Abstract:Single-nanoparticle detection has received tremendous interest due to its significance in fundamental physics and biological applications. Here, we demonstrate an optical nanofibre-enabled microfluidic sensor for the detection and sizing of nanoparticles. Benefitting from the strong evanescent field outside the nanofibre, a nanoparticle close to the nanofibre can scatter a portion of the field energy to the environment, resulting in a decrease in the transmitted intensity of the nanofibre. On the other hand, the narrow and shallow microfluidic channel provides a femtoliter-scale detection region, making nanoparticles flow through the detection region one by one. By real-time monitoring of the transmitted intensity of the nanofibre, the detection of a single polystyrene (PS) nanoparticle as small as 100 nm in diameter and exosomes in solution is realised. Based on a statistical analysis, the mean scattering signal is related to the size of the nanoparticle. Experimentally, a mixture of nanoparticles of different diameters (200, 500, and 1000 nm) in solution is identified. To demonstrate its potential in biological applications, high-throughput counting of yeasts using a pair of microchannels and dual-wavelength detection of fluorescently labelled nanoparticles are realised. We believe that the developed nanoparticle sensor holds great potential for the multiplexed and rapid sensing of diverse viruses. Coiled Optical Nanofiber for Optofluidic Absorbance Detection.

Abstract:A challenge for optofluidic absorbance detection is the high concentration limit of detection due to the short optical path length. Herein, we introduce a concept of utilizing the coiled optical nanofiber for highly sensitive and robust optofluidic absorbance detection. Investigated by measuring the absorbance of FeCl3 solutions, the sensor shows a detection limit down to 10 μM with excellent reversibility in a concentration range of 0-5 mM. The sensitivity is 10-fold higher than that of standard absorbance measurement by using a 1 cm cuvette. Also, highly sensitive chloramphenicol sensing was demonstrated by using the enzyme-linked immunosorbent assay (ELISA) method, achieving a detection limit below 0.5 ng/L. The higher sensitivity and lower detection limit are caused by the large fractional power of evanescent field outside the nanofiber and the long detection length, which can effectively improve the absorption of the evanescent field, while the excellent reversibility is caused by the support of a polydimethylsiloxane (PDMS) pillar rather than by suspending the nanofiber in the microchannel. We envision that the present work may open up new opportunities for ultrasensitive chemical and biological sensing.

Abstract:Conventional fluorescence scanners utilize multiple filters to distinguish different fluorescent labels, and problems arise because of this filter-based mechanism. In this work we propose a line-monitoring, hyperspectral fluorescence technique which is designed and optimized for applications in multi-channel microfluidic systems. In contrast to the filter-based mechanism, which only records fluorescent intensities, the hyperspectral technique records the full spectrum for every point on the sample plane. Multivariate data exploitation is then applied to spectra analysis to determine ratios of different fluorescent labels and eliminate unwanted artifacts. This sensor is designed to monitor multiple fluidic channels simultaneously, providing the potential for multi-analyte biosensing. The detection sensitivity is approximately 0.81 fluors/μm2, and this sensor is proved to act with a good homogeneity. Finally, a model experiment of detecting short oligonucleotides has demonstrated the biomedical application of this hyperspectral fluorescence biosensor.

Abstract:We report a reproducible optofluidic laser (OFL) for multichannel biochemical sensing. A hollow optical fiber (HOF) serves as both microring resonator for lasing and microfluidic channel. The lasing mechanism is analyzed in spectral domain. Thanks to the precise control of the fiber geometry and the rotational symmetry, the HOF-OFL emission is uniformly distributed in the angular direction (σ = 0.6%) and can be employed conveniently for disposable or arrayed use. An array of ten OFLs is demonstrated with good reproducibility ( σ = 3.9%) in laser threshold. The arrayed sensing capability of the OFL is also investigated. The HOF-OFLs are highly reproducible, easy to integrate in chip-scale, and have great potential for biochemical detection.

Abstract:In this paper, we reported an optical fiber optofluidic laser (FOFL) biosensor based on thin-walled hollow optical fiber (HOF). HOFs with wall thickness of several microns were fabricated, which integrated both whispering gallery mode (WGM) optical microcavity and microfluid channel for FOFL. Based on the fluorescence quenching caused by the interaction between rhodamine 6G (R6G) and hemoglobin (Hb), FOFL was used to amplify such quenching effect and an FOFL biosensor was constructed to detect Hb concentration. The experimental results showed that the limit of detection (LOD) of 0.7 nM and the dynamic range of three orders of magnitude were obtained within a short assay time on the order of seconds, which were both great improvements over fluorescence detection results. In addition, the designed sensor has the advantages of low-volume sample consumption, simple homogeneous and wash-free process, and reusability. This work may provide a new approach for Hb detection in blood and has the potential to be applied in the diagnosis of some related diseases.

