Abstract:The long-circulating effect is revisited by simultaneous monitoring of the drug payloads and nanocarriers following intravenous administration of doxorubicin (DOX)-loaded methoxy polyethylene glycol-polycaprolactone (mPEG-PCL) nanoparticles. Comparison of the kinetic profiles of both DOX and nanocarriers verifies the long-circulating effect, though of limited degree, as a result of pegylation. The nanocarrier profiles display fast clearance from the blood despite dense PEG decoration; DOX is cleared faster than the nanocarriers. The nanocarriers circulate longer than DOX in the blood, suggesting possible leakage of DOX from the nanocarriers. Hepatic accumulation is the highest among all organs and tissues investigated, which however is reversely proportionate to blood circulation time. Pegylation and reduction in particle size prove to extend circulation of drug nanocarriers in the blood with simultaneous decrease in uptake by various organs of the mononuclear phagocytic system. It is concluded that the long-circulating effect of mPEG-PCL nanoparticles is reconfirmed by monitoring of either DOX or the nanocarriers, but the faster clearance of DOX suggests possible leakage of a fraction of the payloads. The findings of this study are of potential translational significance in design of nanocarriers towards optimization of both therapeutic and toxic effects.

Photo-crosslinked Poly(ethylene Glycol)-B-poly(ϵ-caprolactone) Nanoparticles for Controllable Paclitaxel Release

Abstract:Novel biodegradable core-crosslinked nanoparticles (CNPs) consisting of methoxy poly(ethylene glycol)-block-poly(E-caprolactone-co--cinnamoyloxy-E-caprolactone) (mPEG-b-P(CL-co-CCL)) were prepared and evaluated for paclitaxel (PTX) delivery. mPEG(113)-b-P(CL65.2-co-CCL10.1) had a higher drug loading efficiency (95%) compared to mPEG(113)-b-PCL93.1 (43%). The stability of NPs has been largely improved and PTX release was significantly inhibited by crosslinking via UV irradiation at =254nm. MTT assays demonstrated that both blank non-crosslinked and crosslinked NPs showed low cytotoxicity to NCL-H460 cells while PTX-loaded non-crosslinked and crosslinked NPs exhibited obvious cytotoxicity against NCL-H460 cells, and the cytotoxicity was both dose-dependent and time-dependent. Furthermore, after 48h incubation the cell viability of PTX-loaded crosslinked NPs was lower compared to that of PTX-loaded non-crosslinked NPs or free PTX. These properties indicated that CNPs prepared from mPEG-b-P(CL-co-CCL) have great potentials as carriers for drug delivery.
Quantification of paclitaxel in rat plasma by LC-MS/MS and pharmacokinetics of paclitaxel thermosensi-tive liposomes in rats

Abstract:OBJECTIVE: To develop and validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of paclitaxel in rat plasma and to study the pharmacokinetics of paclitaxel thermosensitive liposomes in rats. METHODS: The plasma was alkalized with sodium bicarbonate (pH 8.0) and extracted with tert-butyl methyl ether (TBME). Paclitaxel and IS(docetaxel) were separated on an Agilent Zorbax SB C18 column (2.1 mm × 50 mm, 3.5 μm) with the mobile phase consisting of methanol-0.5 mmol · L-1 ammonium acetate (70: 30, V/V) at a flow rate of 300 μL · min-1. The injection volume was 5 μL An Agilent 6410 A QQQ triple-quadrople mass spectrometer equipped with an electrospray ionization (ESI) source was used as detector and was operated in positive ion mode. Selected ion monitoring (SIM) was performed and the m/z of ions selected for quantitation were 876.3 → 308.1 (paclitaxel) and 830.2 → 549.2 (internal standard, docetaxel) . SD rats were injected paclitaxel thermosensitive liposomes and paclitaxel injection via tail vein at the dose of 7 mg · kg-1, respectively. Pharmacokinetic parameters were calculated. RESULTS: The chromatograms showed no endogenous interfering peaks in blank rat plasma. Each analysis was completed within 3.5 min. The linear range of paclitaxel in plasma was 2-1 000 ng · mL-1. The lower limit of quantification was 2 ng · mL-1. The intra-run and inter-run relative standard deviations (RSDs) were less than 15%. The differences of f1/2β, MRT, AUC and CL were significant between the two preparations. CONCLUSION: The method is sensitive, accurate, and convenient for the pharmacokinetic study of paclitaxel thermosensitive liposomes. Compared with paclitaxel injection, paclitaxel thermosensitive liposomes has a controlled release rate, which can alleviate the acute toxicity and improve the tolerance to a certain extent.

