Abstract:Some agronomic characteristics such as height, stem diameter, relative leaf area and leaf length breadth ratio and components such as water content, total polysaccharide, water-soluble polysaccharide, and cellulose contents of 8 kinds Dendrobium officinale from different distribution areas were determined and analyzed. It was found that the differences among these agronomic characteristics have to some extent related to where they are from. Dendrobium officinale from Yandang and Yunnan areas which have moderate plant morphology and robust stem are better then that in other areas for planting in Zhejiang, but for Dendrobium officinale from Yunnan, the greenhouse is nessesary.

Abstract:ObjectiveDendrobium officinale is a perennial epiphytic herb of the Orchidaceae family. It has very strict needs with respect to growth environment and climatic conditions. In order to precisely control the artificial growth environment of Dendrobium candidum, a monitoring system was designed and implemented using a Programmable Logic Controller (PLC) and Supervisory Control and Data Acquisition (SCADA) system.MethodsA platform was created for precise control using a Siemens S7-200CPU224 PLC and different sensors of various environmental parameters as the control unit and SCADA-configured software as the core of the monitoring unit. We here used 4 indexes, soil temperature, soil moisture, humidity, and light, as prediction parameters and established a three-layer feed-forward fuzzy optimization neural network model.ResultsThe system not only allows prediction of the optimal environmental parameters for the growth of Dendrobium candidum, real-time monitoring, and intelligent control but also escapes the shortcomings of traditional back-propagation (BP) neural networks, which suffer from slow convergence, shock, and poor generalization. The current model’s average prediction error is less than 2.5%. It also provides a theoretical basis and decision support for the precision control of planting projects and relevant environment forecasting. The climate in the test area is hot and rainy in summer and colder and drier in winter. The annual precipitation is concentrated in spring and summer, peaking twice, in May and October. The subtropical high temperature was recorded in August, which has little rainfall and is prone to drought. Winter features both cold and warm air and some rainy days, but not as much overall precipitation as summer.

Abstract:Dendrobium Sw.,known as one kind of valuable medicinal herb,are widely used in pharmaceuticals and health products.Under natural conditions,medical Dendrobium endophytes can promote the seedling growth rate,enhance biological resistance and induce synthesis of secondary metabolites.Moreover,medical Dendrobium endophytes can accelerate the growth rate and ensure survival of transplanting.In this paper,the biodiversity of endophytes and the relationship between endophytes and medical Dendrobium were overviewed,and the application of the endophytes was prospected.

Abstract:Herbaceous peony (Paeonia lactiflora Pall.) is a renowned traditional Chinese flower, which has been cherished by people since ancient times. It holds significant value as both an agricultural cash crop and an ornamental and medicinal plant. However, the lack of in vitro propagation system has been long hindered the commercial production of this species and the development of new cultivars. In this study, the low chilling requirement cultivar Paeonia lactiflora 'Hang Baishao' that originated from low latitudes of China, was used to study the in vitro culture of embryos. The results of our experiment showed that the optimal medium for in vitro culture of embryos was a 1/2 Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 6-benzyladenine (6-BA), 1.0 mg/L gibberellic acid (GA3) and 2.0 g/L polyvinyl pyrrolidone (PVP). Additionally, histological observations revealed the crucial role of vascular bundle development in the survival of transplants. Plants with well-developed vascular tissue exhibited better transplanting success. This study presents, for the first time, a protocol for embryos propagation and transplanting of 'Hang Baishao', a cultivar with low chilling requirement (CR) during winter. This study provides an efficient method for in vitro culture of 'Hang Baishao', and makes it possible for large-scale production, utilization, and germplasm preservation of this species.

Abstract:Main conclusionThis review summarizes current knowledge of chromosome characterization, genetic mapping, genomic sequencing, quality formation, floral transition, propagation, and identification in Dendrobium.The widely distributed Dendrobium has been studied for a long history, due to its important economic values in both medicine and ornamental. In recent years, some species of Dendrobium and other orchids had been reported on genomic sequences, using the next-generation sequencing technology. And the chloroplast genomes of many Dendrobium species were also revealed. The chromosomes of most Dendrobium species belong to mini-chromosomes, and showed 2n = 38. Only a few of genetic studies were reported in Dendrobium. After revealing of genomic sequences, the techniques of transcriptomics, proteomics and metabolomics could be employed on Dendrobium easily. Some other molecular biological techniques, such as gene cloning, gene editing, genetic transformation and molecular marker developing, had also been applied on the basic research of Dendrobium, successively. As medicinal plants, insights into the biosynthesis of some medicinal components were the most important. As ornamental plants, regulation of flower related characteristics was the most important. More, knowledge of growth and development, environmental interaction, evolutionary analysis, breeding of new cultivars, propagation, and identification of species and herbs were also required for commercial usage. All of these studies were improved using genomic sequences and related technologies. To answer some key scientific issues in Dendrobium, quality formation, flowering, self-incompatibility and seed germination would be the focus of future research. And genome related technologies and studies would be helpful.

