Abstract:The photoredox-catalyzed 1,6-difluoromethylation of 3-methyl-4-nitro-5-styrylisoxazole with HCF2SO2Na has been developed. Structurally diverse difluoromethylated products were obtained in good yields, and their further transformations were also investigated. The di-, tri-, and monofluoromethylation of the substrates were compared, and the yield of the difluoromethylation was the highest. DFT calculations revealed that in the difluoromethylation reaction the CF2H radical was nucleophilic, and the transition state activation energy was the lowest.

Abstract:The selective incorporation of fluorinated motifs, in particular CF2FG (FG=a functional group) and CF2H groups, into organic compounds has attrracted increasing attention since organofluorine molecules are of the utmost importance in the areas of nuclear imaging, pharmaceutical, agrochemical, and material sciences. A variety of synthetic approaches has been employed in late-stage difluoroalkylation reactions. Visible light photoredox catalysis for the production of CF2FG and CF2H radicals has provided a more sustainable alternative to other conventional radical-triggered reactions from the viewpoint of safety, cost, availability, and "green" chemistry. A wide range of difluoroalkylating reagents has been successfully implemented in these organic transformations in the presence of transition metal complexes or organic photocatalysts. In most cases, upon excitation via visible light irradiation with fluorescent light bulbs or blue light-emitting diode (LED) lamps, these photocatalysts can act as both reductive and oxidative quenchers, thus enabling the application of electron-donor or electron-acceptor difluoroalkylating reagents for the generation of CF2FG and CF2H radicals. Subsequent radical addition to substrates and additional organic transformations afford the corresponding difluoroalkylated derivatives. The present review describes the distinct strategies for the transition metal- and organic-photocatalyzed difluoroalkylation of a broad range of organic substrates by visible light irradiation reported in the literature since 2014.

Abstract:Recent advances in photoinduced trifluoromethylation and difluoroalkylation under photocatalysis are summarized. Most of the photoredox reactions proceed efficiently under mild conditions with simple operation. Various fluorinated reagents are developed and applied in different transformations. Usually, the photocatalyst in a photocatalytic cycle is initially stimulated to the excited state in the presence of visible light, which then provides an electron to the fluorinated reagent to release a CF3 radical or CF2 radical. The subsequent radical addition to an unsaturated bond with further transformations would produce the corresponding fluorinated products. Alkenes, alkynes, isocyanides, and arenes are frequently employed as substrates for access to fluorochemicals. The advantages of trifluoromethylation and difluoroalkylation under photocatalysis including mild reaction conditions, good functional group tolerance, and experimental ease will make the approaches attractive for further applications.

Abstract:A simple and efficient visible-light photoredox one-pot method for difluoromethylation of phenols and thiophenols has been developed. Abstract:Herein, we report a method that combines hydrogen atom transfer and photoredox catalysis to achieve the dehydrogenative difluoroallylation of amides, ethers, and alkyl aldehydes. This operationally convenient method transforms a broad scope of substrates into the corresponding gem-difluoroalkenes via the construction of C(sp3)–C(sp3) or C(sp3)–C(sp2) bonds. Abstract:Photoredox-catalyzed hydrodifluoromethylation of alkenes has become an effective method to introduce difluoromethyl group into organic molecules. As the reported methods involve either photocatalysts or superstoichiometric amounts of additives, we herein describe a simple alternative without using photocatalyst or additive for the hydrodifluoromethylation of alkenes, through photoactivation of difluoromethyltriphenylphosphonium iodide salt. Mechanistic studies shed light on how the transformation takes place.

Abstract:Abstract The discovery of modern medicine relies on the sustainable development of synthetic methodologies to meet the needs associated with drug molecular design. Heterocycles containing difluoromethyl groups are an emerging but scarcely investigated class of organofluoro molecules with potential applications in pharmaceutical, agricultural and material science. Herein, we developed an organophotocatalytic direct difluoromethylation of heterocycles using O 2 as a green oxidant. The C–H oxidative difluoromethylation obviates the need for pre-functionalization of the substrates, metals and additives. The operationally straightforward method enriches the efficient synthesis of many difluoromethylated heterocycles in moderate to excellent yields. The direct difluoromethylation of pharmaceutical moleculars demonstrates the practicability of this methodology to late-stage drug development. Moreover, 2′-deoxy-5-difluoromethyluridine (F 2 TDR) exhibits promising activity against some cancer cell lines, indicating that the difluoromethylation methodology might provide assistance for drug discovery.

