Abstract:One of the major obstacles to successful paclitaxel (PTX) chemotherapy is toxic side effects, which are often due to the conventional surfactants used, such as Cremophor EL. PTX is characterized by its hydrophobicity and insolubility, which limit its application in ovarian cancer therapy. The aim of this study was to develop Cremophor EL-free PTX-loaded methoxy poly(ethylene glycol)-block-(lactic-co-glycolic acid) copolymers (PLGA-mPEG) nanoparticles (NPs) using d-alpha-tocopheryl polyethylene glycol 1000 succinate (TPGS) as a novel emulsifier. The ability of nanoparticles loaded with paclitaxel (NP-PTX) to inhibit tumor growth was assessed in vitro and in vivo. The acute toxicity of NP-PTX was also evaluated in vivo. We found that paclitaxel was efficiently encapsulated into PLGA-mPEG NPs with a low concentration of TPGS as the emulsifier. The synthesized NP-PTX demonstrated the desired diameter of 80 nm as characterized by transmission electron microscopy. The NP-PTX also exhibited a sustained release of loaded PTX over 4 d with the same chemotherapeutic efficiency and reduced side effects. NP-PTX-treated cells showed slightly lower cytotoxic responses compared with those treated with free PTX at the same concentration. In vivo studies confirmed that NP-PTX significantly enhanced the median lethal dose of paclitaxel by 10-fold, and a similar effect on the inhibition of tumor growth was achieved in nude mice.

Abstract:This Letter focuses on the preparation and characterisation of a novel amphiphilic multiblock poly(ether-ester anhydride) copolymer [poly(sebacic acid-b-poly(ethylene glycol)-b-sebacic acid) (PES)], paclitaxel-loaded PES nanoparticles (NPs), in vitro evaluation of the release of paclitaxel and cytotoxicity of NPs. PES was prepared by melt polycondensation and the PES NPs were prepared by nanoprecipitation technique. In vitro release behaviour of the paclitaxel-loaded NPs was investigated by high-performance liquid chromatography (HPLC). In vitro cytotoxicity of NPs was evaluated by MTT assay. The composition and structure of PES are consistent with that of the designed copolymer. The paclitaxel-loaded NPs are of spherical shape and size lower than 200 nm. Paclitaxel can be continuously released from the paclitaxel-loaded NPs. The PES NPs are non-toxic to cells. The results suggest that PES NPs are a potential candidate nanocarrier material for the controlled delivery of paclitaxel and other hydrophobic compounds.
In-vitro evaluation of paclitaxel-loaded MPEG-PLGA nanoparticles on laryngeal cancer cells.

Abstract:Induction chemotherapy or concurrent chemotherapy with radiotherapy can preserve laryngeal function without a detrimental effect on survival in patients with advanced laryngeal cancer. However, systemic chemotherapy with traditional injection often causes side effects. In this study, methoxy poly(ethylene glycol)-poly(lactide-co-glycolide) (MPEG-PLGA) nanoparticles (NPs) were developed as a carrier for paclitaxel, a potent cytotoxic agent. The NPs were prepared by an emulsification-solvent evaporation method with the particle size and zeta potential around 153.3 +/- 41.7 nm and -5.36 mV. Transmission electron microscopy showed that the NPs were homogeneous and spherical in shape. Differential scanning calorimetry and X-ray powder diffractometry did not detect any crystalline drug in the NP samples. High-performance liquid chromatography was used to measure the drug loading, encapsulation efficiency, and in-vitro drug release. The drug loading efficiency was (5.35 +/- 0.75)% and encapsulation efficiency was (75.56 +/- 2.61)%. In the in-vitro drug release study, paclitaxel was released from the NPs; in a slow but time-dependent manner. In-vitro cytotoxicity of the paclitaxel-loaded NPs was investigated by using human laryngeal cancer Hep-2 cells. The anticancer activity of paclitaxel-loaded NPs was comparable with the free paclitaxel. No significant cytotoxicity was observed in blank MPEG-PLGA Nips. Laser scanning confocal microscopy was used to observe the uptake of fluorescent coumarin-6-loaded NPs by Hep-2 cells. We conclude that the formulation of Nips inhibits Hep-2 cell growth to a similar extent as free paclitaxel injection. These results suggest that MPEG-PLGA NPs may have potential as an alternative delivery system for paclitaxel. Photo-crosslinked Poly(ethylene Glycol)-B-poly(ϵ-caprolactone) Nanoparticles for Controllable Paclitaxel Release

