Abstract:A facile promiscuous reaction catalyzed by Acylase ‘Amano’ was developed for the preparation of spirooxindole derivatives. At the optimized reaction condition, the acylase-catalyzed domino reactions could provide a series of different spirooxindole products in moderate yields and good diastereoselectivity. A chemo-enzymatic two-step process was also designed successfully, and several representative spirooxindole derivatives could be obtained in nearly quantitative yields. Additionally, one tentative mechanism for this enzyme-catalyzed process was proposed.

Abstract:A simple and environmentally friendly method to synthesize spiropyrazolo[3,4-b]pyridines derivatives starting from isatin, cyclic-1,3-diketone and 3-methyl-5-aminopyrazole was developed. With the optimized conditions for enzymes, solvents, enzyme loading and reaction time in hand, 9 compounds were obtained in acceptable yields. Abstract:The ubiquitous presence of spirooxindole architectures with several functionalities and stereogenic centers in bioactive molecules has been appealing for the development of novel methodologies seeking their preparation in high yields and selectivities. Expansion and refinement in the field of asymmetric organocatalysis have made possible the development of straightforward strategies that address these two requisites. In this review, we illustrate the current state-of-the-art in the field of spirooxindole synthesis through the use of non-covalent organocatalysis. We aim to provide a concise overview of very recent methods that allow to the isolation of unique, densely and diversified spirocyclic oxindole derivatives with high structural diversity via the use of cascade, tandem and domino processes.

Abstract:An efficient enantioselective synthesis of spiro[indoline-3,4'-pyrano[2,3-c]pyrazole] derivatives by a cascade reaction between pyrazolones and isatylidene malononitriles is described. With only 1 mol% of (DHQD)2PYR, chiral spirooxindole derivatives have been produced in excellent yields (96-99%) with good-to-excellent enantioselectivities (up to 91% ee).

Abstract:A direct method to construct 3,4-dihydropyridin-2-ones by enzymatic condensation of aldehyde with cyanoacetamide and 1,3-dicarbonyl compounds was developed. One ring and four new bonds (two C–C, one C–N, one CC) were formed in one pot. And reaction conditions involving hydrolases, solvents, substrate molar ratios, and hydrolase loading were optimized. A series of new compounds based on the 3,4-dihydropyridin-2-one core were synthesized by the unprecedented three-component domino reaction.

Abstract:The first organocatalytic asymmetric synthesis of a spirooxindole skeleton incorporated with a cyclobutane moiety has been successfully developed on the basis of H-bond-directing dienamine activation. Structurally complex spirocyclobutyl oxindoles, which possess four contiguous stereocenters, including one spiro quaternary center, were obtained in good yields (up to 83%) with excellent β,γ-regioselectivity (>19:1) and stereocontrol (up to >19:1 dr and 97% ee).

Abstract:Spirooxindoles have emerged as promising architec-tures for engineering biologically active compounds. The diaster-eodivergent construction of unique scaffolds of this type with full control of continuous chiral centers including an all-carbon quaternary stereogenic center is yet to be developed. Here, we report an unprecedented diastereodivergent desymmetric [3 + 3] annulation of oxabicyclic alkenes with enals enabled by N-heterocyclic carbene (NHC)/Rh cooperative catalysis, leading to a series of enantiomerically enriched spirooxindole lactones with excellent enantioselectivities (up to >99% ee) and diastereoselectivities (up to >95:5 dr). The combined catalyst system comprises a rhodium complex that controls the configuration at the electrophilic carbon and an NHC catalyst that controls the configuration at the nucleophilic oxindole-containing carbon; thus, four stereoisomers of the spirooxindole products can be readily obtained simply by switching the configurations of the two chiral catalysts. Transformations of the chiral spirooxindoles delivered synthetically useful compounds. Importantly, those chiral spirooxindoles arrested mammalian cells in mitosis and exhibited potent antiproliferative activities against HeLa cells. Significantly, both absolute and relative configurations exert prominent effects on the bioactivities, underscoring great importance of catalytic asymmetric diastereodivergent synthesis beyond creating useful tools for the exploration of structure-activity relationships.

