Abstract:A Pd-catalyzed highly enantioselective three-component coupling of 1,3-dienes with aryl iodines and sodium dialkyl malonates has been successfully established by using a H-8-BINOL-based phosphoramidite ligand. Abstract:1,3-Dienes are easily accessible chemicals that participate in a series of reactions acting on the carbon-carbon double bonds. Catalytic difunctionalization of 1,3-dienes provides a wide scope of functionalized chemicals. Pd(0) catalysts provide a diverse set of principles for the creation of asymmetric catalytic reactions, which are initiated with the oxidative addition and then undergo insertion reaction with one of double bonds of the 1,3-diene to become a p-allyl palladium species that is reactive toward nucleophilic attack. This review summarizes typical advances on the Pd(0)-catalyzed difunctionalization of 1,3-dienes in recent decades, particularly emphasizing the concepts that enable the switch from a racemic reaction to an enantioselective version.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:An enantioselective multicomponent carbonyl allylation reaction of 1,3-butadienes, alkynyl bromides and aldehydes has been established, leading to the generation of densely functionalized homoallylic alcohols in high levels of structural diversity and stereoselectivity. The multicomponent process sequentially comprises the oxidative addition, salt metathesis, insertion/π-allyl-Pd formation, asymmetric allylic borylation, and enantioslecitve carbonyl allylborylation, wherein the alkynyl palladium phosphate in situ generated from the chiral silver phosphate and the alkynyl palladium bromide turns out to be a key intermediate that allows the efficient stereochemical control.

Abstract:Transition-metal-catalyzed reductive coupling reactions have emerged as powerful protocols to construct C-C bonds. However, the development of enantioselective C(sp3)-C(sp3) reductive coupling remains challenging. Herein, we report a highly regio-, diastereo-, and enantioselective reductive homocoupling of allylic acetates through cooperative palladium and photoredox catalysis using diisopropylethylamine or Hantzsch ester as a homogeneous organic reductant. This straightforward protocol enables the stereoselective construction of C(sp3)-C(sp3) bonds under mild reaction conditions. A series of C2-symmetrical chiral 1,5-dienes were easily prepared with excellent enantioselectivities (up to >99% ee), diastereoselectivities (up to >95:5 dr), and regioselectivities (up to >95:5 rr). The resultant chiral 1,5-dienes can be directly used as chiral ligands in asymmetric synthesis, and they can be also transformed into other valuable chiral ligands.

Abstract:An iridium-catalyzed asymmetric allylic substitution reaction with 2-alkoxy carbonyl-1,3-dithianes has been achieved with high regio- and enantioselectivities. The transformation provides a new method for the enantioselective alpha-functionalization of dithianes. The corresponding dithiane-containing products are easily converted into many other derivatives with high yields and enantioselectivities.

Abstract:1,3-Enynes have been widely recognized as a prevalent synthon for the construction of valuable allenes, 1,3-dienes, and propargylic derivatives. However, significant advancements have primarily focused on elucidating the ionic reaction mechanism. The selective difunctionalization of 1,3-enynes involving radicals remains an outstanding challenge in particular within the realm of radical 1,2-difunctionalizations. Herein, we present a comprehensive account of the regio- and diastereoselective radical multicomponent 1,2-dialkylation of 1,3-enynes via photoexcited Hantzsch ester and chromium co-catalysis to furnish highly valuable homopropargylic alcohols. Our protocol shows excellent compatibility and 1,2-regioselectivity. Remarkably, this innovative approach obviates the need of external photocatalysts, stoichiometric metal reductants, or additional additives, offering a novel strategy for the valuable transformation of 1,3-enynes.

Abstract:Sonogashira-type cross-couplings are one of the most significant alkynylations in organic chemistry. One of the first palladium-catalyzed intramolecular Heck/Sonogashira reactions of alkenes with terminal alkynes is now reported. With this method, a variety of uniquely substituted chiral benzene-fused heterocycles bearing a propargyl-substituted all-carbon quaternary stereocenter were obtained in a straightforward, high-yielding, and highly stereoselective manner under mild conditions. Salient features of this process include the use of readily available substrates, high selectivities, a broad substrate scope as well as versatile product functionalizations.

Abstract:Over the past 30 years, Heck-type difunctionalization of unactivated alkenes involving C−H bond activation has emerged as a powerful strategy for the construction of synthetically valuable spirocycles; however, the development of asymmetric version has largely lagged behind. Herein, we demonstrate a robust Heck-type difunctionalization of a broad range of unactivated alkenes enabled by the first palladium/Xu-Phos-catalyzed tandem Heck/remote C–H bond alkylation. Moreover, both enantiomers of the product can be efficiently prepared using the same enantiomer of a chiral ligand via a position of the phenyl ring-dependent enantiodivergent synthesis. The salient features of this methodology include operational simplicity, high chemo- and enantio-selectivities, and broad substrate scope. In addition, we first revealed that the C(sp2)–H activation, alkene insertion, and C−I reductive elimination steps are reversible by experiments.

Abstract:The chiral allylic sulfonylated group is a unique structural motif embedded in a range of natural products and pharmaceuticals. Notably, the synthesis of structurally diverse chiral allylic sulfonylated derivatives via alkylsulfonylation of 1,3-dienes remains underexplored because of its inherent challenges in stereocontrol and regioselectivity. Herein, photoinduced, palladium-catalyzed enantioselective 1,2-alkylsulfonylation of conjugated 1,3-dienes is described. A wide variety of alkyl bromides, 1,3-dienes, and sodium sulfinates could be facilely coupled to generate value-added chiral allylic sulfonylated derivatives with high enantioselectivity. Finally, the utility of this enantioselective difunctionalization protocol was demonstrated in pharmaceutical analogue synthesis.

Abstract:The 1,2-difunctionalization of conjugated dienes is an important homogeneous catalytic reaction. The obtained products through 1,2-difunctionalization are widely existed in natural products and bioactive compounds, and are also sources of important organic intermediates, in addition, the preserved double bond in the difunctionalized product can be further transformed to give the desired structures or be functionalized sequentially to achieve multi-functionalization. The main difficulties focus on the encountered complex selectivities, including the regioselectivity, chemoselectivity and stereoselectivity in reactions. In recent years, with the development of organometallic chemistry, metal palladium, copper, iron or silver catalyzed 1,2-difunctionalizations of conjugated dienes have been reported in succession. In some cases the enantioselective 1,2-difunctionalizations of conjugated dienes were achieved via the introduction of chiral ligands. This review mainly focus on the recent metal catalyzed 1,2-difunctionalizations of conjugated dienes.