Abstract:Novel biodegradable core-crosslinked nanoparticles (CNPs) consisting of methoxy poly(ethylene glycol)-block-poly(E-caprolactone-co--cinnamoyloxy-E-caprolactone) (mPEG-b-P(CL-co-CCL)) were prepared and evaluated for paclitaxel (PTX) delivery. mPEG(113)-b-P(CL65.2-co-CCL10.1) had a higher drug loading efficiency (95%) compared to mPEG(113)-b-PCL93.1 (43%). The stability of NPs has been largely improved and PTX release was significantly inhibited by crosslinking via UV irradiation at =254nm. MTT assays demonstrated that both blank non-crosslinked and crosslinked NPs showed low cytotoxicity to NCL-H460 cells while PTX-loaded non-crosslinked and crosslinked NPs exhibited obvious cytotoxicity against NCL-H460 cells, and the cytotoxicity was both dose-dependent and time-dependent. Furthermore, after 48h incubation the cell viability of PTX-loaded crosslinked NPs was lower compared to that of PTX-loaded non-crosslinked NPs or free PTX. These properties indicated that CNPs prepared from mPEG-b-P(CL-co-CCL) have great potentials as carriers for drug delivery.
Paclitaxel-loaded Nanoparticles of Star-Shaped Cholic Acid-Core PLA-TPGS Copolymer for Breast Cancer Treatment.

Abstract:A system of novel nanoparticles of star-shaped cholic acid-core polylactide-d-α-tocopheryl polyethylene glycol 1000 succinate (CA-PLA-TPGS) block copolymer was developed for paclitaxel delivery for breast cancer treatment, which demonstrated superior in vitro and in vivo performance in comparison with paclitaxel-loaded poly(d,l-lactide-co-glycolide) (PLGA) nanoparticles and linear PLA-TPGS nanoparticles. The paclitaxel- or couramin 6-loaded nanoparticles were fabricated by a modified nanoprecipitation method and then characterized in terms of size, surface charge, surface morphology, drug encapsulation efficiency, and in vitro drug release. The CA-PLA-TPGS nanoparticles were found to be spherical in shape with an average size of around 120 nm. The nanoparticles were found to be stable, showing no change in the particle size and surface charge during 90-day storage of the aqueous solution. The release profiles of the paclitaxel-loaded nanoparticles exhibited typically biphasic release patterns. The results also showed that the CA-PLA-TPGS nanoparticles have higher antitumor efficacy than the PLA-TPGS nanoparticles and PLGA nanoparticles in vitro and in vivo. In conclusion, such nanoparticles of star-shaped cholic acid-core PLA-TPGS block copolymer could be considered as a potentially promising and effective strategy for breast cancer treatment.

Abstract:Purpose: In the present study, to achieve high paclitaxel (PTX) loading in a conjugated drug delivery system with minimal long-term side effects, we formulated a novel degradable stereocomplexed micelle-like particle with a core-shell structure. Materials and methods: In this system, methoxy polyethylene glycol (MPEG) acted as the hydrophilic shell, and the stereocomplex of polylactic acid with PTX (SCPLA-PTX) acted as the hydrophobic core. The MPEG-SCPLA-PTX micelle-like particles were synthesized via the self-assembly of a MPEG-poly L-lactic acid (PLLA) copolymer with a PTX-poly D-lactic acid-PTX copolymer. The resultant copolymers and their intermediates were characterized using 1H nuclear magnetic resonance and GPC. Micelle-like particles with different molecular weight ratios of MPEG and PLLA were synthesized to demonstrate the functions of both components. Results: PTX loading into MPEG2000Da-PLLA6000Da particles reached as high as 20.11%. At 216 h, the cumulative release from MPEG5000Da-PLLA6000Da, MPEG2000Da-PLLA6000Da, and MPEG5000Da-PLLA22000Da particles were 51.5%, 37.7%, and 52.0%, respectively. Conclusions: According to the cell uptake experiments, inhibition of tumor cell growth was satisfactory, indicating that the stereocomplexed particles developed in the present study can be employed as a promising nanocarrier for PTX delivery.
Nanoformulation of Paclitaxel: Exploring the Cyclodextrin / PLGA Nano Delivery Carrier to Slow Down Paclitaxel Release, Enhance Accumulation in Vivo

