Abstract:A highly efficient gold-catalyzed cycloisomerization of 1,5-diynes was developed. Various functional groups are tolerated under the mild reaction conditions, which provides an alternative approach for the synthesis of indeno[1,2-<i>c</i>]furans. On the basis of mechanistic studies, including crossover experiments, deuterium labeling, and computational chemistry, the product formation proceeds via a formal [5,5]-sigmatropic rearrangement, a yet unknown reactivity pattern in gold catalysis. Abstract:Now, Jérôme Waser and colleagues report cyclopropenyl benziodoxoles as stable precursors of σ -type cyclopropenium cations and their subsequent application in catalytic transformations. The cyclopropenyl benziodoxoles were synthesized by deprotonation of cyclopropenes with n -butyllithium followed by addition of hypervalent iodine precursors. Then, σ -type CPC transfer to terminal alkynes and vinylboronic acids was demonstrated using gold catalysis with bench-stable (Me 2 S)AuCl and a bipyridinyl ligand (pictured). Notably, these C–C coupling reactions could not be replicated in this work using palladium or nickel catalysts. Although stoichiometric amounts of iodoarene are generated, this byproduct can be isolated and used again for the synthesis of cyclopropenyl benziodoxoles. The σ -type CPC transfer operates under mild reaction conditions and is compatible with coupling partners bearing diverse functional groups. Moreover, late-stage modification of complex natural products and bioactive molecules was demonstrated. Based on experimental studies and density functional theory calculations, the researchers proposed a catalytic cycle involving a gold(III)–cyclopropenyl intermediate whose existence is supported by mass spectrometric experiments. The hypervalent iodine reagent proved important to oxidize the gold(I) catalyst to generate the gold(III)–cyclopropenyl species, which allows the transfer of the cyclopropenyl moiety to the coupling partner. Moreover, chloride was identified as a supporting ligand and found to be relevant for the efficiency of the σ -type CPC transfer. The employment of cyclopropenyl benziodoxoles as synthetic equivalents of σ -type cyclopropenium cations is an elegant solution to address the stability issue of the latter in organic chemistry. This strategy in combination with gold catalysis provides convenient access to C1/C2-substituted cyclopropenes that are valuable building blocks for further modifications involving ring-opening transformations and C=C bond functionalization.

Abstract:Previously, we developed a gold-catalyzed cycloisomerization of dienediynes to synthesize the fused 6,7,5-tricyclic compounds. This reaction involves aliphatic C-H functionalization under mild conditions with high regio- and diastereoselectivities. Herein, we present a combined density functional theory (DFT) and experimental study to understand its mechanism. The reaction starts with a 6-endo-dig cyclization to generate a cis-1-alkynyl-2-alkenylcyclopropane. Then, a Cope rearrangement takes place to give a seven-membered-ring allene intermediate, whose central carbon atom possesses vinyl cation character and thus is highly reactive toward aliphatic C-H insertion. After the C-H insertion, two successive [1,2]-hydride shifts then occur to give the tricyclic product and to complete the catalytic cycle. Notably, steric effect induced by the bulky ligand is found to be important for the diastereocontrol in the C-H insertion step. DFT calculations suggested that the malonate-tethered substrate utilized in our previous work may undergo an undesired 5-exo-dig cyclization under gold catalysis, which could be the reason why the desired fused 6,7,5-tricarbocyclic product was not generated. These mechanistic insights then guided us to design substrates with a shortened carbon tether in the present work to inhibit the exo-dig cyclization so that the tandem cyclopropanation/Cope rearrangement/C-H functionalization could occur to construct polycarbocycles containing a seven-membered ring. This prediction was supported by new experiments, providing a new strategy to access fused 5,7,5-tricyclic and 5,7,6,6-tetracyclic carbocycles. In addition, how the substituents affect the chemoselectivity was also investigated.

Abstract:The first intramolecular enantioselective cyclopropanation of indenes and trisubstituted alkenes was accomplished by using new chiral phosphine X5 derived gold(I) complexes. This reaction is a straightforward, efficient method for constructing [5-3-6] fused-ring compounds with two vicinal all-carbon quaternary stereogenic centers, a core structure shared by numerous pharmacological products, and bioactive compounds. The salient features of this transformation include high enantioselectivity (up to >98% ee), excellent yield (>97%), and nice functional group tolerance.

Abstract:Cyclopropenes are the smallest unsaturated carbocycles. Removing one substituent from cyclopropenes leads to cyclopropenium cations (C 3 + systems, CPCs). Stable aromatic π -type CPCs were discovered by Breslow in 1957 by removing a substituent on the aliphatic position. In contrast, σ -type CPCs—formally accessed by removing one substituent on the alkene—are unstable and relatively unexplored. Here we introduce electrophilic cyclopropenyl-gold(III) species as equivalents of σ -type CPCs, which can then react with terminal alkynes and vinylboronic acids. With catalyst loadings as low as 2 mol%, the synthesis of highly functionalized alkynyl- or alkenyl-cyclopropenes proceeded under mild conditions. A class of hypervalent iodine reagents—the cyclopropenyl benziodoxoles (CpBXs)—enabled the direct oxidation of gold(I) to gold(III) with concomitant transfer of a cyclopropenyl group. This protocol was general, tolerant to numerous functional groups and could be used for the late-stage modification of complex natural products, bioactive molecules and pharmaceuticals.

