Abstract:A challenge for optofluidic absorbance detection is the high concentration limit of detection due to the short optical path length. Herein, we introduce a concept of utilizing the coiled optical nanofiber for highly sensitive and robust optofluidic absorbance detection. Investigated by measuring the absorbance of FeCl3 solutions, the sensor shows a detection limit down to 10 μM with excellent reversibility in a concentration range of 0-5 mM. The sensitivity is 10-fold higher than that of standard absorbance measurement by using a 1 cm cuvette. Also, highly sensitive chloramphenicol sensing was demonstrated by using the enzyme-linked immunosorbent assay (ELISA) method, achieving a detection limit below 0.5 ng/L. The higher sensitivity and lower detection limit are caused by the large fractional power of evanescent field outside the nanofiber and the long detection length, which can effectively improve the absorption of the evanescent field, while the excellent reversibility is caused by the support of a polydimethylsiloxane (PDMS) pillar rather than by suspending the nanofiber in the microchannel. We envision that the present work may open up new opportunities for ultrasensitive chemical and biological sensing.
An optical nanofibre-enabled on-chip single-nanoparticle sensor

Abstract:Single-nanoparticle detection has received tremendous interest due to its significance in fundamental physics and biological applications. Here, we demonstrate an optical nanofibre-enabled microfluidic sensor for the detection and sizing of nanoparticles. Benefitting from the strong evanescent field outside the nanofibre, a nanoparticle close to the nanofibre can scatter a portion of the field energy to the environment, resulting in a decrease in the transmitted intensity of the nanofibre. On the other hand, the narrow and shallow microfluidic channel provides a femtoliter-scale detection region, making nanoparticles flow through the detection region one by one. By real-time monitoring of the transmitted intensity of the nanofibre, the detection of a single polystyrene (PS) nanoparticle as small as 100 nm in diameter and exosomes in solution is realised. Based on a statistical analysis, the mean scattering signal is related to the size of the nanoparticle. Experimentally, a mixture of nanoparticles of different diameters (200, 500, and 1000 nm) in solution is identified. To demonstrate its potential in biological applications, high-throughput counting of yeasts using a pair of microchannels and dual-wavelength detection of fluorescently labelled nanoparticles are realised. We believe that the developed nanoparticle sensor holds great potential for the multiplexed and rapid sensing of diverse viruses. An nanofibre-enabled microfluidic sensor for detection nanoparticles which hold great potentials for sensing of diverse viruses.

Abstract:Disposable sensors are widely used in biomedical detection due to their inherent safety, ease of use and low cost. An optofluidic laser is a sensitive bioassay platform; however, demonstrating its fabrication cheaply and reproducibly enough for disposable use has been challenging. Here, we report a low-cost, reproducible fiber optofluidic laser (FOFL) using a microstructured optical fiber (MOF). The MOF not only supports the whispering gallery modes for lasing but also serves as a microfluidic channel for sampling the liquid gain medium via capillary force. Because of the precise control of its geometry (δ < 0.4%) during the fiber-drawing process, good reproducibility in laser intensity (δ = 6.5%) was demonstrated by changing 10 sections of the MOF. The strong coupling between the in-fiber resonator and gain medium enables a low threshold of 3.2 μJ mm-2. The angular dependence of the laser emission was observed experimentally and analyzed with numerical simulations. An array of the FOFLs was also demonstrated. This technology has great potential for low-cost bioassay applications.
Nanoparticle-based Fiber Optofluidic Laser for Label-Free Protein Detection

Abstract:Fiber optofluidic laser (FOFL) is a promising technology for investigating biomedical and chemical interactions. However, label-free FOFL for small molecular weight protein analysis remains a major challenge. Here, a nanoparticle-based fiber optofluidic laser is demonstrated to achieve label-free biodetection of small molecular weight protein. The proposed FOFLs amplify the loss inside the optical fiber microring through the nanoparticle enhanced light-matter interaction, enabling the detection of small molecular weight protein. The laser dye and the nanoparticle-based immunoassay kits are homogeneously mixed to achieve the label-free biodetection. The C-reactive protein (CRP) is detected in the dynamic range of 38.1 ng/mL to 381 ng/ml with a limit of detection of 26 ng/ml, which is two orders of magnitude lower than that of methods without nanoparticles. The assay time can be as short as 15 min with a low-volume biosample/reagents consumption of 301 nL. This novel concept not only provides a simple but versatile method to achieve the label-free biodetection of small molecular weight protein, but also deepens insight into how molecules interact with the laser emission.
A Sequentially Bioconjugated Optofluidic Laser for Wash-out-free and Rapid Biomolecular Detection.