Abstract:Background: Improving the aggregation and penetration in tumor sites increases the anti-tumor efficacy of nanomedicine. In the current study, we designed cyclodextrin modified PLGA nanoparticles loaded with paclitaxel to elevate the accumulation and prolong circulation of chemotherapy drugs in vivo. Methods: The PLGA nanoparticles loaded with paclitaxel (PTX PLGA NPs) and cyclodextrin (CD) modified PLGA nanoparticles loaded with paclitaxel (PTX PLGA/CD NPs) were prepared using the emulsification solvent evaporation method. The nanoparticles were characterized by particle size, zeta potential, encapsulation efficiency, infrared spectroscopy analysis and X-Ray diffraction (XRD). Then, drug release of the nanoparticles was evaluated via reverse dialysis method in vitro. Finally, the in vivo distribution fate and pharmacokinetic characteristics of the nanoparticles were assessed in mice and rats. Results: The average particle size, zeta potential, and encapsulation efficiency of PTX PLGA NPs were (163.57±2.07) nm, - (20.53±2.79) mV and (60.44±6.80)%. The average particle size, zeta potential, and encapsulation efficiency of PTX PLGA/CD NPs were (148.57±1.66) nm, - (11.42±0.84) mV and (85.70±2.06)%. In vitro release studies showed that PTX PLGA/CD NPs were released more slowly compared to PTX PLGA NPs under normal blood pH conditions, while PTX PLGA/CD NPs were released more completely under tumor site pH conditions. The modified PLGA nanocarrier (PLGA/CD NPs) increased drug residence time and accumulation than the plain PLGA nanocarrier (PLGA NPs) in vivo distribution. In addition, the elimination half-life, area under the drug-time curve, and maximum blood concentration of the nanoparticle group were higher than those of Taxol®, especially the PTX PLGA/CD NPs group, which was significantly different from Taxol® and plain nanoparticle groups (p<0.001). Conclusions: The 2-HP-β-CD modified PLGA nanoparticles prolonged circulation time and accumulation of the chemotherapy drug paclitaxel in vivo.

Abstract:Breast cancer is one of the most devastating cancers that have no cure. Many therapeutic and diagnostic strategies have been extensively studied in the past decade. Cancer nanotechnology has developed as a promising approach in preclinical examinations, allowing for the early exposure of primary tumors and metastases and the efficient destruction of cancer cells. The current study aims to fabricate and analyze polymeric nanoparticles for breast cancer therapy established on a copolymeric block of α-tocopheryl polyethylene glycol 1000-b-polycaprolactone (TPGS-b-PCL) incorporating paclitaxel (PTX) (termed as T-b-P@PTX). Characterization of the physical, chemical, and structural properties and the in vitro biological activity of TPGS-b-PCL (T-b-P@PTX) obtained by the nanoprecipitation method. The in vitro cytotoxicity of the nanoparticles was explored by MTT assay using MCF-7 breast cancer cells in a time-dependent manner. The IC 50 values T-b-P@PTX for MCF-7 cells 543.7 ± 2.58, 130.2 ± 3.54, 66.99 ± 3.43 in 24, 48, 72-h. Biochemical experiments showed that T-b-P@PTX has a strong ability to induce cell death and to be internalized in the MCF-7 cells. Ethidium bromide (EB)-acridine orange (AO) and nuclear DAPI staining methods investigated the morphological changes of the cells. Considering all the advantages, Paclitaxel-loaded T-b-P@PTX polymeric nanoparticle is a promising approach for treating breast cancer.

