Abstract:Quantifying stochastic processes is essential to understand many natural phenomena, particularly in biology, including the cell-fate decision in developmental processes as well as the genesis and progression of cancers. While various attempts have been made to construct potential landscape in high dimensional systems and to estimate transition rates, they are practically limited to the cases where either noise is small or detailed balance condition holds. A general and practical approach to investigate real-world nonequilibrium systems, which are typically high-dimensional and subject to large multiplicative noise and the breakdown of detailed balance, remains elusive. Here, we formulate a computational framework that can directly compute the relative probabilities between locally stable states of such systems based on a least action method, without the necessity of simulating the steady-state distribution. The method can be applied to systems with arbitrary noise intensities through A-type stochastic integration, which preserves the dynamical structure of the deterministic counterpart dynamics. We demonstrate our approach in a numerically accurate manner through solvable examples. We further apply the method to investigate the role of noise on tumor heterogeneity in a 38-dimensional network model for prostate cancer, and provide a new strategy on controlling cell populations by manipulating noise strength.

Abstract:Stochastic dynamics studies various stochastic dynamical processes and phenomena in nature,engineering and society using probability and statistics.After the developments in the last century,nonlinear stochastic dynamics has plenty of theories and methods,among which the theory and method of diffusion processes are dominant.The present paper outlines this theory and method,especially the Hamiltonian theoretical framework of nonlinear stochastic dynamics and control proposed and developed by the present author.Finally,the collaboration of researchers in various fields of stochastic dynamics and control is suggested.

While various attempts have been made to construct potential landscape in high dimensional systems and to estimate rare transitions, they are practically limited to cases where either noise is small or detailed balance condition holds. A general and practical approach to investigate nonequilibrium systems typically subject to finite or large multiplicative noise and breakdown of detailed balance remains elusive. Here, we formulate a computational framework to address this important problem. The current approach is based on a least action principle to efficiently calculate potential landscapes of systems under arbitrary noise strength and without detailed balance. With the deterministic stability structure preserving A-type stochastic integration, the potential barrier between different (local) stable stables is directly computable. The prevailing methods such as those based on WKB or Freidlin-Wentzell quasi-potential theory can be classified into the case of the present framework under small noise limit. We demonstrate our approach in a numerically accurate manner through explicitly solvable examples. We further apply the method to investigate the role of noise on tumor heterogeneity in a 38 dimensional network model for prostate cancer, and provide a new strategy on controlling cell populations by manipulating noise strength.

Abstract:This paper is devoted to proving the small noise asymptotic behaviour, particularly large deviation principle, for multi-scale stochastic dynamical systems with fully local monotone coefficients driven by multiplicative noise. The main techniques are based on a combination of the weak convergence approach, the time discretization technique and the theory of pseudo-monotone operator. The main results derived in this paper have broad applicability to various multi-scale models, where the slow component could be such as stochastic porous medium equations, stochastic Cahn-Hilliard equations and stochastic 2D Liquid crystal equations.

Abstract:In many instances, the dynamical richness and complexity observed in natural phenomena can be related to stochastic drives influencing their temporal evolution. For example, random noise allied to spatial asymmetries may induce stabilization of otherwise diverging trajectories in dynamical systems. However, to identify how exactly this takes place in actual processes usually is not a simple task. Here we unveil a few trends leading to dynamical stabilization and diversity of behavior by introducing Gaussian white noise to a class of exactly solvable non-linear deterministic models displaying space-dependent drifts. For the resulting nonlinear Langevin equations, the associated Fokker-Planck equations can be solved through the similarity method or the Fourier transform technique. By comparing the cases with and without noise, we discuss the changes in the systems dynamical characteristics. Simple examples of drift and diffusion coefficients are explicitly analyzed and comparisons with some other models in the literature are made. Our study illustrates the rich phenomenology originated from spatially heterogeneous dynamical systems under the influence of white noise.