Abstract:Paclitaxel-loaded poly(ethylene oxide)-b-poly(lactide) (PEO-b-PLA) systems have been observed to assemble into fiber structures with remarkably different properties using different chirality and molecular weight of PLA segments. In this study, dissipative particle dynamics (DPD) simulations were carried out to elaborate the microstructures and properties of pure paclitaxel and paclitaxel-loaded PEO-b-PLA systems. Paclitaxel molecules formed ribbon or fiber like structures in water. With the addition of PEO-b-PDLA, PEO-b-PLLA and their stereocomplex, paclitaxel acted as a template and polymer molecules assembled around the paclitaxel structure to form core/shell structured fibers having a PEO shell. For PEO19-b-PDLA27 and PEO19-b-PLLA27 systems, PLA segments and paclitaxel molecules were distributed homogeneously in the core of fibers based on the hydrophobic interactions. In the stereocomplex formulation, paclitaxel molecules were more concentrated in the inner PLA stereocomplex core, which led to slower release of paclitaxel. By increasing the length of PLA segments (e.g. 8,16,22 and 27), the crystalline structure of paclitaxel was gradually weakened and destroyed, which was further proved by X-ray diffraction studies. All the simulation results agreed well with experimental data, suggesting that the DPD simulations may provide a powerful tool for designing drug delivery systems.

Abstract:This Letter focuses on the preparation and characterisation of a novel amphiphilic multiblock poly(ether-ester anhydride) copolymer [poly(sebacic acid-b-poly(ethylene glycol)-b-sebacic acid) (PES)], paclitaxel-loaded PES nanoparticles (NPs), in vitro evaluation of the release of paclitaxel and cytotoxicity of NPs. PES was prepared by melt polycondensation and the PES NPs were prepared by nanoprecipitation technique. In vitro release behaviour of the paclitaxel-loaded NPs was investigated by high-performance liquid chromatography (HPLC). In vitro cytotoxicity of NPs was evaluated by MTT assay. The composition and structure of PES are consistent with that of the designed copolymer. The paclitaxel-loaded NPs are of spherical shape and size lower than 200 nm. Paclitaxel can be continuously released from the paclitaxel-loaded NPs. The PES NPs are non-toxic to cells. The results suggest that PES NPs are a potential candidate nanocarrier material for the controlled delivery of paclitaxel and other hydrophobic compounds.
Nanoformulation of Paclitaxel: Exploring the Cyclodextrin / PLGA Nano Delivery Carrier to Slow Down Paclitaxel Release, Enhance Accumulation in Vivo

Abstract:Background: Improving the aggregation and penetration in tumor sites increases the anti-tumor efficacy of nanomedicine. In the current study, we designed cyclodextrin modified PLGA nanoparticles loaded with paclitaxel to elevate the accumulation and prolong circulation of chemotherapy drugs in vivo. Methods: The PLGA nanoparticles loaded with paclitaxel (PTX PLGA NPs) and cyclodextrin (CD) modified PLGA nanoparticles loaded with paclitaxel (PTX PLGA/CD NPs) were prepared using the emulsification solvent evaporation method. The nanoparticles were characterized by particle size, zeta potential, encapsulation efficiency, infrared spectroscopy analysis and X-Ray diffraction (XRD). Then, drug release of the nanoparticles was evaluated via reverse dialysis method in vitro. Finally, the in vivo distribution fate and pharmacokinetic characteristics of the nanoparticles were assessed in mice and rats. Results: The average particle size, zeta potential, and encapsulation efficiency of PTX PLGA NPs were (163.57±2.07) nm, - (20.53±2.79) mV and (60.44±6.80)%. The average particle size, zeta potential, and encapsulation efficiency of PTX PLGA/CD NPs were (148.57±1.66) nm, - (11.42±0.84) mV and (85.70±2.06)%. In vitro release studies showed that PTX PLGA/CD NPs were released more slowly compared to PTX PLGA NPs under normal blood pH conditions, while PTX PLGA/CD NPs were released more completely under tumor site pH conditions. The modified PLGA nanocarrier (PLGA/CD NPs) increased drug residence time and accumulation than the plain PLGA nanocarrier (PLGA NPs) in vivo distribution. In addition, the elimination half-life, area under the drug-time curve, and maximum blood concentration of the nanoparticle group were higher than those of Taxol®, especially the PTX PLGA/CD NPs group, which was significantly different from Taxol® and plain nanoparticle groups (p<0.001). Conclusions: The 2-HP-β-CD modified PLGA nanoparticles prolonged circulation time and accumulation of the chemotherapy drug paclitaxel in vivo.