Abstract:Paclitaxel appears to be a potential substrate of the multidrug resistance protein p-glycoprotein, thus preventing itself from entry into the brain and penetrating blood-brain barrier poorly. In this study, the main objective was to design paclitaxel formulation using PLGA-based nanoparticles with different additives and surface coatings to facilitate the paclitaxel transport through MDCK cell monolayer. PLGA-based nanoparticles of around 200 nm without and with additives and surface coatings were developed by direct dialysis. The transendothelial electrical resistance (TEER) variation of MDCK cell monolayer on the cell inserts imposed by paclitaxel-loaded nanoparticles with and without additives was investigated. (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, a tetrazole) (MTT) assay was used to quantify the cell viability of C6 glioma cells after administration of formulations on the topical side. Investigations showed that particles with additives were able to enhance cellular uptake more than surface-coated particles. TEER values dropped upon the introduction of paclitaxel-loaded PLGA nanoparticles to the cell inserts. After incubation for 24 h, the cell viability of C6 glioma cells in the wells treated with nanoparticles was lower than that of the control wells without particles. Taken together, PLGA nanoparticles with vitamin E TPGS and polysorbate 80 as additives were successfully obtained as paclitaxel formulations, demonstrating great potential for the delivery of paclitaxel through MDCK cell monolayer in vitro.
Release of paclitaxel from polylactide-co-glycolide (PLGA) microparticles and discs under irradiation

Abstract:Paclitaxel is a promising anti-cancer drug as well as a radiosensitizer for chemotherapy and radiotherapy applications. Because of the poor solubility of paclitaxel in water and most pharmaceutical reagents, it is usually formulated with an adjuvant called Cremophor EL, which causes severe side effects. This work develops new dosage forms of paclitaxel for controlled release application, which do not require the adjuvant and, thus, can avoid its associated side effects. Paclitaxel was encapsulated into the PLGA matrix with various additives such as polyethylene glycol (PEG), isopropyl myristate (IPM) and d-alpha tocopheryl polyethylene glycol (Vitamin E TPGS). These additives were used to enhance the release rate of paclitaxel from the polymer matrix. Spray-drying and an hydraulic press were used to prepare paclitaxel-PLGA microspheres and discs. The microspheres and discs were given different irradiation doses to investigate their effects on the surface morphology (characterized by SEM, AFM and XPS) and in vitro release properties. There seems to be a small effect of the ionizing radiation on various formulations. Although the irradiation did not cause observable changes on the morphology of the polymer matrix, the release rate can be enhanced by a few per cent. It was found that PEG has the highest enhancement effect for release rate among all the additives investigated in this study.

Abstract:Background: Biodegradable nanoparticles with diameters between 100 nm and 500 nm are of great interest in the contexts of targeted delivery. Objective: The present work provides a review concerning the effect of binary organic solvents together with emulsifier on particle size as well as the influence of particle size on the in vitro drug release and uptake behavior. Methods: The polymeric lipid nanoparticles (PLNs) with different particle sizes were prepared by using binary solvent dispersion method. Various formulation parameters such as binary organic solvent composition and emulsifier types were evaluated on the basis of their effects on particle size and size distribution. PLNs had a strong dependency on the surface tension, intrinsic viscosity and volatilization rate of binary organic solvents and the hydrophilicity/hydrophobicity of emulsifiers. Acetone-methanol system together with pluronic F68 as emulsifier was proved to obtain the smallest particle size. Then the PLNs with different particle sizes were used to investigate how particle size at nanoscale affects interacted with tumor cells. Results: As particle size got smaller, cellular uptake increased in tumor cells and PLNs with particle size of similar to 120 nm had the highest cellular uptake and fastest release rate. The paclitaxel (PTX)-loaded PLNs showed a size-dependent inhibition of tumor cell growth, which was commonly influenced by cellular uptake and PTX release. Conclusion: The PLNs would provide a useful means to further elucidate roles of particle size on delivery system of hydrophobic drugs.

Abstract:Objective To investigate the preparation and in vitro evaluation of paclitaxel nanoparticles with a poly(lactide-co-glycolide) carrier.Methods TAX-PGLA-NPs were prepared by the modified solvent diffusion method.The different type of emulsifier and process parameters involved were investigated with respect to the nanopartide mean size.The properties of nanoparticles were primarily studied by a dynamic laser particle size analyzer,transmission electron microscopy and X-ray powder diffraction (XRD).In addition,in vitro release characteristics of freeze-dried powder was also investigated.Results The preparation process was optimized as follows:the volume ratio of water to organic phase was 20:10,both the surfactant and polymer concentration were 1%,the homogenizer speed was at 16 000 r·min-1,the drug concentration was 0.1%.The optimized result showed that TAX-PLGA-NPs were spherical with an average diameter of 99.0 nm,Zeta potential of 58.3 mV,drug-loading content of 7.10%,encapsulation efficiency of 56.77%.The release behavior of drug in vitro showed a sustained-release,with an initial burst effect,subsequently a slower rate stage.The entrapment of TAX in nanoparticles was ensured by DSC and XRD analysis.Conclusion PLGA-NPs might be used as a new carrier for paclitaxel.
Development and Characterization of Paclitaxel-Loaded Li-pid-Coated Mesoporous Silica Nanoparticles for Ph-Responsive Tumor-Targeted Drug Delivery

