This reaction exhibits good functional group compatibility and proceeds under relatively mild conditions. Abstract:We herein describe a nickel-catalyzed cyanation reaction of aryl/alkenyl halides with alkyl isocyanides. Both aryl/alkenyl iodines and bromides were found to be competent electrophiles that reacted with alkyl isocyanides, affording nitrile compounds in moderate to good yields. A range of functional groups including halogens as well as hydroxyl, formyl, and acetamino groups were fairly compatible with the nickel catalysis. This protocol featured broad functional group tolerance, simple reaction conditions, and gram-scale synthesis.

Abstract:Palladium-catalyzed Suzuki-Miyaura cross-coupling or aryl halides is widely employed in the synthesis of many important molecules in synthetic chemistry, including pharmaceuticals, polymers and functional materials. Herein, we disclose the first palladium-catalyzed decarbonylative Suzuki-Miyaura cross-coupling of amides for the synthesis of biaryls through the selective activation of the N-C(O) bond of amides. This new method relies on the precise sequence engineering of the catalytic cycle, wherein decarbonylation occurs prior to the transmetallation step. The reaction is compatible with a wide range of boronic acids and amides, providing valuable biaryls in high yields (>60 examples). DFT studies support a mechanism involving oxidative addition, decarbonylation and transmetallation and provide insight into high N-C(O) bond activation selectivity. Most crucially, the reaction establishes the use of palladium catalysis in the biaryl Suzuki-Miyaura cross-coupling of the amide bond and should enable the design of a wide variety of cross-coupling methods in which palladium rivals the traditional biaryl synthesis from aryl halides and pseudohalides.

Abstract:Dedicated to Prof. Erick M. Carreira on the occasion of his 60 th birthday. A general Pd-catalyzed approach for the conversion of phenols into benzonitriles via aryl imidazolylsulfonates with a non-toxic cyanide source, potassium ferrocyanide, has been developed. Salient features of this method include low palladium precatalyst loadings (as low as 1.0 mol% total Pd), mild reaction conditions, and environmentally benign by-products compared to other (pseudo)halide aryl electrophiles. The initial scope of the reaction on a range of phenolic precursors is demonstrated including a one-pot, two-step approach to convert phenols directly into benzonitriles. Abstract:A mild and efficient protocol for the high para-selective arylation of monosubstituted arenes with tertiary benzamides has been developed via palladium-catalyzed oxidative coupling reactions. Due to the mild conditions and the easy availability of substrates and oxidant, this method could potentially provide a practical approach for the synthesis of para-substituted biaryl compounds.

Abstract:A practical, efficient Pd(OAc)(2)/Cu(II)/PPh3 cocatalytic system has been developed. With only 1 mol% Pd(OAc)(2), 20 mol% copper(II) salt, and 0.5 equiv of inexpensive PPh3 as ligand, the direct arylation of benzoazoles with aryl bromides could be performed smoothly in mild condition, affording the desired arylated benzoazoles in good yields (75-93%).

Abstract:A chemo- and regioselective olefinic C-H cyanation strategy was developed through palladium-catalyzed C(sp 2 )-S bond cleavage of stable alkenyl sulfonium salts with CuCN, efficiently affording multisubstituted alkenyl nitriles (acrylonitriles). This process features broad substrate scopes, good chemo- and regioselectivities, and excellent functional group tolerance. The present protocol provides an alternative route to alkenyl nitriles from readily available functionalized alkenes by a site-selective interrupted Pummerer activation/palladium-catalyzed olefinic C(sp 2 )-S cyanation sequence.

Abstract:A denitrative cyanation of nitroarenes using organocyanides and a palladium catalyst was developed. The key for this reaction was the utilization of an aminoacetonitrile as a cyano source to avoid the generation of stoichiometric metal- and halogen-containing chemical waste. A wide range of nitroarenes, including heteroarenes and pharmaceutical molecules, can be converted into aryl nitriles.

Abstract:We report herein the application of 1,1‐bis[(pinacolato)boryl]methane as a new type of terminating reagent to realize three‐component coupling reaction with aryl iodide and different electrophiles. A series of ortho‐disubstituted benzylboronates can be synthesized and the ortho‐substituent can be varied by using bromoacetic esters or O‐benzoyl hydroxylamine as electrophiles with slightly modified reaction conditions. The reaction shows good functional group tolerance and the obtained benzyl boronate products can be easily transformed to various synthetically useful compounds.

Abstract:The direct transformation of aryl carboxylic acids to aryl nitrile compounds is an interesting topic because carboxylic acids are not only abundant in nature but are also inexpensive and stable. Here, the synthesis of a series of aryl nitriles by palladium-catalyzed decarbonylative cyanation of carboxylic acids without base has been achieved. The successful decarbonylative cyanation of drug molecules and Gram-scale reaction to verify the practicality and operability of this method are analyzed.
