Abstract:AbstractDie ersten erfolgreichen gold‐katalysierten photoredoxselektiven C(sp2)‐H‐Difluoralkylierungen und ‐Perfluoralkylierungen von Hydrazonen mit gut verfügbaren RF‐Br‐Reagentien werden beschrieben. Die resultierenden gem‐difluormethylierten und perfluoralkylierten Hydrazone sind hoch funktionalisierte, vielseitige Moleküle. Eine milde Reduktion der Kupplungsprodukte liefert effizient gem‐difluormethylierte β‐Aminophosphonsäuren und β‐Aminosäure‐Derivate. In mechanistischen Studien wurde eine Difluoroalkylradikal‐Zwischenstufe in einem EPR‐“Spin‐Trapping”‐Experiment detektiert, was auf einen gold‐katalysierten Radikal‐Mechanismus hinweist.

Abstract:Herein, we report a method that combines hydrogen atom transfer and photoredox catalysis to achieve the dehydrogenative difluoroallylation of amides, ethers, and alkyl aldehydes. This operationally convenient method transforms a broad scope of substrates into the corresponding gem-difluoroalkenes via the construction of C(sp3)–C(sp3) or C(sp3)–C(sp2) bonds. Abstract:An unprecedented visible-light-induced direct C-H bond difluoroalkylation of aldehyde-derived hydrazones was developed. This reaction represents a new way to synthesize substituted hydrazones. The salient features of this reaction include difluorinated hydrazone synthesis rather than classical amine synthesis, extremely mild reaction conditions, high efficiency, wide substrate scope, ease in further transformations of the products, and one-pot syntheses. Mechanistic analyses and theoretical calculations indicate that this reaction is enabled by a novel aminyl radical/polar crossover mechanism, with the aminyl radical being oxidized into the corresponding aminyl cation through a single electron transfer (SET) process.

Abstract:Hexafluoroisopropanol (HFIP)-catalyzed direct dehydroxydifluoro-alkylation of benzylic and allylic alcohols with difluoroenoxysilanes is developed. This procedure enables the synthesis of a broad range of alpha,alpha-difluoroketones, a class of highly valuable intermediates and building blocks in medicinal and organic chemistry. Here, we have demonstrated for the first time that HFIP could act as a powerful catalyst for fluorinated carbon-carbon bond formation. The application of this protocol in late-stage dehydroxydifluoroalkylation of potentially bioactive drugs and natural products has also been carried out.

Abstract:Difluoroenoxysilane, a commonly used difluoroallylating reagent, has attracted considerable attention in recent years. However, its application in the annulation reaction for the construction of fluorinated heterocyclic compounds remains relatively limited. Presented here is the Bronsted acid-catalyzed efficient formal [4 + 2] annulation of difluoroenoxysilanes with alpha-cyano chalcones. The developed protocol demonstrates tolerance to various substituents under mild reaction conditions, providing a reliable approach to construct gem-difluoro-3,4-dihydro-2H-pyrans in good to excellent yields with high diastereoselectivities.

Abstract:The hydroxyl group was discovered to promote gold catalyzed hydrofluorination of alkynes via hydrogen bonding interaction. Based on this strategy, propargyl alcohols could be hydrofluorinated smoothly using Et3N·3HF under acidic additive-free conditions, which provided a straightforward alternative protocol for the synthesis of 3-fluoroallyl alcohols.

Abstract:A controllable mono-and di-multifluoroarylation of acyclic and cyclic N-aryl amines with aryl fluorides by photocatalyzed dual C(sp(3))-H/C(sp(2))-F functionalization has been developed, providing new access to a wide array of valuable alpha-fluoroarylated amines. In addition, the one-pot consecutive hetero-di-multifluoroarylation of N-aryl pyrrolidines and N,N-dimethylanilines was achieved with high to excellent diastereoselectivity. This new defluorinative C(sp(3))-C(sp(2)) coupling is distinguished by a broad scope, good regioselectivity, and mild conditions as well as gram-scale and late-stage applicability, and thus constitutes a significant advance in the arylation of unactivated C(sp(3))-H bonds with aryl fluorides.

Abstract:A photoredox-catalyzed, bromine-radical-mediated C(sp 3 )–H difluoroallylation of amides is disclosed. This modular approach exploits the hydrogen atom transfer (HAT) ability of photooxidatively generated bromine radicals to convert both cyclic and acyclic amides into the corresponding α-amino alkyl radicals that then are coupled with readily available trifluoromethyl alkenes. This method is distinguished by its mild conditions, broad substrate scope (30 examples), and the use of a simple HAT reagent, namely sodium bromide (NaBr). This strategy offers a promising paradigm for the incorporation of carbonyl isosteres into saturated aliphatic amines. Abstract:We report the first example of gold cata- lyzing CF bond activation for perfluoroarenes in the presence of silanes. Tricoordinated gold(I) com- plexes supported by Xantphos-type ligands, such as Xantphos and t BuXantphos ligands, exhibit efficacy in the hydrodefluorination (HDF) of various types of perfluoroarenes. For ( t BuXantphosAuA2)), the highest turnover number is up to 1000 in the HDF of pentafluoronitrobenzene with diphenylsilane. An ex- amination of functional group tolerance shows the orthogonality of this gold(I) catalytic protocol to ketone, ester, carboxylate, alkynyl, alkenyl and amide groups, suggesting its potential application in chemoselective CF activations. Mechanistic studies show that the equilibrium between tetracoordinated (L2Au) + and (LAu) + is important for the reactivity of gold catalysts, which is dependent on the sterically bulky group of Xantphos-type ligands. Furthermore, computational studies for the possible reaction path- ways suggest that direct oxidative addition of CF bonds by gold(I) cation might be the key step during these catalytic reactions.

Abstract:A computational mechanistic study of gold-catalyzed intermolecular alkene difunctionalization/cross-coupling of aryl iodides with methanol was studied. By calculating the corresponding initial oxidative addition step mediated by the related chelating (P,N) ligands, our results uncover the essential roles of hemilabile Me-Dalphos to promote the oxidative addition process by minimizing the distortion of substrate and catalyst. For the nucleophile methanol, by comparing the mechanism of intermolecular alkene difunctionalization versus cross-coupling, the C(sp 2 )–C(sp 3 ) reductive elimination is found more favorable than the corresponding C(sp 2 )–O formation, which contributes to the exclusive intermolecular alkene oxyarylation product. Furthermore, the different reactivity and selectivity of other nucleophiles such as tosyl amine and 1,3,5-trimethoxybenzene were compared. This mechanistic understanding will provide valuable guidance for the further development and wider application of gold-catalyzed redox reactions in synthetic chemistry.