Abstract:A gold-catalyzed asymmetric intermolecular [3 + 2]-cycloaddition of gamma-1-ethoxyethoxy-propiolate and aldehydes by the use of the chiral phosphoramidite (S,R,R)-5a is developed, which provides facile access to highly substituted 2,5-dihydrofurans in up to 84% yield with up to 97% ee. Control experiments support the result that this transformation indeed proceeds via an all-carbon gold 1,3-dipole with an open carbocation rather than a S(N)2 type reaction.

Abstract:The homogeneously catalyzed cycloisomerization reaction of propargylic amides was tested as benchmark reaction for two homologous series of Au(I) and Au(III) complexes with NHC ligands. This reaction is known to afford either aromatic oxazoles or dihydrooxazoles, depending on the oxidation state of the applied gold catalysts. For this purpose, symmetric and unsymmetric NHC ligands with mixed alkyl or benzyl substituents (1,3‐dimethyl‐imidazolyl‐2yl or 1‐benzyl‐3‐methyl‐imidazolyl‐2‐yl) and the corresponding Au(I) and Au(III) derivatives were prepared and characterized by analytical methods, IR and 1H and 13C NMR spectroscopies, and by X‐ray diffraction methods. Ten NHC‐gold(I) and NHC‐gold(III) complexes were tested on the cyclization reactions by adding gold catalysts in the presence and absence of AgPF6.Noteworthy, only the 1‐benzyl‐3‐methyl‐gold(III) trichloride was able to give selectively the aromatic oxazole without the addition of the silver salt. Interestingly, on contrary to what is reported in the literature, the use of gold(I) or gold(III) complexes does not influence the chemoselectivity of the cyclization reaction, while the halide bound to the metal does. In this regard, a mechanism purposing the dihydrooxazole as an intermediate for the formation of aromatic oxazole is proposed.

Abstract:Homogeneous gold-catalyzed cyclopropanation has emerged as a powerful method in organic synthesis due to its rich chemistry and fascinating reactivity. This thriving strategy is remarkable for its mild conditions, good selectivity, and high efficiency, which provides complementarity and orthogonality to traditional metal-catalyzed cyclopropanation. This review summarizes recent advances in gold-catalyzed cyclopropanation divided by the type of carbenoid precursors. Besides the commonly used diazo compounds, current approaches enable readily available enynes, propargyl esters, cyclopropenes, cycloheptatrienes, alkynes, and sulfonium ylides as safer surrogates in the realm of gold carbenoid chemistry. Meanwhile, these reactions allow for the rapid building of molecular complexity including synthetically useful and intricate cyclic, heterocyclic, and polycyclic skeletons. The combination of the new reactivity of gold complexes with their capability to catalyze cyclopropanations may lead to myriad opportunities for the design of new reactions. Furthermore, the synthetic utilities of such superior methods have also been illustrated by the total syntheses of selected natural and biologically interesting products and the asymmetric formation of challenging target molecules.

Abstract:The functional group compatibility of our recently disclosed gold(I)‐catalyzed cycloisomerization of 1‐bromoalkynes is studied in detail. Two main features are addressed, the minimum distances at which polar functional groups are tolerated and the influence of both electronic and steric effects on the performance of aryl ether derivatives. Moreover, robustness assessment experiments have been carried out shedding light on the factors dictating functional group tolerance. In addition, the reaction has been applied to the synthesis of a key intermediate in the total synthesis of a natural product and a heterobifunctional molecule. DFT calculations have shed light on the mechanistic features behind the observed reactivity trends. On one hand, the destabilization of the TS by the proximity of the polar group may hamper the reactivity for proximal functional groups. On the other hand, some polar functional groups are not tolerated regardless of their position due to the preferential coordination of gold to the oxygen atom over the alkyne, resulting presumably in competitive catalyst deactivation pathways.

Abstract:A highly enantioselective oxidative cyclopropanation of 1,6-enynes catalyzed by cationic Au(I)/chiral phophoramidite complexes is presented. The new method provides convenient access to densely functionalized bicyclo[3.1.0]hexanes bearing three contiguous quaternary and tertiary stereogenic centers with high enantioselectivity (up to e.r. 98:2). Control experiments suggest that the quinoline moiety of the β-gold vinyloxyquinolinium intermediate in the reaction plays an important role in promoting good enantioselectivity through a transitional auxiliary effect in the transition state.
