Abstract:A novel cascade radical addition/cyclization reaction of non-activated olefins and oxamic acids has been proposed. Under transition metal-free conditions, 36 quinazolinone derivatives containing an amide moiety were successfully synthesized, with the highest yield being 81%. This method involves the preparation of aminoacyl fused quinazolinone derivatives under mild conditions, offering advantages such as a high yield, a broad substrate compatibility, and a high atom economy.

Abstract:In recent years, much attention has been directed towards the syntheses of 1,4-dihydropyridyl compounds due to the fact that 1,4-dihydropyridyl compounds possess a variety of biological activities.[1] In view of the importance of polyhydroquinoline derivatives, many classical methods for the synthesis of polyhydroquinoline derivatives were reported[2] by conventional heating and refluxing approaches in the presence of organic solvent. These methods, however, involve long reaction time, harsh reaction conditions, the use of a large quantity of organic solvent and unsatisfactory yields. Abstract:An efficient and regioselective method to multisubstituted quinoline and quinazoline derivatives with diaryliodonium salts catalyzed by Cu(OTf)(2) has been presented. The reaction proceeded via cascade annulation in the mode of [2+2+2] form. This novel strategy enables use of readily available materials and bears great flexibility on the substitution groups.

Abstract:Atom- and step-economy in IBX assisted diversity-oriented synthesis is achieved with a versatile AQ auxiliary α-amino acid analogs offering rapid access to polycyclic spiro-quinolines featuring a quaternary stereocenter in 20%–91% yields under mild conditions via 7,8-dearomatization of quinolines. Free of a preinstalled activation group is highlight of this intramolecular oxidation spiroannulation tandem reaction. This type of N-heterospirocycles, traditionally difficult to access, may open the door to a potentially interest scaffold for synthetic and medicinal chemistry.

Abstract:A solvent controlled regioselective metal-free synthesis of iodo-substituted N-heterocycles has been developed. Abstract:We have developed an efficient method for the synthesis of unsymmetrical and symmetrical 2,2′-biquinoline derivatives under 55% HI-catalyzed conditions using oxygen as the oxidant and cheap and readily available aromatic amines as feedstocks in one pot. This transformation undergoes multistep reactions such as multiple radical additions and isomerization, cyclization, and oxidation. Mechanistic investigations indicate that the transformation may involve a free radical cascade cyclization pathway.

Abstract:A facile one-pot, four-component reaction for the synthesis of pyrrolo[1,2-a]quinolines is described. Knoevenagel condensation reaction of malononitrile and aromatic aldehydes gave 2-arylmethylidenemalononitriles which on treatment with quinoline and cyclohexyl isocyanide under solvent-free conditions afforded 1-(cyclohexylamino)-2-arylpyrrolo[1,2-a]quinoline-3-carbonitriles in good yields.

Abstract:Diverse quinazolinone-[2,3]-fused polycyclic skeletons occupy a prominent position in drug discovery. Even with currently available methods there still remain unmet needs for flexible access to such structures. Herein, we have explored a mild "one pot" procedure for the construction of various quinazolinone-[2,3]-fused polycycles. The procedure involves Pd-catalyzed carbonylation of N -(2-iodophenyl)acetamides, release of the masked terminal amine, and two sequential and spontaneous cyclizations. This generally applicable approach features easy assembly of precursors from readily available starting materials, mild reaction conditions, non-cumbersome operation, and polycyclic diversity.

This method is enabled by a three-component cascade annulation of readily available aryl diazonium salts, nitriles, and alkynes. This reaction is catalyst- and additive-free. Abstract:An efficient protocol was developed for the synthesis of fused heterocyclic multiring compounds pyrrolo[1,2-a]quinolin-1(2H)-ones via a AuBr3/AgSbF6-catalvzed cascade transformation. Significantly, the strategy affords a straightforward and efficient approach to construction of tricyclic lactam molecular architectures in which two new C-C bonds and one new C-N bond are formed in a one-pot synthetic operation from simple starting materials. Moreover, a broad spectrum of substrates can participate in the process effectively to produce the desired products in good yields and with excellent regio- and chemoselectivities.