Effect of Polymer Degradation on Prolonged Release of Paclitaxel from Filomicelles of Polylactide/poly(ethylene Glycol) Block Copolymers.

Abstract:Paclitaxel is one of the most efficient anticancer agents, but the conventional dosage formulations cause many side effects. PLA-PEG filomicelles are promising carriers of paclitaxel because high loading capacity and long term release can be achieved. Slow release of cytostatic drugs is very advantageous due to prolonged exposure of tumor cells to cytostatic over multiple cell cycles. The aim of this study was to evaluate the potential of bioresorbable PLA-PEG filomicelles for prolonged delivery of paclitaxel. Paclitaxel is encapsulated in PLLA-PEG filomicelles and PDLLA-PEG spherical micelles. Drug release was studied in PBS at 37°C at various pH values to elucidate the influence of polymer degradation on drug release. NMR, GPC and HPLC were used to follow polymer degradation and drug release. The release of paclitaxel is strongly dependent on the degradation of micelles. A biphasic drug release profile is observed for both PLLA-PEG and PDLLA-PEG micelles: slow release in the first phase and faster release in the second phase. Degradation is faster at acidic pH than at pH7.4, and PLLA-PEG filomicelles degrade less rapidly than PDLLA-PEG spherical micelles, leading to various rates of drug release. The correlation between degradation and drug release is very helpful for the development of novel drug carriers with tailored properties. Importantly, the cytotoxic activity of PLLA-PEG filomicelles was evidenced, thus showing their potential as carrier of antitumor drugs.
Reexamining in Vivo Fate of Paclitaxel-Loaded Polymeric Micelles

Abstract:Polymeric micelle (PM) is one of the most promising drug delivery systems for therapeutic agents with poor solubility. Yet the in vivo fate of PM remains unclear due to the limitations in separation and detection methods. Herein we established a facile method for PEGylated PM separation by combining a single-chain variable antibody fragment against polyethylene glycol (PEG) with Ni-NTA agarose beads. Complete micelle separation was accomplished by mild centrifugation (2000 × g) at 4 ℃. In virtue of this method, we surprisingly discovered that paclitaxel (PTX) was not completely trapped in the hydrophobic cores of micelles in PTX-loaded PMs, and the portion of unencapsulated PTX further increased either with the dilution of micelle solutions or in the presence of serum ascribing to the high binding affinity between PTX and serum proteins. After entering the bloodstream, PTX-loaded PMs rapidly released PTX to form plasma protein-bound form, leaving the empty micelles circulating for a while. Moreover, anti-PEG antibodies, whether stimulated by the administration of PEGylated therapeutics or pre-existing in serum with unknown origin, could exacerbate the accelerated blood clearance phenomenon and induce anaphylactoid reactions via complement activation, which persuaded us to be more cautious in the clinical medication of PEGylated micelles.
In Vitro Evaluation and Finite Element Simulation of Drug Release from Polydiacetylene-Polyethylene Glycol Stearate Nanovesicles

