Abstract:In this work, a photoelectrochemical (PEC) glucose biosensor was synthesized on the basis of a type of Au@C/TiO2 composite by using an unsophisticated secondary hydrothermal strategy. The compounded Au@C/TiO2 material was characterized by XRD, SEM, TEM, UV-vis, and XPS to identify the composition and the purity of the phase. Glucose oxidase (GODx) was immobilized on the Au@C/TiO2. The PEC biosensor presented satisfactory stability and outstanding reproducibility. The range and the sensitivity of the linear measurement were 0.1-1.6 mM and 29.76 μA mM-1 cm-2, respectively, and the detection limit was low (0.049 mM). The GODx/Au@C/TiO2/FTO biosensor presented excellent efficiency in detecting glucose, suggesting the great potential application of this synthesized material in PEC biosensors.

Abstract:Photoelectrochemical detection represents a unique signal transducing modality, where the photogenerated charge carriers are modulated by the redox reactions of molecular targets over the electrode surface. In this work, we report a novel glucose sensor based on hydrothermally-grown, single-crystalline TiO2 nanowires with surface-functionalized glucose oxidase (GOx), which can oxidize glucose to gluconic acid. The key feature of this glucose sensor design is that the photogenerated holes over the TiO2 anode are utilized to form O-2, which directly serves as an efficient electron shuttling mediator between the enzymatic redox center of GOx and the sensor surface, thus leading to an increase of the photocurrent. As a proof-of-concept, the GOx-functionalized TiO2 (TiO2-GOx) nanowires exhibit a high sensitivity of similar to 0.9 nM in the detection of glucose in buffer. The capability of detecting glucose in mouse serum is also demonstrated. Our work suggests the potential of developing these nanowire-based photoelectrochemical biosensors for convenient, efficient and low-cost biomarker detection and disease diagnosis.
Optical and Photoelectrochemical Sensing of Glucose Based on Au Nanohole Arrays Coupled with TiO2/Al Films

Abstract:The mainstream technology used in commercial glucose sensors is enzymatic electrochemical sensing. Although the use of enzymatic electrodes can specifically detect glucose, this sensor has many deficiencies such as large environmental impacts, complex enzyme-modification processes and relatively high cost. Constructing well-designed metal nanostructures can excite environment-dependent surface plasmon resonance (SPR) and generate hot electrons. By monitoring the central wavelength shift of SPR and collecting the photocurrent generated from the hot electrons, label-free, enzyme-free, optical and photoelectrochemical (PEC) dual-mode sensing of glucose can be realized. Here, we combine magnetron sputtering, thermal treatment, nanosphere lithography, electron beam evaporation and lift-off processes to fabricate hexagonally-arranged gold nanohole arrays (Au NHAs) with adjustable diameters on aluminum (Al)/titanium dioxide (TiO2) film substrates, and constructed an enzyme-free glucose sensor based on the Al/TiO2/Au NHAs structure. The sensor is demonstrated to work in both optical sensing and PEC sensing modes. For optical sensing mode, the shift of the SPR peak corresponding to the glucose concentration range of 0–10 mM is observed, with an optical sensing sensitivity of 857 nm/RIU. For PEC sensing mode, the sensor at zero bias shows a linear range of 1 µM– 10 mM for glucose detection under simulated one-sun illumination, along with the detection of limit down to 1 µM and the photocurrent sensing sensitivity of 0.53×logC µA μM-1 cm-2 (where C is the molar concentration of glucose). Furthermore, the as-prepared glucose sensor also shows good stability and specific selection for glucose detection.

Abstract:Controlling the charge transfer and thus enhancing the excitons’ lifetime are the key to the realization of efficient photoelectrochemical (PEC) devices. Moreover, fabrication of flexible and collapsible sensors can greatly facilitate the implementation of smart PEC sensing devices into practical applications. Herein, we sagely designed and successfully fabricated three-dimensional flexible Au nanoparticles-decorated TiO2 nanotube arrays (Au@TiO2) for the efficient PEC biosensing of glucose. The Schottky barrier derived from the Au@TiO2 heterostructure efficiently separates the charge carriers at the junction interfaces, thus greatly increasing the concentration and lifetime of holes left in the valence band of TiO2. The separated holes further evidently generate the active hydroxyl radicals, which can specifically recognize and oxidize glucose. As a result, Au@TiO2 exhibits excellent photoelectric activity and selectivity, far superior to TiO2 without decorated Au nanoparticles. In addition, such asymmetric Au@TiO2 system has been proved to feature the intrinsic flexibility nature, since its PEC biosensing performance is almost unaffected under indirect light irradiation and serious tensile strain.
Study on Enzyme-Free Glucose Sensor Based on Au and CuO Co-Modified TiO2 Films for Photoelectrochemical Detection