Abstract:The in vitro regeneration of Dendrobium nobile white Christmasseagull through bud formation on the stem apex culture was investigated. The effects of sterilization, hormone and hormone combination on the explant survival rate, adventitious bud induction and proliferation were studied. The results indicated that the sterilization by 0.1 % HgCl2 6 min can improve tender stem apex survival rate, but 0.1 % HgCl2 8 min can improve aged stem apex survival rate. The combination of 0.5 mg/L NAA and 1.0 mg/L BA can improve adventitious bud induction. Adding 1.5 mg/L GA3 can improve the buds proliferation and elongation rate. The addition of spermidine to the MS- based medium also significantly improves the percentage of adventitious bud proliferation and the buds growth.

Abstract:Dendrobium officinale Kimura et Migo is a rare medicinal herbs. The paper screened substrate ratio,light intensity,concentration of fertilizer for its cultivation.The results showed that the optimum proportion of matrix for bark:bamboo charcoal:sphagnum=6:2:2,shading rate of 60%shading,fertilizer concentration was 2 000 times.

Abstract:As a well-known Chinese important medicine, Dendrobium resources are increasingly scarce. In order to avoid the quality problems caused by blind cultivation, we predicted the ecologically suitable areas for three species of important Dendrobium species. In this paper, using the maximum entropy model and ArcGIS, we used a database and literature search to collect the known distribution information of Dendrobium, combined with the relevant ecological factors such as climate, elevation and soil, to divide the geographical distribution of Dendrobium into categories, and find out the main ecological factors that affect the distribution of Dendrobium and thus the most suitable regions in China. The results showed that Dendrobium is mainly distributed south of the Yangtze River And the distribution of the most suitable regions was different for each of these three species of Dendrobium species. The potential suitable areas of Dendrobium nobile Lindl, Dendrobium officinale Kimura et Migo, Dendrobium moniliforme (L.) Sw. were approximately 134.19 x 10(4) km(2) (accounting for 13.98 %), 229.08 x 10(4) km(2) (accounting for 23.86 %) and 188.33 x 10(4) km(2) (accounting for 19.62 %) of China's land area, respectively. Based on our model, annual temperature annual range and annual precipitation are the main ecological variables controlling the distribution of the three species of Dendrobium species, but these three species also have slightly different characteristics. All in all, these species prefer a warm, stable and humid climate and loose soil that is sufficiently aerated and with appropriate moisture properties in order to grow. This will provide a reference for the subsequent introduction, cultivation and local adaptation.

This paper provides the basic principles and characteristics of some DNA molecular marker techniques, including RFLP, allele-specific diagnostic PCR, RAPD, AP-PCR, AFLP,ISSR, SRAP, and DNA sequencing analysis, genechip,and reviews their application progresses in Dendrobium. These techniques provide more effective and reliable methods for discrimination of true and false Dendrobium. Abstract:Dendrobium officinale Kimura & Migo (D. officinale) is a well-recognized traditional Chinese medicinal herb that is both medicinal and edible. Contemporary pharmacological studies have revealed that D. officinale contains abundant bioactive compounds, including polysaccharides, flavonoids, alkaloids, and dendrobine, exhibiting diverse pharmacological properties such as antioxidant, anti-inflammatory, and immunomodulatory effects. However, the industrial application of D. officinale faces many problems, such as the scarcity of wild resources, low natural reproduction rate, and slow growth rate as well as the lack of relevant industrial standards. Nevertheless, substantial advancements, including the exploitation of artificial propagation techniques and breeding of new varieties, have been achieved in recent years. These developments have effectively addressed the challenges associated with its low natural reproduction rate and the scarcity of wild resources. This review summarizes the progress in the industrial development, seedling cultivation, and pharmacological exploration of D. officinale in recent years. Furthermore, it analyzes current research inadequacies and offers strategic solutions to enhance its application in healthcare and medicine.

Technology of Tissue Culture and Rapid Propagation of Dendrobium candidum

Abstract:[Objective] The aim was to study the technology of tissue culture and rapid propagation of Dendrobium candidum. [Method]The stems of D. candidum were used as explants to study the effects of different exogenous hormones and its proportion on the buds induction and rooting of D. candidum. [Result]The best medium for bud proliferation was MS + 5 mg / L 6-BA + 0. 4 mg / L NAA,in which the growth of explants was robust,and the number of bud differentiation and induction rate were higher; the best rooting medium was MS + 0. 5 mg / L IBA + 0. 5 mg / L NAA. [Conclusion] The study laid the foundation for the industrialized production of seedlings of D. candidum.

