Abstract:A triphenylphosphine (PPh3)-catalyzed, regioselective and stereoselective hydrohalogenation reaction of 2-(1-alkynyl)-2-alken-1-ones with metal halides in the presence of acetic acid is described, furnishing functionalized 3,4-disubstituted 2-halo-1,3-dienes in moderate to good yields with high stereoselectivity and regiospecificity. The simple execution, mild conditions and the potential utilization of the products make this protocol attractive.

Abstract:The hydrohalogenation reaction of 1,2-allenic carboxylic acids, esters, amides, nitriles, and diphenyl phosphine oxides with HX or MX (X = I, Br, Cl, M = Na, Li) in different solvents, such as HOAc, CF3CO2N, and CF3CO2H-HOAc (1:1) was studied. The reaction is a nucleophilic conjugate addition in nature and provides efficient entries to 3-halo-3-enoic acids, esters, amides, nitriles, and 2-haloallyl diphenyl phosphine oxides. The beta,gamma- over alpha,beta -selectivity and stereoselectivity depend on the solvent(s) and the temperature applied. In most cases, the Z- and E-isomers can be easily separated by chromatography on silica gel. The subsequent cross-coupling reaction with organometallic reagents and terminal alkynes showed the synthetic potential of these products.

Abstract:Summary of main observation and conclusionAn enantioselective desymmetric nucleophilic α‐addition of cyclohexanone to propiolamide has been developed through a 6‐exo‐dig cyclization reaction. By employing simple and readily available L‐proline sodium salt as a bifunctional catalyst, a series of chiral 6,6‐bicyclic bridged products bearing morphan scaffold have been isolated in good yields and excellent enantioselectivities. Density functional theory (DFT) calculations elucidated the origins of the enantioselectivity and regioselectivity of this transformation. A salt bridge that links the amide carbonyl group with proline carboxylate in the transition state was proven to be the driving force for the induction of excellent enantioselectivity.

Abstract:A straightforward approach to synthesizing substituted 1,3-alkadien-2-yl carboxylic acids starting from readily available propargylic alcohols was developed. Based on mechanistic studies, the reaction was found to proceed via regioselective hydrocarbonylation of the C-C triple bonds of the in situ formed 1,3-enyne intermediates, providing 1,3-alkadien-2-yl carboxylic acids with a very high selectivity.

Abstract:The hydrohalogenation reactions of electron-deficient allenes, such as 1,2-allenic ketones, carboxylic acids, esters, amides, nitriles, sulfoxides (with the use of AlX3/H2O), sulfones, and phosphine oxides with MX (X = I, Br, Cl, M = Na, Li) in different solvents, such as HOAc, CF3COOH-HOAc (1 : 1), CF3COOH and THF were studied. Abstract:The first metal/organo cooperatively catalyzed asymmetric reaction of C-alkynyl N-Boc-protected N,O-acetals with in situ generated oxonium ylides has been developed. This new type of propargylation allows for the efficient synthesis of structurally diverse unreported chiral propargylamines bearing oxa-quaternary stereocenters. The reaction features unprecedented substrate scope and high diastereo- and enantioselectivity. Theoretical studies suggest a novel cooperative catalysis model and the unique transfer of R2OH.

Abstract:We herein describe a palladium-catalyzed hydrocyanation of propiolamides for the stereodivergent synthesis of trisubstituted acrylonitriles. This synthetic method tolerated various primary, secondary and tertiary propiolamides. The cautious selection of a suitable ligand is essential to the success of this stereodivergent process. Control experiments indicate the intermediacy of E-acrylonitriles, which lead to Z-acrylonitriles via isomerization. The density functional theory calculations suggests that the bidentate ligand L2 enables a feasible cyclometallation/isomerization pathway for the E to Z isomerization, while the monodentate ligand L1 inhibits the isomerization, leading to divergent stereoselectivity. The usefulness of this method can be demonstrated by the readily derivatization of products to give various E- and Z-trisubstituted alkenes. In addition, the E- and Z-acrylonitrile products have also been successfully employed in cycloaddition reactions.