Abstract:Vesicles comprised of 10,12-pentacosadiynoic acid (PCDA) were modified, using polyethylene glycol 40 stearate (PEG40S), and crosslinked by ultraviolet (UV) irradiation to create polymerized nanovesicles for sustained drug release. Paclitaxel, a water-insoluble compound widely used in cancer chemotherapy, was used as a model drug to examine the physicochemical stability and release profiles of PCDA/PEG40S nanovesicles. TEM analysis revealed the formation of paclitaxel-encapsulated PCDA/PEG40S nanovesicles of 40 to 200 nm in size. Upon the addition of ethanol, instantaneous releases of paclitaxel in the amount of 28 microg/mL from polymerized PCDA/PEG40S nanovesicles and 108 microg/ml from unpolymerized ones were observed. This suggested the non-complete drug release from polymerized PCDA/PEG40S nanovesicles due to their enhanced physicochemical stability by ultraviolet irradiation-induced polymerization, if compared to unpolymerized ones. An in vitro study demonstrated that an accumulative release of 24.1 +/- 3.1% and 8.1 +/- 1.7% of paclitaxel was obtained within 24 hrs from nanovesicles comprised of PCDA/PEG40S at a 9:1 and 7:3 molar ratio, respectively. A finite element model that considered the diffusion-driven releases and the reversible drug-vesicle interaction captured the sustained release of paclitaxel from polymerized PCDA/PEG40S nanovesicles. PCDA/PEG40S nanovesicles capable of sustained release and with enhanced physicochemical stability thus possess great potential for applications in drug release.

Abstract:Polymer-nanoparticle-encapsulated doxorubicin (DOX) and paclitaxel (TAX) have the potential for novel therapeutic use against cancer in the clinic. However, the systemic biological effect of the nanoparticle material, namely, methoxypoly(ethylene glycol)-poly(lactide-co-glycolide) (mPEG-PLGA), and its encapsulated drugs have not been fully studied. We have applied NMR-based metabonomics methodology to characterize and analyze the systemic metabolic changes in mice after being exposed to mPEG-PLGA, mPEG-PLGA-encapsulated DOX and TAX (NP-D/T), and their free forms. The study revealed that mPEG-PLGA exposure only induces temporary and slight metabolic alternations and that there are detoxification effects of nanoparticle packed with D/T drugs on the heart when comparing with free-form D/T drugs. Both NP-D/T and their free forms induce a shift in energy metabolism, stimulate antioxidation pathways, and disturb the gut microbial activity of the host. However, mPEG-PLGA packaging can relieve the energy metabolism inhibition and decrease the activation of antioxidation pathways caused by D/T exposure. These findings provide a holistic insight into the biological effect of polymer nanoparticle and nanoparticle-encapsulated drugs. This study also furthers our understanding of the molecular mechanisms involved in the amelioration effects of mPEG-PLGA packaging on the toxicity of the incorporated drugs.
Nanoparticles of lipid monolayer shell and biodegradable polymer core for controlled release of paclitaxel: effects of surfactants on particles size, characteristics and in vitro performance

Abstract:This work developed a system of nanoparticles of lipid monolayer shell and biodegradable polymer core for controlled release of anticancer drugs with paclitaxel as a model drug, in which the emphasis was given to the effects of the surfactant type and the optimization of the emulsifier amount used in the single emulsion solvent evaporation/extraction process for the nanoparticle preparation on the particle size, characters and in vitro performance. The drug loaded nanoparticles were characterized by laser light scattering (LLS) for size and size distribution, field-emission scanning electron microscopy (FESEM) for surface morphology, X-ray photoelectron spectroscopy (XPS) for surface chemistry, zetasizer for surface charge, and high performance liquid chromatography (HPLC) for drug encapsulation efficiency and in vitro drug release kinetics. MCF-7 breast cancer cells were employed to evaluate the cellular uptake and cytotoxicity. It was found that phospholipids of short chains such as 1,2-dilauroylphosphatidylocholine (DLPC) have great advantages over the traditional emulsifier poly(vinyl alcohol) (PVA), which is used most often in the literature, in preparation of nanoparticles of biodegradable polymers such as poly(D,L-lactide-co-glycolide) (PLGA) for desired particle size, character and in vitro cellular uptake and cytotoxicity. After incubation with MCF-7 cells at 0.250 mg/ml NP concentration, the coumarin-6 loaded PLGA NPs of DLPC shell showed more effective cellular uptake versus those of PVA shell. The analysis of IC(50), i.e. the drug concentration at which 50% of the cells are killed, demonstrated that our DLPC shell PLGA core NP formulation of paclitaxel could be 5.88-, 5.72-, 7.27-fold effective than the commercial formulation Taxol after 24, 48, 72h treatment, respectively.
Preparation and Characterization of Paclitaxel-Loaded DSPE-PEG-liquid Crystalline Nanoparticles (lcnps) for Improved Bioavailability

