Abstract:A facile promiscuous reaction catalyzed by Acylase ‘Amano’ was developed for the preparation of spirooxindole derivatives. At the optimized reaction condition, the acylase-catalyzed domino reactions could provide a series of different spirooxindole products in moderate yields and good diastereoselectivity. A chemo-enzymatic two-step process was also designed successfully, and several representative spirooxindole derivatives could be obtained in nearly quantitative yields. Additionally, one tentative mechanism for this enzyme-catalyzed process was proposed.

Abstract:A simple and environmentally friendly method to synthesize spiropyrazolo[3,4-b]pyridines derivatives starting from isatin, cyclic-1,3-diketone and 3-methyl-5-aminopyrazole was developed. With the optimized conditions for enzymes, solvents, enzyme loading and reaction time in hand, 9 compounds were obtained in acceptable yields. Abstract:The ubiquitous presence of spirooxindole architectures with several functionalities and stereogenic centers in bioactive molecules has been appealing for the development of novel methodologies seeking their preparation in high yields and selectivities. Expansion and refinement in the field of asymmetric organocatalysis have made possible the development of straightforward strategies that address these two requisites. In this review, we illustrate the current state-of-the-art in the field of spirooxindole synthesis through the use of non-covalent organocatalysis. We aim to provide a concise overview of very recent methods that allow to the isolation of unique, densely and diversified spirocyclic oxindole derivatives with high structural diversity via the use of cascade, tandem and domino processes.

Abstract:An efficient method for a domino [3+2] cycloaddition/deamination/imine hydrolysis reaction of D-A cyclopropane with inexpensive thiourea catalyzed by Lu(OTf)3 has been developed. A series of spiro-γ-thiolactone oxidized indole compounds with potential biological activity were obtained up to 98% yields. Experiments to investigate the mechanism showed that γ-thiolactone products were from the hydrolysis of the imine intermediate. In addition, the asymmetric catalytic synthesis of D-A cyclopropane and thiourea has been successfully developed to afford the spiro-γ-thiolactone compounds in good enantioselectivity (up to 98% ee). The utility of this method was showcased by the facile transformation and biological activity of products.

Abstract:A common synthetic method involving two tandem processes to prepare varieties of oxindole derivatives, which are important bioactive cores that exist widely in natural products and drugs, from similar materials under mild conditions is described. Herein a substituent-controlled product-selective strategy succeeded in synthesizing spirooxindoles and oxazolyloxindoles with excellent diastereoselectivity from methyleneindolinones and -bromoamides. The synthesis has clear chemoselectivity, which relies on different substituent groups at the nitrogen atoms of -bromoamides. From mechanism analysis, the spirooxindoles were constructed via elimination-Michael-Michael addition from N-benzyloxy--bromoamides, while oxazolyloxindoles were achieved via cyclization-Michael addition from N-benzoyl--bromoamides, both of which cleverly combined the nucleophilic component formation and nucleophile-triggered cascade reactions in one pot.

Abstract:Asymmetric synthesis of spiro[indoline‐3,4‐pyrrolo [3,4‐b]pyridines] derivatives was first developed through organocatalytic cascade Knoevenagel/Michael/cyclization reaction using a quinidine ‐derived squaramide. Under the optimized conditions, the three‐component reaction of isatins, cyanoacetates, and 3‐aminomaleimides yield the desired heterocycle‐fused spirooxindoles in good yields (78–91%) with 53‐99% enantiomer excess (ee). Notably, this reaction enabled a broad substrate scope under mild conditions, and provided a convenient method for enantioselective construction of diverse spirooxindoles combined with dihydropyridine and maleimide skeleton, which brought great potential to build new bioactive chemical entities.

Abstract:A novel enzyme-initiated multicomponent reaction from readily available aldehyde, nitrostyrene, cyclohexanone and acetamide substrates was discovered, enabling the facile construction of six new CC/N bonds and two rings in single step, one-pot operation, for the synthesis of spirooxazino derivatives in moderate to high yields. Several methods such as isotope labelling and enzyme mutation were used to probe the possible mechanism of this complex synthesis.

Abstract:Palladium-catalyzed intramolecular dearomative reductive-Heck reaction of C2- substituted indoles is developed, which provides access to structurally diverse 3,2'-spiropyrrolidine oxindoles. By changing the hydride source to AcONa base, direct C3-arylation products [2,3-b] quinolinones are achieved in good yields. Abstract:The organocatalyzed asymmetric Mannich-type spirocyclization was employed to access the spiro[pyrrolidin-3,3'-oxindoles], a biologically privileged ring structure, from readily available 2-oxotryptaphan methyl ester and aromatic aldehydes. The stereoselectivity of this cyclization are closely correlated with the organocatalyst and solvent used for the reaction. The best diastereoselectivity (d.r. >= 99%) was achieved using only 5% loading of the bifunctional organocatalyst, Takemoto's thiourea, and dioxane as solvent, resulting in the formation of the the spirocycle with the (2'R, 3R) configuration. The yield could be increased up to 97% when excess of benzaldehyde was used to overcome its trimerization. (C) 2018 Elsevier Ltd. All rights reserved.