Abstract:Immunoassay can be divided into two aspects, one is immobilization of antibodies for the efficient detection of corresponding antigens, and the other is immobilization of antigens to search for antibodies which work against them. In this paper, we demonstrated these two aspects of immunoassay by using the disposable fiber optic biosensors based on surface plasmon resonance (SPR) through surface decoration with half-antibody fragments, which has been scarcely ever reported to the best of our knowledge. We first fabricated the fiber optic SPR biosensor which consists of one gold film coated single-mode fiber sandwiched by two multimode fibers. Then, we decorated the fiber optic SPR biosensors with antibody fragments and antigen fragments, respectively, and compared the specific detection performances of these two kinds of sensors. After surface decoration with half-antibody fragments, the antigen-decorated fiber probe has a demonstrated sensitivity and limit of detection of 0.9771nm/(μg/mL) and 0.1 μg/mL, respectively, which improves by 10 times compared with the performance of the antibody-decorated fiber probe. Additionally, the selective detection results indicate that our proposed biosensor can be employed as a reliable antigen detector or an effective antibody filter. Our proposed sensor has the advantages of miniaturization, low cost, simple usage, label-free detection, high efficiency and sensitivity, and can effectively avoid cross-contamination caused by reuse. Given the reliable and clean detection method for immunoassay, our work should open a new window for the utilization of miniaturized fiber optic sensors in biochemical sensing.
Gliomas: association of histology and molecular genetic analysis of chromosomes 1p, 10q, and 19q.

Abstract:The aim of our study was to evaluate the frequencies of loss of heterozygosity (LOH) on chromosomes 1p, 10q, and 19q in gliomas and to correlate them with the histological diagnosis and with patient age and gender. We found deletions within chromosome 1p to be significantly associated with the histological subtype of glial tumor (P < 0.05); frequency of 1p deletions increased from astrocytoma (0%) through glioblastoma (31%) and oligoastrocytoma (57%) to oligodendroglioma (63%). In patients with 1p LOH, the odds for having astrocytoma or glioblastoma were approximately 10-fold and 4-fold lower, respectively, than oligodendroglioma. The odds for having oligoastrocytoma were similar to oligodendroglioma (OR = 1.3). The frequency of 10q LOH in patients with glioblastoma was significantly higher than in patients with oligodendroglioma (89% vs. 36%; P < 0.005). In patients with oligodendroglioma, most cases with LOH on chromosome 1p also had LOH 19q (90%), one case of 1p LOH also had a deletion on 10q. Statistical analyses revealed a significant association between deletions on 1p and 19q (P < 0.05). Our data provide evidence that use of molecular genetic analyses of chromosomes 1p, 19q, and 10q might improve the diagnosis of gliomas.

Abstract:Fluorescent nanosensors hold promise to address analytical challenges in the biopharmaceutical industry. The monitoring of therapeutic protein critical quality attributes such as aggregation is a long-standing challenge requiring low detection limits and multiplexing of different product parameters. However, general approaches for interfacing nanosensors to the biopharmaceutical process remain minimally explored to date. Herein, we design and fabricate a integrated fiber optic nanosensor element, measuring sensitivity, response time, and stability for applications to the rapid process monitoring. The fiber optic-nanosensor interface, or optode, consists of label-free nIR fluorescent single-walled carbon nanotube transducers embedded within a protective yet porous hydrogel attached to the end of the fiber waveguide. The optode platform is shown to be capable of differentiating the aggregation status of human immunoglobulin G, reporting the relative fraction of monomers and dimer aggregates with sizes 5.6 and 9.6 nm, respectively, in under 5 min of analysis time. We introduce a lab-on-fiber design with potential for at-line monitoring with integration of 3D-printed miniaturized sensor tips having high mechanical flexibility. A parallel measurement of fluctuations in laser excitation allows for intensity normalization and significantly lower noise level (3.7 times improved) when using lower quality lasers, improving the cost effectiveness of the platform. As an application, we demonstrate the capability of the fully integrated lab-on-fiber system to rapidly monitor various bioanalytes including serotonin, norepinephrine, adrenaline, and hydrogen peroxide, in addition to proteins and their aggregation states. These results in total constitute an effective form factor for nanosensor-based transducers for applications in industrial process monitoring.

