental analysis. Its structure was determined by single-crystal X-ray diffraction. It crystallizes in monoclinic, space group P21/c with a = 1.4903(3), b = 1.5230(2), c = 0.9615(16) nm, Z = 4, V = 1.7769(5) nm3, Dc = 1.317 g/cm3, μ = 0.201 mm-1, F(000) = 736, R = 0.0795 and wR = 0.2233. In the title compound, all rings are essentially planar.

Abstract:A synthetic approach to the previously unknown thieno[2,3-f][1,2,4]triazolo[1,5-a]azepine derivatives was efficiently accomplished by cycloaddition of the bicyclic 1-aza-2-azoniaallene salts 6, derived from 6,7-dihydrobenzo[b]thiophen-4(5H)-one hydrazones 4, to the triple bond of nitriles, followed by ring enlargement. The X-ray crystal diffraction analysis of the picrate 10a provided affirmative proof for the structural assignment made.

Abstract:8-chloro-3-((3-chlorobenzyl)thio)-[1,2,4]triazolo[4,3-a]pyridine was synthesized and recrystallized from EtOH. The compound was characterized by H-1 NMR, C-13 NMR, FTIR, MS, elemental analysis and X-ray diffraction. The compound was crystallized in the monoclinic space group P2(1)/c with a = 8.1992(5), b = 21.7731(12), c = 7.8454(6) angstrom, alpha = 90, beta = 108.421(7), gamma = 90 degrees, V = 1328.81 (15) angstrom(3), Z = 4 and R = 0.0351. Theoretical calculation of the title compound was carried out with B3LYP/6-31G. The full geometry optimization was carried out by using the 6-31G basis set. The frontier orbital energy and atomic net charges were discussed. The experimental results of the compound have been compared with theoretical results and it was found that the experimental data shows good agreement with the calculated values.