Abstract:Nanoparticle carriers can selectively deliver the drug cargo to tumor cells, thus having the ability to pre-vent early drug release, reduce non-specific cell binding, and prolong in vivo drug retention. We con-structed paclitaxel (PTX)-loaded lipid-shell mesoporous silica nanoparticles (LMSNs) for targeted an-ti-cancer drug delivery. The physical properties of PTX-LMSNs were analyzed by scanning electron mi-croscopy (SEM) and transmission electron microscopy (TEM). The drug loading (DL%) and entrapment efficiency (EE%) of PTX-LMSNs were measured by high performance liquid chromatography (HPLC). In vitro drug release test, in vivo imaging, tissue distribution and pharmacokinetics of PTX-LMSNs were also evaluated. The SEM examination showed that MSNs were sphere, whereas TEM showed that they were rich in fine pores. The uniform core-shell structure of PTX-LMSNs was also verified by TEM. The DL capacity of PTX-LMSN was as high as 21.75%, and PTX was released from the nanoparticles in vitro in a pH-dependent manner. The cumulative amount of free PTX increased at lower pH, which is conducive to selective drug release from LMSNs in the acidic tumor tissues. In vivo imaging showed prolonged reten-tion of PTX-LMSNs, which is beneficial to their therapeutic efficacy. In addition, PTX-LMSNs were primarily concentrated in the liver. Pharmacokinetic experiments showed that the half-life of PTX-LMSNs was 23.21% longer and 79.24% higher than that of Taxol. Together, LMSNs are a highly promising antineoplastic drug carrier system.

Abstract:Microspheres of a new kind of copolymer, poly(lactic acid)-poly(ethylene glycol)-poly(lactic acid) (PLA-PEG-PLA), are proposed in the present work for clinical administration of an antineoplastic drug paclitaxel with hypothesis that incorporation of a hydrophilic PEG segment within the hydrophobic PLA might facilitate the paclitaxel release. Paclitaxel-loaded PLA-PEG-PLA microspheres of various compositions were prepared by the solvent extraction/evaporation method. Characterization of the microspheres was then followed to examine the particle size and size distribution, the drug encapsulation efficiency, the colloidal stability, the surface chemistry, the surface and internal morphology, the drug physical state and its in vitro release behavior. The effects of polymer types, solvents and drug loading were investigated. It was found that in the microspheres the PEG segment was homogeneously distributed and caused porosity.Significantly faster release from PLA-PEG-PLA microspheres resulted in comparison with the PLGA counterpart. Incorporation of water-soluble solvent acetone in the organic solvent phase further increased the porosity of the PLA-PEG-PLA microspheres and facilitated the drug release. A total of 49.6% sustained release of paclitaxel within I month was achieved. Potentially, the presence of PEG on the surface of PLA-PEG-PLA microspheres could improve their biocompatibility. PLA-PEG-PLA microspheres could thus be promising for the clinical administration of highly hydrophobic antineoplastic drugs such as paclitaxel. (C) 2003 Elsevier Ltd. All rights reserved.

Abstract:Background: As a promising anticancer drug, severe side-effects of current clinical formulations for paclitaxel have restricted its use, developing a better technical-economical formulation for paclitaxel delivery is needed. Method: In this study, the compound of folate-poly(ethylene glycol) (PEG)-phosphatidylethanolamine was synthesized and characterized with Fourier transform infrared spectroscopy and nuclear magnetic resonance spectroscopy. The solid-liquid lipid nanoparticle (SLLN) for paclitaxel modified with folate and poly(ethylene glycol) (folate-PEG-SLLN) was prepared and characterized. Morphology of folate-PEG-SLLN was examined by transmission electron microscopy. The particle size and zeta potential were performed by Zetapals. Encapsulation efficiency was analyzed by HPLC. The in vitro drug release of paclitaxel was investigated via membrane dialysis. The in vivo pharmacokinetics was measured with male Sprague-Dawley rats. Treatment efficiency was investigated with the mouse with sarcoma180 ascites tumor. Results: Paclitaxel loaded on the newly designed binary SLLN showed a longer and sustained in vitro releasing property. More importantly, S180 tumor-bearing mice treated with paclitaxel-loaded SLLN exhibited higher tumor inhibition rate, comparing with animals administered with paclitaxel injection alone (45.3% and 37.3%, respectively). Conclusion: The newly developed paclitaxel delivery system may have improved in vivo antitumor activity. The results demonstrated a great interest to use folate-mediated SLLN as a prospective drug delivery system for paclitaxel.</.

