Abstract:The lamellarins, a group of natural pyrrole alkaloids with interesting anticancer activities, were firstly isolated from a prosobranch mollus Lamellaria sp.. Due to their multidrug resistance (MDR) reversal in some cancer cell lines, total synthesis and structure modifications of lamellarins to provide drug candidates for the treatment of cancer have become one of the most promising project in the area of novel anticancer drug discovery. In this article, the latest results and progresses regarding the synthesis and modification of lamellarins are reviewed.
Synthesis and Antitumor Activity Study of Nitrogen-Containing Curcumin Derivatives

Abstract:Research on new antitumor drugs is of great significance since most of the currently available drugs had the disadvantages of high toxicity,low effection and multidrug resistance against subset of solid tumors.Structure modification of natural products may still deliver new hope for antitumor drug discovery.Curcumin and flavonoids are two kinds of important natural products with antitumor activity for recent years.Based on SAR of curcuminoids and our previous work,15 new novel curcumin analogues were designed and synthesized according to pharmacophore and scaffold hopping principle.Structurally,the new analogues are 1,5-diarylpentadienones which possess one nitrogen-containing substitution at the position 5 on one of the aromatic ring.The target compounds 6a-6n and 6o were obtained using 4-hydroxy-3-methoxybenzaldehyde and 2,4-dihydroxybenzaldehyde as starting materials,respectively.After a series of reactions,and the new compounds were finally obtained via Mannich reaction.The structure of new compounds were confirmed by MS(ESI) and 1H-NMR.All of the new compounds were screened for antiproliferation activity against two human tumor cell lines in vitro.Most of the new analogues showed more potent antiproliferation activitities against both tested cells,than that of curcumin,with IC50 values below 10 μmol · L-1.The substituted groups introduced in the aromatic ring didn't make significant effect on their activity.The IC50 value of compound 6c and 6b was lower than that of curcumin and chalcone.The results indicated that compounds with alkaline groups,especially morpholine group,resulted in stronger potent activities.Compounds with cyclohexone(6m,6n) showed decreased activities than that with straight carbon chain(6a).Also,compounds with the second alkaline side chain(6h,6k) did not enhance the activities.The compound(6l) introduced a methy group at 4-position obviously resulted in the decreased antitumor activity.In summary,new compounds with five-carbon chain exhibited more potent activities than those with seven-carbon or three-carbon chain.However,substituted groups were introduced at the 4-positon and 2-position resulted in the decreased bioactivities of compounds.And compounds with a chlorine at the 2-position or trifluoromethyl at the 4-position of ring B enhanced antitumor activities.And the'drug-like' properties predicted by Discovery Studio 2.0 suggested that most of them may be well absorbed by intestine and have good bioavailability.

Abstract:Novel heteroatom-incorporated antofine and cryptopleurine analogues were designed, synthesized, and tested against a panel of five cancer cell lines. Two new S-13-oxo analogues (11 and 16) exhibited potent cell growth inhibition in vitro (GI(50): 9 nM and 20 nM). Interestingly, both compounds displayed improved selectivity among different cancer cell lines, in contrast to the natural products antofine and cryptopleurine. Mechanism of action (MOA) studies suggested that R-antofine promotes dysregulation of DNA replication during early S phase, while no similar effects were observed for 11 and 15 on corresponding replication initiation complexes. Compound 11 also showed greatly reduced cytotoxicity against normal cells and moderate antitumor activity against HT-29 human colorectal adenocarcinoma xenograft in mice without overt toxicity.

Abstract:In this work, a green, efficient and catalyst-free synthesis of a series of structurally novel (E)-diethyl 2-styrylquinoline-3,4-dicarboxylates via a direct olefination reaction between diethyl 2-methylquinoline-3,4-dicarboxylate and various aromatic aldehydes was successfully accomplished by employing eco-friendly 1,3-dimethylurea/l-(+)-tartaric acid (DMU/LTA) as an inexpensive, non-toxic and reusable reaction medium. This methodology has the attractive advantages of mild reaction conditions, simple experimental operation, and the absence of any dangerous catalysts or unsafe volatile organic solvents, with satisfactory to good yields. Subsequently, a primary in vitro evaluation for their anti-proliferative activity against human cancer cell lines A549, HT29 and T24 revealed that the compound with the 3,4,5-trimethoxystyryl moiety exhibited potent anti-tumor activity with IC50 values of 2.38, 4.52 and 9.86 μmol L-1, respectively, thereby being equipotent or even better than the reference cisplatin.

Abstract:A series of novel N-1-(5-substituted-1,3,4-thiadiazole-2-yl)-N-3-substituted phenyl-urea derivatives have been synthesized and evaluated for their anti-tumor activity. The effect of the title compounds on tumor metastasis was analyzed by Lewis-lung-carcinoma model. The bioassay showed that some title compounds dramatically reduced the numbers of lung metastasis and depressed the primary tumor growth. The structures of compounds were identified from the elemental analysis, IR and H-1 NMR.

Abstract:Quinoxaline and its derivatives exhibit a broad spectrum of biological activity, making them valuable for various therapeutic applications. However, most quinoxalines are synthetically produced due to their scarcity in nature. In this article, a series of unsymmetric benzils were synthesized and subsequently condensed with 1,2‐diaminobenzene to produce unsymmetric quinoxalines. The novel synthetic benzils and quinoxalines were evaluated for their anticancer activities against human non‐small‐cell lung cancer (NSCLC) cells harboring different gene mutations, to explore their potential as anticancer agents. Among these synthesized molecules, compound 5g demonstrated inhibitory effects comparable to those of cisplatin.

Abstract:Selenium-containing small molecules have attracted considerable attention of chemical and medicinal researchers owing to their various biological activities, such as antitumor effects, cardiovascular protection, antibacterial or antiviral effects, immunoregulation and nerve protection, among which the most promising area is antineoplasm. In the past several decades, different kinds of organoselenium compounds, such as selenides, seleno(iso)cyanates, substituted selenoureas, selenious esters and Se-containing heterocycles have been reported as candidates of anti-cancer agents. Current reviews of Se-containing anticancer compounds mainly concerned about the investigation of their bioactivities, whereas, few attention has been addressed on their synthetic approaches. Herein, we summarized methodologies recently developed to synthesize organoselenium compounds with potent antineoplastic properties, which would be helpful for further design and synthesis of new bioactive Se-containing molecules with diverse structural features.