Abstract:Difluoromethylation reactions are increasingly important for the creation of fluorine-containing heterocycles, which are core groups in a diverse range of biologically and pharmacologically active ingredients. Ideally, this typically challenging reaction could be performed photocatalytically under mild conditions. To achieve this separation of redox processes would be required for the efficient generation of difluoromethyl radicals and the reduction of oxygen. A covalent organic framework photocatalytic material was, therefore, designed with dual reactive centers. Here, anthracene was used as a reduction site and benzothiadiazole was used as an oxidation site, distributed in a tristyryl triazine framework. Efficient charge separation was ensured by the superior electron-donating and -accepting abilities of the dual centers, creating long-lived photogenerated electron-hole pairs. Photocatalytic difluoromethylation of 16 compounds with high yields and remarkable functional group tolerance was demonstrated; compounds included bioactive molecules such as xanthine and uracil. The structure-function relationship of the dual-active-center photocatalyst was investigated through electron spin resonance, femtosecond transient absorption spectroscopy, and density functional theory calculations.

Abstract:A photoredox-catalyzed approach for the difluoroalkylation of amino acids was achieved through simultaneous decarboxylation and defluorination processes. This innovative protocol employs commonly available amino acids and trifluoroacetophenones as the primary starting materials, eliminating the necessity for preactivation. This strategy has enabled the synthesis of several difluoroketone functionalized amines in moderate to impressive yields. These synthesized compounds are presented as foundational molecules for subsequent modification. The underlying mechanism for the transformation is anchored in a single electron transfer (SET) radical pathway.

Abstract:Controlling regioselectivity during difunctionalization of alkenes remains a significant challenge, particularly when the installation of both functional groups involves radical processes. In this aspect, methodologies to install trifluoromethane (-CF3) via difunctionalization have been explored, due to the importance of this moiety in the pharmaceutical sectors; however, these existing reports are limited, most of which affording only the corresponding β-trifluoromethylated products. The main reason for this limitation arises from the fact that -CF3 group served as an initiator in those reactions and predominantly preferred to be installed at the terminal (β) position of an alkene. On the contrary, functionalization of the -CF3 group at the internal (α) position of alkenes would provide valuable products, but a meticulous approach is necessary to win this regioselectivity switch. Intrigued by this challenge, we here develop an efficient and regioselective strategy where the -CF3 group is installed at the α-position of an alkene. Molecular complexity is achieved via the simultaneous insertion of a sulfonyl fragment (-SO2R) at the β-position. A precisely regulated sequence of radical generation using red light-mediated photocatalysis facilitates this regioselective switch from the terminal (β) position to the internal (α) position. Furthermore, this approach demonstrates broad substrate scope and industrial potential for the synthesis of pharmaceuticals under mild reaction conditions.

Abstract:The incorporation of difluoromethylated (CF2) moiety into potentially useful parent molecules lead to significant changes in metabolic stability, lipophilicity, and solubility of the molecules. Over the past several years, with the advent of new difluoromethylating reagents, great progress has been made in the development of a protocol for the direct incorporation of the CF2H group into organic molecules. Among them, difluroalkylation induced by visible light has emerged as an efficient strategy over the past few years. In particular, this protocol provides a more sustainable alternative to other traditional radical‐triggered reactions in terms of environment, energy, step‐economy, health, and safety. The present review mainly focuses on the development of the photocatalytic difluoroalkylation to olefinic moiety using transition‐metal complexes, organic dyes as the photocatalyst; and some organic compounds as a medium of photocatalysis.

Abstract:Despite the importance of difluoromethylene fragments in medicine, agrochemistry, and materials science, the precise and sustainable C−H difluoromethylation of aryl rings remains a challenge. Studies of metal-free difluoromethylation would be advanced if methods were as reliable and modular as metal catalysis. Herein, we present a modular platform for the sustainable synthesis of para-difluoromethylation of readily available aromatic amines via organophotoredox catalysis. The reaction exhibits excellent functional group tolerance and broad substrate scope, delivering a variety of valuable difluoromethylated products. Furthermore, the derivatization of various complex molecules further demonstrates the synthetic utility and practicality of this protocol.

Abstract:A mild and general methodology for the difluoromethylthiolation of cycloalkanols has been developed, employing N‐difluoromethylthiophthalimide as the SCF2H radical source, in combination with an acridinium-derived organo-photosensitizer, under redox-neutral conditions. This reaction protocol demonstrates high efficiency, scalability, and oxidant free, thus presenting a green approach for the rapid synthesis of distal difluoromethylthiolated alkyl ketones that are challenging to synthezise through alternative means.