Abstract:Purpose The aim of this study is to develop novel polylactide/poly(ethylene glycol) (PLA/PEG) micelles as carrier of hydrophobic drug (paclitaxel) by direct dissolution method without using any organic solvents. The in vitro and in vivo release properties were studied in comparison with micelles prepared by dialysis. Methods Drug encapsulation efficiency (EE) and loading content (LC) of the micelles were evaluated by high-performance liquid chromatography. Micelle diameters and structures were determined by dynamic light scattering and transmission electron microscopy. In vitro release was performed in phosphate-buffered saline (pH 7.4) at 37°C, and in vivo experiments were realized in lung cancer-bearing mice. Results Similar EE and LC values were obtained for micelles by direct dissolution method and those by dialysis. L- and D-PLA/PEG mixed micelles present higher drug encapsulation ability than separate micelles due to stereocomplexation. Micelle diameters are enlarged by drug-loading. Faster drug release was obtained for micelles by direct dissolution than those by dialysis. Compared with current clinical formulation and micelles by dialysis, paclitaxel-loaded micelles by direct dissolution showed the highest antitumor ability. Conclusion The L- and D-PLA/PEG mixed micelles by direct dissolution method present many advantages such as easy formulation and absence of toxic organic solvents, which shows great potential as carrier of hydrophobic drugs.

Abstract:Novel nanocarriers for paclitaxel (PTX) were developed based on Pluronic-b-poly(ε-caprolactone) bearing pendant benzyl-oxycarbonylmethyl (BOM) groups and carboxyl groups (Pluronic-b-P(CL-co-BCL), FB)/(Pluronic-b-P(CL-co-CCL), FC). The formation and the physicochemical properties of paclitaxel-loaded polymeric micelles, including FB/PTX micelles and FC/PTX micelles, were investigated by various methods. The results demonstrated that paclitaxel was amorphous in the micellar core and the micelles were on nanoscale. Besides the hydrophobic-hydrophobic interaction in both micelles, there was π-π interaction in the FB/PTX micelles while hydrogen bonding interaction existed in the FC/PTX micelles. Thus the different interaction between the drug and the polymer endowed the polymers with different binding forces which determined the various properties of the nanocarriers. The comparative study revealed that the compatibility was improved due to the introduction of the pendant groups according to the calculation based on the Flory-Huggins interaction parameter (χdc). The two nanocarriers also displayed high encapsulation efficiency, which could reach 88.61 ± 5.33% and 90.7 ± 2.08%, and they could also provide a continuous and sustained in vitro PTX release and the release half time was greatly enhanced in comparison with commercial Taxol®. Furthermore, the in vitro anti-tumor efficiency revealed that the FC PTX-loaded micelles had the best anti-tumor activity against C6 glioma cells inducing cell apoptosis and the in vitro blood compatibility and the in vivo long-circulation characteristics were well retained for the FB and FC PTX-loaded micelles in comparison with the original Pluronic-b-PCL. Therefore, these findings indicated that the functionalized Pluronic-b-poly(ε-caprolactone) micelles would be efficient nanocarriers for paclitaxel.