Abstract:Dendrobium species(Dendrobium SW.) with high ornamental value and medicinal value,the growth in natural conditions,seeds germination of Dendrobium,vegetative growth,reproductive growth are necessary symbiosis with mycorrhizal fungi.Beneficial to the formation of mycorrhizal fungi and Dendrobium plants with the promotion of seedling growth,increase plant resistance and other functions,is to speed up plant growth rate Dendrobium species and key to ensuring their survival.The microstructure,characteristics and function of mycorrhiza of Dendrobium were reviewed in this paper.The identification and classification of mycorrhiza fungi in Dendrobium and their reciprocity were summarized.The prospect and the field of mycorrhiza research of Dendrobium in the future were put forward.

Abstract:The new in situ seed baiting method using seed packets to assess germination of orchid species within soil provides a means of locating, collecting, and identifying specific fungi that are involved in the lifecycle of orchids in the wild and investigating their relationship with the orchids under natural field conditions. Two isolates (SHH44, SHH53) originating from the seedlings found in the seeds packets in situ were demonstrated to support seed germination and seedling development (in vitro) of two endangered Chinese endemic herbs, Dendrobium officinale and Dendrobium nobile (Orchidaceae). Advanced protocorm development (Stage 3 and greater) in this study of these species of Dendrobium only occurred under the 12/12 h L/D photo-period indicated that illumination may play an important role in seedling recruitment of terrestrial orchid species in their natural environment. The information provided in this study may prove invaluable in the conservation of D. officinale and D. nobile and congener species in China, especially given their rare status.

Abstract:Dendrobium cariniferum is a valuable ornamental and medicinal plant rich with polysaccharides, alkaloid, and other bioactive compounds, which are potential raw materials for pharmacological utilization. In this study, an efficient protocol for the rapid propagation of D. cariniferum was developed. By using the tissue culture protocol, the effects of pH, hormone combinations, temperatures, light intensity, culture time protocorm proliferation, seedlings rooting, and accumulation of biomass with bioactive compounds were investigated. The experiments showed that the medium [1/2 MS + activated carbon1.0 g/L+ agar strip 7.5 g/L + sucrose 25 g/L] effectively promoted the germination of D. cariniferum seeds. The optimal culture conditions were found at pH 5.7, temperature 23 ± 2°C, and light intensity of 1000 Lx in the protocorm proliferation stage. Adding 1.5 g/L peptone in the medium effectively promoted the seedling rooting. The optimal culture conditions for accumulation of bioactive compounds (polysaccharides and alkaloids) of seedlings were found at temperature of 25 ± 2°C, light intensity of 1500-2000 Lx after the 60-day (d). Our study constructed a rapid propagation system in vitro for D. cariniferum, as well as the methods for efficient accumulation of active substances in seedling culture, which will serve as guidance for industrial production of D. cariniferum seedlings for both medicinal raw materials and ornamental plants. In addition, our study provided a new idea that we can directly use the high bioactive compound seedlings to extract medicinal components in industry conditions without transferring to the field.
Efficience Evaluation and Application Research of Dendrobium Candidum Wall with Bag-Type Tissue Culture System in Rooting Stage.

Abstract:The rooting culture of Dendrobium candidum wall was carried out with differentiation seedlings as test materials,by means of conventional bottles-type tissue culture system and bag-type tissue culture system.The results showed that tissue culture seedling produced from conventional bottles culture system and bag culture system showed no significant differences and they had some common characteristics: developed root system,100 percent rooting rate,strong root,good quality,over 90 % survival rate of transplants,and so on.However,some characteristics of bag-type tissue culture system were better than that of the conventional bottles-type tissue culture system,such as sterilizing of culture medium,the seeding efficiency,contamination rate,survival rate of transplants,the utility of cultivate space,the efficiency of seedling exercising,and so on.The results of this study have confirmed the fact that the bag-type tissue culture system could significantly improve the production efficiency as well as could achieve the effects of saving energy increasing efficiency.
[Field experiment of F1 generation and superior families selection of Dendrobium officinale]

Abstract:Based on randomized block design of experiment, agronomic traits and yields of 14 F1 generations of Dendrobium officinale were determined. The results showed that the differences in agronomic traits and yields among families were significant, and the hybrid vigor was obvious. Families of 6b x 2a, 9 x 66 and 78 x 69 were selected with the remarkable superiority of yields, agronomic traits and product customization. Correlation analysis between agronomic traits and yields showed that plant height, stem diameter, leaf number, blade length and blade width were all significantly correlated with biological yields and economic yields. Among which, stem diameter, leaf number and blade length were the most significant, and an optimal linear regression model could be established. When the number of shoots was fewer than 4.5, both biological yields and economic yields increased with the increasing number of shoots, but it could not much affect yields when the number of shoots was larger than 4.5. Shoots number, stem diameter and leaf index were basic stability when compared biennial traits to annual, which could be used for early selection.