Abstract:Currently, widely used glucose sensors rely on enzyme catalysts, which suffer from issues such as enzyme inactivation, high production costs, and insufficient stability. To address these challenges, this study fabricated TiO2 films on Ti sheets using radio frequency magnetron sputtering and subsequently modified Au nanoparticles (NPs) and CuO films via direct current magnetron sputtering. Then, Au and CuO co-modified TiO2 (CuO-Au@TiO2) photoelectrode with a Z-type band structure was prepared, enabling enzyme-free glucose detection via photoelectrochemical (PEC) sensing technology. The optimized photoelectrode exhibited high sensitivity for glucose detection, with values of 7300 μA μM-1 cm-2 (0–1 mM) and 500 μA μM-1 cm-2 (1–10 mM) under AM 1.5G illumination at a bias of 0.2 V (vs. Ag/AgCl). The detection limit was determined to be 58.14 μM (Signal/Noise = 3), with excellent stability, repeatability, and selectivity. Spectral and PEC performance analyses revealed that the outstanding sensing performance of the CuO-Au@TiO2 photoelectrode can be attributed to: (1) unique Z-Type band structure promoting carrier separation; (2) the local surface plasmon resonance (LSPR) effect induced by Au NPs enhancing light absorption; (3) CuO can undergo a specific enzyme-like catalytic reaction with glucose molecules under alkaline conditions. This research offers a novel, convenient, and accurate monitoring approach for diabetes management and prevention.

Abstract:TiO2 single-crystal nanomaterials with highly reactive surfaces have attracted widespread attention due to their fundamental aspects and industrial applications. However, many previous studies have ignored the poor photoelectric efficiency of TiO2 nanomaterials originating from a mismatch in the diffusion distance of electrons and holes as well as migration rates of holes and electrons. Therefore, suppression of hole migration could enhance the photoelectrochemical performance of TiO2. In this work, single-crystal anatase TiO2 nanomaterials with (001) facets were successfully prepared. The results suggested that some (101) facets were retained by the modified fluoride ion crystal facet control process. Subsequently, fluorine-free and Au-containing crystal facets were obtained by annealing and electrochemical deposition. In addition, the propensity for adsorption of Au on a TiO2 (101) surface was verified by first-principles quantum chemical calculations. Electrochemical impedance spectroscopy and UV–visible spectrophotometry showed the presence of a large area (001), conducive to better enzyme affinity. Biosensors prepared by carrier self-separation derived from natural different facets and Au nanoparticles (TiO2 (001)/Au/GOx) achieved high sensitivities reaching 16.86 μA mM–1 cm–2, an extended linear range (0.01–3 mM), and a low detection limit (0.83 μM). In summary, the proposed route allowed for the first time the use of nanocrystal engineering in the construction of glucose biosensors with satisfactory performances, which is promising for the future fabrication of high-performance biosensors.

Abstract:Developing stable PEC glucose biosensors with high sensitivity and low detection limit is highly desirable in the biosensor field. Herein, a highly stable and sensitive enzymatic glucose photoelectrochemical biosensor is rationally designed and fabricated by constructing TiO2/PDA core/shell nanorod arrays. The TiO2 nanorod as the core has the advantages of increasing charge transportation towards interfaces and enhancing the absorption of incident sunlight due to its single-crystal nature and one dimensional array structure. The PDA shell not only induces a rapid charge transfer across the interfaces but also stabilizes the biosensor performance by avoiding the decomposition of enzymes induced by the strong oxidizing holes from the TiO2 core. A remarkable performance with an ultrahigh sensitivity of 57.72 μA mM-1 cm-2, a linear range of 0.2-1.0 mM, a glucose detection limit of 0.0285 mM (S/N = 3) and a high sensitivity of 8.75 μA mM-1 cm-2 in a dynamic range of 1.0-6.0 mM were obtained for the glucose detection. This study might provide a strategy for constructing inorganic/organic core/shell structures with a satisfactory PEC performance.