Abstract:Optical fiber provides a unique and versatile platform for developing point-of-care optical sensing systems. Here we first propose a Fabry-Pérot (FP) based flow-through optofluidic biosensor, and then construct an all-fiber system which fully utilizes optical fibers to achieve rapid, sensitive, label-free biomolecular detection. This sensor consists of two single mode fibers (SMFs) with reflecting surfaces and a photonic crystal fiber (PCF) vertically sandwiched by them. Firstly, the SMFs act as waveguides for delivering light into and out of an optofluidic device (like PCF); secondly, instead of using the optical properties of the PCF, we take advantage of its inherent multiple fluidic channels and large sensing surface; thirdly, the two reflective surfaces and the PCF form a Fabry-Pérot microresonator and its resonance mode is sensitive to the change in the fluidic channels, which can be used to detect the substances flowing through the fluidic channels or adsorbing on the channel surface. In this paper, we explore the operating principle of the FP-based optofluidic biosensor, theoretically and experimentally investigate its feasibility and capability. The results show that the all-fiber optofluidic sensor is a promising technology platform for rapid, sensitive and high-throughput biological and chemical sensing.

Abstract:The novel 3D microfluidic concept of "lab-on-hollow fiber membrane (HFM)" was presented for multifunctional and rapid biological assays, integrating sample size sieving and colorimetric quantification in a HFM. Herein, micro-porous HFMs with a gradient pore size and high hydrophilic flux were used as microfluidic device substrates. The membrane pores selectively trapped macromolecules within the inner surface, while allowing free diffusion of smaller molecules, including glucose and protein. The microfluidic flow rate in HFM closely followed the Lucas-Washburn and Laplace's models, indicating that the microfluidics facilitated the upward flow of the fluid by micro-capillary action without external pumping. Subsequently, for sensing of different biomolecules, a highly sensitive fluorescent or optical chromogenic reagent was immobilized in HFM by an electrostatic interaction. Pyronin G fluorescence reagent was quenched by blood glucose and the quenching efficiency showed a good linear correlation with glucose concentration (1~22 mM, R2=0.997). Moreover, this sensing platform was then further applied for the analysis of urine protein or glucose in the visible spectrum, with wide testing range. Compared to traditional 2D flat membrane devices, this 3D-HFM microfluidic device exhibited excellent sensing versatility and color rendering uniformity with enhanced sensitivity. Target molecules screening, conditioning, enzymatic recognition, and signal readout of biomolecules have all been implemented on this device, which has paved the way to highly sensitive assays on POCT. Cyanophage Diversity, Inferred from g20 Gene Analyses, in the Largest Natural Lake in France, Lake Bourget

Abstract:ABSTRACT The genetic diversity of the natural freshwater community of cyanophages and its variations over time have been investigated for the first time in the surface waters of the largest natural lake in France. This was done by random screening of clone libraries for the g20 gene and by denaturing gradient gel electrophoresis (DGGE). Nucleotide sequence analysis revealed 35 distinct cyanomyovirus g20 genotypes among the 47 sequences analyzed. Phylogenetic analyses showed that these sequences fell into seven genetically distinct operational taxonomic units (OTUs). The distances between these OTUs were comparable to those reported between marine clusters. Moreover, some of these freshwater cyanophage sequences were genetically more closely related to marine cyanophage sequences than to other freshwater sequences. Both approaches for the g20 gene (sequencing and DGGE analysis) showed that there was a clear seasonal pattern of variation in the composition of the cyanophage community that could reflect changes in its biological, chemical, and/or physical environment.