Abstract:Asymmetric hydroxycarbonylation is one of the most fundamental yet challenging methods for the synthesis of carboxylic acids. Herein, we reported the development of a palladium-catalyzed highly enantioselective Markovnikov hydroxycarbonylation of vinyl arenes with CO and water. A monodentate phosphoramidite ligand L6 plays vital role in the reaction. The reaction tolerates a range of functional groups, and provides a facile and atom-economical approach to an array of 2-arylpropanoic acids including several commonly used non-steroidal anti-inflammatory drugs. The catalytic system has also enabled an asymmetric Markovnikov hydroalkoxycarbonylation of vinyl arenes with alcohols to afford 2-arylpropanates. Mechanistic investigations suggested that the hydropalladation is irreversible and is the regio- and enantiodetermining step, while hydrolysis/alcoholysis is probably the rate-limiting step.

Abstract:The most prevalent strategies for preparing halogenated alkyl aromatics face one remarkable challenge: low regioselectivity. Therefore, the highly regioselective halogenation of alkyl arenes is still an unsolved problem. Herein, a highly regioselective halogenation of alkyl aromatics through nitromethane-assisted Bronsted acid catalysis is disclosed. Under the mild reaction conditions, a wide diversity of halogenated alkyl aromatics is obtained in high yields with excellent regioselectivity. The hydrogen bonding of protonic acid with the solvent nitromethane promotes this transformation.
Tuning of Regioselectivity in the Coupling Reaction Involving Allenic/propargylic Palladium Species.

Abstract:Two different types of coupling patterns for the Pd(0)-catalyzed coupling reaction of allenic/propargylic zinc reagents with organic halides or propargylic carbonates (acetate) with the corresponding organometallic reagents were observed. After studying the controlling factors on the regioselectivity of this reaction, we demonstrated that the steric hindrance of both reactants and the types of organic halides determine the regioselectivity of this coupling reaction. By subtle choosing of the substrates, the regioselectivity can be tuned. On the basis of these results, new methodologies for the highly regio- and stereoselective synthesis of 6-substituted hex-5-yn-2-enoates and 4,6-dialkylhexa-2,4,5-trienoates have been developed. Some of the products synthesized by the carbonate protocol cannot be prepared by the lithiation protocol because the regioselectivity of lithiation of dialkyl-substituted internal alkynes is an intrinsic problem.

Abstract:Allylic alcohols were found to undergo a semipinacol-type rearrangement induced by halogen cation generated from the chloramine-T/ZnX2 combination, which provided a highly efficient and stereoselective method for the preparation of alpha-quaternary beta-haloketo compounds. This reaction is valuable and versatile since a quaternary carbon could be constructed effectively and three kinds of beta-haloketo compounds (X = Cl, Br, I) could be achieved readily.

Abstract:A stereoselective hydroxyallylation reaction of cyclopropenes with cyclopropanols is achieved under zinc-mediated conditions, affording densely functionalized cyclopropanes with excellent diastereocontrol over three contiguous stereocenters within and outside the cyclopropane ring. A racemic variant of the reaction is synergistically promoted by catalytic N-heterocyclic carbene (NHC) and organic base (DBU), whereas chiral amino alcohol-derived bifunctional NHC enables a catalytic enantioselective variant. Mechanistically, the reaction features transient generation of enolized zinc homoenolate via ring-opening of zinc cyclopropoxide and enolization of the resulting homoenolate, followed by its addition to the cyclopropene as a prochiral allylzinc nucleophile.

Abstract:The hydrohalogenation reaction of 1‐alkyl substituted 1,2‐allenyl carboxylic acid esters (1) with MX (M= Na, or Li, X= Cl, Br, I) afforded a mixture of β,γ‐unsaturated 3‐halo–3‐alkenoates (2) and Q,P unsaturated 3‐halo‐2‐alkenoates (3) in HOAc, while using a mixture of HOAC‐CF 3 CO 2 H (1:1) or CF 3 C 2 H as the reaction medium the corresponding reaction cleanly produced β,γ ‐unsaturated 3‐hale3‐alkenoates (2) as the sole products in high yields. The subsequent coupling reactions were studied.