Abstract:Developing stable photoelectrochemistry (PEC) glucose biosensors with high sensitivity and a low detection limit is highly desirable in the biosensor field. Herein, a highly sensitive and stable enzymatic glucose PEC biosensor is rationally designed and fabricated using a TiO2NTs/Au/Pt/GOx electrode. First, we prepared one-dimensional TiO2 nanotube arrays which could realize the orthogonalization of the light-incident direction and the carrier diffusion direction via anodization. Subsequently, we used the method of photoassisted deposition for anchoring Pt nanoparticles on TiO2NTs after electrodepositing Au nanoparticles. Among them, Au nanoparticles promote light absorption via the surface plasmon resonance effect and the separation of photogenerated carriers through forming a Schottky junction. Moreover, the Pt nanoparticles on the electrode surface can react with hydrogen peroxide (H2O2) generated from glucose (Glu) oxidation by glucose oxidase (GOx), accelerating the electron-transfer process during glucose oxidation and greatly improving the sensitivity of the glucose biosensor. As a result, TiO2NTs/Au/Pt/GOx exhibited excellent PEC performance, achieving a high sensitivity of 81.93 μA mM-1 cm-2 and a low detection limit (1.39 μM), far exceeding the performance of TiO2NTs/M/GOx (M = Au, Pt). Therefore, the introduction of Pt nanoparticles as active substances to promote enzymatic reactions is important for designing high-performance enzyme biosensors.

Abstract:A new photoelectrochemical (PEC) sensing platform comprising TiO2 nanotube arrays (TiONTAs), polyaniline (PANI), and gold nanoparticles (AuNPs) was successfully fabricated. After loading the enzyme, this Au-PANI-TiONTA electrode showed excellent response to glucose at a linear range of 2-36 mM with a 0.02 mM detection limit. Good PEC performance was obtained due to the following advantages of the material: high visible-light harvesting ability for excellent light trapping capacity of PANI and AuNPs, good separation of the photo-induced charges related to the specific Au-PANI-TiONTA heterostructure, efficient electrode surface reaction kinetics derived from the large specific surface area of TiONTAs and improved electrode catalytic activity. This work proposed a new and general PEC enzymatic format and can be extended to prepare different PEC biosensors for biomolecules such as DNA, proteins and substrates of oxidases.
Electrochemical nonenzymatic glucose sensors catalyzed by Au nanoclusters on metallic nanotube arrays and polypyrrole nanowires

Abstract:Monitoring blood glucose level is critical, since its abnormality leads to diabetes and causes death, even though glucose is essential for human living. Herein, the sensing study was performed on electrochemical nonenzymatic glucose sensors, which are composed of an Au nanocluster (AuNC) catalyst deposited on a metallic nanotube array (MeNTA) and polypyrrole nanowire (PPyNW). The AuNC was produced by irradiating a femtosecond pulse laser to the Au precursor solution, and it is a simple and facile method. The successful deposition of AuNC on both MeNTA and PPyNW was confirmed by means of the surface morphology and the Au content increase. On the exploration by cyclic voltammetry in alkaline condition, AuNC/MeNTA electrodes showed better performance than AuNC/PPyNW electrodes: The former was a remarkable electrocatalytic detector towards glucose oxidation with better sensitivity, lower detection limit, wider linear range, and longer-term stability without interference from potential interfering agents such as ascorbic acid, urea, NaCl, KCl, etc. Moreover, nonenzymatic AuNC/MeNTA electrodes exhibited high precision and accuracy in real human blood samples and, thus, can be a promising candidate in glucose sensing applications.

Abstract:In this communication, we report on a novel green photocatalytic route to synthesize Au nanoparticles/multi-walled carbon nanotubes (AuNPs/MWCNTs) nanocomposites with the use of Sn-porphyrin (SnP) as a high-efficiency photocatalyst to reduce Au3+ to form AuNPs onto MWCNTs. Such AuNPs/MWCNTs nanocomposites exhibit good catalytic performance toward both oxidation and reduction of H2O2. An electrochemical glucose biosensor was further constructed by dropping glucose oxidase on the surface of AuNPs/MWCNTs nanocomposites modified glassy carbon electrode. The biosensor shows linear response toward different concentrations of glucose from 1 to 33 mM (r = 0.998) and the detection limit at 0.5 V is estimated to be 240 mu M at a signal-to-noise ratio of 3.

