Abstract:We have developed a novel and efficient method for the preparation of 2,4-disubstituted-1,2,3-triazoles. These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. The reaction proceeds under mild conditions and is compatible with various functional groups.

The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:A novel straightforward synthesis of both symmetrical and unsymmetrical 2,4-disubstituted-1,3,5-triazines via aerobic copper-catalyzed cyclization of amidines with DMF as a one-carbon synthon has been developed. The presented method allows synthesizing the products that are currently inaccessible or challenging to prepare with the advantages of operational simplicity, broad substrate scope, and no need for prefunctionalized reagents, making it a highly practical approach to access various 2,4-disubstituted-1,3,5-triazines.

Abstract:The solid-phase synthesis of 1,3-disubstituted and 1,3,5-trisubstituted 1,3,5-triazine-2,4,6-triones from MBHA and Wang resin is described. Reaction of resin-bound amino acids with isocyanates yield resin-bound ureas, which further react with chlorocarbonyl isocyanate in toluene at 65 degreesC to selectively afford the resin-bound 1,3-disubstituted 1,3,5-triazine-2,4,6-triones. Selective alkylation at the N-5 position of the resin-bound 1,3-disubstituted 1.3,5-triazine-2,4,6-triones was accomplished by treatment with alkyl halides in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU). The desired products were cleaved from their solid support and obtained in good yield and purity. The method can be employed in production of toltrazuril analogue libraries for identification of new anticoccidial agents.

Abstract:Based on an efficient in situ metal/ligand reaction, a serial of 3,5-disubstituted-1,2,4-triazoles were synthesized via cycloaddition of organonitriles and ammonia under microwave conditions. Abstract:In this work, a series of 4-aryl-3-(trifluoromethyl)-1 H -1,2,4-triazol-5(4 H )-one derivatives is synthesized through nucleophilic intramolecular cyclization reactions of ethyl 2-(2,2,2-trifluoro-1-(arylimino)ethyl)hydrazine-1-carboxylate intermediates, which are themselves prepared in high yields via reactions of ethyl chloroformate and N -aryl-2,2,2-trifluoroacetimidoyl chloride derivatives. Some of the merits of the reported procedure are an operationally simple and concise method, the ready availability of starting materials, excellent product yields, no formation of harmful by-products and easy purification of the products. Abstract:A novel approach has been reported for the synthesis of 1,3,5-triazine with three distinct substituents. The synthesis of the pivotal intermediate 2-chloro-4-R 1 -6-R 2 -1,3,5-triazine plays a critical role in achieving 1,3,5-triazine with three different substituents. This article presents two innovative one-pot processes for synthesizing various substituted 2-chloro-4-R 1 -R 2 -1,3,5-triazines. In the first method, substituted benzoyl isocyanates were initially synthesized by reacting different amides with oxalyl chloride. Subsequently, 21 target products of 2-chloro-4-R 1 -R 2 -1,3,5-triazines compounds were obtained through the cyclization of substituted benzoyl isocyanates with benzamidine hydrochloride at relatively low temperatures and a one-pot chlorination reaction. The second method involved the synthesis of the same 21 compounds of 2-chloro-4-R 1 -R 2 -1,3,5-triazines by reacting benzamide with oxalyl chloride followed by cyclization with different amidine hydrochloride and then a one-pot chlorination reaction. Finally, a total of 35 different unsymmetrical trisubstituted 1,3,5-triazine compounds were successfully synthesized using various reactions.