Abstract:Lipid-based liquid crystalline nanoparticles (LCNPs) have attracted growing interest as a new drug nanocarrier system for improving bioavailability for both hydrophilic and hydrophobic drugs. In this study, self-assembled LCNPs based on soy phosphatidyl choline and glycerol dioleate, which was known possessing low toxicity and negligible hemolysis, were prepared using poly(ethylene glycol)-grafted 1,2-distearoyl-sn-glycero-3-phosphatidylethanolamine (DSPE-PEG) as the dispersing agent. Paclitaxel (PTX) was used as a model hydrophobic drug. The particle size of the optimized DSPE-PEG-LCNPs and PTX-loaded DSPE-PEG-LCNPs were around 70nm. Crossed polarized light microscopy was used to characterize the phase behavior of liquid crystalline (LC) matrices, which showed a fan-like birefringent texture in dark background indicating the coexistence of reversed cubic and hexagonal phase in the optimized LC matrix. Transmission electron microscopy and cryo-field emission scanning electron microscopy revealed its internal water channel and “twig-like” surface morphology. PTX-loaded DSPE-PEG-LCNPs exhibited a biphasic drug sustained release pattern with a relatively fast release at the initial stage and a sustained release afterwards. PTX-loaded DSPE-PEG-LCNPs presented higher AUC (410.942±72.522μg/Lh) when compared with commercial product Taxol (212.670±41.396μg/Lh). These results indicated that DSPE-PEG-LCNPs might serve as a potential sustained release system for poorly water-soluble agents.

Abstract:This paper aims to improve the systemic circulation of paclitaxel (PTX) by modifying nanocrystals with DSPE-PEG 2000. PTX nanocrystals (PNCs) were prepared by an anti-solvent method and DSPE-PEG 2000 was inserted into PNCs by hybridization. The average size of DSPE-PEG-PNCs did not change obviously compared to the naked PNCs, but the negative charge increased after hybridization. The saturated attachment ratio of DSPE-PEG 2000 was 0.4%. In vitro release rate of DSPE-PEG-PNCs was significantly slower than that of PTX solution and the naked PNCs, which indicated that DSPE-PEG 2000 hindered the release of PTX. Moreover, pharmacokinetics results showed that DSPE-PEG-PNCs achieved a higher area-under-the-curve (AUC, 4.43 +/- 0.19 mg/L*h) and lower clearance rate (1.08 +/- 0.16 L/h/kg) compared to PNCs with an AUC of 2.48 +/- 0.18 mg/L*h and a clearance rate of 1.89 +/- 0.15 L/h/kg. Therefore, DSPE-PEG-PNCs could have a significant potential in clinical use by improving the systemic circulation of the drug.
Near-Critical Fluid Micellization for High and Efficient Drug Loading: Encapsulation of Paclitaxel into Peg-B-Pcl Micelles