Abstract:Based on the enhanced charge separation efficiency of the one-dimensional structure and strong surface plasmon resonance (SPR) of gold, a gold modified TiO2 nanotube (Au/TiO2NTs) glucose photoelectrochemical (PEC) sensor was prepared. It could be activated by visible red light (625 nm). Under optimal conditions, the Au/TiO2NTs sensor exhibited a good sensitivity of 170.37 μA·mM−1·cm−2 in the range of 1–90 μM (R2 = 0.9993), and a detection limit of 1.3 μM (S/N = 3). Due to its high selectivity, good anti-interference ability, and long-term stability, the fabricated Au/TiO2NTs sensor provides practical detection of glucose. It is expected to be used in the construction of non-invasive PEC biosensors.

Abstract:Optical and electrochemical sensors have been widely employed to detect biochemical substances, but each one has its own strengths and weaknesses. In this study, an optical and photoelectrochemical optional-mode sensor based on the nanopatterned metal-semiconductor-metal (MSM) structure is proposed for glucose detection. The MSM structure is designed and prepared by sequentially stacking a TiO 2 film and the orderly Au nanodisks on an Al film, and the Au and TiO 2 contact forms Schottky junction. In the optical term, the nanopatterned Au-TiO 2 Schottky junction coupled with the Al film can produce enhanced surface plasmon resonance effect, which is used to construct a refractive-index sensor. In the electrical term, the Au-TiO 2 Schottky junction can collect photogenerated carriers via the oxidation of glucose, resulting in an observable photocurrent changing with glucose concentration. That is the prepared sensor can optionally work in optical and PEC modes. In the optical mode, the sensitivity for the design and experiment is respectively 968.8 nmRIU -1 and 364.9 nmRIU -1 . In the PEC mode, the sensitivity at relatively zero bias and under one-sun illumination is 1050 μAM -1 cm -2 within the glucose concentration range of 0–10 mM. This work provides an alternate route to the achievement of multi-mode sensor for the large-concentration-range and low-detection-limit detection.

Abstract:A simple procedure for constructing the photoelectrochemical biosensor based on TiO2/CdSe@CdS QD nanocomposite modified electrode was developed. The TiO2/CdSe@CdS QD nanocomposite modified electrode was obtained by alternately depositing water-soluble CdSe@CdS QD and a mixture of [cobalt(o-phen)3]2+/3+ and poly(ethyleneimine) (PEI) over mesoscopic TiO2 films. The obtained TiO2/CdSe@CdS QD nanocomposite modified electrode exhibits much higher and stable photocurrent intensity than that of the photoelectrochemical biosensors reported so far. After the TiO2/CdSe@CdS QD nanocomposite modified electrode was coated with glucose oxidase, the photocurrent intensity was enhanced towards the addition of glucose. Thus, a photoelectrochemical biosensor for the detection of glucose was developed by monitoring the changes in the photocurrent intensity. Moreover, the proposed biosensor possessed good reproducibility and storage stability. Combining TiO2/CdSe@CdS nanocomposite and other biomolecules could be extended readily for sensing other biorecognition events.
Enhanced Photoeletrochemical Biosensing Performance from Rutile Nanorod/anatase Nanowire Junction Array

Abstract:In this work, a novel enzymatic glucose photoelectrochemical biosensor was successfully fabricated based on rutile/anatase Ti-O2(R/A-TiO2) heterostructure which was composed of anatase nanowires and rutile nanorods. Compared to a rutile nanorod electrode, an obvious enhanced photocurrent was observed on R/A-TiO2 electrode in photoelectrochemical test. Excellent photoeletrochemical performance was obtained due to these follow points: higher optical absorptivity for the larger surface area, better separation of the photogenerated carriers related to the specific R/A-TiO2 heterostructure and faster electrical transmission stemmed from the unique conductive structure. This R/A-TiO2 heterostructure array was applied to glucose detection by loading glucose oxidase on R/A-TiO2 electrode. A remarkable performance with a 5.71 mu AmM(-1)cm(-2) sensitivity, 1-20mM linear range and a 0.019 mM glucose detection limit (S/N = 3) were obtained in the glucose detection.

