Abstract:A silver catalyzed domino reaction of N-cyanamide alkenes and 1,3-dicarbonyls including 1,3-diketones and ethyl acetoacetate has been developed for the facile synthesis of quinazolinones. In the presence of AgNO3/K2S2O8, the diketones could be converted to radicals and coupled with N-cyanamide alkenes to undergo a cyclization cascade for accessing quinazolinones. This method features mild reaction conditions, readily available starting materials, and valuable synthetic utility. Moreover, the products could be further transformed into various heterocycles.

Abstract:A novel cobalt(II)-catalyzed domino alkene activation/isocyanide insertion/cyclization/directing group migration sequence has been developed for the construction of valuable 2-aminoquinoline scaffolds. By using Co(OAc)2·4H2O as the non-noble catalyst, the reaction of N-(2-vinylphenyl)picolinamides proceeded smoothly to give various 2-aminoquinolines in good yields (up to 91%).

Abstract:A cobalt‐catalyzed annulation of o‐phenylenediamines and internal alkynes for the synthesis of quinoxalines is developed. This method provides a variety of quinoxalines in good to high yields under a simple and mild reaction conditions with molecular O2 as a terminal oxidant. Preliminary mechanistic studies show that a novel reaction mechanism is involved in the current reaction, which is different from the previous reported dicarbonyl compound mechanism via the oxidation of the alkyne.

Abstract:An efficient and practical method for the synthesis of 1,2,4-triazolo[5,1-b]quinazolin-9(3H)-ones has been developed via the copper-catalyzed domino reactions of readily available substituted N-acetyl-2-bromobenzohydrazides with cyanamide. The protocol uses inexpensive CuI as the catalyst, and no other ligand or additive was required. The target products were prepared in good to excellent yields with tolerance of various functional groups.

Abstract:A silver(I)-mediated phosphorylation/cyclization radical cascade of N-cyanamide alkenes has been developed. The addition of in situ generated phosphorus radical to N-cyanamide alkenes triggers the cascade, resulting in late-stage cyclization toward divergent access to 4-quinazolinones and dihydroisoquinolinones. Both terminal and internal N-cyanamide alkenes are applicable in this protocol, and the cyclizations are consistent with Baldwin's rule.

Abstract:Copper‐catalyzed and visible‐light catalytic difluoroalkylation reactions of N‐cyanamide alkenes have been developed. Various α‐bromodifluoromethyl substituted esters, amides and heterocycles were employed as the CF2 source. Both protocols are characterized by medium to high yields, good functional group compatibility, and applicable to gram‐scale synthesis. Comparable studies of copper catalysis and photoredox catalysis demonstrate minimal differences between the two protocols. Preliminary mechanism suggest that both transformations involve tandem radical cyclization process.

Abstract:Multi-heteroatom heterocycle synthesis through direct C-H bond activation is methodologically appealing but synthetically challenging. An efficient double C-N bond formation sequence to prepare quinazolinones utilizing primary amides and oxadiazolones in a catalytic redox-neutral [CoCp*(CO)I-2]/AgSbF6 system, where oxadiazolone could function as an internal oxidant to maintain the catalytic cycle, is reported. Amide-directed C-H bond activation and oxadiazolone decarboxylation are key to the success of this traceless, atom- and step-economic, and cascade approach for the construction of the quinazolinone skeleton.

Abstract:An efficient and rapid synthesis of multiply substituted quinolines is described. This method is enabled by a three-component cascade annulation of readily available aryl diazonium salts, nitriles, and alkynes. This reaction is catalyst- and additive-free. Various aryl diazonium salts, nitriles, and alkynes can participate in this transformation, and the yields are up to 83%.

Abstract:A concise and efficient method for the synthesis of 1,4-naphthoquinones has been successfully developed involving a CuI/Cu(OTf)2/DMSO system-catalyzed intramolecular oxidative cyclization of (o-alkynyl)arylketones. The present protocol provided a novel approach to access functionalized 1,4-naphthoquinones from non-naphthoquinone precursors with good selectivity and functional group tolerance.

Abstract:Via an imine-protection strategy, we herein present an unprecedented copper-catalyzed oxidative multicomponent annulation reaction for direct synthesis of quinazolinones. The construction of various products is achieved via formation of three C-N and one C-C bonds in conjunction with the benzylic functionalization. The merits of easily available feedstocks, naturally abundant catalyst, good functional group and substrate compatibility, and release of H2O as the byproduct make the developed chemistry a practical way to access quinazolinones.
