Abstract:Copper-mediated cyanation of aryl C-H bond with removable bidenate auxiliary as a directing group has been developed, in which green and cheap acetonitrile is used as the "CN" source. By switching the reaction conditions, the C-H cyanated products and N-Ar-phthalimides can be provided in acceptable yields with high chemoselectivity, respectively. The use of quaternary ammonium salt is essential for cyanation reaction.

Abstract:A novel approach to the Cu-catalyzed cyanation of simple arenes using acetonitrile as an attractive cyano source has been documented. The C-H functionalization of arenes without directing groups involves a sequential iodination/cyanation to give the desired aromatic nitriles in good yields. A highly efficient Cu/TEMPO system for acetonitrile C-CN bond cleavage has been discovered. TEMPO is used as a cheap oxidant and enables the reaction to be catalytic in copper. Moreover, TEMPOCH2CN 6 has been identified as the active cyanating agent and shows high reactivity for forming the -CN moiety.

Abstract:Nitriles are important structural motifs found in agrochemicals, pharmaceuticals, and natural products. Furthermore, nitriles are versatile synthetic precursors for organic synthesis because they can be easily converted into various functionalities, such as amides, ketones, esters, primary amines, aldehydes, carboxylic acids, and nitrogen-containing heterocycles. Therefore, the development of efficient methods for the synthesis of nitrile compounds has attracted much attention from synthetic chemists. Cyanomethylation of various substrates is a synthetically useful reaction because a variety of diversely cyano-containing compounds could be readily prepared. Acetonitrile is the simplest commercially available alkyl nitrile, which can act as the cyanomethyl carbanion source. The traditional method for the cyanomethylation of organic molecules is deprotonation of acetonitrile in the presence of strong base. Alternatively, transition-metal-catalyzed C-H bond activation of acetonitrile represents an attractive approach to cyanomethylated compounds due to its atom and step economy. In this communication, we developed a simple and highly efficient method for the synthesis of cyanated difluorostyrene derivatives by cyanomethylation of alpha-(trifluoromethyl)styrenes using cheap and commercially available acetonitrile as the CH2CN- source. The reaction proceeded smoothly in the presence of LiHMDS at room temperature and was finished within 1 h, affording the cyanated gem-difluoroalkenes in moderate to good yields. Furthermore, the cyanomethylation reaction exhibited good substrate scope and functional group compatibility. A general procedure for the cyanomethylation of alpha-(trifluoromethyl)styrenes with acetonitrile is as following: alpha-(trifluoromethyl)styrenes 1 (0.5 mmol) was dissolved in acetonitrile 2a (4 mL) at room temperature under argon atmosphere. Subsequently, a solution of the LiHMDS in THF (1.5 mL, 1.0 mol/L, 1.5 mmol, 3.0 equiv.) was added dropwise within 50 min and stirring was continued for further 10 min (monitored by TLC). After completion of the reaction, the reaction mixture was quenched with saturated aqueous solution of NH4Cl (15 mL) and extracted with ethyl acetate (5 mL X 3). The combined organic layer was dried over anhydrous Na2SO4, filtered, and concentrated under vacuum. The crude residue was then purified by column chromatography on silica gel [(V(hexane)/V(ethyl acetate)= 10 : 1 similar to 6 : 1] directly to afford the pure target compounds.

Abstract:Aryl nitriles can be efficiently synthesized through transition-metal-mediated cyanation of aryl halides, arenes, aryl boronic acids, and so on. Among those most commonly used transition-metals, copper catalysts are surely much more inexpensive and easier to handle, compared to any other metal catalysts. Considering the high-efficiency of copper catalysts in the activation of C–X, C–H, and C–B bond as well as in the formation of C–CN bond, this Letter summarizes various copper-mediated cyanations based on the different kinds of cyanide reagents, such as metal cyanides, potassium hexacyanoferrate(II), acetone cyanohydrins, DDQ, AIBN, benzyl cyanide, malononitrile, nitromethane, and DMF. Our group’s recent contributions to this area are also demonstrated.

Abstract:A Cu(I)‐catalyzed three‐component reaction of 2‐iodo‐2,2‐difluoroacetophenones, alkynes and TMSCN is described. The reaction provided a facile method for the synthesis of difluoroacyl‐substituted nitriles, which might be served as potentially useful fluoroorganic intermediates for further transformation in drug discovery. This method has broad substrate scope, good efficiency and excellent stereoselectivity . Preliminary mechanistic investigation indicated that radical‐mediate process was involved in this cyanodifluoroalkylation reaction.

Abstract:A novel copper-catalyzed radical cyanotrifluoromethylation has been achieved through a multicomponent reaction of isocyanides, Togni's reagent, and trimethylsilyl cyanides, affording trifluoroacetimidoyl nitriles in good yields. This reaction demonstrates a unique feature of merging two valuable functional groups-trifluoromethyl (CF) and cyan (CN)-onto the same C atom. The transformation proceeds by the initial addition of the CF radical to isocyanide and the subsequent intermolecular C-CN formation. The products can be successfully transformed to a series of CF-containing amines and imines that may serve in the synthesis of valuable pharmaceuticals and agrochemicals.