Abstract:An efficient method for a domino [3+2] cycloaddition/deamination/imine hydrolysis reaction of D-A cyclopropane with inexpensive thiourea catalyzed by Lu(OTf)3 has been developed. A series of spiro-γ-thiolactone oxidized indole compounds with potential biological activity were obtained up to 98% yields. Experiments to investigate the mechanism showed that γ-thiolactone products were from the hydrolysis of the imine intermediate. In addition, the asymmetric catalytic synthesis of D-A cyclopropane and thiourea has been successfully developed to afford the spiro-γ-thiolactone compounds in good enantioselectivity (up to 98% ee). The utility of this method was showcased by the facile transformation and biological activity of products.

Abstract:A catalytic asymmetric nitroso aldol intramolecular transesterification cyclization domino reaction between a C3 alkyl ester substituted oxindole and nitrosobenzene for the construction of chiral spirooxindoles was achieved in the presence of a chiral bifunctional tertiary amine thiourea catalyst and water. Optically active spirooxindoles were obtained in moderate yields (up to 76 %) and excellent enantioselectivities (up to 95 % ee).

Abstract:A novel enzyme-initiated multicomponent reaction from readily available aldehyde, nitrostyrene, cyclohexanone and acetamide substrates was discovered, enabling the facile construction of six new CC/N bonds and two rings in single step, one-pot operation, for the synthesis of spirooxazino derivatives in moderate to high yields. Several methods such as isotope labelling and enzyme mutation were used to probe the possible mechanism of this complex synthesis.

Abstract:Asymmetric synthesis of spiro[indoline‐3,4‐pyrrolo [3,4‐b]pyridines] derivatives was first developed through organocatalytic cascade Knoevenagel/Michael/cyclization reaction using a quinidine ‐derived squaramide. Under the optimized conditions, the three‐component reaction of isatins, cyanoacetates, and 3‐aminomaleimides yield the desired heterocycle‐fused spirooxindoles in good yields (78–91%) with 53‐99% enantiomer excess (ee). Notably, this reaction enabled a broad substrate scope under mild conditions, and provided a convenient method for enantioselective construction of diverse spirooxindoles combined with dihydropyridine and maleimide skeleton, which brought great potential to build new bioactive chemical entities.

Abstract:A common synthetic method involving two tandem processes to prepare varieties of oxindole derivatives, which are important bioactive cores that exist widely in natural products and drugs, from similar materials under mild conditions is described. Herein a substituent-controlled product-selective strategy succeeded in synthesizing spirooxindoles and oxazolyloxindoles with excellent diastereoselectivity from methyleneindolinones and -bromoamides. The synthesis has clear chemoselectivity, which relies on different substituent groups at the nitrogen atoms of -bromoamides. From mechanism analysis, the spirooxindoles were constructed via elimination-Michael-Michael addition from N-benzyloxy--bromoamides, while oxazolyloxindoles were achieved via cyclization-Michael addition from N-benzoyl--bromoamides, both of which cleverly combined the nucleophilic component formation and nucleophile-triggered cascade reactions in one pot.

Abstract:The novel construction method of 3,4'-pyran spirooxindole heterocyclic skeleton has always been the focus of attention. Herein we report the highly enantioselective inverse-electron-demand oxa-Diels−Alder cycloaddition reaction of β,γ-unsaturated pyrazole amide and N-diphenyl isatin-derived oxodiene using a bifunctional catalyst. In addition, large-scale experiments confirmed the reliability of the reaction. The resultant products derived from this study can be further transformed.

Abstract:An enantioselective domino reaction of 4-bromo-3-oxobutanoates with isatin-derived ketimines was developed to construct 3-amino-2-oxindoles with a quaternary stereocentre.