Abstract:Microstructures can improve both sensitivity and assay time in heterogeneous assays (such as ELISA) for biochemical analysis; however, it remains a challenge to perform the essential wash process in those microstructure-based heterogeneous assays. Here, we propose a sequential bioconjugation protocol to solve this problem and demonstrate a new type of fiber optofluidic laser for biosensing. Except for acting as an optical microresonator and a microstructured substrate, the miniaturized hollow optical fiber (HOF) is used as a microfluidic channel for storing and transferring reagents thanks to its capability in length extension. Through the capillary action, different reagents were sequentially withdrawn into the fiber for specific binding and washing purposes. By using the sequentially bioconjugated FOFL, avidin molecules are detected based on competitive binding with a limit of detection of 9.5 pM, ranging from 10 pM to 100 nM. It is demonstrated that a short incubation time of 10 min is good enough to allow the biomolecules to conjugate on the inner surface of the HOF. Owing to its miniaturized size, only 589 nL of liquid is required for incubation, which reduces the sample consumption and cost for each test. This work provides a tool to exploit the potential of microstructured optical fibers in high-performance biosensing.
In-fiber Optofluidic Michelson Interferometer for Detecting Small Volume and Low Concentration Chemicals with a Fiber Ring Cavity Laser

Abstract:An in-fiber optofluidic Michelson interferometer fabricated by femtosecond laser micromachining technology was presented. The interferometer structure was prepared by a special optical fiber with two cores and a microchannel. One core was connected to the core of a single-mode fiber (SMF) and functioned as a sensing arm on the microchannel inner wall, the other core worked as a reference arm and was connected to the optical waveguide introduced by femtosecond laser processing. By combining with a ring cavity laser, the detection limit (DL) of the interferometer could be improved. The laser sensor presented a high linear response with a refractive index (RI) sensitivity of 1039.77 nm/RIU and the calculated DL could reach as low as 1.675 x 10(-5) RIU. Then, the sensor measured different concentrations of vitamin C in tablets in the concentration range of 0-10 mg/mL with a detection limit of 0.06 mg/mL. Due to the advantages of optofluidic structure, the volume of the detection solution could be as low as few tens of nanoliters, and the time to replace and detect the solution only took a few seconds. In addition, the laser sensor realized label-free DNA hybridization detection through surface func-tionalization, and could detect 1 mu M target DNA solution with high specificity. This sensing device had great potential for small volume and low concentration sample detection in the biochemical field.

Abstract:Abstract Biomarker detection is key to identifying health risks. However, designing sensitive and single-use biosensors for early diagnosis remains a major challenge. Here, we report submonolayer lasers on optical fibers as ultrasensitive and disposable biosensors. Telecom optical fibers serve as distributed optical microcavities with high Q-factor, great repeatability, and ultralow cost, which enables whispering-gallery laser emission to detect biomarkers. It is found that the sensing performance strongly depends on the number of gain molecules. The submonolayer lasers obtained a six-order-of-magnitude improvement in the lower limit of detection (LOD) when compared to saturated monolayer lasers. We further achieve an ultrasensitive immunoassay for a Parkinson’s disease biomarker, alpha-synuclein (α-syn), with a lower LOD of 0.32 pM in serum, which is three orders of magnitude lower than the α-syn concentration in the serum of Parkinson’s disease patients. Our demonstration of submonolayer biolaser offers great potentials in high-throughput clinical diagnosis with ultimate sensitivity.

Abstract:Disposable biosensors are of great importance in disease diagnosis due to their inherent merits of no cross-contamination and ease of use. Optofluidic laser (OFL) sensors are a new category of sensitive biosensors; however, it is challenging to cost-effectively mass-produce them to achieve disposability. Here, we report a disposable optofluidic laser immunosensor based on thin-walled hollow optical fibers (HOFs). Using a fiber draw tower, the fabrication parameters, including drawing speed and gas flow rate, are explored, and the HOF geometry is precisely controlled, which allows identical laser microring resonators to be distributed along the fibers. The disposable OFL immunosensor detects the protein concentration in the HOF through a wash-free immunoassay. Enabled by the disposable sensors, the statistical characteristics of 80 tests for each concentration greatly reduces the bioassay uncertainty. A low coefficient of variation (CV) of 3.3% confirms the high reproducibility of the disposable HOF-OFL sensors, and the mean of the normal distribution of the logarithmic OFL intensity serves as the sensing output. A limit of detection of 11 nM within a short assay time of 15 min is achieved. These disposable immunosensors possess the advantages of low cost, high reproducibility, fast assay, and low-volume consumption of sample and reagents. We believe that this work will inspire disposable optofluidics through the mass production of multifunctional microstructured optical fibers.