Abstract:Paclitaxel, an expensive first-line anticancer drug, is known to have better pharmacokinetics and therapeutic efficacy if encapsulated in polymeric micelles. However, the conventional encapsulation methods using incompressible aqueous solutions are limited to low drug loading, less than 3% of micelle weight, and low efficiency, more than two-thirds of the drug in solution remains unencapsulated, and hence wasted, not to mention the burst release problems. This work demonstrates that expansion of near-critical fluid solutions, for example in compressible dimethyl ether and trifluoromethane not too far from their critical region, can lead to a much higher drug loading, for example in micelles formed from poly(ethylene glycol)-block-poly(epsilon-caprolactone) (PEG-b-PCL). By controlling the drug precipitation within the micellar solution region, the loading of paclitaxel in PEG-b-PCL can reach over 12% with a loading efficiency of 87%, which is unattainable by conventional methods. Moreover, the burst release fraction of the drug can be reduced despite the higher drug loading. This means that the new near-critical fluid micellization (NCFM) method will allow not only for a lower exposure of the body to the copolymer at the same treatment drug rate, due to the high drug loading, but also for less waste of the expensive drug, due to the high efficiency.
Preparation and Characterization of Paclitaxel Delivery System Based on Semi-Solid Lipid Nanoparticles Coated with Poly (ethylene Glycol).

Abstract:The current formulation of paclitaxel causes significant side effects, so there is a need to develop a safer delivery system. In this study, novel semi-solid lipid nanoparticles (PEG-PE-SSLN) were prepared, and evaluated as delivery vehicles for paclitaxel. The PEG-PE-SSLN are approximately 213 nm in size with a zeta-potential of -37 mV, compared to Span60-SSLN with a particle size of over 275 nm and zeta-potential of -29 mV. The encapsulation efficiency of PEG-PE-SSLN could be as high as 98%. Furthermore, differential scanning calorimetry (DSC) analysis indicated that paclitaxel in the nanoparticles existed in an amorphous form. A sustained release of 85% of the loaded paclitaxel from PEG-PE-SSLN within 12 days was observed while release from Span60-SSLN particles was complete within 6 days. PEG-PE-SSLN can effectively decrease phagocytosis of murine peritoneal macrophages. Lyopholization of SSLNs in the presence of trehalose tended to preserve their stability. Therefore, PEG-PE-SSLN represents an improved and promising paclitaxel delivery system.

Abstract:Paclitaxel (PTX) is a broad‐spectrum alkaloid anticancer drug with high therapeutic efficacy. However, PTX has a very low water solubility and its metabolism demonstrates nonlinear pharmacokinetic characteristics, resulting in systemic adverse reactions in clinical applications. Many endeavors are devoted to develop new PTX preparations in response to these obstacles, and some are used in clinical phase of cancer treatment. In particular, highly specific medical intervention at nanoscale is an emerging research field focusing on nanoapproaches to solve clinical problems. In a perspective of nanomedicine, the recent covalent and noncovalent approaches toward PTX formulation on various substrates, as well as their advantages and disadvantages during preclinical researches and clinical applications are herein summarized. It is anticipated that nanotechnology‐based drug delivery systems can further enhance therapeutic efficiencies of PTX, by reducing systemic side effects and alleviating severely negative effects on patients. Recent development of paclitaxel delivery systems is summarized, focusing on noncovalent or covalent strategies to immobilize paclitaxel on various carrier substrates such as vesicles, micelles, hydrogels, synthetic and bio‐macromolecules, as well as inorganic materials.
A High Capacity Polymeric Micelle of Paclitaxel: Implication of High Dose Drug Therapy to Safety and in Vivo Anti-Cancer Activity

Abstract:The poor solubility of paclitaxel (PTX), the commercially most successful anticancer drug, has long been hampering the development of suitable formulations. Here, we present translational evaluation of a nanoformulation of PTX, which is characterized by a facile preparation, extraordinary high drug loading of 50% wt. and PTX solubility of up to 45 g/L, excellent shelf stability and controllable, sub-100 nm size. We observe favorable in vitro and in vivo safety profiles and a higher maximum tolerated dose compared to clinically approved formulations. Pharmacokinetic analysis reveals that the higher dose administered leads to a higher exposure of the tumor to PTX. As a result, we observed improved therapeutic outcome in orthotopic tumor models including particularly faithful and aggressive “T11” mouse claudin-low breast cancer orthotopic, syngeneic transplants. The promising preclinical data on the presented PTX nanoformulation showcase the need to investigate new excipients and is a robust basis to translate into clinical trials.
Free paclitaxel loaded PEGylated-paclitaxel nanoparticles: preparation and comparison with other paclitaxel systems in vitro and in vivo

