Abstract:2-Trifluoromethyl and 2-difluoromethyl substituted benzimidazole, benzoxazole and benzothiazole derivatives were efficiently prepared through a one-pot reaction of trifluoroacetic acid and difluoroacetic acid, respectively, with commercially available o-phenylenediamines, 2-aminophenols, and 2-aminobenzenethiols in good to excellent yields. Subsequential bromination of 2-difluoromethyl groups by photolysis with NBS led to the formation of bromodifluoromethyl benzo-1,3-diazoles which may be utilized to prepare the new generation of gem-difluoromethylene linked identical or non-identical twin molecules for drug synthesis.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:The synthesis of monosubstituted 1-aryl-1H-1,2,3-triazoles was achieved in a one-pot reaction from arylboronic acids and prop-2-ynoic acid or calcium acetylide (=calcium carbide), respectively, as a source of acetylene, with yields ranging from moderate to excellent (Scheme 1, Table 2). The reaction conditions were successfully applied to arylboronic acids, including analogs with various functionalities. Unexpectedly, the 1,2,3-triazole moiety promoted a regioselective hydrodebromination (Scheme 2).

Abstract:Reactions of easily accessible chloropyrimidinyl hydrazones with bromine were investigated. Oxidative cyclization followed by concomitant bromination led to the formation of [1,2,4]triazolo[4,3-c]pyrimidines, which then underwent the Dimroth rearrangement to afford highly functionalized 2-substituted [1,2,4]triazolo[1,5-c]pyrimidines.

Abstract:1-(2-Cyan)-phenyl-3-methyl-5-pyrazolone is synthesized from 2-aminobenzonitrile by diazotization,reduction with stannous chloride in the presence of hydrochloric acid to give 2-cyanphenylhydrazine hydrochloride,which is subjected to neutralization with sodium methoxide and nucleophilic reaction with methyl acetoacetate.The method has been improved in the process of synthesis of 2-cyanphenylhydrazine hydrochloride and 1-(2-cyan)-phenyl-3-methyl-5-pyrazolone and the total yield can be increased to 79.4%.

Abstract:In this work, a series of 4-aryl-3-(trifluoromethyl)-1 H -1,2,4-triazol-5(4 H )-one derivatives is synthesized through nucleophilic intramolecular cyclization reactions of ethyl 2-(2,2,2-trifluoro-1-(arylimino)ethyl)hydrazine-1-carboxylate intermediates, which are themselves prepared in high yields via reactions of ethyl chloroformate and N -aryl-2,2,2-trifluoroacetimidoyl chloride derivatives. Some of the merits of the reported procedure are an operationally simple and concise method, the ready availability of starting materials, excellent product yields, no formation of harmful by-products and easy purification of the products. Abstract:The synthesis of 1-monosubstituted 1,2,3-triazoles was achieved using azides and propiolic acid by copper-catalyzed click cycloaddition/decarboxylation, which was easily carried out in N, N-dimethylformamide at room temperature or 60 degrees C with yields ranging from moderate to excellent. Abstract:8-Bromo-7-chloro[1,2,4]triazolo[4,3-c]pyrimidines were prepared by bromine-mediated oxidative cyclization of the aldehyde-derived hydrazones. The structure of one such product was unambiguously confirmed by single-crystal X-ray analysis. While the C-5 unsubstituted 1,2,4-triazolo[4,3-c]pyrimidine chemotype is extremely susceptible to ring isomerization at ambient conditions, the C-5-substituted analogues were found to be quite stable, permitting isolation in pure form. Nevertheless, they can still be converted into the 1,2,4-triazolo[1,5-c]pyrimidines by base-or acid-promoted Dimroth rearrangement. The presence of halogen functionalities on the pyrimidine nucleus renders the products useful as versatile synthetic intermediates for the easy diversification, as evidenced by palladium-catalyzed Kumada cross-couplings and Buchwald-Hartwig amination, as well as direct aromatic substitution.

Abstract:A new methodology for the Cu-catalyzed synthesis of 1H-1,2,3-triazoles from 1,1-dibromoalkenes and sodium azide is presented. Aryl dibromoolefins were efficiently converted into the corresponding 1,2,3-triazoles. A comprehensive number of functional groups were compatible with this reaction. 1,2,3-Triazoles were obtained in moderate to excellent yields.

Abstract:A novel strategy for catalytic synthesis of 2-aryl-2H-benzo[d][1,2,3]triazoles by non-noble molybdenum-catalyzed deoxygenative heterocyclization of 2-nitroazobenzenes is described, bearing a wide range of functional groups in good to excellent yields. The salient features of the transformation include the use of readily available substrates, valuable products and ease of scale-up. The mechanistic study indicates the reaction occurred via twice deoxygenation by Mo(VI)/Mo(IV) catalytic cycle from 2-nitroazobenzene, through the formation of 2-aryl-2H-benzo[d][1,2,3]triazole-N1-oxide or nitrene intermediates.

Abstract:Many [1, 2, 4] triazolobenzoheteroazepine derivatives show interesting biological activities. In this paper, the geminal arylazo isocyanato compounds 3 were prepared from chroman-4-ones in three steps, i.e. condensation with arylhydrazine, cycloaddition of the resulting hydrazone with HNCO, and oxidative ring cleavage by KMnO4. Compounds 3 were allowed to react with HBF4, producing the respective 3-spiro substituted 1-aryl-4, 5-dihydro-5-oxo-3H-1, 2, 4-triazolium salts 4. The concurrent rearrangement of compound 4 with insertion of the nitrogen atom into the carbon skeleton provided, after the basic work-up, 2-oxo-[1, 2, 4] triazolo [3, 2-d] [1, 5] benzoxazepines 5a-5h. An unambigous structutal proof was obtained from the X-ray diffraction analysis for compound 5h.