Abstract:Optofluidic lasers (OFLs) are an emerging technological platform for biochemical sensing, and their good performance especially high sensitivity has been demonstrated. However, high-throughput detection with an OFL remains a major challenge due to the lack of reproducible optical microcavities. Here, we introduce the concept of a distributed fibre optofluidic laser (DFOFL) and demonstrate its potential for highthroughput sensing applications. Due to the precise fibre geometry control via fibre drawing, a series of identical optical microcavities uniformly distributed along a hollow optical fibre (HOF) can be achieved to obtain a one-dimensional (1D) DFOFL. An enzymatic reaction catalysed by horseradish peroxidase (HRP) can be monitored over time, and the HRP concentration is detected by DFOFL-based arrayed colorimetric detection. Experimentally, five-channel detection in parallel with imaging has been demonstrated. Theoretically, spatial multiplexing of hundreds of channels is achievable with DFOFL-based detection. The DFOFL wavelength is tuned over hundreds of nanometers by optimizing the dye concentration or reconfiguring the liquid gain materials. Extending this concept to a two-dimensional (2D) chip through wavelength multiplexing can further enhance its multi-functionality, including multi-sample detection and spectral analysis. This work opens the door to high-throughput biochemical sensing.

Abstract:Optofluidic laser is an emerging method for biochemical sensing. The topic on, which kind of structure is the best to integrate the laser resonator and microfluidic channel, is worth to be explored. Here, we report a hollow optical fiber optofluidic laser based on a dye-doped polymer film. This fiber microstructure is a promising candidate that serves both as a laser microresonator and a compact microfluidic channel. The refractive index of the polymer film is higher than silica so that the whispering gallery mode can be supported. The coating process is explored in detail. The laser spectra and threshold properties of the HOF microlaser are investigated. The dye-doped polymer film can be controlled to be as thin as 1.6 μm and a threshold of below 1 μJ/mm2 can be reached. Thanks to the strong light-liquid interaction enabled by the polymer coating on the inner interface of HOF, sensitive refractive index detection is demonstrated by monitoring the laser wavelength shift, which has the potential for wavelength division multiplexing. This laser sensor is promising for label-free biochemical detection.

Abstract:We have developed a chip-scale optofluidic sensor for biomolecular detection, by tapering laterally aligned silica microfiber and capillary to form a modal interferometer. With the pre-immobilization of DNA probes, the sensor is capable of selectively detecting single-stranded microRNA-let7a (molecular weight: 6.5 k) by measuring the spectral shift of the interferometric spectrum. A log-linear response from 2 nM to 20 μM and a minimum detectable concentration of 212 pM (1.43 ng/mL) have been achieved. The sensor is promising for future diagnosis applications due to its high sensitivity, resistance to environmental perturbations, improved portability, and intrinsic connection to fiber optic measurement.

Abstract:An optical fiber optofluidic biosensor for the detection of DNA hybridization and methylation has been proposed and experimentally demonstrated. An in-line fiber Michelson interferometer was formed in the photonic crystal fiber. A micrhole in the collapsed region, which combined the tunable mode coupler and optofluidic channel, was fabricated by using femtosecond laser micromachining. The mode field diameter of the guided light is changed with the refractive index in the optofluidic channel, which results in the tunable coupling ratio. Label-free detections of the DNA hybridization and methylation have been experimentally demonstrated. The probe single stranded DNA (ssDNA) was bound with the surface of the optofluidic channel through the Poly-l-lysine layer, and the hybridization between a short 22-mer probe ssDNA and a complementary target ssDNA was carried out and detected by interrogating the fringe visibility of the reflection spectrum. Then, the DNA methylation was also detected through the binding between the methylated DNA and the 5-methylcytosine (5-mC) monoclonal antibody. The experiments results demonstrate that the limit of detection of 5nM is achieved, establishing the tunable mode coupler as a sensitive and versatile biosensor. The sensitive optical fiber optofluidic biosensor possesses high specificity and low temperature cross-sensitivity.

Abstract:We report a reproducible optofluidic laser (OFL) for multichannel biochemical sensing. A hollow optical fiber (HOF) serves as both microring resonator for lasing and microfluidic channel. The lasing mechanism is analyzed in spectral domain. Thanks to the precise control of the fiber geometry and the rotational symmetry, the HOF-OFL emission is uniformly distributed in the angular direction (σ = 0.6%) and can be employed conveniently for disposable or arrayed use. An array of ten OFLs is demonstrated with good reproducibility ( σ = 3.9%) in laser threshold. The arrayed sensing capability of the OFL is also investigated. The HOF-OFLs are highly reproducible, easy to integrate in chip-scale, and have great potential for biochemical detection.