Abstract:Fiber optic (FO) sensors have great potential for low-cost, microsample testing and compact medical diagnostics. In this work, a FO sensor based on dielectric nanostructure is proposed and demonstrated based on the unique characteristics of dielectric materials, such as low ohmic loss and good biocompatibility. Specifically, we fabricate a SPR-FO sensor by inserting a sleeve with a large-scale, highly ordered dielectric nanodisk array at the end of the fiber probe. And we systematically investigate the sensing performance of the FO sensor including bulk and surface sensitivities. It is found that due to very low absorption losses, the FO sensor based on dielectric nanodisk exhibits a narrow resonance, thus leading to a sensing figure-of-merit almost twice larger than that of based on the metal plasmonic nanodisk. Also, the FO sensor shows a linear response to adsorbate layer thickness changes, which is clearly advantageous for quantitative analysis. Due to the highly localized electromagnetic fields, it can detect small molecules with a concentration of 1 μM, in which a molecular layer with thickness less than 1 nm can produce resonance changes as large as 6 nm. Our work could open a new opportunity for future bio-compatible, robust and low-loss medical diagnostic devices.

Abstract:In situ acquisition of spatial distribution of biochemical substances is important in cell analysis, cancer detection and other fields. Optical fiber biosensors can achieve label-free, fast and accurate measurements. However, current optical fiber biosensors only acquire single-point of biochemical substance content. In this paper, we present a distributed optical fiber biosensor based on tapered fiber in optical frequency domain reflectometry (OFDR) for the first time. To enhance evanescent field at a relative long sensing range, we fabricate a tapered fiber with a taper waist diameter of 6 μm and a total stretching length of 140 mm. Then the human IgG layer is coated on the entire tapered region by polydopamine (PDA) -assisted immobilization as the sensing element to achieve to sense anti-human IgG. We measure shifts of the local Rayleigh backscattering spectra (RBS) caused by the refractive index (RI) change of an external medium surrounding a tapered fiber after immunoaffinity interactions by using OFDR. The measurable concentration of anti-human IgG and RBS shift has an excellent linearity in a range from 0 ng/ml to 14 ng/ml with an effective sensing range of 50 mm. The concentration measurement limit of the proposed distributed biosensor is 2 ng/ml for anti-human IgG. Distributed biosensing based on OFDR can locate a concentration change of anti-human IgG with an ultra-high sensing spatial resolution of 680 μm. The proposed sensor has a potential to realize a micron-level localization of biochemical substances such as cancer cells, which will open a door to transform single-point biosensor to distributed biosensor.

Abstract:A critical challenge to realize ultra-high sensitivity with optical fiber interferometers for label free biosensing is to achieve high quality factors (Q-factor) in liquid. In this work a high Q-factor of 105, which significantly improves the detection resolution is described based on a structure of single mode -core-only -single mode fiber (SCS) with its multimode (or Mach-Zehnder) interference effect as a filter that is integrated into an erbium-doped fiber laser (EDFL) system for excitation. In the case study, the section of core-only fiber is functionalized with porcine immunoglobulin G (IgG) antibodies, which could selectively bind to bacterial pathogen of Staphylococcus aureus (S. aureus). The developed microfiber-based biosensing platform called SCS-based EDFL biosensors can effectively detect concentrations of S. aureus from 10 to 105 CFU/mL, with a responsivity of 0.426 nm wavelength shift in the measured spectrum for S. aureus concentration of 10 CFU/mL. The limit of detection (LoD) is estimated as 7.3 CFU/mL based on the measurement of S. aureus with minimum concentration of 10 CFU/mL. In addition, when a lower concentration of 1 CFU/mL is applied to the biosensor, a wavelength shift of 0.12 nm is observed in 10% of samples (1/10), indicating actual LoD of 1 CFU/mL for the proposed biosensor. Attributed to its good sensitivity, stability, reproducibility and specificity, the proposed EDFL based biosensing platform has great potentials for diagnostics.

Abstract:An optofluidic microlaser with a unique dual‐band emission is realized by using a photonic bandgap fiber. The periodic resonances in one band are used for immunosensing of microalbumin, while the location‐dependent random resonances in another band generate a 2D laser tag. The photonic bandgap fiber microlaser integrates dual functions of high‐capacity tagging and biosensing with promising applications in disease screening. Lasers are emerging as novel photonic tags for single‐cell labeling, anticounterfeiting, and encryption technology due to their narrow linewidth, high spectral multiplexing capacity, and superior stimuli‐responsiveness. These laser‐encoded photonic tags mostly distinguish the heterogeneity but do not yet provide both tagging and sensing of biosamples, which is highly desirable for disease screening. Here, a photonic bandgap (PBG) fiber microlaser that works as a 2D tag and an immunosensor is developed. The tubular PBG structure allows strong light–matter interaction and supports dual‐band lasing in the same optical fiber, enabling massive biosample tagging and sensitive biodetection. By encoding the random resonant peaks in the short‐wave band and multiplexing in the spatial domain, a 2D laser tag is generated with a large encoding capacity (>28500). Immunosensing of microalbumin is realized by using the periodic resonant peaks in the long‐wave band, and a limit of detection of 0.06 ng μL−1 is achieved. This work is inspiring for the development of high‐performance, multifunctional integrated devices for disease screening.
A Self-Assembled Plasmonic Optical Fiber Nanoprobe for Label-Free Biosensing

