Abstract:The goal of this study was to investigate surface ion conductance variation in an anion-exchange membrane (AEM) using current-sensing scanning probe microscopy. No direct correlation was found between the membrane surface topography and local ion conductance. Smaller and larger conducting areas associated with ion channels and ionic clusters were identified in images of membrane surface ion conductance. Both the size of ion channels and the density of ionic clusters tended to increase significantly at higher relative humidity (RH) conditions. The ionic conductance of the AEM was one order of magnitude lower than its proton-exchange counterpart (Nafion®) at 100% relative humidity. This decrease may be due to the rate-limiting properties of the studied AEM, such as lower mobility of anions (HCO3− and OH−), smaller size of ionic channels, lower density of ionic clusters, and lower capability for water uptake as compared to Nafion® 212. Nevertheless, the ionic conductance was found to be higher on the AEM when the RH was lower than 45%. These results have direct implications for the application of the AEM in fuel cells without humidification systems.
Correlating Humidity-Dependent Ionically Conductive Surface Area with Transport Phenomena in Proton-Exchange Membranes.

Abstract:The objective of this effort was to correlate the local surface ionic conductance of a Nafion 212 proton-exchange membrane with its bulk and interfacial transport properties as a function of water content. Both macroscopic and microscopic proton conductivities were investigated at different relative humidity levels, using direct-current voltammetry and current-sensing atomic force microscopy (CSAFM). We were able to identify small ion-conducting domains that grew with humidity at the surface of the membrane. Numerical analysis of the surface ionic conductance images recorded at various relative humidity levels helped determine the fractional area of ion-conducting active sites. A simple square-root relationship between the fractional conducting area and observed interfacial mass-transport resistance was established. Furthermore, the relationship between the bulk ionic conductivity and surface ionic conductance pattern of the Nafion membrane was examined.

Abstract:Over the last 2 decades, scanning electrochemical microscopy (SECM) has been extensively employed for topographic imaging and mapping surface reactivity on the micrometer scale. We used flat, polished nanoelectrodes as SECM tips to carry out feedback mode imaging of various substrates with nanoscale resolution. Constant-height and constant-current images of plastic and Au compact disc surfaces and more complicated objects (computer chips and wafers) were obtained. The possibility of simultaneous imaging of surface topography and electrochemical reactivity was demonstrated. Very fast mass transfer at nanoelectrodes allowed us to obtain high-quality electrochemical images in viscous media under steady-state conditions, e.g., in 1-methyl-3-octylimidazolium-bis(tetrafluoromethylsulfonyl)imide (C(8)mimC(1)C(1)N) ionic liquid. Ion-transfer-based imaging was also performed using nanopipets as SECM tips.

Abstract:A new mode of the scanning electrochemical microscope (SECM) operation was developed that combines reagent delivery from the nanopipette with electron transfer at the conductive substrate and ion transfer across the liquid/liquid interface supported at the nanopipette tip. This approach offers potential advantages for measurements of heterogeneous electron transfer kinetics and reaction rate imaging by enabling straightforward separation of the contributions of surface topography and reactivity features to the tip current. It addresses some other long-standing technical issues, including sensitive probing of low signal sources (e. g., immobilized enzymes or catalyst particles) without diffusional broadening and the elimination of the elevated background signal in generation/collection-type experiments. The high spatial resolution attainable in electron transfer/ion transfer mode experiments and the absence of redox mediator species in the bulk solution are advantageous for studies of biological cells.

Abstract:The distribution of ion conductive channels on the Nafion membrane surface, which determines the formation of the three-phase boundary, plays a very important role in improving the performance of proton-exchange membrane fuel cells. Therefore, understanding the microstructures at the catalyst layer/membrane interfaces of proton-exchange membranes is essential. Although current-sensing atomic force microscopy (AFM) can present some surface conductance data, localized impedance measurement providing more accurate proton-transport information is desirable. To obtain this information, in our study, localized electrochemical impedance spectroscopy was measured automatically with a home-built AFM-electrochemical impedance spectroscopy setup in which AFM was coupled with an impedance tester by a customized procedure. By this method, the localized proton-transport resistance at different humidities was observed in spatially diverse locations, and the value decreased as the membrane became hydrated. Furthermore, the microstructure of the Nafion membrane was numerically reconstructed at different hydration levels to examine the relationship between the membrane microstructural morphology and proton-transport resistance. The results showed that the spatial diversity of proton-transport resistance arose from the variable concentration of hydrophilic groups at the contact location of the AFM tip and the membrane, and from the heterogeneity of dry sulfonic acid groups in the membrane that creates local variation in water content.