Abstract:Biomarker detection is the key to identifying health risks. However, designing sensitive biosensors in a single-use mode for disease diagnosis remains a major challenge. Here, we report sub-monolayer biolasers with remarkable repeatability for ultrasensitive and disposable biomarker detection. The biolaser sensors are designed by employing the telecom optical fibers as distributed optical microcavities and pushing the gain molecules down to the sub-monolayer level. We observe a status transition from the monolayer biolaser to the sub-monolayer biolaser by tuning the specific conjugation. By reducing the fluorophores down to the threshold density (~ 3.2 x 10-13 mol/cm2), we demonstrate an ultimate sensitivity of sub-monolayer biolaser with six orders of magnitude enhancement compared with the monolayer biolasers. We further achieved ultrasensitive immunoassay for Parkinson's disease biomarker, alpha-synuclein, with a lower limit of detection of 0.32 pM in serum. This biosensor with massive fabrication capability at ultralow cost provides a general method for the ultrasensitive disposable biodetection of disease biomarkers.
Enzyme Catalyzed Optofluidic Biolaser for Sensitive Ion Concentration Detection

Abstract:The enzyme horseradish peroxidase (HRP) has been extensively used in biochemistry for its ability to amplify a weak signal. By using HRP catalyzed substrate as the gain medium, we demonstrate sensitive ion concentration detection based on the optofluidic laser. The enzyme catalyzed reaction occurs in bulk solution inside a Fabry-Perot laser cavity, where the colorless, non-fluorescent 10-Acetyl-3,7-dihydroxyphenoxazine (ADHP) substrate is oxidized to produce highly fluorescent resorufin. Laser emission is achieved when pumped with the second harmonic wave of a Q-switched YAG laser. Further, we use sulfide anion (S2-) as an example to investigate the sensing performance of enzyme catalyzed optofluidic laser. The laser onset time difference between the sample to be tested and the reference is set to be the sensing output. Thanks to the amplification effects of both the enzymatic reaction and laser emission, we achieve a detection limit of 10 nM and a dynamic range of 3 orders of magnitude.
Detection of Streptavidin Using Liquid Crystal Based Whispering Gallery Mode Microbubble

Abstract:Protein is a complex chemical substance essential for human survival. Traditional protein detection methods, such as colorimetry, electrochemical analysis, and enzyme-linked immunosorbent assays, have shown good specificity and accuracy for the protein detection. However, all these methods require specialized instruments, and the detection procedures are laborious and time-consuming. As a result, a rapid, sensitive, label-free protein detection method is urgently needed. Herein, we have developed an ultra-sensitive biosensor for the detection of low-concentration protein molecules, employing liquid crystal (LC)-amplified optofluidic resonator. Since the orientations of LCs highly depend on the surface biomolecular binding processes, LCs can be employed to realize the extremely sensitive detection of biomolecules. Immobilized protein molecules interfere with the orientation of LCs by reducing the vertical anchoring force from the alignment layer in which the spectral wavelength shift was monitored as a sensing parameter. A biosensing platform based on an LC-amplified optofluidic whispering gallery mode (WGM) resonator was designed and studied accordingly. Due to the simultaneous interaction of the WGM and the LCs in the optofluidic resonator, the changes caused by the injection of protein molecules will be amplified, resulting in a shift in the resonance wavelength. Total wavelength shifts scale proportionally to the molecular concentrations of the protein within a certain range. The detection limit for streptavidin (SA) can reach as low as the femtometer level, which is significantly higher than the detection limit in the classic polarized optical microscope (POM) method visible with the naked eye. In addition to SA, the LC-based optofluidic resonator can also be applied to detect a variety of protein molecules. Our study demonstrates that LC-amplified optofluidic resonator can provide a novel solution for ultrasensitive real-time characterization of biosensing and biomolecular interactions.

Abstract:Optofluidic laser (OFL) is an emerging technology that integrates microfluidics, optical microcavity, and the liquid gain medium. Thanks to the strong light-matter interaction in the laser cavity, OFL could perform bio-chemical sensing with high performance. Here, we review the recent advances in optofluidic lasers and highlight their applications in biochemical analysis with unique properties. We introduce the physical mechanisms as well as the structure and material employed in the optofluidic laser to achieve high sensitivity, disposability, fast response and high throughput. In addition, we discuss the cross properties between these performance indicators to show how to achieve overall high performance. The effects of new materials including both gain materials and enhancer materials in laser-based sensors are briefly discussed. Furthermore, the comparison with other optical biochemical sensors and the prospects of OFLs are also given.