Abstract:The plasmonic optical fiber sensors have attracted wide attention for label-free biosensing application because of their high integration, small footprint and point-of-care measurement. However, the integration of plasmonic nanostructures on optical fiber probes always relies on the top-down nanofabrication approaches, which have several inherent shortcomings, including high cost, time-consuming, and low yields. Here, we develop a plasmonic nanohole-patterned multimode optical fiber probe by self-assembly nanosphere lithography technique with low fabrication cost and high yields. The multimode optical fiber possesses large facet area and high numerical aperture, which not only simplifies fabrication process, but also increases coupling efficiency of incident light. Originating from the resonant coupling of plasmonic modes, the plasmonic fiber nanoprobe has a distinct reflection dip in the spectrum and exhibits strong near-field electromagnetic enhancement. We experimentally investigate the sensing performances of plasmonic fiber nanoprobe, and further demonstrate it in real-time monitoring specific binding of protein molecules. The experimental results imply that the nanohole-patterned multimode optical fiber probe is a good candidate for developing miniaturized and portable biosensing systems.
Highly sensitive CD40L protein discrimination via label-free fiber sensing technologies

Abstract:The recombinant Cluster of Differentiation 40 Ligand (CD40L) can be expressed in various cells and is closely related to various types of cancer. This association underscores the critical need for expedited and precise measurement of CD40L levels in clinical fluid specimens. A novel optical fiber biosensor has been devised, employing single-mode fibers that are sandwiched around a coreless fiber, with the diameter refined by etching with hydrogen fluoride. This innovative configuration allows for light transmission through the evanescent field, thereby enhancing the sensor's sensitivity to changes in the surrounding refractive index. Employing chemical binding techniques, CD40 was securely immobilized onto the fiber's surface, facilitating the detection of CD40L. The sensor exhibited a sensitivity of 1.126 nm (μg mL-1)-1 and a detection limit of 0.68 nM. Furthermore, the sensor's specificity for CD40L was validated using authentic clinical serum samples spiked with artificial analytes. In addition, the specificity of CD40L of the proposed sensor was proved using natural clinical serum samples with added artificial analyte, assisted by the ELISA method, and the results ideally conformed with the detection of standard samples. With the aid of the ELISA method, the outcomes were found to be in excellent agreement with those from standard sample detection. Consequently, the findings indicate that this sensor provides a specific, label-free, and highly sensitive method for CD40L detection, showcasing its significant potential for applications in molecular biology research.

Abstract:Modification-free biosensing with high specificity and sensitivity is essential for miniaturized, online, integrated, and rapid, or even real-time molecular analyses. However, most optical biosensors are based on surface pre-modification or fluorescent labeling, and have either low sensitivity or low quality factor (Q). To address these difficulties, in this study, an optical sensor prototype was developed with a microbubble optofluidic channel integrated inside a Fabry-Pérot cavity to three-dimensionally tailor the intra-cavity light field via the intra-cavity lensing (microbubble) configuration. A high Q-factor (∼105), small mode volume, and high light energy density were experimentally achieved with this "stereo-sensor" while maintaining an ultrahigh refractive index (RI) sensitivity (679 nm/RIU) and ultra-small RI resolution (∼10-7 RIU at 950 nm). Moreover, specific detection of very low concentration of biomolecules (5 fg/mL for human IgG and 0.5 pg/mL for human serum albumin (HSA)) and wide range of protein concentrations (e.g., fg/mL-ng/mL for human IgG and pg/mL-ng/mL for HSA) without probe pre-modification were achieved owing to the RI change specifically associated with the probe-target binding and the corresponding bio-macromolecular conformation change. This modification-free stereosensing scenario is applicable to continuous, real-time, and multiplexed operations, thus showing potential for online, integrated, dynamic, biomolecular analyses in vitro or in vivo, such as the dynamic metabolic analysis of single cells or organoids and point-of-care tests.