Abstract:Traditional enzyme-based blood glucose sensors demand stringent environmental conditions, while the current nonenzymatic blood glucose sensors exhibit low sensitivity and a limited detection range, hindering the accurate and swift analysis of minute liquid samples. In this study, the TiO2 hierarchical nanotubes (HNTs) were modified with Au and CuO nanoparticles (NPs) in sequence, and the as-prepared CuO-Au@TiO2 HNTs were constructed into a sensing photoelectrode and integrated into a self-designed microfluidic photoelectrochemical (PEC) cell. Accurate monitoring of glucose concentrations in artificial blood, sweat, and saliva was achieved by merely using a microliter analyte, along with satisfactory selectivity and repeatability. Notably, the fit sensitivity in the concentration range of 1-10 mM was over 29-fold higher than the traditional cell (i.e., nonmicrofluidic configuration) with the same sensing photoelectrode under one-sun illumination, and a detection limit was as low as 4.1 mu M (Signal/Noise =3). The excellent sensing performances are ascribed to the synergistic effect of the substantial enhancement in optical absorption and carrier transfer and the dynamic flow of glucose confined in micro-channel. This study demonstrates an enzyme-free sensing photoelectrode integrated with a microfluidic cell for high-performance glucose detection, and presents an alternative avenue toward noninvasive and portable diabetes diagnosis.
In-situ construction of Au/Cu2O nanowire arrays for sensitive glucose sensing

Abstract:Architecture design is widely regarded as a rational strategy to enhance the sensing performance of electrocatalysts. Herein, the novel three-dimensional hybrids based on Au and Cu2O were successfully synthesized via steps of in-situ growth, including anodic oxidation, annealing and galvanic displacement. Cu2O appeared in the morphology of nanowire array on conductive substrate, and was decorated by Au nanoparticles. Benefiting from the unique architecture and binder-free fabrication process, the Au/Cu2O nanowire arrays possessed high conductivity and abundant exposed active sites, as well as facilitated the direct electron transfer among detection object, electrocatalyst and current collector. Moreover, Au/Cu2O particles as contrast were fabricated to clarify the effect of structure on sensing ability. The Au/Cu2O nanowire arrays drove the glucose electro-oxidation reaction with great catalytic activity, in which a potential as low as 0.4 V was needed to reach a high sensitivity of 2.098 mA mM-1 cm-2. The excellent selectivity, stability and reproducibility were also obtained by the sensor. Furthermore, the quantitative detection of glucose level in diluted human serum were performed and the satisfactory result make the obtained sensor have the potential for practical applications.
Towards Self-Driven and Enzyme-Free Sweat Glucose Photoelectrochemical Sensing Via Decorating Cuo Nanoparticles on Tio2 Hierarchical Nanotubes

Abstract:It is of great importance to develop a non-invasive and portable glucose sensor to address challenges and obstacles brought by invasive glucose sensors, such as negative physical and psychological impacts to patients during the fingertip blood collection, limited environment of use for enzyme-based electrodes, and cumbersome active-driven integrated devices. Photoelectrochemical (PEC) sensors have great potential in this regard. In this work, self-driven and enzyme-free sweat glucose PEC sensing is realized by constructing TiO2 hierarchical nanotubes modified by CuO nanoparticles (CuO@TiO2 HNT). A sensitivity of 138.9 mu A mM(-1) cm(-2) in the low concentration range (< 200 mu M) and a detection limit of 0.7 mu M (Signal/Noise=3) are achieved by the asprepared CuO@TiO2 HNT photoelectrode at relatively zero bias. The testing results for sweat glucose detection before and after meal consumption are consistent with those from commercial glucometers. The CuO@TiO2 HNT photoelectrode works well at absolutely zero bias and in a wide temperature range (e.g., 0-60 degrees C). The extraordinary sensing performance can be mainly attributed to the following two factors: (1) the TiO2 hierarchical nanotubes have an excellent light-capturing property and large specific surface area, and (2) the CuO nanoparticles substantially facilitate surface-carrier transfer for the glucose oxidation reaction. This work provides an enzyme-free sensing photoelectrode for self-driven sweat glucose detection and an alternative route to portable and noninvasive sensing of glucose.