Abstract:The distribution of surface charge and potential of cell membrane plays an indispensable role in cellular activities. However, probing surface charge of live cells under physiological conditions, until recently, remains an arduous challenge owing to the lack of effective methods. Scanning ion conductance microscopy (SICM) is an emerging imaging technique for imaging a live cell membrane in its native state. Here, we introduce a simple SICM based imaging technique to effectively map the surface charge contrast distribution of soft substrates including cell membranes by utilizing the higher surface charge sensitivity of the ionic current when the nanopipette tip is close to the substrate with a relatively high current change. This technique was assessed on charged model substrates made of polydimethylsiloxane, and the surface charge sensitivity of ionic current change was supported by finite element method simulations. With this method, we can distinguish the surface charge difference between the cell membrane and the supporting collagen matrix. We also observed the surface charge change induced by the small membrane damage after 1% dimethyl sulfoxide (DMSO) treatment. This new SICM technique provides opportunities to study interfacial and cell membrane processes with high spatial resolution.

Abstract:Scanning ion conductance microscopy (SICM) is one type of scanning probe microscopy (SPM) techniques. By measuring the ionic current of an ultra-micropipette, it can be used to map the surface topography of a sample at high resolution without any contact. SICM has many remarkable advantages, such as high spatial resolution, simple preparation of the probe and no damage to the sample surface, particularly suitable for imaging living cells under physiological condition. Therefore, SICM is an important scanning probe microscopy technique complementary to scanning electrochemical microscopy and atomic force microscopy. SICM has been employed to image soft interfaces and surfaces, such as cell surface and its microstructure. In addition, SICM can be used in conjunction with other techniques to study the relationship between cell topography and function; it can also control the deposition of specific molecules to achieve nanometer-scale microscopic operation and fabrication. Herein, the history, principle, instrumentation, applications and prospects of SICM have been reviewed.

Abstract:Scanning ion conductance microscopy (SICM) is a scanning probe technique that utilizes the increase in access resistance that occurs if an electrolyte filled glass micro-pipette is approached towards a poorly conducting surface. Since an increase in resistance can be monitored before the physical contact between scanning probe tip and sample, this technique is particularly useful to investigate the topography of delicate samples such as living cells. SICM has shown its potential in various applications such as high resolution and long-time imaging of living cells or the determination of local changes in cellular volume. Furthermore, SICM has been combined with various techniques such as fluorescence microscopy or patch clamping to reveal localized information about proteins or protein functions. This review details the various advantages and pitfalls of SICM and provides an overview of the recent developments and applications of SICM in biological imaging. Furthermore, we show that in principle, a combination of SICM and ion selective micro-electrodes enables one to monitor the local ion activity surrounding a living cell.

Abstract:A portable vacuum interface allowing direct probing of the electrode-electrolyte interface was developed. A classical electrochemical system consisting of a gold working electrode, platinum counter electrode, platinum reference electrode, and potassium iodide electrolyte was used to demonstrate real-time observation of the gold iodide adlayer on the electrode and chemical species as a result of redox reactions using cyclic voltammetry (CV) and time-of-flight secondary ion mass spectrometry (ToF-SIMS, a vacuum-based surface technique) simultaneously. This microfluidic electrochemical probe provides a new way to investigate the surface region with adsorbed molecules and the region of the diffused layer with chemical speciation in liquids in situ by surface sensitive techniques.

Abstract:Integration of dual-barrel membrane patch-ion channel probes (MP-ICPs) to scanning ion conductance microscopy (SICM) holds promise of providing a revolutionized approach of spatially resolved chemical sensing. A series of experiments are performed to further the understanding of the system and to answer some fundamental questions, in preparation for future developments of this approach. First, MP-ICPs are constructed that contain different types of ion channels including transient receptor potential vanilloid 1 and large conductance Ca2+ -activated K+ channels to establish the generalizability of the methods. Next, the capability of the MP-ICP platforms in single ion channel activity measurements is proved. In addition, the interplay between the SICM barrel and the ICP barrel is studied. For ion channels gated by uncharged ligands, channel activity at the ICP barrel is unaffected by the SICM barrel potential; whereas for ion channels that are gated by charged ligands, enhanced channel activity can be obtained by biasing the SICM barrel at potentials with opposite polarity to the charge of the ligand molecules. Finally, a proof-of-principle experiment is performed and site-specific molecular/ionic flux sensing is demonstrated at single-ion-channel level, which show that the MP-ICP platform can be used to quantify local molecular/ionic concentrations.

Abstract:Scanning ion conductance microscopy (SICM) offers the ability to obtain nanoscale resolution images of the membranes of living cells. Here, we show that a dual-barrel nanopipette probe based potentiometric SICM (P-SICM) can simultaneously map the topography and surface potential of soft, rough and heterogeneously charged surfaces under physiological conditions. This technique was validated and tested by systematic studies on model samples, and the finite element method (FEM) based simulations confirmed its surface potential sensing capability. Using the P-SICM method, we compared both the topography and extracellular potential distributions of the membranes of normal (Mela-A) and cancerous (B16) skin cells. We further monitored the structural and electrical changes of the membranes of both types of cells after exposing them to the elevated potassium ion concentration in extracellular solution, known to depolarize and damage the cell. From surface potential imaging, we revealed the dynamic appearance of heterogeneity of the surface potential of the individual cell membrane. This P-SICM method provides new opportunities to study the structural and electrical properties of cell membrane at the nanoscale.

