Abstract:RNA interference (RNAi) was discovered as a cellular defense mechanism more than decade ago. It has been exploited as a powerful tool for genetic manipulation. Characterized with specifically silencing target gene expression, it has great potential application for disease treatment. Currently, there are human clinical trials in progress or planned. Despite the excitement regarding this prominent technology, there are many obstacles and concerns that prevent RNAi from being widely used in the therapeutic field. Among them, the non-spatial and non-temporal control is the most difficult challenge, as well as off-target effects and triggering type I immune responses. Inducible RNAi technology can effectively regulate target genes by inducer-mediated small hairpin RNA expression. Combination with inducible regulation systems this makes RNAi technology more sophisticated and may provide a wider application field. This review discusses approaches of inducible RNAi systems, the potential problem areas and solutions and their therapeutic applications. Given the limitations discussed herein being resolved, we believe that inducible RNAi will be a major therapeutic modality within the next several years.

Abstract:RNA interference(RNAi) refers to the process where by double-stranded RNA(dsRNA) induces sequence-specific post-transcriptional gene silencing(PTGS).RNAi is an ancient cell response circuit,which has an important role in viral defense,transposon silencing,and endogenous gene regulation.Recently,short interfering RNAs(siRNAs) has been successfully used in the mammalian cell systems.Advances in the research and applications of gene-silencing technologies induced by siRNAs will be reviewed here.

Abstract:Variation of metabolic pathways result from RNA interference and related RNA silencing pathways have revolutionized our understanding of gene regulation. Gene silencing has been used as a research tool to control the expression of specific genes in numerous experimental organisms. Importing the different cells with specific siRNA need different transfection reagent to achieve maximum transfection efficiency, and will not produce larger cell toxicity. In recent years, new nano-material such as carbon nanotubes exert interference effect expanding us the traditional liposomes and virus’ understanding. The rapid progress in the field of RNA interference was summarized, and then compare different RNA interference experiments used several different carriers. Further, the perspectives of future research on RNA interference mediated from traditional liposomes to virus and polymer nano-material in disease treatment and clinic diagnose were proposed.

Abstract:RNA interference (RNAi) is the process of using specific sequences of double-stranded RNA (dsRNA) to knock down the expression level of sequence-homologous genes. Such ability of small interfering RNA (siRNA) in mammalian cells will undoubtedly revolutionize the study of functional genomics, the discovery of drug targets and even the treatment of human diseases. In this review we briefly describe the history of RNAi discovery, the RNAi mechanism and the general guideline for siRNA design as well as various methods for siRNA production and delivery. We also introduce the potential applications of siRNA, inducible siRNA and siRNA library in speeding up basic biomedical research and in acting as potential therapeutic agents for treatment of numerous human diseases.

Abstract:The ability to knockdown the expression of an endogenous gene by RNAi has emerged as a powerful strategy for the rapid identification of specific gene functions. Vector-based constitutive expression of shRNA can result in stable and efficient knockdown of target genes. However, constitutive expression of shRNA imposes major limitations when analyzing the function of genes whose expression is vital for the survival of an organism. Inducible RNAi systems can circumvent this limitation by enabling the inhibition of expression of an essential gene only when the inducing agent is present, and the level of knockdown of the essential gene can be controlled and adjusted by the concentration of inducing agent. In this review, we briefly summarize the recent development of various inducible RNAi systems and their potential applications in drug target validation.

Abstract:RNAi is a phenomenon of gene silence induced by double-stranded RNA(dsRNA) that is homologous to target gene. Now it is easy to obtain small interference RNA(siRNA), and siRNA is (often)transmitted into organisms by microinjection. RNAi includes encoding region RNAi and promoter region RNAi, which can be used to investigate gene function by degrading a special mRNA target in a cell or organism and thus knocking out or knocking down the level of the encoded protein. They have been used to the analysis of silkworm gene function. With the development of the technique, RNAi can be widely used to genomic analysis, medication target screening, disease treatment and so on.

Abstract:During recent decades there have been remarkable advances in biology, in which one of the most important discoveries is RNA interference (RNAi). RNAi is a specific post-transcriptional regulatory pathway that can result in silencing gene functions. Efforts have been done to translate this new discovery into clinical applications for disease treatment. However, technical difficulties restrict the development of RNAi, including stability, off-target effects, immunostimulation and delivery problems. Researchers have attempted to surmount these barriers and improve the bioavailability and safety of RNAi-based therapeutics by optimizing the chemistry and structure of these molecules. This paper aimed to describe the principles of RNA interference, review the therapeutic potential in various diseases and discuss the new strategies for in vivo delivery of RNAi to overcome the challenges.

Abstract:During the five years since its discovery, great progress has been made in the research of RNA interference (RNAi). especially in techniques of the production and introduction of siRNA, which has made possible the stable expression of the introduced siRNA. The discovery of the germ-line transmission of RNAi has further expanded its potential of application. Besides silencing the target genes, however, the introduction of siRNA can also result in sequence-specificity of siRNA, instead of target gene-specific silencing effect, i. e. , off-target effect, suggesting that care should be exercised in the interpretation of the silencing effect of specific siRNA, especially in the treatment of diseases.

Abstract:Small interfering RNA (siRNA) is emerging as a powerful tool for in vivo research and for use as potential therapeutic agents. This review demonstrates the examples of the applications of siRNA as a new modality in gene therapy which can elicit down-regulation of gene expression and has prodigious potential in the treatment of diseases in mammals. Before siRNA technique is used in vivo, efficient strategies of delivery must be designed, such as vectors and routes of delivery. The problems that should be considered before applying this technology are outlined.

Abstract:RNA interference (RNAi), an effective technique for regulating or silencing specific genes, can be applied to treat various diseases. Multiple clinical trials using RNAi are ongoing, and molecular imaging can serve as a powerful tool in RNAi-based therapies. This brief review will highlight the current progress on in vivo imaging of RNAi delivery and silencing effects. Incorporation of suitable molecular imaging techniques into future RNAi-based clinical trials will provide more pieces of the puzzle, thus facilitating the transformation of RNAi into a powerful therapeutic modality in the clinic.

Abstract:RNA interference (RNAi) is a new genetic interference technology, which specially degradates the corresponding sequence mRNA by the mediate of double-stranded RNA (dsRNA) to induce the post-transcriptional gene silencing (PTGS ). Since the first report of RNAi in 1998, there has been a great deal of significant progress. Up to now, RNAi phenomena have been discovered one after another in drosophila, edlworm, trypanosomatid and some mammalians, indicating the evolution conservatism. From the studies about RNAi in the past eight years, it is concluded that RNAi will be the powerful technology for human genome analysis and play an important role in gene expression imaging and gene therapy of human diseases.

Abstract:RNAi is a kind of peculiar gene silencing phenomenon induced by double strand RNA(dsRNA) homologous with the sequence of target gene.It is a conservative evolution mechanism of living things resisting the infections of outside genes and viruses in the process of evolution,and is a monitoring mechanism against invasion of viruses,transposition activity,and regulating gene expression.Therefore,it is of biological significance.The finding of RNAi technology and it successful application in gene knockout,mode organism,mammalian cells,in animal bodies has proved its important role in pharmaceutical researches.More and more studies show that RNAi has been demonstrated extremely large potentials in the aspect of drug researches.At present,there have been some siRNAs medicines in clinical experimental stages.This paper discusses its application in pharmacy in two aspects: one is identifying drug target,and the other is its application as medicines to treating diseases.