Abstract:Previously, PEGylated paclitaxel (PEG-PTX) was found not favorable as a polymer prodrug because of its poor antitumor efficiency. But surprisingly, it was found in our study that PEG-PTX could form a novel nanoparticle system with free PTX. To address how this system works, we compared PTX loaded PEG-PTX nanoparticles (PEG-PTX/PTX) with PTX loaded PEG-PLA micelles (PEG-PLA/PTX) or PTX injection available (Taxol(®)) in vitro and in vivo. Firstly, it was found that PEG-PTX/PTX was more stable in aqueous solution than PEG-PLA/PTX in terms of PTX crystal formation and drug release. Then it was demonstrated that coumarin loaded PEG-PTX nanoparticles had a much higher uptake in MCF-7 cells compared to coumarin loaded PEG-PLA micelles. The in vivo imaging study revealed that DIR or DID (near infrared fluorescent substances) loaded PEG-PTX nanoparticles distributed more in tumors in MCF-7 tumor bearing mice than DIR or DID loaded PEG-PLA micelles and solvent system of Taxol(®). In the efficacy study with MCF-7 tumor bearing mice, PEG-PTX/PTX showed significantly higher antitumor activity than PEG-PLA/PTX at the same PTX dosage. At the dose of 10mg free PTX per kg, PEG-PTX/PTX displayed similar efficacy as Taxol(®) but less toxicity evaluated by the loss of body weight. With the increase of free PTX to 15 mg/kg, PEG-PTX/PTX showed significantly better efficacy than Taxol(®). In conclusion, with favorable characteristics in stability, cellular uptake, cytotoxicity, biodistribution, safety and efficacy, PEG-PTX/PTX seems highly potential as a nanocarrier for PTX delivery.

Abstract:Polymeric micelles hold great potential for anticancer drug delivery. Sufficient integrity of polymeric micelles after intravenous injection is critical for successful drug delivery to solid tumors, but accurate measurement of the in vivo micellar integrity remains challenging. Methods based on Förster resonance energy transfer (FRET) to monitor in vivo micellar integrity are frequently used. However, the self-quenching effect of the FRET fluorophores used has been improperly ignored and has caused inaccurate measurements. Herein, we report a FRET-based approach using the large Stokes shift (LSS) fluorophores NBD-X and MS735 as the donor and acceptor, respectively, to investigate the integrity of polyethylene glycol-block-poly(ε-caprolactone) (PEG-PCL) micelles. We established a mathematical formula for the integrity calculation, and an in vitro verification experiment showed that the formula results exactly matched the simulated results. Our results demonstrated that the PEG-PCL micelles gradually dissociated in blood circulation, but approximately 60% of the micelles in plasma remained intact 72 h after intravenous (i.v.) injection. This LSS fluorophore-based FRET approach can be used to accurately monitor the integrity of nanoparticles, and this study demonstrates that most of PEG-PCL micelles maintain their aggregation state during blood circulation for anticancer drug delivery.

Abstract:Paclitaxel is one of the most widely used anticancer agents, but strong side effects and low bioavailability limit its clinical efficacy. The use of tumor microenvironment-responsive prodrugs is promising to solve these problems, and a smart linkage is crucial to achieve the efficient release of paclitaxel from such prodrugs in tumor. Herein, an acid-responsive acetone-based acyclic ketal linkage is used to construct paclitaxel prodrugs with different length of poly(ethylene glycol) (PEG). The PEGylated acetone-based acyclic-ketal-linked prodrugs of paclitaxel (PKPs) self-assembled into nanoparticles that were stable in normal physiological environment but released paclitaxel rapidly in mildly acidic environment in tumor. The length of PEG had considerable impact on size and critical micelle concentration of PKP nanoparticles, thereby affecting prodrug hydrolysis kinetics, pharmacokinetics, biodistribution, and antitumor activity for PKP nanoparticles. In an A2780 xenograft mouse model, PKP nanoparticles displayed improved pharmacokinetics and superior antitumor efficacy against Taxol. Our results demonstrate that acyclic-ketal-based prodrugs are useful for the development of acid-responsive anticancer nanomedicines.