Abstract:Electric Scanning Probe Microscopies are used to characterize the surface behavior of ferroelectric materials. The effects of local charge density on the chemistry and physics of ferroelectric surfaces are investigated. The kinetics and thermodynamics parameters of adsorption are assessed by variable temperature Scanning Surface Potential Microscopy. Contrast formation mechanism of Piezoresponse Force Microscopy (PFM) is analyzed in detail, and the contributions of electroelastic constants of the material to response amplitude are determined. The effect of experimental conditions including indentation force and tip radius of curvature is elucidated using PFM Contrast Mechanism Maps. Simple quantitative criterion for non-local cantilever-surface interactions in PFM is developed.

Abstract:During the dielectric breakdown process of thin solid-state nanopores, the application of high voltages may cause the formation of multi-nanopores on one chip, which number and sizes are important for their applications. Here, simulations were conducted to mimic the investigation of in situ nanopore detection with scanning ion conductance microscopy (SICM). Results show that SICM can provide accurate nanopore location and relative pore size. Detection resolution is influenced by the dimensions of the applied probe and separation between the probe and membranes, which can be enhanced under large voltages or a concentration gradient.

Abstract:An apparatus for explorations of ion transport in a medium and across an interface has been constructed. The ion soft-landing technique is used to deposit low-energy ions onto a pre-adsorbed medium layer on a metal substrate. The designed low-energy ion source can produce a mass-filtered ion beam with tens of nanoampere from solid sources such as bulk metals and salts. The kinetic energy of the ion beam can be lower than 1.0 eV, enabling the ions to be soft-landed onto the medium at the surface. A Kelvin probe with a resolution of less than 32 mV is incorporated to measure the surface potential (SP) variation of the ion-landed sample to monitor the ion transport process in the medium. Temperature-programmed SP measurements on an Ag+-adsorbed ice film prepared on Pt(111) reveal that the temperature threshold for the Ag+-induced SP change of the ice film is about 110 K. The apparatus performance demonstrates its potential in studies of ion transport and related phenomena at both macroscopic and microscopic levels.

Abstract:Clemmer and Beebe have pointed out that surface structures on graphite substrates can be misinterpreted as biopolymer images in STM experiments. We have been using electrochemical methods to react DNA fragments onto gold electrodes for STM and AFM imaging. The adsorbates produced in this way are only homogeneous in special circumstances. Searching an inhomogeneous substrate for ‘desired’ images limits the value of the data. Here, we report on a reversible method for imaging adsorbates. The molecules can be lifted onto and off the substrate during imaging. This leaves no doubt about the validity or statistical significance of the images. Furthermore, environmental effects (such as changes in electrolyte or surface charge) can be investigated easily.

Abstract:Applications of SICM extend well beyond topographic imaging. Opportunities to measure ion concentrations or electron transfer reactions with SICM probes have proven particularly powerful approaches. Electrochemical imaging applications of SICM have advanced rapidly and benefit from simplicity in probe (electrode) design, advanced methods of feedback, and developments in instrumentation for imaging. Here we consider select analytical applications of the scanning ion conductance microscope, considering first methods to measure ions, and second methods to measure electrons. Special attention is paid to the design of the probe and analytical caveats considered in such measurements.

Abstract:A new experimental approach to probe the structure of a water/nitrobenzene (W/NB) interface by scanning ion conductance microscopy (SICM) is presented. A nanopipette filled with aqueous solution serves as a SICM tip. The tip current is produced by the transfer of chloride (Cl-) and tetraphenylarsonium (TPAs+) ions between the aqueous solution inside the pipette and outer organic solution. The tip current increases instantaneously as a sign of touching the interface when such a tip approaches the W/NB interface. The continuous change of tip current suggests ion penetrations into the interfacial region which is less than 1 nm, in accordance with the mixed solvent model. Ion distribution information can also be extracted from the current-distance (approach) curves obtained from solutions with a series of electrolyte concentrations. The experimental results show that the SICM approach curves are sensitive to the thickness of diffuse layers. This type of technique is actually a modified version of SICM, and is similar to the operation of ion transfer mode of scanning electrochemical microscopy (SECM) with a SICM instrumentation.

Abstract:The development of ion-selective membranes for the selective response of a particular ion has been studied for many years; however, imaging the response of the membrane with a low detection limit is challenging. Here, high spatial-resolution electrochemical imaging of this response down to picomolar is achieved using scanning ion conductive microscopy. The detection strategy relies on the exclusion of a small amount of counter ions from the membrane in the presence of a low concentration of target ions in the solution. These excluded counter ions are adsorbed at the membrane-solution interface, leading to more positive charges at the surface. The resultant elevation of the ionic current in the approach curve behaves as the response for the target ions down to 10-11 M, which is much more sensitive than that using potentiometric measurement. The constant-current scanning of the membrane exhibits the fluctuation of the apparent surface height that is correlated with the ionic concentration, permitting the imaging of the response at the nanoscale. The achievement of highly sensitive and spatial-resolution imaging for the ionic response enable the collection of spatial response at the ion-selective membrane, which will greatly advance the study of ion-selective electrodes.
