Abstract:A highly enantioselective gold(I)-catalyzed intermolecular tandem cyclization/[3 + 2] cycloaddition of 2-(1-alkynyl)-2-alken-1-ones with 3-stylindoles was achieved by using Ming-Phos as a chiral ligand. A variety of chiral highly substituted cyclopenta[c]furans were obtained in good yields (up to 99%) with excellent diastereoselectivities (>20:1) and enantioselectivities (up to 97% ee). The salient features of the present protocol include mild conditions, excellent yields, and high diastereo- and enantioselectivities, using readily available starting materials and a chiral ligand.

Abstract:The construction of fused [5.5.0] bicyclic heterocycles with precise regio-, stereo-, and enantioselective control remains a significant challenge in asymmetric catalysis. In this work, we introduce a novel gold-catalyzed asymmetric [8+4] cycloaddition of 1-(1-alkynyl)cyclopropyl ketones with simple tropones, yielding highly functionalized cyclohepta[b]furo[3,4-d]oxepine derivatives with excellent diastereo- and enantioselectivity (38 examples, all >20:1 dr, up to 95% ee). Additionally, an efficient kinetic resolution (KR) process is achieved (s factor up to 104). The gram-scale synthesis and subsequent synthetic transformations of the cycloadducts further demonstrate the synthetic potential of this method. Furthermore, the cycloadduct can undergo a [1,5]-H shift under acid catalysis, adding another dimension to the structural diversity.

Abstract:Chiral eight-membered heterocycles comprise a diverse array of natural products and bioactive compounds, yet accessing them poses significant challenges. Here we report a gold-catalyzed stereoselective (4 + 4) cycloaddition as a reliable and divergent strategy, enabling readily accessible precursors (anthranils and ortho -quinone methides) to be intercepted by in situ generated gold-furyl 1,4-dipoles, delivering previously inaccessible chiral furan/pyrrole-containing eight-membered heterocycles with good results (56 examples, all >20 : 1 dr, up to 99% ee). Moreover, we achieve a remarkably efficient kinetic resolution (KR) process (s factor up to 747). The scale-up synthesis and diversified transformations of cycloadducts highlight the synthetic potential of this protocol. Computational calculations provide an in-depth understanding of the stereoselective cycloaddition process.

Abstract:Herein, we report a gold-catalyzed formal dipolar cycloaddition approach to dihydrofurans and dihydropyrans using polarized alkynes (propiolates) as dipolarophiles and butenediol or pentenediol derivatives as 1,3- or 1,4-dipoles. The reaction proceeded via a multistep cascade process with a broad spectrum of substrate scope. A temporary silyl protection strategy was applied to solve the regioselective problem of unsymmetrical diols. Moreover, the reaction could be readily scaled up and the products could be further elaborated into a diverse collection of highly substituted and functionalized cyclic complex structures.

Abstract:The mechanisms of gold(I)-catalyzed cycloaddition of 1-(1-alkynyl) cyclopropyl ketones with nucleophiles have been investigated using density functional theory calculations at the B3LYP/6-31G (d, p) level of theory. A polarizable continuum model (PCM) has been established in order to evaluate the effects of solvents on the reactions. The results of the calculations indicate that the first step of the catalytic cycle is the cyclization of the carbonyl oxygen onto the triple bond which forms a new and stable resonance structure of an oxonium ion and a carbocation intermediate. The subsequent ring expansion step results in the formation of the final product and regeneration of the catalyst. Furthermore, the regioselectivity and effect of substituents has been discussed, including an analysis of energy, bond length, and natural bond orbital (NBO) charge distributions in the rate-determining step. Our computational results are consistent with earlier experimental observations.

Abstract:A gold-catalyzed asymmetric intermolecular [3 + 2]-cycloaddition of gamma-1-ethoxyethoxy-propiolate and aldehydes by the use of the chiral phosphoramidite (S,R,R)-5a is developed, which provides facile access to highly substituted 2,5-dihydrofurans in up to 84% yield with up to 97% ee. Control experiments support the result that this transformation indeed proceeds via an all-carbon gold 1,3-dipole with an open carbocation rather than a S(N)2 type reaction.

Abstract:A highly efficient gold-catalyzed cycloisomerization of 1,5-diynes was developed. Various functional groups are tolerated under the mild reaction conditions, which provides an alternative approach for the synthesis of indeno[1,2-<i>c</i>]furans. On the basis of mechanistic studies, including crossover experiments, deuterium labeling, and computational chemistry, the product formation proceeds via a formal [5,5]-sigmatropic rearrangement, a yet unknown reactivity pattern in gold catalysis. Abstract:Herein, we reported a gold-catalyzed formal cycloaddition reaction of readily available alkynes using amino but/pent-2-enoate derivatives as annulating reagents. The new strategy provides a straightforward and convergent route towards the synthesis of highly functionalized 2,3-dihydropyrroles and 1,2,3,4-tetrahydropyridines with high efficiency and a broad spectrum of substrate scope. Moreover, the reaction could be facilely performed at millimole scale and the products could be further transformed into other valuable classes of N-heterocycles.