Abstract:Highly ordered mesoporous silica nanoparticles (MSNs) with pore diameter of 2.754nm and particle size of 115±15nm were prepared with etching method. Homogeneous PEGylated lipid bilayer with 10-15nm thickness was coated around the surface of MSNs using film hydration method. Systematic optimization and characterization of co-encapsulation process of paclitaxel (Tax) and curcumin (Cur) into PEGylated lipid bilayer coated mesoporous silica nanoparticles (PLMSNs) were performed carrying out single factor test, associated with Box-Behnken Design. The concentration of encapsulated drugs was measured by reversed phase high performance liquid chromatography (RP-HPLC) method. Optimal factor settings were as follows: 50mg MSNs, ratio of MSNs to lipid (w/w)=1:1.11, and ratio of lipid to CHO (w/w)=3.93:1. The average experimental EETax, EECur and stability score value were (77.48±2.73) %, (30.70±3.56) % and 4 point respectively based on the conditions mentioned above. Morphology determination of Tax-Cur-PLMSNs revealed that the composite nanoparticles were spherical particals with uniform dispersion. In vitro release experiment indicated that PLMSNs improved dissolution of Tax compared to Tax powder suspension and exhibited sustained release property. Tax-Cur-PLMSNs manifested definite and persistently promoted cytotoxic effect against canine breast cancer cells. This prolonged and enhanced activity of Tax-Cur-PLMSNs might contribute to its sustained release effect.
Synthesis of Nanoparticles of Star-Shaped Mannitol-Core Pla-Tpgs Copolymer for Delivery of Paclitaxel and Activity of Anti-Prostate Cancer

Abstract:The objective of this research is to fabricate novel nanoparticles (NPs) of star-shaped mannitol-functionalized polylactic acid-D-alpha-tocopheryl polyethylene glycol 1000 suclinat(PLA-TPGS) copolymer for paclitaxel delivery for prostate cancer treatment, and evaluate their therapeutic effects in prostate cancer cell line in comparison with the linear PLGA NPs and PLA-TPGS NPs. The paclitaxel-loaded M-PLA-TPGS NPs, prepared by a modified nano-precipitation method, were observed by field emission scanning electron microscopy (FESEM) to be near-spherical shape with narrow size distribution. The drug-loaded NPs were further characterized in terms of size, surface charge, drug content, encapsulation efficiency and in vitro drug release. In vitro drug release exhibited biphasic pattern with initial burst release followed by slow and continuous release. The cellular uptake level of M-PLA-TPGS NPs was demonstrated higher than linear PLGA NPs and PLA-TPGS NPs in PC-3 cells. The data also showed that the paclitaxel-loaded M-PLA-TPGS nanoparticles have higher antitumor efficacy than that of linear PLA-TPGS nanoparticles and commercial Taxol in vitro. The star-shaped copolymer M-PLA-TPGS could be used as a potential and promising molecular biomaterial applied in developing novel nanoformulation for prostate cancer treatment.
The Preparation, Characterization, and Pharmacokinetic Studies of Chitosan Nanoparticles Loaded with Paclitaxel/Dimethyl-Beta-Cyclodextrin Inclusion Complexes

Abstract:A novel biocompatible and biodegradable drug-delivery nanoparticle (NP) has been developed to minimize the severe side effects of the poorly water-soluble anticancer drug paclitaxel (PTX) for clinical use. PTX was loaded into the hydrophobic cavity of a hydrophilic cyclodextrin derivative, heptakis (2,6-di-O-methyl)-β-cyclodextrin (DM-β-CD), using an aqueous solution-stirring method followed by lyophilization. The resulting PTX/DM-β-CD inclusion complex dramatically enhanced the solubility of PTX in water and was directly incorporated into chitosan (CS) to form NPs (with a size of 323.9-407.8 nm in diameter) using an ionic gelation method. The formed NPs had a zeta potential of +15.9-23.3 mV and showed high colloidal stability. With the same weight ratio of PTX to CS of 0.7, the loading efficiency of the PTX/DM-β-CD inclusion complex-loaded CS NPs was 30.3-fold higher than that of the PTX-loaded CS NPs. Moreover, it is notable that PTX was released from the DM-β-CD/CS NPs in a sustained-release manner. The pharmacokinetic studies revealed that, compared with reference formulation (Taxol(®)), the PTX/DM-β-CD inclusion complex-loaded CS NPs exhibited a significant increase in AUC(0→24h) (the area under the plasma drug concentration-time curve over the period of 24 hours) and mean residence time by 2.7-fold and 1.4-fold, respectively. Therefore, the novel drug/DM-β-CD inclusion complex-loaded CS NPs have promising applications for the significantly improved delivery and controlled release of the poorly water-soluble drug PTX or its derivatives, thus possibly leading to enhanced therapeutic efficacy and less severe side effects.
